A highly sensitive and stable amperometric tyrosinase biosensor has been developed based on multiwalled carbon nanotube (MWCNT) dispersed in mesoporous composite films of sol–gel‐derived titania and perfluorosulfonated ionomer (Nafion). Tyrosinase was immobilized within a thin film of MWCNT–titania–Nafion composite film coated on a glassy carbon electrode. Phenolic compounds were determined by the direct reduction of biocatalytically‐liberated quinone species at ?100 mV versus Ag/AgCl (3 M NaCl) without a mediator. The present tyrosinase biosensor showed good analytical performances in terms of response time, sensitivity, and stability compared to those obtained with other biosensors based on different sol–gel matrices. Due to the large pore size of the MWCNT–titania–Nafion composite, the present biosensor showed remarkably fast response time with less than 3 s. The present biosensor responds linearly to phenol from 1.0×10?7 M to 5.0×10?5 M with an excellent sensitivity of 417 mA/M and a detection limit of 9.5×10?8 M (S/N=3). The enzyme electrode retained 89% of its initial activity after 2 weeks of storage in 50 mM phosphate buffer at pH 7.0.  相似文献

Abstract

A novel approach to assemble an H2O2 amperometric biosensor was introduced. The biosensor was constructed by entrapping horseradish peroxidase (HRP) labeled nano‐scaled particulate gold (nano‐Au) (HRP‐nano‐Au electrostatic composite) in a new silica sol‐gel/alginate hybrid film using glassy carbon electrode as based electrode. This suggested strategy fully merged the merits of sol‐gel derived inorganic‐organic composite film and the nano‐Au intermediator. The silica sol‐gel/alginate hybrid material can improve the properties of conventional sol‐gel material and effectively prevent cracking of film. The entrapment of HRP in the form of HRP‐nano‐Au can not only factually prevent the leaking of enzyme out of the film but also provide a favorable microenvironment for HRP. With hydroquinone as an electron mediator, the proposed HRP electrode exhibited good catalytic activity for the reduction of H2O2. The parameters affecting both the qualities of sol‐gel/alginate hybrid film and the biosensor response were optimized. The biosensor exhibited high sensitivity of 0.40 Al mol?1 cm?2 for H2O2 over a wide linear range of concentration from 1.22×10?5 to 1.46×10?3 mol L?1, rapid response of <5 s and a detection limit of 0.61×10?6 mol L?1. The enzyme electrode has remarkable stability and retained 86% of its initial activity after 45 days of storage in 0.1 mol L?1 Tris‐HCl buffer solutions at pH 7.  相似文献

Mesoporous titania‐Nafion composite doped with carbon nanotube (CNT) has been used for the immobilization of tris(2,2′‐bipyridyl)ruthenium(II) (Ru(bpy)32+) and alcohol dehydrogenase on an electrode surface to yield a highly sensitive and stable electrogenerated chemiluminescence (ECL) ethanol biosensor. The presence of CNT in the composite film increases not only the sensitivity of the ECL biosensor but also the long‐term stability of the biosensor. The present biosensor responds linearly to ethanol in the wide concentration ranges from 1.0×10?5 M to 1.0×10?1 M with a detection limit of 5.0×10?6 M (S/N=3). The present ECL ethanol biosensor exhibited higher ECL response compared to that obtained with the ECL biosensor based on the corresponding composite without CNT. The present CNT‐based ECL biosensor showed good long‐term stability with 75% of its initial activity retained after 2 weeks of storage in 50 mM phosphate buffer at pH 7.0.  相似文献

A new type of amperometric hydrogen peroxide biosensor was constructed based on horseradish peroxidase (HRP) immobilized on Fe3O4/chitosan modified glassy carbon electrode. The effects of some experimental variables such as the concentration of supporting electrolyte, pH, enzyme loading, the concentration of the mediator of methylene blue (MB) and the applied potential were investigated. The linear range of the calibration curve for H2O2 was 2.0×10?4–1.2×10?2 M with a detection limit of 1.0×10?4 M (S/N=3). The response time was less than 12 s. The apparent Michaelis‐Menten constant Km was 21.4 mM and it illustrated the excellent biological activity of the fixed enzyme. In addition, the biosensor had long‐time stability and good reproducibility. And this method has been used to determine H2O2 concentration in the real sample.  相似文献

