A neutral naphthyl methacrylate‐based monolith (NMM) was introduced for RP‐CEC of various aromatic compounds via hydrophobic and π interactions. It was characterized over a wide range of elution conditions to gain insight into its RP retention mechanism toward the various solute probes under investigation. First, the NMM column exhibited cathodal EOF at various mobile phase compositions and pH suggesting that although the NMM column is void of fixed charges, it acquires a negative zeta potential. It is believed that the negative zeta potential is imparted by the adsorption of mobile phase ions to the NMM surface. The NMM column exhibited π–π interactions in addition to hydrophobic interactions due to the aromatic and nonpolar nature of its naphthyl ligands. In all cases, the retention of the various aromatic test solutes including PAHs, benzene derivatives, toluene derivatives, anilines and toluidine, tolunitrile and nitrotoluene positional isomers on the NMM column were compared to their retention on an octadecyl acrylate‐based monolithic column. Not only were the values of the retention factors of the various solutes on the NMM column higher than those obtained on the octadecyl acrylate‐based monolithic column under otherwise the same CEC conditions, but the elution orders were also different on both columns with a superior and unique selectivity exhibited by the NMM column.  相似文献

Terpolymers composed of Nn‐propylacrylamide (NPAAm), butyl methacrylate (BMA), and N,N‐diethylaminoethyl methacrylate (DEAEMA) were prepared in an attempt to investigate the temperature‐induced phase transition and its mechanism. Poly(NPAAm) showed the lower critical solution temperature (LCST) around 24°C in water. With the incorporation of DEAEMA with NPAAm, the LCST change was characterized by an initial increase. However, the LCST was shifted to the lower temperature at the later stage. This might be explained in terms of hydrophilic/hydrophobic contribution of DEAEMA to the LCST. The swelling behavior of copolymer gel in the various solvents and spin‐lattice relaxation time (T1) study by NMR strongly suggested the hydrophilic/hydrophobic contribution of DEAEMA to the LCST depending on the local environment. © 1999 John Wiley & Sons, Inc. We recently presented a short communication on the preparation of epoxy‐based monoliths possessing highly ordered structures by polymerization induced phase separation based on the spinodal decomposition. In this article, we describe in detail on reaction mechanisms and structural properties of the epoxy‐based monoliths with well‐controlled macropores in the micrometer range. We prepared epoxy‐based monoliths based on diglycidyl ether of bisphenol A, bis(4‐aminocyclohexyl)methane, and polyethylene glycol with a bicontinuous structure by in situ step‐growth polymerization. Different morphology of epoxy‐based monoliths could be obtained by changing formulation of monomers and porogenic solvents. Characterizations of their morphologies were performed using scanning electron microscopy, mercury intrusion porosimetry, small angle X‐ray scattering, and gas adsorption measurement (BET method). © 2008 Wiley Periodicals, Inc. Monolithic columns were synthesized inside 1.02 mm internal diameter fused‐silica lined stainless‐steel tubing. Styrene and butyl, hexyl, lauryl, and glycidyl methacrylates were the functional monomers. Ethylene glycol dimethacrylate and divinylbenzene were the crosslinkers. The glycidyl methacrylate polymer was modified with gold nanoparticles and dodecanethiol (C12). The separation of alkylbenzenes was investigated by isocratic elution in 60:40 v/v acetonitrile/water. The columns based on polystyrene‐co‐divinylbenzene and poly(glycidyl methacrylate)‐co‐ethylene glycol dimethacrylate modified with dodecanethiol did not provide any separation of alkyl benzenes. The poly(hexyl methacrylate)‐co‐ethylene glycol dimethacrylate and poly(lauryl methacrylate)‐co‐ethylene glycol dimethacrylate columns separated the alkyl benzenes with plate heights between 30 and 60 μm (50 μL min?1 and 60°C). Similar efficiency was achieved in the poly(butyl methacrylate)‐co‐ethylene glycol dimethacrylate column, but only at 10 μL min?1 (0.22 mm s?1). Backpressures varied from 0.38 MPa in the hexyl methacrylate to 13.4 MPa in lauryl methacrylate columns (50 μL min?1 and 60°C). Separation of proteins was achieved in all columns with different efficiencies. At 100 μL min?1 and 60°C, the lauryl methacrylate columns provided the best separation, but their low permeability prevented high flow rates. Flow rates up to 500 μL min?1 were possible in the styrene, butyl and hexyl methacrylate columns.  相似文献

