Plants with beneficial properties are known in traditional medicine. Nowadays, in spite of widespread availability of synthetic compounds, the search goes towards natural compounds to lower cost and few side effects. The increasing interest in preventive medicine encourages use of nutraceuticals, bioactive compounds of vegetable origin with important nutritional values. Among the medicinal plants, Opuntia ficus-indica (L.) Miller (Family Cactaceae, subfamily Opuntiodeae, Genus Opuntia, subgenus Platyopuntia, species Opuntia ficus-indica (L.) Miller) is widely known for its beneficial properties. The aim of the present review is to stress the major classes of Opuntia components and their medical interest through emphasis on some of their biological effects, particularly those having the most promising expected health benefits and therapeutic impacts on fish and mammals.  相似文献

Opuntia ficus-indica (L.) Mill. is the Cactaceae plant with the greatest economic relevance in the world. It can be used for medicinal purposes, animal nutrition, production of biofuels and phytoremediation of soils. Due to its high content of bioactive compounds, the prickly pear has antioxidant, antimicrobial and anticancer properties. The aim of this study was to determine the polyphenolic, fatty acid and amino acid profile and characterize the antioxidant capacity of seeds of seven Spanish prickly pear cultivars. A total of 21 metabolites, mainly phenolic acids and flavonols, were identified using ultraperformance liquid chromatography photodiode detector quadrupole/time-of-flight mass spectrometry (UPLC-PDA-Q/TOF-MS). Significant differences were found in the phenolic concentrations of the investigated varieties. The highest amount of phenolic compounds (266.67 mg/kg dry matter) were found in the “Nopal espinoso” variety, while the “Fresa” variety was characterized by the lowest content (34.07 mg/kg DM) of these compounds. In vitro antioxidant capacity was positively correlated with the amount of polyphenols. The amino acid composition of protein contained in prickly pear seeds was influenced by the variety. Glutamic acid was the predominant amino acid followed by arginine, aspartic acid and leucine, independent of prickly pear variety. Overall, 13 different fatty acids were identified and assessed in prickly pear seeds. The dominant fatty acid was linoleic acid, with content varying between 57.72% “Nopal ovalado” and 63.11% “Nopal espinoso”.  相似文献

Woodfordia fruticosa (L.) Kurz is a widely used plant in traditional medicine systems. The tribal communities of Amarkantak, Madhya Pradesh (India) are using this plant for the treatment of general weakness, blood related complications like blood deficiency, blood purification and for the treatment of symptoms related to sickle cell disease (SCD). SCD is a genetic disease with life threatening complications. In the absence of any drugs without any side effects, the alternative plant based therapies that may either reduce/ reverse the sickling of the red blood cells can be safe and effective therapeutic agents. We evaluated W. fruticosa extracts for phytoconstituents, anti-oxidant and anti-inflammatory properties. Anti-sickling properties of the extracts were evaluated by estimation of reverse sickling, polymerization inhibition and osmotic fragility assays. Chemical profiling of the methanol extract was done using LC-MS analysis. Phytochemicals such as alkaloids, steroids, tannin, and saponins were present in all the extracts. Methanol extract displayed maximum reversal (66 ± 1%) of sickled Red blood Cells (RBC) and significantly inhibited Hb polymerization. The hexane and methanol extracts led to minimum hemolysis of sickled RBC in the osmotic fragility assays. Total tannin (365 ± 2.4 TAE) content was highest in acetone extract, while the total flavonoid and phenolic content (156.9 ± 2.0 QE) and (113.7 ± 0.7 GAE) were highest in methanol extract. The methanol extract displayed minimum IC50 (8.1 ± 1.5) in 2, 2-diphenylpicrylhydrazyl (DPPH) while the acetone extract had minimum IC50 (215.8 ± 5.7) in 2,2′-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assay. The hexane extract displayed maximum Ferric reducing anti-oxidant power (FRAP) value (1.5 ± 0.5 mM Fe(II)/mg dry weight) that were higher than methanol and aqueous extracts (1.45 ± 0.1, 1.45 ± 0.05 mM Fe(II)/mg dry weight). The methanol extract provided maximum RBC protection from hemolysis (73.8 ± 0.8%). Maximum Lipoxygenase (LOX) inhibition was observed by the acetone and methanol extracts at 400 μg/mL while the hexane extract displayed maximum Xanthine oxidase (XO) inhibition (57.0 ± 0.5%). LC-MS profiling of the methanol extract identified several secondary metabolites that might be responsible for the observed activities. The results validate the traditional use of W. fruticosa and present us with potential compounds for further development of novel anti sickling agents.  相似文献