A cationic quinine‐imide dye brilliant cresyl blue (BCB) and horseradish peroxidase (HRP) were co‐immobilized within ormosil on multiwalled carbon nanotubes modified glassy carbon electrode for the fabrication of highly sensitive and selective hydrogen peroxide biosensor. The presence of epoxy group in ormosil as organic moiety improves the mechanical strength and transparency of the film and amino group provides biocompatible microenvironment for the immobilization of enzyme. The presence of MWCNTs improved the conductivity of the nanocomposite film. The surface characterization of MWCNT modified ormosil nanocomposite film was performed with scanning electron microscopy (SEM) and atomic force microscopy (AFM). Cyclic voltammetry and amperometry measurements were used to study and optimize the performance of the resulting peroxide biosensor. The apparent Michaelis–Menten constant was determined to be 1.5 mM. The proposed H2O2 biosensor exhibited wide linear range from 3×10?7 to 1×10?4 M, and low detection limit 1×10?7 M (S/N=3) with fast response time <5 s. The probable interferences in bio‐matrix were selected to test the selectivity and no significant response was observed in the biosensor. This biosensor possessed good analytical performance and long term storage stability.  相似文献

The third generation amperometric biosensor for the determination of hydrogen peroxide (H2O2) has been described. For the fabrication of biosensor, o-aminobenzoic acid (oABA) was first electropolymerized on the surface of platinum (Pt) electrode as an electrostatic repulsion layer to reject interferences. Horseradish peroxidase (HRP) absorbed by nano-scaled particulate gold (nano-Au) was immobilized on the electrode modified with polymerized o-aminobenzoic acid (poABA) with L-cysteine as a linker to prepare a biosensor for the detection of H2O2. Amperometric detection of H2O2 was realized at a potential of +20 mV versus SCE. The resulting biosensor exhibited fast response, excellent reproducibility and sensibility, expanded linear range and low interferences. Temperature and pH dependence and stability of the sensor were investigated. The optimal sensor gave a linear response in the range of 2.99×10^-6 to 3.55×10^-3 mol·L^-1 to H2O2 with a sensibility of 0.0177 A·L^-1·mol^-1 and a detection limit (S/N = 3) of 4.3×10^-7 mol·L^-1. The biosensor demonstrated a 95% response within less than 10 s.  相似文献

A tyrosinase (Tyr) biosensor was fabricated by immobilizing Tyr on the surface of multiwalled carbon nanotubes (MWNTs)‐chitosan (Chit) composite modified glassy carbon electrode (GCE). The MWNTs‐Chit composite film provided a biocompatible platform for the Tyr to retain the bioactivity and the MWNTs possessed excellent inherent conductivity to enhance the electron transfer rate. The Tyr/MWNTs‐Chit/GCE biosensor showed high sensitivity (412 mA/M), broad linear response (1.0×10?8–2.8×10?5 M), low detection limit (5.0 nM) and good stability (remained 93% after 10 days) for determination of phenol. The biosensor was further applied to rapid detection of the coliforms, represented by Escherichia coli (E. coli) in this work. The current responses were proportional to the quantity of coliforms in the range of 104–106 cfu/mL. After 5.0 h of incubation, E. coli could be detected as low as 10 cfu/mL.  相似文献

Amperometric lactate biosensor based on a carbon paste electrode modified with benzo[c]cinnoline and multiwalled carbon nanotubes is reported. Incorporation of benzo[c]cinnoline acting as a mediator and multiwalled carbon nanotubes providing a conduction pathway to accelerate electron transfer due to their excellent conductivity into carbon paste matrix resulted in a high performance lactate biosensor. The resulting biosensor exhibited a fast response, high selectivity, good repeatability and storage stability. Under the optimal conditions, the enzyme electrode showed the detection limit of 7.0×10?8 M with a linear range of 2.0×10?7 M–1.1×10?4 M. The usefulness of the biosensor was demonstrated in serum samples.  相似文献

