建立了花生中涕灭威及其代谢物涕灭威砜、涕灭威亚砜的高效液相色谱-线性离子阱三级质谱分析方法。样品经环己烷饱和的乙腈提取,凝胶渗透色谱净化后,用高效液相色谱-线性离子阱三级质谱法对样品中的目标物进行定性确证和定量分析。在Capcell PAK CR色谱柱上以含5 mmol/L NH4Ac-HAc的乙腈为流动相进行梯度洗脱分离。采用电喷雾离子源正离子模式进行三级选择离子监测,以涕灭威-d3作为3个目标物的内标物。通过比较基质匹配曲线和纯溶剂标准曲线计算回收率评估基质效应。方法的线性范围为10~500 μg/L,检出限为4~5 μg/kg。涕灭威、涕灭威砜、涕灭威亚砜在3个加标水平(10、20、40 μg/kg)的回收率为81.5%~115%,相对标准偏差为6.35%~15.1%。应用该方法对花生样品进行了测定,结果令人满意。  相似文献

建立了毛细管电泳-场强放大样品堆积测定染发剂中4,4′-二氨基二苯甲烷、苯胺、邻甲氧基苯胺、对氨基苯甲醚、3,4-二甲基苯胺、间氨基苯酚、1-萘胺7种苯胺类物质的分析方法。在优化的缓冲溶液体系(0.15 mol/L NaH2PO4,0.015 mol/L 三乙醇胺, pH 2.3)下7种分析物在6.5 min内实现基线分离。考察了样品中添加的磷酸浓度和乙腈浓度、水柱长度、电动进样时间与电压对场强放大富集效率及重现性的影响。最佳的富集条件为: 水柱注入3.45 kPa(0.5 psi)×6 s,样品中添加40%(v/v)乙腈和0.6×10~3mol/L磷酸,进样电压与进样时间为10 kV×10 s。线性范围为3~1000 μg/L(R2>0.996),检出限为0.26~2.75 μg/L,将已有方法的检测灵敏度提高了1~3个数量级。在2种市售黑色染发剂中均检测到间氨基苯酚,含量分别为7.32 mg/g和1.34 mg/g。平均加标回收率为74%~108%。该方法灵敏度高、快速、重现性好、成本低,可供多种样品基质中痕量苯胺类污染物及其他阳离子物质的测定借鉴使用。  相似文献

A simple, rapid, and sensitive non-aqueous capillary electrophoresis procedure with head-column field-amplified sample stacking concentration for the analysis of fangchinoline and tetrandrine is established. Optimum separation and stacking conditions were obtained when the sample was injected at 8 kV for 50 s after preliminary pressure injection of ethanol (16.9 kPa) for 0.6 s and separated with the buffer containing 50 mM ammonium acetate, 0.5% (v/v) acetic acid, and 50% (v/v) acetonitrile in methanol medium at 24 kV applied voltage. The analytes were detected by UV at 214 nm. The two bisbenzylisoquinoline alkaloids can be separated within 6 min and quantified with high sensitivity. The detection limits were 0.30 ng mL(-1) for fangchinoline and 0.34 ng mL(-1) for tetrandrine, which indicated that the sensitivities were at least 1000-fold enhanced over those reported in the literature as obtained by UV detection. The method was applied to the analysis of fangchinoline and tetrandrine in Radix Stephaniae tetrandrae and its medicinal preparations with good results.  相似文献

Microchip capillary electrophoresis in mixed hydro-organic solvent combined with laser-induced fluorescence detection was developed for the separation and detection of physcion and rhein in rhubarb. In contrast to the conventional capillary electrophoresis method, ammonium acetate-dimethyl sulfoxide was used as the basic buffer system in this method. The effects of background buffer, buffer apparent pH*, buffer concentration, water ratio, sample preparation method, and separation voltage on separation and detection were investigated. Optimized separation and detection conditions were obtained: the buffer consisted of 20 mmol/L of ammonium acetate in hydro-organic solvent composed dimethyl sulfoxide, formamide, and water mixed at 60/20/20 (v/v/v) ratio. The separation voltage was 1.9 kV. Under these conditions, the physcion, rhein, and other components of rhubarb can be completely separated within 150 s. Under the methodological verification, good linearity (R ≥ 0.9995) for physcion and rhein, and low limits of detection (0.085 μg·mL−1 and 0.077 μg·mL−1, respectively), satisfactory peak area precisions, migration time precisions (1.74%–3.09%), and accuracy (recovery rate 97.8% and 101.4%) were achieved. It is shown that the proposed method is simple, efficient, fast, sensitive, simple instrument, consumes few samples, has low operating cost, and is linear.  相似文献