Nettle (Urtica dioica L.) is a herbaceous perennial that has been used for centuries in folk medicine. More recently, nettle extracts have also been used in cosmetics because of the many benefits of their topical application for skin health. Their potential anti-aging action is of particular interest and is primarily ascribed to their antioxidant capacity. Here, using an experimental design approach and a clustering analysis, we linked the phytochemical composition of nettle extracts to their biological activities. This approach confirmed the antioxidant capacity of nettle extracts as well as providing the first evidence of another mechanism for their anti-aging potential involving the inhibition of enzyme activities, such as elastase and collagenase. We attributed these inhibitory effects to ursolic acid and quercetin present in the nettle extracts. Our results also demonstrated the possibility of extracting ursolic acid, quercetin and other phenolic compounds differentially to obtain an extract with a strong antioxidant capacity and anti-aging activities toward both elastase and collagenase. This could be of particular interest for cosmetic applications of nettle extracts.  相似文献

Grape plant (Vitis vinifera L.) is a species of the family Vitaceae. It is an economically important fruit plant abundantly grown all over the world. Vitaceae comprised of 900 species distributed across tropical, sub-tropical and partially temperate sorts of climatic zones. In the current study we aimed to evaluate the antioxidant activities of grape leaves extract from different Pakistani varieties. We have analyzed the crude methanolic extract from the leaves of Pakistani grape varieties (i.e. NARC Black, Sundar Khani, Shogran-1, Shogran-2, Haita, Sheghali, Perlette, and Sultania-C) cultivated in various soil compositions. Total phenolic contents (TPC) of the extract were determind by using Folin-ciocalteu reagent method. Total flavonoid contents (TFC) were determind by the Aluminum Chloride Calorimetric method. Antioxidant activities of the extracts of different concentration were evaluated using Phosphomolybdenum method, Reducing power and DPPH (2, 2-disphenyl-1-picryl-hydrazyl) assays. Assays performed for grape varieties showed great antioxidant potential. All varieties significantly depicted with variation in polyphenolic contents and antioxidant potential with changes in the soil conditions. The highest TPC and TFC were quantified in Shogran-1 97.58 ± 0.09 (GAE, g−1) and Sultania-C 97.47 ± 0.12 (µg QE/g DE) varieties respectively which were grown in (T6) and (T5), while lower were recorded in Sundar khani 5.16 ± 0.23 in (T6). Likewise, Shogran-1 128.26 ± 0.23 (µg AAE/g extract) variety grown in (T6) depicted highest total antioxidant capacity, NARC black 182.06 ± 0.24 (µg AAE/g extract) variety grown in (T6) resulted in maximum reducing power while DPPH free radical scavenging potential were determinded in Sultania-C 80.91 ± 0.10 (T4) equivalent µg/g. In the terms of bioassays, the promising results were showed by compost + sand + clay (T4), compost + clay + dung (T5) and sand + clay + dung (T6). Therefore, we conclude that variation in soil composition has a significant contribution in the quantity of phytochemicals and antioxidants expression. Our study further suggest that use of organic fertilizer (Compost) and loamy sort of soil enhance functioning of most of the varieties in terms of important phytochemicals.  相似文献