A polar polymethacrylate‐based monolithic column was introduced and evaluated as a hydrophilic interaction CEC stationary phase. The monolithic stationary phase was prepared by in situ copolymerization of a neutral monomer 2‐hydroxyethyl methacrylate and a polar cross‐linker N,N′‐methylene bisacrylamide in a binary porogenic solvent consisting of dodecyl alcohol and toluene. The hydroxyl and amino groups at the surface of the monolithic stationary phase provided polar sites which were responsible for hydrophilic interactions. The composition and proportion of the polymerization mixture was investigated in detail. The mechanical stability and reproducibility of the obtained monolithic column preformed was satisfied. The effects of pH and organic solvent content on the EOF and the separation of amines, nucleosides, and narcotics on the optimized monolithic column were investigated. A typical hydrophilic interaction CEC was observed on the neutral polar stationary phase. The optimized monolithic column can obtain high‐column efficiencies with 62 000–126 000 theoretical plates/m and the RSDs of column‐to‐column (n = 9), run‐to‐run (n = 5), and day‐to‐day (n = 3) reproducibility were less than 6.3%. The calibration curves of these five narcotics exhibited good linearity with R in the range of 0.9959–0.9970 and linear ranges of 1.0–200.0 μg/mL. The detection limits at S/N = 3 were between 0.2 and 1.2 μg/mL. The recoveries of the separation of narcotics on the column were in the range of 84.0–108.6%. The good mechanical stability, reproducibility, and quantitation capacity was suitable for pressure‐assisted CEC applications.  相似文献

Metal‐organic frameworks consisting of amino‐modified MIL‐101(M: Cr, Al, and Fe) crystals have been synthesized and subsequently incorporated to glycidyl methacrylate monoliths to develop novel stationary phases for nano‐liquid chromatography. Two incorporation approaches of these materials in monoliths were explored. The metal‐organic framework materials were firstly attached to the pore surface through reaction of epoxy groups present in the parent glycidyl methacrylate‐based monolith. Alternatively, NH2‐MIL‐101(M) were admixed in the polymerization mixture. Using short time UV‐initiated polymerization, monolithic beds with homogenously dispersed metal‐organic frameworks were obtained. The chromatographic performance of embedded UV‐initiated composites was demonstrated with separations of polycyclic aromatic hydrocarbons and non‐steroidal anti‐inflammatory drugs as test solutes. In particular, the incorporation of the NH2‐MIL‐101(Al) into the organic polymer monoliths led to an increase in the retention of all the analytes compared to the parent monolith. The hybrid monolithic columns also exhibited satisfactory run‐to‐run and column‐to‐column reproducibility.  相似文献

In this work, a novel polysaccharide‐based chiral stationary phase, cellulose tris(4‐chloro‐3‐methylphenylcarbamate), also called Sepapak 4 has been evaluated for the chiral separation of amlodipine (AML) and its two impurities. AML is a powerful vasodilatator drug used for the treatment of hypertension. Capillary columns of 100 μm id packed with the chiral stationary phase were used for both nano‐LC and CEC experiments. The optimization of the mobile phase composed of ACN/water, (90:10, v/v) containing 15 mM ammonium borate pH 10.0 in nano‐LC allowed the chiral separation of AML and the two impurities, but not in a single run. With the purpose to obtain the separation of the three pairs of enantiomers simultaneously, CEC analyses were performed in the same conditions achieving better enantioresolution and higher separation efficiencies for each compound. To fully resolve the mixture of six enantiomers, parameters such as buffer pH and concentration sample injection have been then investigated. A mixture of ACN/water (90:10, v/v) containing 5 mM ammonium borate buffer pH 9.0 enabled the complete separation of the three couples of enantiomers in less than 30 min. The optimized CEC method was therefore validated and applied to the analysis of pharmaceutical formulation declared to contain only AML racemate.  相似文献

This paper describes the synthesis and chromatographic evaluation of a new polar‐embedded stationary phase, which utilized 2,4,6‐trichloro‐1,3,5‐triazine as the spacer. The resulting materials were characterized by elemental analysis, IR, and solid‐state 13C NMR. Empirical test mixtures were utilized to evaluate the column, and showed that it had good performance for basic compounds and high selectivity for polyaromatic hydrocarbons. Moreover, the novel stationary phase has unique property, especially in the separation of “homologous alkaloids” from natural products.  相似文献

