Despite the extensive use of porphyrins in photodynamic therapy (PDT), tetraplatinated porphyrins have so far not been studied for their anticancer properties. Herein, we report the synthesis of such novel platinum–porphyrin conjugates as well as their photophysical characterization and in vitro light‐induced anticancer properties. These conjugates showed only minor cytotoxicity in the dark, but IC50 values down to 19 nM upon irradiation with light at 420 nm.These values correspond to an excellent phototoxic index (PI=IC50 in the dark/IC50 in light), which reached 5000 in a cisplatin‐resistant cell line. After incubation with HeLa cells, nuclear Pt concentrations were 30 times higher than with cisplatin. All of these favorable characteristics imply that tetraplatinated porphyrin complexes are worthy of exploration as novel PDT anticancer agents in vivo.  相似文献

A key step in cytochrome P450 catalysis includes the spin‐state crossing from low spin to high spin upon substrate binding and subsequent reduction of the heme. Clearly, a weak perturbation in P450 enzymes triggers a spin‐state crossing. However, the origin of the process whereby enzymes reorganize their active site through external perturbations, such as hydrogen bonding, is still poorly understood. We have thus studied the impact of hydrogen‐bonding interactions on the electronic structure of a five‐coordinate iron(III) octaethyltetraarylporphyrin chloride. The spin state of the metal was found to switch reversibly between high (S=5/2) and intermediate spin (S=3/2) with hydrogen bonding. Our study highlights the possible effects and importance of hydrogen‐bonding interactions in heme proteins. This is the first example of a synthetic iron(III) complex that can reversibly change its spin state between a high and an intermediate state through weak external perturbations.  相似文献

A feasible two‐step synthesis and characterization of a full series of hexaarylbenzene (HAB) substituted porphyrins and tetrabenzoporphyrins is presented. Key steps represent the microwave‐assisted porphyrin condensation and the statistical Diels–Alder reaction to the desired HAB‐porphyrins. Regarding their applications, they proved to be easily accessible and effective high molecular mass calibrants for (MA)LDI mass spectrometry. The free‐base and zinc(II) porphyrin systems, as well as the respective tetrabenzoporphyrins, demonstrate in solid state experiments strong red‐ and near‐infrared‐light emission and are potentially interesting for the application in “truly organic” light‐emitting devices. Lastly, they represent facile precursors to large polycyclic aromatic hydrocarbon (PAH) substituted porphyrins. We prepared the first tetra‐hexa‐peri‐hexabenzocoronene substituted porphyrin, which represents the largest prepared PAH‐porphyrin conjugate to date.  相似文献

The synthesis, structure, and properties of bischloro, μ‐oxo, and a family of μ‐hydroxo complexes (with BF4?, SbF6?, and PF6? counteranions) of diethylpyrrole‐bridged diiron(III) bisporphyrins are reported. Spectroscopic characterization has revealed that the iron centers of the bischloro and μ‐oxo complexes are in the high‐spin state (S=5/2). However, the two iron centers in the diiron(III) μ‐hydroxo complexes are equivalent with high spin (S=5/2) in the solid state and an intermediate‐spin state (S=3/2) in solution. The molecules have been compared with previously known diiron(III) μ‐hydroxo complexes of ethane‐bridged bisporphyrin, in which two different spin states of iron were stabilized under the influence of counteranions. The dimanganese(III) analogues were also synthesized and spectroscopically characterized. A comparison of the X‐ray structural parameters between diethylpyrrole and ethane‐bridged μ‐hydroxo bisporphyrins suggest an increased separation, and hence, less interactions between the two heme units of the former. As a result, unlike the ethane‐bridged μ‐hydroxo complex, both iron centers become equivalent in the diethylpyrrole‐bridged complex and their spin state remains unresponsive to the change in counteranion. The iron(III) centers of the diethylpyrrole‐bridged diiron(III) μ‐oxo bisporphyrin undergo very strong antiferromagnetic interactions (J=?137.7 cm?1), although the coupling constant is reduced to only a weak value in the μ‐hydroxo complexes (J=?42.2, ?44.1, and ?42.4 cm?1 for the BF4, SbF6, and PF6 complexes, respectively).  相似文献

The J aggregates of 4‐sulfonatophenyl meso‐substituted porphyrins are non‐covalent polymers obtained by self‐assembly that form nanoparticles of different morphologies. In the case of high aspect‐ratio nanoparticles (bilayered ribbons and monolayered nanotubes), shear hydrodynamic forces may modify their shape and size, as observed by peak force microscopy, transmission electron microscopy of frozen solutions, small‐angle X‐ray scattering measurements in a disk‐plate rotational cell, and cone–plate rotational viscometry. These nanoparticles either show elastic or plastic behaviour: there is plasticity in the ribbons obtained upon nanotube collapse on solid/air interfaces and in viscous concentrated nanotube solutions, whereas elasticity occurs in the case of dilute nanotube solutions. Sonication and strong shear hydrodynamic forces lead to the breaking of the monolayered nanotubes into small particles, which then associate into large colloidal particles.  相似文献