The catalytic properties of peroxidase basic isoenzymes (PBI's) from Brassica napus towards trans‐resveratrol (t‐Res) oxidation were demonstrated by the first time by conventional UV‐visible spectroscopic measurements. The enzymatic reaction rate was studied under different experimental conditions and the kinetics parameters were determined. An amperometric biosensor based on Brassica napus PBI's to determine t‐Res is also proposed by the first time. The method employs a dialysis membrane covered, PBI's entrapped and ferrocene (Fc)‐embedded carbon paste electrode (PBI's‐Fc‐CP) and is based on the fact that the decreased amount of H2O2 produced by the action of PBI's is proportional to the oxidised amount of t‐Res in the solution. Comparative amperometric experiments showed that, in spite of PBI's activity was much lower than commercial horseradish peroxidase (HRP) activity, t‐Res was a much better substrate for PBI's biosensors than those biosensors constructed by using HRP. The PBI's‐Fc‐CP biosensors showed a very good stability during at least twenty days. The reproducibility and the repeatability were 4.5% and 8.3%, respectively, showing a good biosensor performance. The calibration curve was linear in the t‐Res concentration (ct‐Res) range from 1×10?6 to 2.5×10?5 M, with a sensibility of (2.31±0.05)×106 nA M?1. The lowest ct‐Res value measured experimentally for a signal to noise ratio of 3 : 1 was 0.83 μM.  相似文献

The electrochemical properties of hydrazine studied at the surface of a carbon paste electrode spiked with p‐bromanil (tetrabromo‐p‐benzoquinone) using cyclic voltammetry (CV), double potential‐step chronoamperometry and differential pulse voltammetry (DPV) in aqueous media. The results show this quinone derivative modified carbon paste electrode, can catalyze the hydrazine oxidation in an aqueous buffered solution. It has been found that under the optimum conditions (pH 10.00), the oxidation of hydrazine at the surface of this carbon paste modified electrode occurs at a potential of about 550 mV less positive than that of a bar carbon paste electrode. The electrocatalytic oxidation peak current of hydrazine showed a linear dependent on the hydrazine concentrations and linear analytical curves were obtained in the ranges of 6.00×10?5 M–8.00×10?3 M and 7.00×10?6 M–8.00×10?4 M of hydrazine concentration with CV and differential pulse voltammetry (DPV) methods, respectively. The detection limits (3σ) were determined as 3.6×10?5 M and 5.2×10?6 M by CV and DPV methods. This method was also used for the determination of hydrazine in the real sample (waste water of the Mazandaran wood and paper factory) by standard addition method.  相似文献

A carbon paste electrode spiked with 1‐[4‐ferrocenyl ethynyl) phenyl]‐1‐ethanone (4FEPE) was constructed by incorporation of 4FEPE in graphite powder‐paraffin oil matrix. It has been shown by direct current cyclic voltammetry and double step chronoamperometry that this electrode can catalyze the oxidation of tryptophan (Trp) in aqueous buffered solution. It has been found that under optimum condition (pH 7.00), the oxidation of Trp at the surface of such an electrode occurs at a potential about 200 mV less positive than at an unmodified carbon paste electrode. The kinetic parameters such as electron transfer coefficient, α and rate constant for the chemical reaction between Trp and redox sites in 4FEPE modified carbon paste electrode (4FEPEMCPE) were also determined using electrochemical approaches. The electrocatalytic oxidation peak current of Trp showed a linear dependent on the Trp concentrations and linear calibration curves were obtained in the ranges of 6.00×10?6 M–3.35×10?3 M and 8.50×10?7 M–6.34×10?5 M of Trp concentration with cyclic voltammetry (CV) and differential pulse voltammetry (DPV) methods, respectively. The detection limits (3σ) were determined as 1.80×10?6 M and 5.60×10?7 M by CV and DPV methods. This method was also examined as a selective, simple and precise new method for voltammetric determination of tryptophan in real sample.  相似文献

A label‐free DNA biosensor based on three‐dimensional reduced graphene oxide (3D‐rGO) and polyaniline (PANI) nanofibers modified glassy carbon electrode (GCE) was successfully developed for supersensitive detection of breast cancer BRCA1. The results demonstrated that 3D‐rGO and PANI nanofibers had synergic effects for reducing the charge transfer resistance (Rct), meaning a huge enhancement in electrochemical activity of 3D‐rGO‐PANI/GCE. Probe DNA could be immobilized on 3D‐rGO‐PANI/GCE for special and sensitive recognition of target DNA (1.0×10?15–1.0×10?7 M) with a theoretical LOD of 3.01×10?16 M (3S/m). Furthermore, this proposed nano‐biosensor could directly detect BRCA1 in real blood samples.  相似文献