Porous polymer monoliths have been used to develop an online solid‐phase extraction with liquid chromatography method for determination of dopamine in urine as well as for a continuous monitoring of dopamine in flowing system. A polymerization mixture containing 4‐vinylphenylboronic acid monomer has been used to prepare a trapping column based on specific ring formation reaction with dopamine cis‐diol functionality. Additionally, a monolithic stationary phase with zwitterion functionality has been used to prepare capillary column for the separation of dopamine. Experimental conditions including molarity, pH, and flow rate of the loading buffer together with a valve switching time have been optimized to provide the highest recovery for dopamine. Experimental setup has been used to determine dopamine in a urine. By using both calibration curve and standard addition method, the dopamine level was determined to be 1.19 and 1.28 mg/L, respectively. Further, we have used experimental design to optimize coupling of two extraction monolithic loops to separation capillary column with monolithic phase for a comprehensive monitoring of dopamine. After multivariate analysis, sample loading flow‐rate and a flow‐rate of flushing buffer were selected as the most significant variables. Optimized experimental setup was applied to continuously monitor dopamine degradation.  相似文献

Aiming to develop a suitable ion exchange membrane for vanadium redox flow battery (VRB), a new kind of imidazolium salt type anion exchange membrane based on the copolymer of N‐vinylimidazole and 2,2,2‐trifluoroethyl methacrylate has been prepared. The membrane is characterized by means of water uptake, ion‐exchange capacity, ionic conductivity, and thermal stability. Furthermore, a VRB with this membrane is assembled, and the performance of such VRB is evaluated. The permeability experiments show that this membrane has reasonable low permeability of vanadium ions. The coulombic efficiency (CE) and energy efficiency (EE) of VRB with the synthesized membrane are 99.5% and 75.0%, whereas the CE and EE of the VRB with Nafion® 117 membrane are 82.6% and 72.6%, respectively. The synthesized membrane shows good chemical stability in VRB via more than 4000 cycles test. Therefore, this membrane shows good applicable potential in VRB. A novel polymethacrylate‐based monolithic column with covalently bonded zwitterionic functional groups was prepared by in situ copolymerization of N,N‐dimethyl‐N‐methacryloxyethyl N‐(3‐sulfopropyl) ammonium betaine (SPE), pentaerythritol triacrylate (PETA), and vinylsulfonic acid (VS) in a binary porogenic solvent consisting of cyclohexanol and ethylene glycol. This monolith was developed as a separation column for CEC. While SPE functioned as both an electrostatic interaction stationary phase and the polar ligand provider, VS was employed to generate EOF. PETA, which has much more hydrophilicity due to a hydroxyl sub‐layer, was used to replace ethylene dimethacrylate as a cross‐linker. The monolith provided an adequate EOF when VS level was maintained at 0.6% w/w. Different monolithic stationary phases were easily prepared by adjusting the ratio of PETA/SPE in the polymerization solution as well as the composition of the porogenic solvent. The observed RSD were ≤3.6, ≤4.3 and ≤5.6% for the EOF velocity, retention time, and column efficiency, respectively. The column efficiencies greater than 145 000 theoretical plates/m for thiourea and 132 000 theoretical plates/m for charged cytidine were obtained. The poly(SPE‐co‐PETA‐co‐VS) monolith showed good selectivity for neutral and charged polar analytes. It was found that the separation mechanism of charged polar solutes was attributed to a mixed mode of hydrophilic interaction and electrostatic interaction, as well as electrophoresis. No peak tailing was observed for the separation of basic compounds, such as basic nucleic acid bases and nucleoside on the monolith.  相似文献

A chiral capillary monolithic column for enantiomer separation in capillary electrochromatography was prepared by coating cellulose tris(3,5‐dimethylphenylcarbamate) on porous glycidyl methacrylate‐co‐ethylene dimethacrylate monolith in capillary format grafted with chains of [2(methacryloyloxy)ethyl] trimethylammonium chloride. The surface modification of the monolith by the photografting of [2(methacryloyloxy)ethyl] trimethylammonium chloride monomer as well as the coating conditions of cellulose tris(3,5‐dimethylphenylcarbamate) onto the grafted monolithic scaffold were optimized to obtain a stable and reproducible chiral stationary phase for capillary electrochromatography. The effect of organic modifier (acetonitrile) in aqueous mobile phase for the enantiomer separation by capillary electrochromatography was also investigated. Several pairs of enantiomers including acidic, neutral, and basic analytes were tested and most of them were partially or completely resolved under aqueous mobile phases. The prepared monolithic chiral stationary phases exhibited a good stability, repeatability, and column‐to‐column reproducibility, with relative standard deviations below 11% in the studied electrochromatographic parameters.  相似文献