In Mexico, the mango crop is affected by anthracnose caused by Colletotrichum species. In the search for environmentally friendly fungicides, chitosan has shown antifungal activity. Therefore, fungal isolates were obtained from plant tissue with anthracnose symptoms from the state of Guerrero in Mexico and identified with the ITS and β-Tub2 genetic markers. Isolates of the Colletotrichum gloeosporioides complex were again identified with the markers ITS, Act, β-Tub2, GADPH, CHS-1, CaM, and ApMat. Commercial chitosan (Aldrich, lot # STBF3282V) was characterized, and its antifungal activity was evaluated on the radial growth of the fungal isolates. The isolated anthracnose-causing species were C. chrysophilum, C. fructicola, C. siamense, and C. musae. Other fungi found were Alternaria sp., Alternaria tenuissima, Fusarium sp., Pestalotiopsis sp., Curvularia lunata, Diaporthe pseudomangiferae, and Epicoccum nigrum. Chitosan showed 78% deacetylation degree and a molecular weight of 32 kDa. Most of the Colletotrichum species and the other identified fungi were susceptible to 1 g L−1 chitosan. However, two C. fructicola isolates were less susceptible to chitosan. Although chitosan has antifungal activity, the interactions between species of the Colletotrichum gloeosporioides complex and their effect on chitosan susceptibility should be studied based on genomic changes with molecular evidence.  相似文献

Euphorbia hirta (L.) plant is traditionally used in Malaysia for the treatment of gastrointestinal, bronchial and respiratory ailments caused by nosocomial infectious agents. Bioactivity-guided fractionation of the methanol extract of the aerial parts of E. hirta and analysis using high-performance liquid chromatography have led to the isolation of two antibacterial compounds. These compounds were identified as caffeic acid (CA) and (–)-epicatechin 3-gallate (ECG) based on spectroscopic analyses and comparison with previously published data. Using broth microdilution method, both ECG and CA had demonstrated significant minimum inhibitory concentration of 15.6 and 31.3 μg/mL respectively, against Pseudomonas aeruginosa. Time-kill assessment of ECG and CA displayed bactericidal effect on P. aeruginosa cells.  相似文献

Toddalia asiatica (Linn.) Lam. is a medical plant traditionally used to treat coughs, fevers, and various diseases. Alkaloids are the main active ingredients in Toddalia asiatica (Linn.) Lam., but traditional methods for screening and separation are complex and labor‐intensive. In this work, an efficient strategy was developed to rapidly screen, identify, and separate neuraminidase inhibitors from Toddalia asiatica (Linn.) Lam. Ultrafiltration, high performance liquid chromatography, and time‐of‐flight mass spectrometry were employed for rapid screening and identification of neuraminidase inhibitors. A two‐phase solvent system comprising n‐hexane/ethyl acetate/methanol/water (5:5:3:7, v/v) was then selected for separation by high‐speed counter‐current chromatography. A sample loading of 200 mg and a stepwise flow rate were achieved by increasing the flow rate from 2 to 4 mL/min after 4 h. Three main fluoroquinoline alkaloids (haplopine, skimmianine, and 5‐methoxydictamnine) along with two coumarins were obtained via one‐step separation and their structures were determined by mass spectrometry and nuclear magnetic resonance. In vitro assays revealed skimmianine with half‐maximal inhibitory concentration of 16.2 ± 0.7 µmol/L was selected as the potential highest neuraminidase inhibitor. The results suggest that ultrafiltration high performance liquid chromatography–mass spectrometry combined with high‐speed counter‐current chromatography is efficient for the screening and isolation of neuraminidase inhibitors from complex natural products.  相似文献