The highly efficient H2O2 biosensor was fabricated on the basis of the complex films of hemoglobin (Hb), nano ZnO, chitosan (CHIT) dispersed solution and nano Au immobilized on glassy carbon electrode (GCE). Biocompatible ZnO‐CHIT composition provided a suitable microenvironment to keep Hb bioactivity (Michaelis‐Menten constant of 0.075 mmol L?1). The presence of nano Au in matrix could effectively enhance electron transfer between Hb and electrode. The electrochemical behaviors and effects of solution pH values were carefully examined in this paper. The (ZnO‐CHIT)‐Au‐Hb/GCE demonstrated excellently electrocatalytical ability for H2O2. This biosensor had a fast response to H2O2 less than 4 s and excellent linear relationships were obtained in the concentration range from1.94×10?7 to 1.73×10?3 mol L?1 with the detection limit of 9.7×10?8 mol L?1 (S/N=3) under the optimum conditions. Moreover, the stability and reproducibility of this biosensor were evaluated with satisfactory results.  相似文献

A chemically modified carbon paste electrode with 2,7‐bis(ferrocenyl ethyl)fluoren‐9‐one (2,7‐BFEFMCPE) was employed to study the electrocatalytic oxidation of ascorbic acid in aqueous solution using cyclic voltammetry, differential pulse voltammetry and chronoamperometry. The diffusion coefficient (D=1.89×10?5 cm2 s?1), and the kinetic parameter such as the electron transfer coefficient, α (=0.42) of ascorbic acid oxidation at the surface of 2,7‐BFEFMCPE was determined using electrochemical approaches. It has been found that under an optimum condition (pH 7.00), the oxidation of ascorbic acid at the surface of such an electrode occurs at a potential about 300 mV less positive than that of an unmodified carbon paste electrode. The catalytic oxidation peak currents show a linear dependence on the ascorbic acid concentration and linear analytical curves were obtained in the ranges of 8.0×10?5 M–2.0×10?3 M and 3.1×10?5 M–3.3×10?3 M of ascorbic acid with correlation coefficients of 0.9980 and 0.9976 in cyclic voltammetry and differential pulse voltammetry, respectively. The detection limits (2δ) were determined to be 2.9×10?5 M and 9.0×10?6 M with cyclic voltammetry and differential pulse voltammetry, respectively. This method was also examined for determination of ascorbic acid in pharmaceutical preparations.  相似文献

A novel enzyme immobilization technique based on thionine‐bovine serum albumin conjugate (Th‐BSA) and gold colloidal nanoparticles (nano‐Au) was developed. Thionine was covalently bound onto the BSA film with glutaraldehyde(GA) as cross‐linker to achieve Th‐BSA conjugate. The free amino groups of thionine were then used to attach nano‐Au for the immobilization of horseradish peroxidase (HRP). Such nano‐Au/Th‐BSA matrix shows a favorable microenvironment for retaining the native activity of the immobilized HRP and thionine immobilized in this way can effectively shuttle electrons between the electrode and the enzyme. The proposed biosensor displays excellent catalytic activity and rapid response for H2O2. The linear range for the determination of H2O2 is from 4.9×10?7 to 1.6×10?3 M with a detection limit of 2.1×10?7 M at 3σ and a Michaelies‐Menten constant K value of 0.023 mM.  相似文献

A 1‐[2‐hydroxynaphthylazo]‐6‐nitro‐2‐naphthol‐4‐sulfonate/ CuO nanoparticles modified carbon paste electrode (HNNSCCPE) was constructed and the electro‐oxidation of isoprenaline at the surface of the modified electrode was studied using cyclic voltammetry (CV), chronoamperometry (CHA), and square wave voltammetry (SWV). Under the optimized conditions, the square wave voltammetric peak current of isoprenaline increased linearly with isoprenaline concentrations in the range of 1.0×10?7 to 7.0×10?4 M and detection limit of 5.0×10?8 M was obtained for isoprenaline. The prepared modified electrode exhibits a very good resolution between the voltammetric peaks of isoprenaline, acetaminophen and N‐acetyl‐L‐cysteine which makes it suitable for the detection of isoprenaline in the presence of acetaminophen and N‐acetyl‐L‐cysteine in real samples.  相似文献