建立了鸡肉中二硝托胺、尼卡巴嗪、地克珠利、妥曲珠利、莫能菌素及盐霉素6种抗球虫药物的超高效液相色谱-高分辨串联质谱多残留检测方法。经QuEChERS样品净化,首先使用含有1%(v/v)三氯乙酸的乙腈-水(3 : 7, v/v)溶液提取样品中的被测物,再加入氯化钠,使用50 mg/mL N-丙基乙二胺(PSA)+50 mg/mL中性氧化铝(Alumina-N)的混合分散固相萃取(dispersive solid phase extraction, DSPE)粉末净化提取,过0.22 μ m滤膜后以超高效液相色谱-高分辨串联质谱检测。选择Waters Acquity UPLC® BEH C8色谱柱(100 mm×2.1 mm, 1.7 μ m),以甲醇-5 mmol/L醋酸铵水溶液为流动相进行梯度洗脱。使用正、负离子同时扫描模式,基质外标法定量。研究表明,6种目标化合物的线性范围为:二硝托胺,1.0~30.0 μ g/L;尼卡巴嗪,0.2~6.0 μ g/L;地克珠利、妥曲珠利,2.0~60.0 μ g/L;莫能菌素、盐霉素,4.0~120.0 μ g/L。空白样品中添加低、中、高3个水平的混合标准溶液,回收率在67.7%~126.8%之间,相对标准偏差(RSD)≤10.4%。6种抗球虫药物的定量限分别为:二硝托胺,2.50 μ g/kg;尼卡巴嗪,0.50 μ g/kg;地克珠利、妥曲珠利,5.00 μ g/kg;莫能菌素、盐霉素,20.00 μ g/kg。该方法操作简便,灵敏度高,且能够满足日常检测要求。  相似文献

建立了基于月桂酰基苹果酸酯的胶束电动色谱-质谱法同时分离测定牛黄上清片中黄连碱、小檗碱、药根碱、黄柏碱以及川芎嗪5种有效成分含量的新方法。以7.5 mmol/L月桂酰基苹果酸酯-15 mmol/L氨水-50 mmol/L醋酸铵(含12.5%的乙腈,pH=7.0)为电泳介质,未涂层弹性石英毛细管(88 cm×50 μm)为分离通道,50%异丙醇水溶液(含3 mmol/L乙酸)为鞘液,在25 kV的运行电压下,各组分可以在18 min内得到基线分离。各组分的浓度与其峰面积呈现良好的线性关系,迁移时间和峰面积的相对标准偏差均小于5%,样品中5种生物碱成分的样品加标回收率在96.0%~105%之间。该法简便、快速、准确、重现性好,可用于牛黄上清片中这5种有效成分含量的同时测定。  相似文献

A nonaqueous capillary electrophoresis with laser-induced fluorescence detection method was developed for the quantification of ephedrine and pseudoephedrine after derivatization with 4-chloro-7-nitrobenzo-2-oxa-1, 3-diazol in nonaqueous media. The derivatization was made in off-line mode. By a series of optimizations, a derivatization buffer composed of 40 mm ammonium acetate and 20% acetonitrile and a running buffer composed of 80 mm ammonium acetate and 3% acetic acid were applied for the derivatization and separation of ephedrine and pseudoephedrine, respectively. Linear relationships for ephedrine and pseudoephedrine were obtained in the range 1.23-19.60 mg/L (correlation coefficients 0.9970 for ephedrine and 0.9994 for pseudoephedrine), and the detection limits for ephedrine and pseudoephedrine were 0.014 and 0.011 mg/L, respectively. The method was applied to the analysis of ephedrine and pseudoephedrine in four preparations with recoveries in the range 93.9-105.1%.  相似文献