Five new Cu(II), Zn(II), and Ni(II) 3-(4-nitrophenyl)acrylic acid complexes were synthesized and evaluated for inhibitory activity on jack bean urease. All five complexes were structurally determined by single crystal X-ray analysis. Compared with the positive reference acetohydroxamic acid (IC50?=?13.25?μM), Cu(II) complexes 3 and 4 showed the strongest inhibitory activity against jack bean urease (IC50?=?1.23 and 1.17?μM). Ni(II) and Zn(II) complexes also exhibited inhibitory activities (IC50?=?10.09–13.10?μM).  相似文献

In Iran and other parts of Western Asia, the oleaster (Elaeagnus angustifolia L.) fruit is processed in the dried powdery form, and in recent times, increasingly applied/sprinkled in fruit juices such as those made from oranges (Citrus sinensis L.). To our best knowledge, the effectiveness of oleaster fruit extract in fortifying the orange juice has not yet been reported and the knowledge of this will greatly benefit the consumers, particularly those around the Western Asia region. This current work, therefore, investigated the changes in physicochemical, free radical activity, total phenolic compounds, and sensory properties of orange juice fortified with different oleaster fruit extracts. The orange juice mix formulation comprised different concentrations (5, 10, 15, 20, and 25%) of oleaster (alcoholic, aqueous, and hydro-alcoholic) extracts. The control comprised orange concentrate (4% w/v), sugar (8.5% w/v), and citric acid (0.1% w/v) brought to the desirable volume with water. As the free radical activity depicted the antioxidant properties, the physicochemical aspects of this work involved the determinations of Brix, density, ash, pH, total acidity, sucrose, and total sugar, whereas the sensory aspects involved the determinations of color and taste. Whilst the aqueous oleaster 20 and 25% extracts produced notable physicochemical differences in the orange juice mix, both free radical activity, and phenolic compounds significantly increased (p < 0.05) after 30 days despite resembling (p > 0.05) those of control at day 1. More so, the increases in aqueous, alcoholic, and hydro-alcoholic oleaster extracts would decrease (p < 0.05) the sensory color and taste of the orange juice mix in this study.  相似文献

Acetone extract of faba bean (Vicia faba L.) was found to be highest total phenol and flavonoid content among all extracts. Antioxidant activity for inhibition percentage (free radical scavenging activity) had 86.47% for acetone extract, and 97.36% for ascorbic acid respectively. IC50 value of ascorbic acid and acetone extact were found to be 9 μg/mL ± 0.20 and 30 μg/mL ± 0.21. Faba bean seeds had catechin, epicatechin, gallic acid and ellagic acid which on molecular docking study revealed that it binds effectively with xanthine oxidase by binding energy of –7.78, –6.11, –6.39, –5.78 kcal/mol respectively compared to allopurinol drug having binding energy of –4.94 kcal/mol. Gallic acid, ellagic acid, catechin, epicatechin (polyphenols) and allopurinol bind other than catalytic residues (Glu-1261) of xanthine oxidase. In vitro and in silico analysis recommended that mode of enzyme inhibition was mixed type.  相似文献

The various health challenges posed by conventional candies necessitate the development of health-promoting and phytochemicals supplemented varieties. The current study explored medicinal properties of date palm and tamarind in the formulation of a starch-based candy, using response surface method for the production optimization. Experimental design variables include the starch (90–100%), date palm (0–10%), and tamarind (0–10%), while the functional, phytochemical, and antioxidant properties were the responses. The candy modification response results are as follows: pH (2.80–3.00), crude fibre (4.41–6.43%), soluble sugar (91.69–126.96%), total titrable acidity (90.00–120.00%), total phenolics (0.039–0.063 mg GA/mL), total flavonoids (0.0006–0.001 mg RU/mL), vitamin C (0.362–0.516 mg/mL), hydroxyl radical scavenging activities (14.28–26.67%), 1, 1- diphenyl-2-picryhydrazyl (1.12–3.88%) and ferric reducing property (0.09–0.31 mg/mL). The experiments revealed that most of the nutritional properties of the candies increased favorably with the incorporation of date palm and tamarind pulps and decreasing starch content. Significant (p ≤ 0.05) interactive effects were revealed between the diverse variables. The experiments also revealed the positive influence of the date palm and tamarind fruit pulps on the functional, phytochemical, and antioxidant properties of starch-based candies; hence demonstrating the possibility of developing nutraceutical based-candies with a more beneficial health effect.  相似文献