Composite solution of sol–gel‐derived titania and perfluorosulfonated ionomer (Nafion) was used as a solubilizing agent for multiwalled carbon nanotubes (CNT) as well as an encapsulation matrix for alcohol dehydrogenase (ADH) for the fabrication of a highly sensitive and stable amperometric ethanol biosensor. ADH was immobilized within a thin film of CNT–titania–Nafion composite film coated on a glassy carbon electrode. Because of the mesoporous nature of the CNT–titania–Nafion composite film, the present biosensor exhibited remarkably fast response time within 2 s. The presence of CNT in the composite film increases not only the sensitivity of the ethanol biosensor but also the long‐term stability of the biosensor. The present biosensor responds linearly to ethanol in the wide concentration ranges from 1.0×10?5 M to 3.0×10?3 M with the sensitivity of 51.6 mA M?1cm?2. The present biosensor showed good long‐term stability with 75% of its activity retained after 4 weeks of storage in 50 mM phosphate buffer at pH 7.0.  相似文献

The precursor film was first formed on the Au electrode surface based on the self‐assembly of L ‐cysteine and the adsorption of gold colloidal nanoparticles (nano‐Au). Layer‐by‐layer (LBL) assembly films of toluidine blue (TB) and nano‐Au were fabricated by alternately immersing the electrode with precursor film into the solution of toluidine blue and gold colloid. Cyclic voltammetry (CV) and quartz crystal microbalance (QCM) were adopted to monitor the regular growth of {TB/Au} bilayer films. The successful assembly of {TB/Au}n films brings a new strategy for electrochemical devices to construct layer‐by‐layer assembly films of nanomaterials and low molecular weight materials. In this article, {TB/Au}n films were used as model films to fabricate a mediated H2O2 biosensor based on horseradish peroxidase, which responded rapidly to H2O2 in the linear range from 1.5×10?7 mol/L to 8.6×10?3 mol/L with a detection limit of 7.0×10?8 mol/L. A new enzyme‐based amperometric biosensor for hydrogen peroxide was developed relying on the efficient immobilization of horseradish peroxidase (HRP) to a nano‐scaled particulate gold (nano‐Au) film modified glassy carbon electrode (GC). The nano‐Au film was obtained by a chitosan film which was first formed on the surface of GC. The high affinity of chitosan for nano‐Au associated with its amino groups resulted in the formation of nano‐Au film on the surface of GC. The film formed served as an intermediator to retain high efficient and stable immobilization of the enzyme. H2O2 was detected using hydroquinone as an electron mediator to transfer electrons between the electrode and HRP. The HRP immobilized on nano‐Au film maintained excellent electrocatalytical activity to the reduction of H2O2. The experimental parameters such as the operating potential of the working electrode, mediator concentration and pH of background electrolyte were optimized for best analytical performance of amperometry. The linear range of detection for H2O2 is from 6.1×10?6 to 1.8×10?3 mol L?1 with a detection limit of 6.1 μmol L?1 based on signal/noise=3. The proposed HRP enzyme sensor has the features of high sensitivity (0.25 Almol?1cm?2), fast response time (t90%≤10 s) and a long‐term stability (>1 month). As an extension, glucose oxidase (GOD) was chemically bound to HRP‐modified electrode. A GOD/HRP bienzyme‐modified electrode formed in this way can be applied to the determination of glucose with satisfactory performance.  相似文献

Three different recently synthesized aza‐thioether crowns containing a 1,10‐phenanthroline sub‐unit (L1–L3) and a corresponding acyclic ligand (L4) were studied to characterize their abilities as silver ion ionophores in PVC‐membrane electrodes. Novel conventional silver‐selective electrodes with internal reference solution (CONISE) and coated graphite‐solid contact electrodes (SCISE) were prepared based on one of the 15‐membered crowns containing two donating S atoms and two phenanthroline‐N atoms (L1). The electrodes reveal a Nernstian behavior over wide Ag+ ion concentration ranges (1.0×10?5?1.0×10?1 M for CONISE and 5.0×10?8?4.0×10?2 M for SCISE) and very low limits of detection (8.0×10?6 M for CONISE and 3.0×10?8 M for SCISE). The potentiometric response is independent from pH of the solution in the pH range 3.0–8.0. The electrodes manifest advantages of low resistance, very fast response and, most importantly, good selectivities relative to a wide variety of other cations. The electrodes can be used for at least 2 months (for CONISE) and 4 months for (SCISE) without any appreciable divergence in potentials. The electrodes were used as an indicator electrode in the potentiometric titration of Ag+ ion and in the determination of silver in photographic emulsions and in radiographic and photographic films.  相似文献
