The behavior of seven hydroxy anthraquinone derivatives was studied by capillary zone electrophoresis and micellar electrokinetic chromatography. The effects of buffer pH (6.5–10.8) and sodium dodecyl sulfate concentration (10–20 mmol L?1) on the effective mobilities of the analytes and their separation were tested. A comparison of the two optimized separation systems showed that micellar electrokinetic chromatography was superior as it permits separation of all the seven analytes within 15 min, using 15 mmol L?1 sodium dodecyl sulfate in 10 mmol L?1 tetraborate buffer, pH 8.5, at a voltage of 20 kV. The calibration curves were linear in the concentration range from 5.0 · 10?7 to 5.0 · 10?4 mol L?1 for most of the analytes, at a detection wavelength of 254 nm. LOD and LOQ values of the analytes were in the ranges of 2.10 · 10?7–1.28 · 10?6 mol L?1and 6.99 · 10?7–4.25 · 10?6 mol L?1, respectively. The proposed separation conditions were applied to determination of 1,2-dihydroxy anthraquinone (alizarin) and 1,2,4-trihydroxy anthraquinone (purpurin) in Rubia tinctorum aglycone and of the recently described 1-acetyl-2,4,5,7-tetrahydroxy-9,10-anthraquinone and 1-acetyl-2,4,5,7,8-pentahydroxy-9,10-anthraquinone in the mycelium of fungi Geosmithia lavendula.  相似文献

采用毛细管电泳-电化学检测法(CE-ED)同时分离测定了中药杠板归中的阿魏酸、香草酸、槲皮素、咖啡酸、原儿茶酸等主要生物活性成分的含量。考察了工作电极电位、运行缓冲液的pH值和浓度、分离电压和进样时间等实验参数对实验结果的影响。在优化的实验条件下,以直径300 μm的碳圆盘电极为工作电极,检测电位为+0.95V(vs. SCE),在10mmol/L磷酸盐(pH 9.2)的运行缓冲溶液中,五个分析物能够在17min内实现很好的基线分离,被测物浓度与峰电流在3个数量级呈良好的线性,检测限(S/N=3)范围从7.1×10-8 到 9.3×10-8g mL-1。该方法已应用于实际样品的分析,样品处理简单,获得了令人满意的结果。  相似文献

A method based on capillary electrophoresis with electrochemical detection has been developed for the separa-tion and determination of epicatechin,kaempferol,chlorogenic acid,4-hydroxybenzoic acid,quercetin and proto-catechuic acid in hawthorn for the first time.The effects of working electrode potential,pH and concentration ofrunning buffer,separation voltage and injection time on CE-ED were investigated.Under the optimum conditions,the analytes could be separated in a 60 mmol·L~(-1) borate buffer(pH 8.7)within 21 min.A 300 μm diameter carbondisk electrode has a good response at 0.95 V(vs.SCE)for all analytes.The response was linear over three ordersof magnitude with detection limits(S/N=3)ranging from 3×10~(-8) to 2×10~(-7) g·mL~(-1) for the analytes.The methodhas been successfully applied to the analysis of real sample,with satisfactory results.  相似文献

The electrocatalysis oxidation of guanosine‐5′‐monophosphate (GMP) was investigated on Mg‐Al layered double hydroxide (LDH) functionalized with sodium dodecyl sulfate (SDS) and room temperature ionic liquid (RTIL) modified glass carbon electrode (GCE). The cyclic voltammogram of GMP on the modified electrode (RTIL/ LDH‐SDS/GCE) exhibited a well defined anodic peak at 1.091 V in 0.2 mol·L?1 pH 4.4 acetate buffer solution. The GMP oxidation was enhanced in the presence of anionic surfactant in the ?lms. The results suggest that the surfactant molecules intercalate the LDH layers to preconcentrate GMP molecules and the RTIL showed good ionic conductivity. The experimental parameters were optimized, the kinetic parameters were investigated and the probable oxidation mechanism was proposed. Under the optimized conditions, the oxidation peak current was proportional to GMP concentration in the range from 5.0×10?7 to 1.0×10?4 mol·L?1 with the correlation coefficient of 0.9987 and the detection limit was 1.0×10?7 mol·L?1. The RTIL/LDH‐SDS/GCE showed a good electrochemical response to the oxidation of GMP and would be developed into a new biosensor.  相似文献