This paper for the first time describes the development of micelle to solvent stacking (MSS) to nonaqueous capillary electrophoresis (NACE). In this proposed MSS-NACE, sodium dodecyl sulfate (SDS) micelles transport, release, and focus analytes from the sample solution to the running buffer using methanol as their solvent. After the focusing step, the focused analytes were separated via NACE. The focusing mechanism and influencing factors were discussed using berberine (BBR) and jatrorrhizine (JTZ) as model compounds. And the optimum condition was obtained as following: 50 mM ammonium acetate, 6% (v/v) acetic acid and 10 mM SDS in redistilled water as sample matrix, 50 mM ammonium acetate and 6% (v/v) acetic acid in pure methanol as the running buffer, -20 kV focusing voltage with 30 min focusing time. Under these conditions, this method afforded limits of detection (S/N=3) of 0.002 μg/mL and 0.003 μg/mL for BBR and JTZ, respectively. In contrast to conventional NACE, the concentration sensitivity was improved 128-153-fold.  相似文献

An ion chromatographic method with conductivity detection for the simultaneous determination of ephedrine, pseudoephedrine and norephedrine was developed. A mixture of 2.0 mmol/L HNO3 and 2% (v/v) acetonitrile was used as eluent. The three ephedrine-like compounds were separated and determined within 20 min. The linear ranges were 0.08-50 microg/mL for ephedrine, 0.08-40 microg/mL for pseudoephedrine and 0.06-40 microg/mL for norephedrine. The detection limits were 0.03 microg/mL for ephedrine and pseudoephedrine, and 0.02 microg/mL for norephedrine. The method has been applied successfully to the determination of these sympathomimetics in pharmaceutical preparations and in Ephedra herbs.  相似文献

建立了复方化学消毒剂中常用有效成分醋酸洗必泰和苯扎氯铵(C12-BAC、C14-BAC及C16-BAC)同时分离测定的毛细管电泳(CE)方法。以37 cm×50 μm未涂层熔融石英毛细管为分离柱,以150 mmol/L磷酸二氢钠-62.5 mmol/L磷酸(pH 2.5)缓冲液(含体积分数为40%的乙腈)为分离缓冲溶液,50 mmol/L醋酸-乙腈(体积比为1:1)为样品介质,检测波长为214 nm。方法的日内及日间精密度分别小于3.0%及3.7%。醋酸洗必泰、C12-BAC、C14-BAC及C16-BAC的检出限(信噪比为3)分别为0.3、0.5、0.5、0.5 mg/L,定量限(信噪比为10)分别为1.0、1.5、1.5和1.5 mg/L,在1.0~400、1.5~200、1.5~200和1.5~200 mg/L范围内,4种有效成分的校正峰面积与相应质量浓度均具有良好的线性关系,相关系数分别为0.9995、0.9998、0.9997和0.9998。加标回收率为93.83%~104.97%。将该法用于实际样品分析,并与液相色谱的分析结果进行比对,获得满意结果。  相似文献

采用在线柱浓缩-超高效液相色谱联用技术测定水体中痕量甲萘威和呋喃丹。水样过滤后直接进样,采用固相萃取小柱富集待测物,梯度洗脱后,利用阀切换技术将待测物反冲至分析柱Acclaim RSLC C18(100 mm×2.1 mm, 2.2 μm)上进行色谱分离,以10 mmol/L醋酸铵缓冲溶液(pH 5.0,用醋酸调节)和乙腈分别为流动相A和B,梯度洗脱,泵流速为0.8 mL/min,检测波长为280 nm,二极管阵列检测器检测。甲萘威和呋喃丹在1.0~100 μg/L范围内线性良好(相关系数r2 > 0.9999),检出限(S/N=3)分别为0.5和0.25 μg/L,加标回收率为76.0%~120.0%。用所建立的方法测定了水中痕量的甲萘威与呋喃丹的含量,结果令人满意。  相似文献

采用液相色谱-串联质谱法(LC-MS/MS)同时测定水产品中的孔雀石绿、结晶紫以及它们的隐色代谢物残留。匀质后的水产品样品用乙腈和乙酸铵缓冲液提取。合并提取液,用二氯甲烷反提取,经中性氧化铝柱和PRS柱固相萃取净化。采用ZORBAX SB-C18色谱柱,并以0.5 mmol/L乙酸铵-乙腈(体积比为10∶90)混合溶液为流动相,无需使用氧化铅柱在线氧化,色谱分离后直接进入串联质谱检测器检测。采用电喷雾离子源,正离子多反应监测(MRM)模式检测。方法的检测限(S/N=3)可达0.5 ng/g,平均加标回收率为77.6%~98.1%,相对标准偏差均小于8.2%。大量实际水产品样品的检测结果表明,此方法适合于对水产品中孔雀石绿、结晶紫以及它们的隐色代谢物的残留检测。  相似文献

