High-performance liquid chromatography (HPLC) fingerprint has been commonly used in the quality control and assessment of herbal medicines, and two-dimensional (2D) fingerprint obtained by means of HPLC-diode array detector (HPLC/DAD) can provide higher reliability. In this paper, an approach to the analysis of the 2D HPLC/DAD fingerprints, which was based on digital image recognition techniques, was developed for the first time. First, wavelet transform was employed to eliminate noise signal in the 2D fingerprint, and then the 2D fingerprint was converted into grayscale image. Second, several features of the image were calculated, and hierarchical clustering. This approach was applied to the qualitative analysis of the different samples of coptis chinensis, and the clustering result of samples was all highly consistent with the real situation. Based on the densities in grayscale image, three components in standard samples were quantitative analyzed, and the obtained correlation coefficients between concentration and grayscale density were more than 0.999. Our study indicated that the analysis of the 2D HPLC/DAD fingerprint was successful based on the idea and techniques of digital image recognition techniques, and this proposed approach provided a new pathway for the analysis of two-dimensional spectrums.  相似文献

The objective of this paper was to establish an efficient method for quality control of Lidanpaishi (LDPS) tablet and keli, two famous traditional Chinese medicines. A simple and reliable high-performance liquid chromatography (HPLC) coupled with photodiode array detector (DAD) method was developed both for fingerprint analysis (FA) and quantitative determination. In quantitative analysis, linear regressions, limit of detection (LOD) and quantification (LOQ), intra-day and inter-day precisions, recovery, repeatability and stability were all tested and good results were obtained to simultaneously determine the 15 marker compounds, namely chlorogenic acid, rhaponticin, 6,7-dimethoxycoumarin, naringin, hesperidin, neohesperidin, baicalin, wogonoside, baicalein, wogonin, chrysin, honokiol, magnolol, emodin, and chrysophanol in the herbal drugs. In fingerprint analysis, 34 peaks were selected as the characteristic peaks to evaluate the similarities of different samples collected from different pharmaceutical companies in China according to the State Food and Drug Administration (SFDA) requirements, and two kinds of data, relative retention time (RRT) and relative peak area (RPA) were used to identify the common peaks in 15 samples for investigation. Furthermore, hierarchical cluster analysis (HCA) was also performed to evaluate the variation of the herbal drugs. The present approach, i.e. HPLC coupled with multiple compounds determination (MCD) and FA is a powerful and meaningful tool to comprehensively conduct the quality control of traditional Chinese medicines.  相似文献

Shuanghuanglian injection, a famous composite formula in China, is a combination of Flos lonicerae japonicae, Radix scutellariae and Fructus forsythiae. To develop a method to overall evaluate the correlation between the formula and its three raw herbs, the representative LC fingerprint of Shuanghuanglian injection was established from 12 batches of samples. Ten peaks were selected as the common peaks to evaluate the similarities of different samples from different manufacturers. Hierarchical clustering analysis was used as a powerful pattern recognition tool to identify and classify Shuanghuanglian injection from different factories. In addition, chemical correlation between the formula and the three raw herbs was evaluated qualitatively through comparison of their chromatograms under the same HPLC conditions. The results showed that peak 1, 2, 3 and 9 in the formula fingerprint were characteristic components of Flos lonicerae japonicae, peaks 5, 6 and 8 were that of F. forsythiae, peak 10 was that of R. scutellariae, while peak 7 was the common component of Flos lonicerae japonicae and F. forsythiae, peak 4 was addressed to F. forsythiae and it was also the product of the manufacturing procedure.  相似文献