The present work describes the individual, selective and simultaneous quantification of acetaminophen (ACP) and tramadol hydrochloride (TRA) using a modification‐free boron‐doped diamond (BDD) electrode. Cyclic voltammetric measurements revealed that the profile of the binary mixtures of ACP and TRA were manifested by two irreversible oxidation peaks at about +1.04 V (for ACP) and +1.61 V (for TRA) in Britton‐Robinson (BR) buffer pH 3.0. TRA oxidation peak was significantly improved in the presence of anionic surfactant, sodium dodecyl sulfate (SDS), while ACP signal did not change. By employing square‐wave stripping mode in BR buffer pH 3.0 containing 8×10?4 mol L?1 SDS after 30 s accumulation under open‐circuit voltage, the BDD electrode could be used for quantification of ACP and TRA simultaneously in the ranges 1.0–70 μg mL?1 (6.6×10?6–4.6×10?4 mol L?1) and 1.0–70 μg mL?1 (3.3×10?6–2.3×10?4 mol L?1), with detection limits of 0.11 μg mL?1 (7.3×10?7 mol L?1) and 0.13 μg mL?1 (4.3×10?7 mol L?1), respectively. The practical applicability of the proposed approach was tested for the individual and simultaneous quantification of ACP and/or TRA in the pharmaceutical dosage forms.  相似文献

The voltammetric behavior of camptothecin (CPT) in Britton-Robinson (B-R) buffer solutions (pH 2.09-9.07) was studied by the means of linear sweep voltammetry (LSV), cyclic voltarnmetry (CV) and normal pulse voltammetry (NPV) at a hanging mercury drop electrode. In different pH range of B-R buffer solutions, CPT could cause three reduction waves. In B-R buffer solutions (pH 2.09-5.46), wave P1 yielded by CPT was a two-electron wave. Between pH 6.01 and 9.07, CPT could yield two reduction waves P2 and P3. In addition, the pure CPT obtained from camptotheca acumina grown only in China was determined by NPV, and a linear response was observed in the range of 2.0 × 10^-3-4.0 × 10^-2 mmol·L^-1 with a 0.9991 correlation coefficient and a 8.0 × 1^-4 mmol·L^-1 detection limit for CPT.  相似文献

A composite electrode was fabricated from Cu2O powder, carboxyl-functionalized multi-wall carbon nanotubes (MWCNT-COOH), and paraffin oil in the proportions 51:17:32 (w/w). This composite electrode was used for amperometric detection (CZE–AD) in simultaneous capillary zone electrophoretic analysis of chlorogenic acid, rutin, sucrose, glucose, mannose, and fructose in tobacco samples. Under the optimum conditions, the six analytes could be separated in 100 mmol L?1 NaOH buffer within 30 min. Good linearity was achieved in the range 1 × 10?7–1 × 10?4 mol L?1 for the two polyphenols and 5 × 10?6–1 × 10?3 mol L?1 for the four sugars. The detection limits (S/N = 3) for the polyphenols and sugars were as low as 10?8 mol L?1 and 10?6 mol L?1, respectively.  相似文献

The electrochemical behavior of cationic tetradecyltrimethylammonium bromide (TTABr), anionic sodium dodecylsulfate (SDS), cationic‐anionic (catanionic) mixed surfactant and self‐assembled solutions at Pt wire electrode has been studied by cyclic voltammetry (CV). On the basis of the cyclic voltammograms and determining the self‐assembled structures by using freeze‐fracture transmission electron microscopy (FF‐TEM), the mechanisms of the electrochemical reactions near the electrode for the two surfactant self‐assembled solutions, i.e., micelles and vesicles, are presented. When mixing TTABr and SDS, at the right mixing ratio of TTABr:SDS, vesicles are established spontaneously. The redox behavior of two vesicle‐phase solutions at a constant total concentration of 25 mmol·L?1 with the ratios of TTABr:SDS 9.35:0.65 of positive charges of bilayer membranes and 1.25:8.75 of negative charges of bilayer membranes are investigated by cyclic voltammetry. These cyclic voltammograms of vesicles with different charges are compared with those of 100 mmol · L?1 TTABr and 100 mmol · L?1 SDS micelle solutions. This CV study on surfactant self‐assembled solutions should open up a new method of study in surfactant science.  相似文献

A microemulsion electrokinetic chromatographic (MEEKC) method has been developed and validated for determination of resibufogenin and cinobufagin in toad venom and in traditional Chinese medicine prepared from the venom. The MEEKC method involved use of sodium dodecyl sulfate (SDS) as surfactant, heptane as organic solvent, and butan-1-ol as co-solvent. To improve the separation, the effect of temperature and running buffer pH were evaluated. The optimized conditions (heptane 0.81% (w/w), SDS 3.31% (w/w), butan-1-ol 6.61% (w/w), and 10 mmol L−1 sodium tetraborate buffer, pH 9.2, and 298 nm as the detection wavelength) enabled useful and repeatable separation of the analytes.  相似文献