以在线扫集-胶束毛细管电动色谱法(Sweeping-MEKC)测定了复方氨酚烷胺胶囊中的马来酸氯苯那敏、咖啡因和对乙酰氨基酚3种有效成分。考察了缓冲溶液pH值、SDS浓度、分离电压及进样时间等对分离效果的影响。优化条件:以未涂层熔融石英毛细管(55 cm×50μm,有效柱长35 cm)为分离柱;环境温度25℃;80 mmol/L十二烷基磺酸钠+20 mmol/L NaH2PO4(pH 2.2)+15%乙腈为缓冲体系,分离电压-20kV,进样时间60 s(H=20.0 cm),测量波长210 nm。在该条件下氯苯那敏、咖啡因和对乙酰氨基酚在25min内出峰,峰面积RSD均小于4%;线性范围分别为2.45~39.17、1.61~25.76、1.58~25.28 mg/L;检出限(S/N=3)分别达139、34、24μg/L,回收率分别为96%~101%、98%~102%、96%~102%。  相似文献

A novel method based on separation by nonaqueous capillary electrophoresis (NACE) combined with laser-induced fluorescence (LIF) detection was developed and compared with classic aqueous modes of electrophoresis in terms of resolution of solutes of interest and sensitivity of the fluorescence detection. Catecholamines derivatized with 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBD-Cl) were chosen as test analytes for their subtle fluorescence properties. In aqueous systems, capillary zone electrophoresis (CZE) was not suitable for the analysis of test analytes due to complete fluorescence quenching of NBD-labeled catecholamines in neat aqueous buffer. The addition of micelles or microemulsion droplets into aqueous running buffer can dramatically improve the fluorescence response, and the enhancement seems to be comparable for micellar electrokinetic chromatography (MEKC) and microemulsion electrokinetic chromatography (MEEKC). As another alternative, NACE separation was advantageous when performing the analysis under the optimum separation condition of 20 mM sodium tetraborate, 20 mM sodium dodecyl sulfate (SDS), 0.1% (v/v) glacial acetic acid, 20% (v/v) acetonitrile (ACN) in methanol medium after derivatization in ACN/dimethyl sulfoxide (DMSO) (3:2, v/v) mixed aprotic solvents containing 20 mM ammonium acetate. Compared with derivatization and separation in aqueous media, NACE-LIF procedure was proved to be superior, providing high sensitivity and short migration time. Under respective optimum conditions, the NACE procedure offered the best fluorescence response with 5-24 folds enhancement for catecholamines compared to aqueous procedures. In addition, the mechanisms of derivatization and separation in nonaqueous media were elucidated in detail.  相似文献

建立了畜禽产品中维吉尼亚霉素M1和S1药物残留检测的液相色谱-串联质谱分析方法。样品以甲醇-乙腈溶液(1:1, v/v)提取,上清液经0.01 mol/L磷酸二氢铵溶液稀释后,Oasis HLB固相萃取小柱净化,Luna C18色谱柱分离,以乙腈和含0.1%(体积分数)甲酸的5 mmol/L乙酸铵水溶液作为流动相进行梯度洗脱,电喷雾正离子模式电离(ESI+),多反应监测(MRM)模式检测,外标法定量。该方法对两物质线性范围均为0.15~10.0 μg/L,相关系数r2均大于0.999;定量下限均为0.25 μg/kg。在不同基质中,0.25、0.50、2.5 μg/kg3个添加水平的平均回收率范围为71.2%~98.4%,精密度范围为3.6%~15.4%。该方法具有快速简便、灵敏度高、准确性强等特点,适用于畜禽产品中维吉尼亚霉素的检测。  相似文献

In this work, a fast analytical method based on hydrophilic interaction liquid chromatographic-Ultraviolet detection (HILIC-UV) using a short narrow bore cyano-bonded silica column packed with fully porous sub-2?µm particles has been developed for simultaneous determination of eight pharmaceuticals in wastewater. The method involved pre-concentration and clean-up by solid phase extraction using Oasis HLB extraction cartridges. The analytes were separated using a mobile phase consisted of acetonitrile and 5?mM ammonium acetate buffer (95:5?v/v) with a flow rate of 0.6?mL/min. The chromatographic separation was optimized in order to achieve short analysis time and good resolution for all analytes in a single run. Each analyte was detected at its maximum wavelength for higher sensitivity. All analytes could be separated in 5.7?min with resolution ≥2.7. The optimized method was validated based on linearity, precision, detection and quantification limits, selectivity and accuracy. The detection limits of the studied pharmaceuticals ranged from 0.6 to 3?µg/L, while limits of quantification were in the range from 2 to 10?µg/L with UV detection. The developed method is fast, reliable, cost-effective and could be used for the analysis of the studied analytes in other matrices such as food, pharmaceutical preparations and biological fluids.  相似文献

