Two members of a family of pyrrole–imidazole marine alkaloids, (+)‐dibromophakellin and the nonnatural congener (+)‐phakellin, were synthesized enantioselectively from 4‐hydroxy‐L ‐proline. The chiral aminal at C10 was constructed efficiently by means of an Overman‐type [3,3] sigmatropic rearrangement of an enamide (see scheme).
Economical atoms : 2‐Cyclopenten‐1‐ones, 5‐alkylidenefuran‐2(5 H)‐ones and indan‐1‐ones have been synthesized by atom‐economic reductive cyclocarbonylation of internal alkynes with carbon monoxide catalyzed by [{RhCl(CO)2}2]/CO(NH2)2 in the presence of water (see scheme).
A bicycle built for tubulin : The total synthesis of (+)‐chamaecypanone C has been achieved by using a tandem retro‐Diels–Alder/Diels–Alder cascade reaction (see scheme). Initial biological studies demonstrate that (+)‐chamaecypanone C is an inhibitor of tubulin assembly and binds at the colchicine site.
Panacea for aldol desymmetrizations : We describe an easy entry for the desymmetrization of 4‐substituted‐cyclohexanones catalyzed by proline, using as cocatalysts different hydrogen‐bonding donors (see scheme), which dramatically improves the catalytic efficiency of proline in desymmetrization reactions.
Going through the phases : The title reaction was found to proceed by an initial base‐mediated isomerization to allenyl esters and subsequent phase transfer catalyzed alkylation at the α position of the ester (see scheme).