Controlled synthesis of asymmetrical gold nanokites was realized by reducing HAuCl4 with polyvinyl alcohol (PVA) in PVA‐sodium dodecyl sulfate (SDS) aggregations aqueous solution at room temperature without any additional reducing agents. The crystal structures, optical property and growth process of the gold nanokites were investigated by X‐ray diffraction (XRD), Vis‐NIR spectrum and transmission electron microscope (TEM). HAuCl4 concentration dramatically influenced the morphologies of the products. When HAuCl4 concentration was increased from 0.5 mmol·L−1 to 2.2 mmol·L−1, the products changed from network‐like nanostructures to nanokites, nanoleaves and microplates. SDS was indispensable for the formation of gold nanokites and without SDS super‐branched structures became the dominant products.  相似文献

The aim of this study was to investigate the correlation between levels of a marker of oxidative DNA damage, 8-hydroxy-2′-deoxyguanosine (8-OHdG), in human saliva and urine, and to explore its potential application in fast diagnosis of many diseases (especially cancer) associated with oxidative damage. A simple and time-efficient method, based on capillary electrophoresis with electrochemical detection, was developed for the determination of salivary and urinary 8-OHdG. Under the optimum conditions, 8-OHdG and its coexisting analytes could be well separated within 16 min at a voltage of 14 kV in 60 mmol L?1 borax running buffer (pH 8.2). A good linear relationship was established between peak current and concentration of analytes over three orders of magnitude with detection limits (S/N = 3) ranging from 0.41 × 10?7 to 2.50 × 10?7 mol L?1 for all analytes.  相似文献

A sensitive, straightforward electroanalytical method for determining pyrogallol (PY) in biodiesel in the presence of a surfactant was developed using a voltammetric technique and screen‐printed electrodes. The influence of surfactant addition (sodium dodecyl sulfate (SDS), Triton X‐100 (TX‐100), cetyltrimethylammonium bromide (CTAB), tetrabutylammonium iodide (TBAI), tetrabutylammonium bromide (TBAB), or tetraethylammonium bromide (TEAB)) on the supporting electrolyte (0.04 mol L?1 Britton? Robinson buffer) was evaluated. Only CTAB significantly increased the oxidation peak current. Under optimal conditions, the method demonstrated linearity over the concentration range of 0.8–9.0×10?6 mol L?1, with limits of detection and quantification of 4.9×10?7 and 1.5×10?6 mol L?1, respectively. The results were satisfactory, relative to those obtained using high‐performance liquid chromatography (HPLC).  相似文献

The anti-cancer synthetic drug irinotecan (CPT-11) and its active metabolite SN-38 have been determined by micellar electrokinetic capillary chromatography (MEKC). The detection of the analytes was made at 368 nm and their separation took less than 7 min using a borate buffer (pH 8.8 at 25 mmol L?1) solution containing sodium dodecyl sulfate (45 mmol L?1) and acetonitrile (13.5% v/v). On-line analyte concentration (normal stacking mode) and the use of a highly sensitive cell (Z shaped cell) improved detection limits (at the 10?8 mol L?1 level). Recovery in fortified human saliva was 108 ± 5%, in agreement with the result achieved with the reference HPLC method. For the analysis of urine from rats submitted to a single dose of CPT-11 and SN-38, camptothecin was used as internal standard enabling recoveries close to 100% when compared to the results achieved using HPLC.  相似文献

The interaction between mercuric ion and apoCopC in the absence or presence of cupric ion was investigated through difference UV spectra in Hepes buffer (10 mmol·L^-1) at pH 7.4. The results suggest that mercuric ion can bind to C- and N-terminal binding sites of apoCopC, and the conditional binding constants were calculated to be kN=(6.79± 1.12)× 10^6 mol^-1·L and kc=(3.06±0.05)× 10^5 mol^-1·L. Using urea as a chemical agent, the conformational stabilities of apoCopC and HgN^2+ -CopC-Hgc^2+ were monitored by fluorescence spectrum in Hepes buffer (50 mmol·L^-1) at pH 7.4. The free energy of stabilization is (14.69±0.85) and (16.66±0.55) kJ.mol^-1, respectively. HgN^2+ -CopC-Hgc^2+ is more stable than apoCopC.  相似文献