Molecular structures for three oxidation forms (anion, radical, and cation) of terbium(III) bis(porphyrinato) double‐decker complexes have been systematically studied. We found that the redox state controls the azimuthal rotation angle (φ) between the two porphyrin macrocycles. For [TbIII(tpp)2]n (tpp: tetraphenylporphyrinato, n=?1, 0, and +1), φ decreases at each stage of the oxidation process. The decrease in φ is due to the higher steric repulsion between the phenyl rings on the porphyrin macrocycle and the β hydrogen atoms on the other porphyrin macrocycle, which results from the shorter interfacial distance between the two porphyrin macrocycles. Conversely, φ=45° for both [TbIII(oep)2]?1 and [TbIII(oep)2]0 (oep: octaethylporphyrinato), but φ=36° for [TbIII(oep)2]+1. Theoretical calculations suggest that the smaller azimuthal rotation angle of the cation form is due to the electronic interaction in the doubly oxidized ligand system.  相似文献

The physicochemical characterization, that is, ground and excited state, of a new series of dendronized porphyrin/fullerene electron donor–acceptor conjugates in nonaqueous and aqueous environments is reported. In contrast to previous work, we detail the charge‐separation and charge‐recombination dynamics in zinc and copper metalloporphyrins as a function of first‐ and second‐generation dendrons as well as a function of ortho, meta, and para substitution. Both have an appreciable impact on the microenvironments of the redox‐active constituents, namely the porphyrins and the fullerenes. As a matter of fact, the resulting charge‐transfer dynamics were considerably impacted by the interplay between the associated forces that reach from dendron‐induced shielding to dipole–charge interactions.  相似文献

The recognition of 4‐alkylpyridines by water‐soluble poly(ethylene oxide)–zinc porphyrin conjugates was studied with a focus on the thermodynamic parameters of binding. Microcalorimetric studies indicated that binding of the alkyl group of the guest in water is driven by the entropic term (δΔH0H0(4‐pentylpyridine)? ΔH0(4‐methylpyridine)=+1.7 kJ mol?1, δTΔS0=TΔS0(4‐pentylpyridine)? TΔS0(4‐methylpyridine)=+11.8 kJ mol?1 at 298 K), thus showing the significance of water reorganization during host–guest interaction. The enthalpy–entropy compensation temperature of binding of 4‐alkylpyridines was as low as 38 K; only below this temperature could the enthalpic term be a driving force. The binding affinity was modulated by the addition of cations and by varying the degree of polymerization of poly(ethylene oxide), which suggests that guest binding is coupled with polymer conformation.  相似文献

New covalently C60‐connected zinc phthalocyanine (ZnPc) derivatives have been synthesized by utilizing successive cycloaddition reactions of C60 with a ZnPc derivative containing a pyridazine moiety employing Komatsu’s method in reaction of C60 with phthalazine. The UV/Vis absorption spectrum of the fused conjugate ( 5 ) shows red shifts from the corresponding absorption of ZnPc derivative ( 8 ), indicating interactions between the ZnPc and C60 moieties. The DFT calculations under non‐polar medium predict that the HOMO and LUMO of 5 localize on the ZnPc moiety, whereas LUMO+1 localizes on the C60 moiety, which reasonably explain the magnetic circular dichroism (MCD) and absorption spectra in toluene. Electrochemical redox potentials of 5 in polar solvents indicate the first‐oxidation potential arises from the ZnPc moiety, whereas the first reduction potential is associated with the C60 moiety, suggesting the LUMO localizes on the C60 moiety in polar solvent. This reversal of the LUMO is supported by the ZnPc‐fluorescence quenching with a nearby C60 moiety in benzonitrile, which leads to the charge‐separation via the excited singlet state of the ZnPc moiety. In toluene on the other hand, such a ZnPc‐fluorescence quenching owing to the photoinduced charge separation is not observed as predicted by the DFT‐calculated LUMO on the ZnPc moiety.  相似文献

The two different coordination spheres afforded by Pacman architectures offer cooperativity derived from two different metal centers. A modular strategy is developed to produce a hetero‐Pacman scaffold featuring a porphyrin and terpyridine for metal‐ion binding. A double Suzuki reaction was employed to first attach a terpyridine moiety to a xanthene backbone and then attach a porphyrin. The new hetero‐Pacman scaffold has been characterized and all building blocks have been isolated and structurally characterized. The principle objective to incorporate different metal centers was confirmed by isolating a trinuclear complex comprising two porphyrinic units and a bis(terpyridine)–iron unit. The compounds described herein expand the Pacman scaffold concept by allowing for the incorporation of a terpyridine–metal complex proximate to a porphyrin‐cofactor active site for small‐molecule activation.  相似文献