A high-performance liquid chromatography coupled with diode array detector and mass spectrometry (HPLC-DAD-MS) method was developed to evaluate the quality of Rhizoma Belamcandae (Belamcanda chinensis (L.) DC.) through establishing chromatographic fingerprint and simultaneous determination of seven phenolic compounds. The analysis was achieved on an Alltima C(18) analytical column (250 mm x 4.6 mm i.d. 5 microm) using linear gradient elution of acetonitrile-0.1% trifluoroacetic acid. The correlation coefficients of similarity were determined from the HPLC fingerprints, and they shared a close similarity. By using an online APCI-MS/MS, twenty phenols were identified. In addition, seven of these phenols including mangiferin, 7-O-methylmangiferin, tectoridin, resveratrol, tectorigenin, irigenin and irisflorentin were quantified by the validated HPLC-DAD method. These phenols are considered to be major constituents in Rhizoma Belamcandae, and are generally regarded as the index for quality assessment of this herb. This developed method by having a combination of chromatographic fingerprint and quantification analysis could be applied to the quality control of Rhizoma Belamcandae.  相似文献

The feasibility of early disease detection in potato seeds storage monitoring of volatile organic compounds (VOCs) and plant physiological markers was evaluated using 10 fungal and bacterial pathogens of potato in laboratory-scale experiments. Data analysis of HS-SPME-GC-MS revealed 130 compounds released from infected potatoes, including sesquiterpenes, dimethyl disulfide, 1,2,4-trimethylbenzene, 2,6,11-trimethyldodecane, benzothiazole, 3-octanol, and 2-butanol, which may have been associated with the activity of Fusarium sambucinum, Alternaria tenuissima and Pectobacterium carotovorum. In turn, acetic acid was detected in all infected samples. The criteria of selection for volatiles for possible use as incipient disease indicators were discussed in terms of potato physiology. The established physiological markers proved to demonstrate a negative effect of phytopathogens infecting seed potatoes not only on the kinetics of stem and root growth and the development of the entire root system, but also on gas exchange, chlorophyll content in leaves, and yield. The negative effect of phytopathogens on plant growth was dependent on the time of planting after infection. The research also showed different usefulness of VOCs and physiological markers as the indicators of the toxic effect of inoculated phytopathogens at different stages of plant development and their individual organs.  相似文献

Pot experiments were conducted to study the efficacy of a slow sulfur-releasing fertilizer, sulfur glass fritz (SGF 1), on growth, photosynthesis, and sulfur, and nitrogen assimilation potentials of brown mustard (Brassica juncea L. Czern. & Coss. cv. Pusa Jaikisan). Growth as indicated by biomass accumulation slowed down in response to the application of sulfur glass fritz. A similar trend was observed in the case of photosynthesis rate. The activity of two marker enzymes, ATP-sulfurylase and nitrate reductase, showed very low levels of activity, indicating poor assimilation of sulfur and nitrogen by the plant under sulfur glass fritz. It is therefore concluded that the release of sulfur by sulfur glass fritz is too slow and that the initial nonavailability of sulfur to the plants could lead to suboptimization of both sulfur- and nitrogen-assimilating enzymes. These factors may contribute to low rates of photosynthesis and poor growth.  相似文献