Si-Mo-Tang(SMT) oral liquid preparation, a traditional Chinese medicine, was prepared from four crude herbal drugs, Fructus Aurantii Submaturus, Radix Aucklandiae, Semen Arecae and Radix Linderae Aggregatae. A combinative method using HPLC fingerprint and quantitative analysis was developed and validated for quality consistency evaluation of SMT. Individual HPLC chromatograms were evaluated against the mean chromatogram generated via a similarity evaluation computer program. Data from chromatographic fingerprints were also processed with principal component analysis(PCA) and hierarchical cluster analysis(HCA). Additionally, six components (naringin, isonaringin, hesperidin, neohesperidin, norisoboldine and potassium sorbate) in SMT were simultaneously determined to interpret the quality consistency. For fingerprint analysis, 20 peaks were selected as the characteristic peaks to evaluate the similarities of 26 SMT collected from different manufacturers. Among the 20 characteristic peaks, 10 peaks were assigned to be naringin, hesperidin, neohesperidin, isonaringin, neoeriocitrin, tangeretin, nobiletin, norisoboldine, 5-(ethoxymethyl)furan-2-carbaldehyde and potassium sorbate, respectively. The results of similarity analysis, PCA and HCA, indicate that the samples from different manufacturers were consistent with each other in composition. The results from the quantitative data show that the contents of six compounds were significantly different in SMT oral liquid preparations from different manufacturers. The combinative method of chromatographic fingerprint with quantitative analysis developed here offered an efficient way for the quality consistency evaluation of the traditional Chinese medicine SMT.  相似文献

Traditionally, pomegranate (Punica granatum L.) has been consumed as fresh fruit or as pomegranate juice. Pomegranate peel, the dried husk of P· granatum, is an important herbal medicine for treating diarrhea, hemostasis and insect‐induced abdominal pain in China. However, the quality control methods for pomegranate peel remain unsatisfactory. In this work, a new HPLC‐based qualitative and quantitative method for quality control of pomegranate peel was developed and validated for the simultaneous determination of polyphenols and triterpenes (including punicalagins A and B, ellagic acid, oleanolic acid and ursolic acid) by solvent extraction and ratio blending method in tandem with wavelength switching. The average recoveries were 98.07–100.61% with relative standard deviation no more than 4.27%. In addition, the fingerprint analysis was conducted to interpret the consistency of the quality test. Thirteen characteristic peaks were selected to evaluate the similarities of 16 batches of pomegranate peel. The similarities of samples were all more than 0.80, indicating that the samples from different areas of China were consistent. The results demonstrated that quantitative analysis and the HPLC fingerprint as a characteristic distinguishing method combining similarity evaluation can be successfully used to assess the quality and to identify the authenticity of pomegranate peel.  相似文献

HPLC fingerprint analysis, principle component analysis (PCA), and cluster analysis were introduced for quality assessment of Cortex cinnamomi (CC). The fingerprint of CC was developed and validated by analyzing 30 samples of CC from different species and geographic locations. Seventeen chromatographic peaks were selected as characteristic peaks and their relative peak areas (RPA) were calculated for quantitative expression of the HPLC fingerprints. The correlation coefficients of similarity in chromatograms were higher than 0.95 for the same species while much lower than 0.6 for different species. Besides, two principal components (PCs) have been extracted by PCA. PC1 separated Cinnamomum cassia from other species, capturing 56.75% of variance while PC2 contributed for their further separation, capturing 19.08% variance. The scores of the samples showed that the samples could be clustered reasonably into different groups corresponding to different species and different regions. The scores and loading plots together revealed different chemical properties of each group clearly. The cluster analysis confirmed the results of PCA analysis. Therefore, HPLC fingerprint in combination with chemometric techniques provide a very flexible and reliable method for quality assessment of traditional Chinese medicines.  相似文献

A novel method combining high performance liquid chromatography (HPLC) fingerprint and simultanous quantitative analysis of multiple acitve components was developed and validated for quality evaluation of one type of traditional Chinese medicine preparations: Shuang-huang-lian (SHL) oral liquid formulation. For fingerprint analysis, 45 peaks were selected as the common peaks to evaluate the similarities among several different SHL oral liquid preparations collected from manufacturers. Additionally, simultanous quantification of eleven markers, including chlorogenic acid, caffeic acid, rutin, forsythiaside, scutellarin, baicalin, forsythin, luteoloside, apigenin, baicalein and wogonin, was performed. Statistical analysis of the obtained data demonstrated that our method has achieved desired linearity, precision and accuracy. Finally, concentrations of these eleven markers in SHL oral liquid prepared by different manufacturers in China were determined. These results demonstrated that the combination of HPLC chromatographic fingerprint and simultaneous quantification of multi-ingredients offers an efficient and reliable approach for quality evaluation of SHL oral liquid preparations.  相似文献