A new humic acid stationary phase was prepared by immobilizing humic acid onto aminopropyl silica via an amide linkage formation and used, for the first time, for the separation and quantification of the tocopherol compounds in cold‐pressed oil samples under normal‐phase high‐performance liquid chromatography conditions. Parameters affecting the chromatographic separation such as mobile phase composition and flow rate were optimized. By evaluating the calculations of capacity factor, asymmetry factor, resolution, selectivity factor, and theoretical plate number, the best separation was obtained with isocratic elution of n‐hexane and isopropyl alcohol (99:1% v/v) at a flow rate of 1.0 mL/min. The effluent was monitored by a fluorescence detector set at excitation and emission wavelengths 295 and 330 nm, respectively. All compounds were separated in 20 min. The method was validated according to international guidelines and found to be linear in a wide concentration range, also the mean recovery of the compounds ranged from 97.9 to 99.2%, with a CV less than 2.7% in all cases. The results showed that the developed stationary phase is suitable for the separation and quantification of the tocopherol compounds in real oil samples.  相似文献

Protein retention is very sensitive to the change of solvent composition in reversed‐phase liquid chromatography for so called “on–off” mechanism, leading to difficulty in mobile phase optimization. In this study, a novel 3‐chloropropyl trichlorosilane ligand bonded column was prepared for protein separation. The differences in retention characteristics between the 3‐chloropropyl trichlorosilane ligand bonded column and n‐alkyl chain modified (C2, C4, C8) stationary phases were elucidated by the retention equation . Retention parameters (a and c) of nine standard proteins with different molecular weights were calculated by using homemade software. Results showed that retention times of nine proteins were similar on four columns, but the 3‐chloropropyl trichlorosilane ligand bonded column obtained the lowest retention parameter values of larger proteins. It meant that their retention behavior affected by acetonitrile concentration would be different due to lower |c| values. More specifically, protein elution windows were broader, and retentions were less sensitive to the change of acetonitrile concentration on the 3‐chloropropyl trichlorosilane ligand bonded column than that on other columns. Meanwhile, the 3‐chloropropyl trichlorosilane ligand bonded column displayed distinctive selectivity for some proteins. Our results indicated that stationary phase with polar ligand provided potential solutions to the “on–off” problem and optimization in protein separation.  相似文献

Negative ion CID spectra of N‐linked glycans released from glycoproteins contain many ions that are diagnostic for specific structural features such as the detailed arrangement of antennae and the location of fucose residues. Identification of such ions requires reference glycans that are often difficult to acquire in a pure state. The recent acquisition of a sample of N‐glycans from a patient lacking the enzyme N‐acetylglucosaminyltransferase‐2 provided an opportunity to investigate fragmentation of glycans lacking a 6‐antenna. These glycans contained one or two galactose‐N‐acetylglucosamine‐chains attached to the 3‐linked mannose residue of the trimannosyl‐chitobiose core with and without fucose substitution. The spectra from the patient sample clearly defined the antenna distribution and showed striking differences from the spectra of isomeric compounds obtained from normal subjects. Furthermore, they provided additional information on previously identified antenna‐specific fragment ions and indicated the presence of additional ions that were diagnostic of fucose substitution. Glycans obtained from such enzyme‐deficient patients can, thus, be a valuable way of obtaining spectra of specific isomers in a relatively pure state for interpretation of mass spectra. A novel method for the sensitive and selective identification and quantification of N‐acylphosphatidylethanolamine molecular species was developed. Samples were prepared using a combination of liquid–liquid and solid‐phase extraction, and intact N‐acylphosphatidylethanolamine species were determined by reversed‐phase high‐performance liquid chromatography coupled to positive electrospray tandem mass spectrometry. As a result of their biological functions as precursors for N‐acylethanolamines and as signaling molecules, tissue concentrations of N‐acylphosphatidylethanolamines are very low, and their analysis is additionally hindered by the vast excess of other sample components. Our sample preparation methods are able to selectively separate the analytes of interest from any expected biological interferences. Finally, the highest selectivity is achieved by coupling chromatographic separation and two N‐acyl chain specific selected reaction monitoring scans per analyte, enabling identification of both the N‐acyl chain and the phosphatidylethanolamine moiety. The validated method is suitable for the reliable quantification of N‐acylphosphatidylethanolamine species from rat brain with a lower limit of quantification of 10 pmol/g and a linear range up to 2300 pmol/g. In total, 41 N‐acylphosphatidylethanolamine molecular species with six different N‐acyl chains, amounting to a total concentration of 3 nmol/g, were quantified.  相似文献