A simple, rapid and sensitive CE-ESI-MS method for the simultaneous analysis of seven stimulants and narcotics (amphetamine, ephedrine, methadone, pethidine, tetracaine, codeine and heroin) was developed. The CE-ESI-MS experimental conditions were optimized as follows: 20?mmol/L ammonium acetate with pH 9.0 as running buffer, the separation voltage of 22?kV and the sheath liquid of isopropanol/water (1:1 v/v) containing 7.5?mmol/L acetic acid with 3.0?μL/min flow rate. Under the optimized conditions, the stimulants and narcotics were well separated within 4.6?min using a 70-cm length fused-silica capillary (50?μm id). The detection limits (S/N=3) of the CE-ESI-MS analysis were in the range of 0.40-1.0?ng/mL. Method repeatability of intra-day and inter-day was satisfactory. The recoveries obtained from the analysis of spiked urine samples were between 84.1 and 108%. The developed method was successfully applied for the simultaneous analysis of methadone, pethidine and codeine and their in vitro metabolites.  相似文献

基于酸性介质中甲酸对高锰酸钾-苯二酚氧化发光反应的增敏作用建立了高效液相色谱-化学发光柱后检测苯二酚的新方法。优化了高锰酸钾-苯二酚氧化发光反应及高效液相色谱分离苯二酚的条件,用甲醇-0.1 mmol/L β-环糊精水溶液(体积比为30∶70) 作为流动相可实现对水中3种苯二酚异构体的分离,且能与高锰酸钾-苯二酚氧化化学发光反应条件很好地偶合。对所测定的苯二酚异构体,方法的线性范围达两个数量级;以信噪比为3测得邻、间、对苯二酚的检出限(n=3)分别为:5.2,4.7,3.2 μg/L,对质量浓度均为0.10 mg/L的3种苯二酚混合溶液连续测定11次,邻、间、对苯二酚的相对标准偏差分别为2.8%,3.4%,6.5%。将该方法与固相萃取技术相结合,对河水中的痕量苯二酚进行了测定,加标回收率为92.1%~95.4%。  相似文献

Eight textile dye compounds including five cationic dyes, namely, basic blue 41, basic blue 9, basic green 4, basic violet 16 and basic violet 3, and three anionic dyes, acid green 25, acid red 1 and acid blue 324, were separated and detected by non-aqueous capillary electrophoresis (NACE) with electrochemical detection. Simultaneous separations of acid and basic dyes were performed using an acetonitrile-based buffer. Particular attention was paid to the determination of basic textile dyes. The optimized electrophoresis buffer for the separation of basic dyes was a solvent mixture of acetonitrile/methanol (75:25, v/v) containing 1 M acetic acid and 10 mM sodium acetate. The limits of detection for the basic dyes were in the range of 0.1–0.7 μg mL−1. An appropriate solid-phase extraction procedure was developed for the pre-treatment of aqueous samples with different matrices. This analytical approach was successfully applied to various water samples including river and lake water which were spiked with textile dyes.  相似文献

建立了毛细管电泳-电喷雾电离质谱联用法同时测定清肺抑火片中苦参碱、药根碱、氧化苦参碱、栀子苷4种药效成分含量的分析方法.采用未涂层石英毛细管,以35 mmol/L乙酸铵(含20%乙腈,pH=6.5)为缓冲溶液、60%异丙醇(含3.3 mmol/L乙酸)为鞘液,分离电压为28 kV时,各组分在11 min内达到基线分离.苦参碱、药根碱、氧化苦参碱、栀子苷的线性范围分别为0.030~150 mg/L、0.060~20 mg/L、0.060~80 mg/L、0.60~300mg/L,检出限分别为0.010、0.020、0.020、0.20mg/L.样品的加标回收率在91.0%~107%之间,相对标准偏差均小于4.9%.方法简便、快速、灵敏度高,已成功用于清肺抑火片中的苦参碱、药根碱、氧化苦参碱、栀子苷含量的同时测定.  相似文献