A method based on high performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD) was developed for the quantitative analysis of three active compounds and chemical fingerprint analyses of saccharides in Morindae officinalis radix. Ten batches of Morindae officinalis radix were collected from different plantations in the Guangdong region of China and used to establish the fingerprint. The samples were separated with a COSMOIL Sugar-D column (4.6 mm × 250 mm, 5 μm) by using gradient elution with water (A) and acetonitrile (B). In addition, Trapped-Ion-Mobility (tims) Time-Of-Flight (tims TOF) was used to identify saccharides of Morindae officinalis radix. Fingerprint chromatogram presented 26 common characteristic peaks in the roots of Morinda officinalis How, and the similarities were more than 0.926. In quantitative analysis, the three compounds showed good regression (r = 0.9995–0.9998) within the test ranges, and the recoveries of the method were in the range of 96.7–101.7%. The contents of sucrose, kestose and nystose in all samples were determined as 1.21–7.92%, 1.02–3.37%, and 2.38–6.55%, respectively. The developed HPLC fingerprint method is reliable and was validated for the quality control and identification of Morindae officinalis radix and can be successfully used to assess the quality of Morindae officinalis radix.  相似文献

A capillary electrophoresis fingerprint was constructed for Sanhuang tablet, a Chinese traditional patent medicine, that was commonly used in clinical practice, where the isosceles trapezoid method was first applied for the optimization of background electrolyte solution, and the resolution index was performed to assess the experimental conditions; furthermore, a novel linear quantitative fingerprint method was established for accurate qualitative and quantitative discrimination of the test samples from diverse commercial brands. The fingerprint analysis coupled with quantitative determination of two components was employed to elucidate that the quality consistency of the products was relatively good within one manufactory, but poor among different companies for the 30 batches of samples. In addition, the fingerprint–efficacy relationship between chemical components and antioxidant activity in vitro was investigated using partial least squares analysis, and the calibration and prediction of the antioxidant activity of the selected samples via fingerprint data were presented with the desired results. This work illustrates that the proposed fingerprint analysis based on linear quantitative fingerprint method can be applied for the quality evaluation of traditional Chinese medicine and herbal preparations as part of their quality control, and the constructed mathematical model is particularly suitable for depicting the fingerprint–efficacy relationship.  相似文献

(1)Objective: In this study, a quantitative analysis of chemical groups (the triterpenoids, water-soluble polysaccharides, and acidic polysaccharides) and quantitative high liquid performance chromatography (HPLC) fingerprint of Poria cocos (Schw.) Wolf (PC) for quality control was developed. (2) Methodology: First, three main chemical groups, including triterpenoids, water-soluble polysaccharides, and acidic polysaccharides, in 16 batches of PC were evaluated by ultraviolet spectrophotometry. Afterward, the quantitative fingerprint of PC was established, and the alcohol extract of PC was further evaluated. The method involves establishing 16 batches of PC fingerprints by HPLC, evaluating the similarity of different batches of PC, and identifying eight bioactive components, including poricoic acid B (PAB), dehydrotumulosic acid (DTA), poricoic acid A (PAA), polyporenic acid C (PAC), 3-epidehydrotumulosic acid (EA), dehydropachymic acid (DPA), dehydrotrametenolic acid (DTA-1), and dehydroeburicoic acid (DEA), in PC by comparison with the reference substance. Combined with the quantitative analysis of multi-components by a single marker (QAMS), six bioactive ingredients, including PAB, DTA, PAC, EA, DPA, and DEA, in PC from different places were established. In addition, the multivariate statistical analyses, such as principal component analysis and heatmap hierarchical clustering analysis are more intuitive, and the visual analysis strategy was used to evaluate the content of bioactive components in 16 batches of PC. Finally, the analysis strategy of three main chemical groups in PC was combined with the quantitative fingerprint strategy, which reduced the error caused by the single method. (3) Results: The establishment of a method for the quantification of chemical groups and quantitative HPLC fingerprint of PC was achieved as demonstrated through the quantification of six triterpenes in PC by a single marker. (4) Conclusions: Through qualitative and quantitative chemical characterization, a multi-directional, simple and efficient routine evaluation method of PC quality was established. The results reveal that this strategy can provide an analytical method for the quality evaluation of PC and other Chinese medicinal materials.  相似文献