An ionic liquid was incorporated into the porous polymer monoliths to afford stationary phases with enhanced chromatographic performance for small molecules in reversed‐phase high‐performance liquid chromatography. The effect of the ionic liquid in the polymerization mixture on the performance of the monoliths was studied in detail. While monoliths without ionic liquid exhibited poor resolution and low efficiency, the addition of ionic liquid to the polymerization mixture provides highly increased resolution and high efficiency. The chromatographic performances of the monoliths were demonstrated by the separations of various small molecules including aromatic hydrocarbons, isomers, and homologues using a binary polar mobile phase. The present column efficiency reached 27 000 plates/m, which showed that the ionic liquid monoliths are alternative stationary phases in the separation of small molecules by high‐performance liquid chromatography  相似文献

A systematic study of the behavior of several common mobile‐phase volume markers using traditional and polar‐group‐containing reversed‐phase stationary phases is presented. Examined mobile‐phase volume markers include two neutral molecules, uracil and thiourea, concentrated (0.10 M) and dilute (0.0001 M) KNO3, and D2O. Mobile‐phase volumes are examined over the entire reversed‐phase mobile‐phase range of 100% water to 100% methanol or acetonitrile. The behavior of these mobile‐phase volume markers is compared with a maximum theoretical value (i.e. the void volume), as determined by pycnometry. The data suggest that: (i) uracil begins to fail as a mobile‐phase volume marker in mobile phases below about 40% strong solvent for polar group containing phases; (ii) in nearly all cases, the mobile‐phase volume measured dynamically is smaller than the pycnometric void volume; (iii) a significant dependence of measured mobile‐phase volume on salt concentration is seen on the polar endcapped phase, which is not observed on the traditional and embedded polar group phase; and (iv) D2O does not work well as a mobile‐phase volume marker with polar‐group‐containing phases, possibly due to interaction with the stationary phase polar group.  相似文献

A novel o‐phenanthroline‐immobilized ionic‐liquid‐modified hybrid monolith for capillary electrochromatography was synthesized based on chloropropyl‐silica, which was prepared by the in situ polymerization of tetramethoxysilane and 3‐chloropropyltrimethoxysilane via a sol–gel process. The morphology of the hybrid monolith was characterized by scanning electron microscopy, and relatively stable anodic electroosmotic flow was observed under a broad pH ranged from pH 3.0 to 9.0. The separation mechanism was investigated by separating four neutral molecules (toluene, dimethylformamide, formamide, and thiourea). The obtained hybrid monolith possessed an obviously reversed‐phase retention mechanism, but when the acetonitrile content in the mobile phase was >90% v/v, a weak hydrophilic mechanism was observed on the resultant o‐phenanthroline‐modified chloropropyl‐silica hybrid monolith. The reproducibility of the column was also investigated by measuring relative standard deviations of the migration time for four neutral molecules. Relative standard deviations of run to run (n = 3), day to day (n = 3), and column to column (n = 3) were in the range of 0.4–0.7, 0.9–2.1, and 1.4–3.3%, respectively. Basic separations of various polar analytes including phenols and aromatic amines were successfully achieved.  相似文献

Analysis of N‐glycans released enzymatically from patients’ sera or other clinical samples may provide diagnostically and prognostically important information on human disease. Permethylation of these biomolecules simultaneously increases their hydrophobicity and substantially improves their detection parameters in the following mass spectrometric analyses. The overall procedure, from the glycan cleavage to the final mass spectrometric determinations, includes several steps involving extraction, derivatization, and purification. During these steps, certain polymeric contaminants that may have been coincidentally introduced could hamper the final measurements. To understand and counter these interferences and further fractionate or preconcentrate these glycans, we introduce here an effective microgradient chromatographic technique that employs a small reversed‐phase microcolumn connected to a gas‐tight microsyringe delivering a mobile‐phase gradient. After loading the glycan fraction onto the microcolumn, three elution steps are recommended: (1) remove polar contaminants; (2) recover permethylated glycans for either liquid chromatography with electrospray ionization mass spectrometry or matrix‐assisted laser desorption/ionization mass spectrometry; and (3) remove larger polymeric contaminants and regenerate the precolumn. We further demonstrate that the trapped second fraction can be beneficially preconcentrated and further separated to achieve matrix‐assisted laser desorption/ionization mass spectrometric detection of the derivatized N‐glycans up to 6300 Da. The enhanced detection capabilities for tetra‐antennary N‐glycans are of increasing interest in disease biomarker discovery.  相似文献