To identify the natural antifungal agents, the antifungal activities of Melaleuca leucadendra (L.) L. essential oil (ML-EO) from Fujian Province of China were assayed. Treatment of ML-EO in combination with the front-line using antibiotics against Candida led to synergistic effects. Electron microscopy analysis on the oil treated C. albicans cells revealed the formation of mesosome-like structures, suggesting well the membrane damage caused by the essential oil. The Griess assay by monitoring NO production in LPS-stimulated RAW264.7 cells indicated the potent anti-inflammatory activity of ML-EO. In comparison with the marked essential oil of Melaleuca alternifolia EO, ML-EO had almost the same chemical components. In general, the antifungal activity of ML-EO and its synergistic interactions with conventional antibiotics were able to lead the development of new treatment strategies on Candida infection.  相似文献

A sensitive and reliable ultra-high performance liquid chromatography-electrospray ionization-tandem mass spectrometry has been developed and partially validated to evaluate the quality of Semen Cassiae (Cassia obtusifolia L.) through simultaneous determination of 11 anthraquinones and two naphtha-γ-pyrone compounds. The analysis was achieved on a Poroshell 120 EC-C(18) column (100 mm × 2.1 mm, 2.7 μm; Agilent, Palo Alto, CA, USA) with gradient elution using a mobile phase that consisted of acetonitrile-water (30 mM ammonium acetate) at a flow rate of 0.4 mL/min. For quantitative analysis, all calibration curves showed perfect linear regression (r(2) > 0.99) within the testing range. This method was also validated with respect to precision and accuracy, and was successfully applied to quantify the 13 components in nine batches of Semen Cassiae samples from different areas. The performance of developed method was compared with that of conventional high-performance liquid chromatography method. The significant advantages of the former include high-speed chromatographic separation, four times faster than high-performance liquid chromatography with conventional columns, and great enhancement in sensitivity. This developed method provided a new basis for overall assessment on quality of Semen Cassiae.  相似文献

The local botanical Imperata cylindrica in Cameroon was investigated for its antibacterial potency. The methanol extract afforded a total of seven compounds, including five hitherto unreported compounds comprising three flavonoids (1–3) and two C-15 isoprenoid analogues (4 and 5) together with known derivatives (6 and 7). The novelty of the flavonoids was related to the presence of both methyl and prenyl groups. The potential origin of the methyl in the flavonoids is discussed, as well as the chemophenetic significance of our findings. Isolation was performed over repeated silica gel and Sephadex LH-20 column chromatography and the structures were elucidated by (NMR and MS). The crude methanol extract and isolated compounds showed considerable antibacterial potency against a panel of multi-drug resistant (MDR) bacterial strains. The best MIC values were obtained with compound (2) against S. aureus ATCC 25923 (32 µg/mL) and MRSA1 (16 µg/mL).  相似文献

A method of microwave-assisted extraction coupled with high-speed counter-current chromatography was established for separation and purification of isopimpinellin, pimpinellin and phellopterin from Toddalia asiatica (L.) Lam. The conditions of MAE including the extraction solvent, size of sample, solid/liquid ratio, extraction temperature and extraction time were optimized by a mono-factor test. That is, 2.0 g dried powder of T. asiatica (L.) Lam of 0.30-0.15 mm size was extracted with 20 mL (solid/liquid ratio of 1:10, g/mL) methanol under 50 °C for 1 min. The crude extract was separated and purified by high-speed counter-current chromatography with hexane-ethyl acetate-methanol-water (5:5:5.5:4.5, v/v/v/v) solvent system. 0.85 mg/g of isopimpinellin, 2.55 mg/g of pimpinellin and 0.95 mg/g of phellopterin were obtained from original sample in one-step within 240 min, the purity determined by high performance liquid chromatography was 95.0%, 99.1% and 96.4%, respectively. Their chemical structures were further identified by mass spectroscopy and nuclear magnetic resonance spectroscopy. The results demonstrated that microwave-assisted extraction coupled with high-speed counter-current chromatography was a feasible, economical and efficient technique for rapid extraction, separation and purification of effective compounds from natural products.  相似文献