A novel model of evaluating the quality and identifying the origins of Longjing green tea was established for the first time in this study. The analysis procedures in the model included the structure speculation of the major compounds and the identification of the bioactive components, the establishment of the fingerprint, the multiple analysis of the fingerprint, and the quantitative determination of the bioactive components. All the analysis procedures were conducted on an HPLC coupled with diode array detection (DAD) and an ion-trap coupled with a time-of-flight mass spectrometry (IT-TOF-MS). In this study, 13 batches of Longjing green tea from five traditional production sites and a new production site in Hangzhou, China were selected to establish the model. The result showed that using the model, the difference between Longjing green tea from the traditional and the new production sites was revealed successfully. It indicated that the model could be successfully applied to evaluate the quality and identify the origin of unknown Longjing green tea.  相似文献

Because almost every traditional Chinese medicine (TCM) is a multicomponent system, QC of TCMs always involves various difficulties. As a current popular quality assessment approach, focusing on qualitative and quantitative analysis of certain compounds contained in herbal medicine has been widely used for the sake of expediency rather than being a practical and realistic way. However, this method does not take the existence of other constituents into account. Comparatively, the chromatographic fingerprint of the components is a more suitable approach to holistically assess the quality of herbal drugs. Fructus xanthii is a well-known herbal drug listed in all editions of the Chinese Pharmacopoeia. However, there is no quality evaluation method given in its monograph, even for the above-mentioned expediency. This paper reports an HPLC fingerprinting method for quality evaluation of F. xanthii. The HPLC profiles of 27 batches of commercial samples were further analyzed using chemometric methods, including similarity evaluation and principal component analysis. As a result, the established HPLC fingerprint contained 23 characteristic peaks; therein, 13 peaks were unambiguously assigned by comparing their retention times and UV spectra with those of reference compounds, and five peaks were tentatively identified on the basis of their MS/MS fragmentation patterns and UV spectra. Moreover, it could be clearly observed that caffeoylquinic acid and its analogs predominate in F. xanthii. Except for three samples identified as outliers, 24 other commercial samples displayed similar HPLC profiles, indicating that the quality of the herbs from different markets is stable and consistent.  相似文献

通过建立复方甘草片标准制剂(SP)控制模式和定量高效液相色谱指纹图谱,结合5个质量标志物的精准定量评价了9个厂家共145批复方甘草片质量一致性。首先建立了复方甘草片标准制剂的标准指纹图谱(SP-RFP),然后以SP-RFP作为评价标准,采用系统指纹定量法对145批复方甘草片进行整体定性和整体定量评价。用双标校正法校正定量指纹图谱的系统误差,结果表明所检样品质量均合格。此外,在统一化色谱条件下测定各原料药和模拟样品,对制剂指纹进行归属相关度和准确度评判,得到原料指纹与制剂指纹的相关性,从而实现智能预测制剂或原料药质量和阻止低劣原料入药。同时用紫外全指纹溶出度法测定5个厂家的复方甘草片的溶出度曲线,用以评价制剂工艺的合理性。以上方法可行且准确度高,实现了对复方甘草片质量和工艺的一致性评价。该文为中药质量一致性评价提供了基础评价模式和基本操作思路以及具体实例。  相似文献

To overcome the instability of traditional Chinese medicine (TCM) fingerprint spectra (FPS) and to build up absolute identification standard of TCM, the construction method of combinational numeral fingerprint spectra (CNFPS) was set up. The analysis of invariableness based on CNFPS was carried out. It can be used as absolute quantitative standard to identify the difference of TCM samples. According to this method, the HPLC FPS of components extracted from Glycyrrhiza root samples by ethanol was analyzed. Stable results and accurate quality evaluation were obtained perfectly. The common peak ratio invariableness is an absolute standard in identifying TCM samples, which is better than the similarity method of the FPS.  相似文献

