在B3PW91/6-311+G(d)计算水平上, 计算并讨论了Ni4Ti2, [Ni4Ti2]2+, [Ni4Ti2]2-与Ni4Ti4, [Ni4Ti4]2+, [Ni4Ti4]2-团簇的几何结构和芳香性. 在构型优化过程中得到了Ni4Ti2(D4h), [Ni4Ti2]2+(D4h), [Ni4Ti2]2-(D4h)和Ni4Ti4(D2h)4个稳定构型, 发现当引入上下2个Ti原子后, Ni4环成为了平面正方形构型. 核无关化学位移(NICS)计算结果表明, Ni4Ti2(D4h)与Ni4Ti4(D2h)的NICS值为正, 而[Ni4Ti2]2+(D4h)和[Ni4Ti2]2-(D4h)的NICS值为负, 且[Ni4Ti2]2-(D4h)的NICS值更负. 同时还发现, 由s与d轨道参与形成的反磁性环流是引起[Ni4Ti2]2+(D4h)和[Ni4Ti2]2-(D4h)具有较大芳香性的主要原因; 其中Ti原子主要提供dz2与s轨道, 而Ni原子主要利用其dz2与dx2-y2轨道形成正方形环, 它们之间构成了球状的d轨道环流, 且[Ni4Ti2]2+(D4h)和[Ni4Ti2]2-(D4h)中还有非常明显的π轨道环流.  相似文献

Griseofulvin is an antifungal polyketide metabolite produced mainly by ascomycetes. Since it was commercially introduced in 1959, griseofulvin has been used in treating dermatophyte infections. This fungistatic has gained increasing interest for multifunctional applications in the last decades due to its potential to disrupt mitosis and cell division in human cancer cells and arrest hepatitis C virus replication. In addition to these inhibitory effects, we and others found griseofulvin may enhance ACE2 function, contribute to vascular vasodilation, and improve capillary blood flow. Furthermore, molecular docking analysis revealed that griseofulvin and its derivatives have good binding potential with SARS-CoV-2 main protease, RNA-dependent RNA polymerase (RdRp), and spike protein receptor-binding domain (RBD), suggesting its inhibitory effects on SARS-CoV-2 entry and viral replication. These findings imply the repurposing potentials of the FDA-approved drug griseofulvin in designing and developing novel therapeutic interventions. In this review, we have summarized the available information from its discovery to recent progress in this growing field. Additionally, explored is the possible mechanism leading to rare hepatitis induced by griseofulvin. We found that griseofulvin and its metabolites, including 6-desmethylgriseofulvin (6-DMG) and 4- desmethylgriseofulvin (4-DMG), have favorable interactions with cytokeratin intermediate filament proteins (K8 and K18), ranging from −3.34 to −5.61 kcal mol−1. Therefore, they could be responsible for liver injury and Mallory body (MB) formation in hepatocytes of human, mouse, and rat treated with griseofulvin. Moreover, the stronger binding of griseofulvin to K18 in rodents than in human may explain the observed difference in the severity of hepatitis between rodents and human.  相似文献

在RHF/6-311G**,RHF/6-311+G**和B3LYP/6-311+G**水平优化得到3,4-二硫方酸(3,4-二巯基-3-环丁烯-1,2-二酮)三种平面构象异构体的平衡几何构型.用MP2(Full)/6-311G**//RHF/6-311G**方法计算单点能量,发现ZZ型异构体是能量最低构象,且ZZ和ZE型能量非常接近.用优化的最稳定构象ZZ型异构体在RHF/6-311G**//RHF/6-311G**,RHF/6-311G**//RHF/6-311G**,MP2(Full)/6-311G**//RHF/6-311G**和B3LYP/6-311G**//B3LYP/6-311G**水平计算其气相酸性(ΔG0)和同键反应芳香性稳定化能(HASE).用基团加和法(Group Increment Approach)在RHF/6-311G**//RHF/6-311G**和B3LYP/6-311G**//B3LYP/6-311G**水平计算其磁化率增量(Λ).计算结果表明,标题化合物的同键反应芳香性稳定化能和磁化率增量均为负值,表明它具有芳香性,实现了标题化合物芳香性的几何、能量和磁性的判定.  相似文献