The influence of sodium dodecyl sulfate(SDS) on the cloud point temperature(Tcp) of the aqueous solution of thermoresponsive hyperbranched polyethylenimine derivative HPEI-IBAm was studied systematically. When p H was below 8.5, HPEI-IBAm was positively-charged. Initially, the Tcp of HPEI-IBAm decreased significantly, followed by an obvious increase with the increase of SDS concentration. The lower the p H was, the higher the SDS concentration was required to achieve the minimum Tcp. When p H was above 8.5, HPEI-IBAm was neutral and raising the SDS concentration led to the gradual increase of Tcp. Compared to linear poly(N-isopropyl acrylamide)(PNIPAm), the Tcp of the current hyperbranched HPEI-IBAm was more sensitive to SDS. The thermoresponsive HPEI-IBAm/SDS complex was used as host to accommodate the non-polar pyrene in water. The lowest SDS concentration for effectively enhancing the solubility of pyrene in water was around 6.4 mmol·L~(-1). When HPEI-IBAm was present, the SDS concentration threshhold was decreased to about 0.31 mmol·L~(-1). Fluorescence technique with pyrene as the hydrophobic probe demonstrated that the SDS concentration of 7.2 mmol·L~(-1) was required to form the hydrophobic domain to accommodate pyrene guests without HPEI-IBAm, while only 0.2 mmol·L~(-1) of SDS was required in the presence of HPEI-IBAm.  相似文献

A simple, accurate method based on capillary electrophoresis with electrochemical detection (CE–ED) has been developed to determine loureirin A, loureirin B and dracorhodin for differentiation of Resina Draconis from Sanguis Draconis. The effects of some important factors such as acidity and concentration of running buffer, separation voltage, injection time, and applied potential on the CE–ED working electrode were investigated. Under the optimum conditions, the three analytes could be well separated within 30 min in a 75 cm capillary at a separation voltage of 14 kV in a 80 mmol L?1 borate buffer (pH 9.24). The working electrode was a 300-μm-diameter carbon disc electrode positioned opposite the outlet of the capillary in a wall-jet configuration and was set at a potential of 0.90 V (vs. SCE). Excellent linearity was established over two orders of magnitude with detection limits (S/N = 3) ranging from 3 × 10?7 g mL?1 to 1 × 10?6 g mL?1 for all three analytes. The relative standard deviations of peak current and migration times of loureirin A, loureirin B and dracorhodin were 2.1, 1.7, 4.4 and 2.9, 2.8, 3.3% (n = 5), respectively. The recoveries of three constituents ranged from 98.8 to 101.8%. The methodology has been successfully applied to analyze and differentiate the actual samples with satisfactory assay results.  相似文献

The activation of bovine liver arginase, which catalyzes the hydrolysis of L‐arginine to L‐ornithine and urea, by glycine was studied by thermokinetic methods at 37°C in 40 mmol·L?1 sodium barbiturate‐HCl buffer solution (pH 9.4). Results of this experiment indicate that an appropriate concentration of glycine can enhance the activity of arginase, and the relative activation rate reached its maximum value, 74%, when the concentration of glycine in reaction system was 1 mmol·L?1 and the initial concentration of arginine was 5 mmol·L?1. With the increase of substrate concentration, the relative activation rate decreased in a definite glycine concentration. Michealis constant Km of reaction decreased from 5.53 to 3.31 mmol·L?1 and inhibition constant of product L‐ornithine Kp increased from 1.18 to 3.73 mmol·L?1 when glycine concentration was 1 mmol·L?1. For these reasons one possible activation mechanism of arginase by glycine was suggested that the activation effect results from the competition of glycine and arginine to enzyme activity position. When one or two of the activity positions of arginase are occupied by glycine, it is propitious for the enzyme to complex with substrate and obstruct L‐ornithine from combining with enzyme, and when all of the activity positions are occupied by glycine, the activation effect vanishs and the inhibition effect appears.  相似文献

Abstract

A sensitive and simple micellar electrokinetic chromatography (MEKC) method was developed for the determination of the antiepileptic drug carbamazepine (CBZ) using a sweeping on‐line concentration method with photodiode array detection. The effect of pH, concentration of the running buffer solution, organic modifier, applied voltage and injection time on the concentration efficiency and separation was investigated. An untreated fused‐silica capillary was used (50 cm; effective length, 40 cm, 75 µm i.d.) for the analysis. The background solution (BGS) was 50 mmol · L?1 NaH2PO4 (pH 3.0) containing 100 mmol · L?1 SDS and 20% acetonitrile (5.82 ms · cm?1) with an applied voltage of ?20 kV at 25°C. Sample introduction was performed at 0.5 psi for 90 s with diode array detection at 214 nm. For the method, the calibration curve was linear over a range of 0.5–40 µg · mL?1 for CBZ with a correlation coefficient of 0.998. The detection limit (S/N=3∶1) of CBZ was 0.10 µg · mL?1. About 100‐fold improvement in concentration sensitivity was achieved in terms of peak height by the sweeping method compared to conventional injection method. The sweeping‐MEKC method has been successfully applied to the analysis of CBZ in tablet and human serum.  相似文献