建立了三波长融合高效液相色谱指纹图谱,并结合6组分定量和主成分分析(PCA)评价25批银翘解毒片的质量一致性。采用反相高效液相色谱法(RP-HPLC)分别于230、279、327 nm下检测。运用多波长融合指纹图谱技术建立银翘解毒片三波长融合指纹图谱,采用系统指纹定量法对其进行定性和整体定量评价。结果鉴别出25批银翘解毒片样品完全合格且区分良好。同时测定6组分含量,与指纹图谱结合,从整体和局部角度评价银翘解毒片质量。此外,运用PCA法对融合指纹图谱进行分析,通过主成分得分图可以明显区分来自两个厂家的25批银翘解毒片样品。方法综合性较强且有效,为科学评价与有效控制银翘解毒片的质量提供了可靠的参考。  相似文献

Flavonoids are the main active components of natural medicinal plants with many physiological functions. In this study, an HPLC fingerprinting method based on the distribution and relative amount of 11 bioactive flavonoids was established for the quality evaluation of commercially available wild Jujube leaf tea (JLT) from China. Separation of the crude flavonoid extract was achieved on a column filled with C18 material with a high carbon content. The flavonoids in wild JLT were identified based on UV spectroscopy and accurate mass measurements by TOF‐MS. Twenty‐one batches of practical samples collected from different habitats were analyzed by using the developed HPLC method to construct the HPLC characteristic fingerprint of wild JLT. Then, combined with clustering and similarity analyses, the HPLC characteristic fingerprint was used for the authentication and quality evaluation of commercial wild JLT. Results indicated that the proposed HPLC characteristic fingerprint reflected the inherent characteristics of wild JLT collected from different regions. Authenticity identification and quality control of commercially available wild Jujube tea were achieved based on the HPLC characteristic fingerprint analysis. This new approach to bioactive component profiling provided a promising reference method for the quality evaluation of commercial wild flower and plant tea.  相似文献

A simple, sensible, and reliable HPLC–DAD method was first developed for fingerprint analysis of Alismatis Rhizoma, and then applied to analyze 85 samples from three main cultivated areas. In all, 40 common fingerprint peaks were designated, and six of which were definitely identified. Then, the combinatory analysis using similarity evaluation, principal component analysis, and orthogonal partial least square discriminant analysis revealed clear chemical consistency between samples from Fujian and Jiangxi provinces and substantial differences between those from Fujian/Jiangxi and Sichuan provinces. Furthermore, six components were dug out as potential chemical markers for distinguishing Alismatis Rhizoma from different areas, among which five were qualified for quantitative analysis. In conclusion, the combination of chemical fingerprint, multiple components quantification, and pattern recognition analysis was rather powerful and useful in discriminating Alismatis Rhizoma from different regions, which was a benefit for quality control.  相似文献

Using high-performance liquid chromatography (HPLC), a chemical fingerprint method was developed for investigating and demonstrating the variance of flavonoids among different origins of sea buckthorn berries. Thirty-four samples were analyzed including 15 RS (Hippophae rhamnoides ssp. sinensis) samples, 7 RY (H. rhamnoindes ssp. yunnanensis) samples, 5 RW (H. rhamnoides ssp. wolongensis) samples, 4 NS (H. neurocarpa ssp. stellatopilosa) samples and 3 TI (H. tibetana) samples. In the HPLC chromatograms, 12 compounds were identified as flavonoids, including quercetin 3-O-sophoroside-7-rhamnoside, kaempferol 3-O-sophoroside-7-O-rhamnoside, isorhamnetin 3-O-sophoroside-7-O-rhamnoside, isorhamnetin 3-O-glucoside-7-O-rhamnoside, quercetin 3-O-rutinoside, quercetin 3-O-glucoside, isorhamnetin 3-O-rutinoside, isorhamnetin 3-O-glucoside, quercetin, kaempferol 7-O-rhamnoside, kaempferol and isorhamnetin. Both correlation coefficient of similarity in chromatograms and relative peak areas of characteristic compounds were calculated for quantitative expression of the HPLC fingerprints. Our results revealed that the chromatographic fingerprint combining similarity evaluation could efficiently identify and distinguish sea buckthorn berries from different species. However, no obvious difference between RS and RY suggested that the two subspecies might have very close relationship in terms of chemotaxonomy. The established method was considered to be suitable for fingerprint analysis to check the genuine origin and control the quality of sea buckthorn berries and extracts.  相似文献