Azlactone anions—the key intermediates in the classical Erlenmeyer synthesis of amino acids—apparently possess aromatic stabilization, as indicated by the relative rate of base catalyzed deuterium exchange in the following analogs: 1-methyl-2-phenyl-5(4H)-imidazolone > 2-phenyl-5(4H)-oxazolone (azlactone) > 3,3-dimethyl-2-phenyl-4(3H)-pyrrolone. This is paralleled by the relative rate of condensation of these compounds with hexadeuteroacetone. Reported pKa data also suggest that the azlactone products of the Erlenmeyer synthesis are analogs of the fulvenes.  相似文献

Interactions of dsDNA and ssDNA, at the surface of gold and silver electrodes, with three novel anthraquinone derivatives: 3‐(9′,10′‐dioxo‐9′,10′‐dihydro‐anthracen‐1‐yl)‐7,11‐di(carboxymethyl)‐3,7,11‐triazatridecanedioic acid, (AQ‐1); 1‐(9′,10′‐dioxo‐9′,10′‐dihydro‐anthracen‐1yl)‐9‐carboxymethyl‐5‐methyl‐1,5,9‐triazaundecanoicacid, (AQ‐2); and N‐(2‐(9,10‐dioxo‐9,10‐dihydro‐anthracen‐1‐ylamino)ethyl)‐2‐(1,4,10,13‐tetraoxa‐7,16‐diazacyclooctadecan‐7‐yl)acetamide, (AQ‐3) are studied. These derivatives are well soluble in water and phosphate buffer solutions. The square wave voltammetric behavior of these redox indicators is described and the parameters of interactions with DNA are reported. It is also pointed out that these compounds can be employed as the hybridization indicators. The difference in the binding ability of the particular redox indicator to single and double stranded DNA can be used for the detection of the complementary nucleic acids.  相似文献

Xanthine, caffeine and three isomeric dimethyl xanthines (theobromine, theophylline and paraxanthine) are often described as aromatic on various criteria. Here we complete the picture by assessing these molecules for aromaticity on the ring-current criterion. Magnetic response calculations are performed at the B3LYP and CHF/6-31G∗∗ ipsocentric levels of theory on structures optimised at the B3LYP/6-31G∗∗ level. All five systems display consistent π-electron ring-currents delocalised around the imidazole moiety in all cases; these are accompanied by localised features on the six-membered rings attributed to nitrogen and oxygen ‘lone-pair’ π-electron circulations. All are therefore aromatic on the magnetic criterion, with a ‘locally delocalised’ ring current in the imidazole moiety, similar to those in the isolated imidazole and methylimidazole molecules.  相似文献

首次报道通过充石蜡石墨电极电化学氧化腐蚀制备的微孔穴电极,成功地用于 ss-DNA的固载和杂化反应,证明了这种微孔穴石墨充石蜡电极可以固载较多量的 ss-DNA(包括解链的小牛胸腺DNA和15-mer ss-DNA),可以成为性能良好的DNA杂 化探针。用Co(phen)_3~(3+)电化学指示剂和循环伏安方法,讨论了电极预氧化腐 蚀的时间和ss-DNA固载时间的影响,提出了DNA探针在微孔穴三维结构中固载的可 能机理。  相似文献

An updated review of emerging plant proteases with potential biotechnological application is presented. Plant proteases show comparable or even greater performance than animal or microbial proteases for by-product valorization through hydrolysis for, for example, cheese whey, bird feathers, collagen, keratinous materials, gelatin, fish protein, and soy protein. Active biopeptides can be obtained as high added value products, which have shown numerous beneficial effects on human health. Plant proteases can also be used for wastewater treatment. The production of new plant proteases is encouraged for the following advantages: low cost of isolation using simple procedures, remarkable stability over a wide range of operating conditions (temperature, pH, salinity, and organic solvents), substantial affinity to a broad variety of substrates, and possibility of immobilization. Vegetable proteases have enormous application potential for the valorization of industrial waste and its conversion into products with high added value through low-cost processes.  相似文献
