S‐((Phenylsulfonyl)difluoromethyl)thiophenium salts were designed and prepared by a triflic acid catalyzed intramolecular cyclization of ortho‐ethynyl aryldifluoromethyl sulfanes. The thiophenium salts were found to be efficient as electrophilic difluoromehtylating reagents for introduction of a CF2H group to sp3‐hybridized carbon nucleophiles such as of β‐ketoesters and dicyanoalkylidenes. The (phenylsulfonyl)difluoromethyl group can be readily transformed into CF2H under mild reaction conditions. Enantioselective electrophilic difluoromethylation was also achieved in the presence of bis(cinchona) alkaloids.  相似文献

The new electrophilic trifluoromethylating 1‐(trifluoromethyl)‐benziodoxole reagents A and B (Scheme 1) have been used to selectively attach CF3 groups to the S‐atom of cysteine side chains of α‐ and β‐peptides (up to 13‐residues‐long; products 7 – 14 ). Other functional groups in the substrates (amino, amido, carbamate, carboxylate, hydroxy, phenyl) are not attacked by these soft reagents. Depending on the conditions, the indole ring of a Trp residue may also be trifluoromethylated (in the 2‐position). The products are purified by chromatography, and identified by 1H‐, 13C‐, and 19F‐NMR spectroscopy, by CD spectroscopy, and by high‐resolution mass spectrometry. The CF3 groups, thus introduced, may be replaced by H (Na/NH3), an overall Cys/Ala conversion. The importance of trifluoromethylations in medicinal chemistry and possible applications of the method (spin‐labelling, imaging, PET) are discussed.  相似文献

The first highly efficient and stereoselective difluoromethylation of structurally diverse N‐tert‐butylsulfinyl ketimines has been achieved with an in situ generated PhSO2CF2? anion, which provides a powerful synthetic method for the preparation of a variety of structurally diverse homochiral α‐difluoromethyl tertiary carbinamines, including α‐difluoromethyl allylic amines and α‐difluoromethyl propargylamines. The stereocontrol mode of the present diastereoselective difluoromethylation of ketimines was found to be different from that of other known fluoroalkylations of Ntert‐butylsulfinyl aldimines, which suggests that a cyclic six‐membered transition state may be involved in the reaction.  相似文献

The reactions of Zn(CF3)Br · 2 CH3CN, Cd(CF3)2 · 2 CH3CN or Bi(CF3)3/AlCl3 with tertiary amines lead to the formation of quaternary ammonium salts of the general formula [R3NCF2H]X. The reaction of 4‐N,N‐dimethylaminopyridine with Zn(CF3)Br · 2 CH3CN yields (N‐difluoromethyl)‐4‐N,N‐dimethylaminopyridinium bromide. Bi(CF3)3/AlCl3 reacts with 1,4‐diazabicyclo[2.2.2]octane to form a mixture of mono‐ and bis(difluoromethylammonium) salts.  相似文献

We have developed a novel and simple protocol for the direct incorporation of a difluoromethyl (CF2H) group into alkenes by visible‐light‐driven photoredox catalysis. The use of fac‐[Ir(ppy)3] (ppy=2‐pyridylphenyl) photocatalyst and shelf‐stable Hu's reagent, N‐tosyl‐S‐difluoromethyl‐S‐phenylsulfoximine, as a CF2H source is the key to success. The well‐designed photoredox system achieves synthesis of not only β‐CF2H‐substituted alcohols but also ethers and an ester from alkenes through solvolytic processes. The present method allows a single‐step and regioselective formation of C(sp3)–CF2H and C(sp3)?O bonds from C=C moiety in alkenes, such as hydroxydifluoromethylation, regardless of terminal or internal alkenes. Moreover, this methodology tolerates a variety of functional groups.  相似文献

The salts 3‐[(2,2,3,3‐tetrafluoropropoxy)methyl]pyridinium saccharinate, C9H10F4NO+·C7H4NO3S, (1), and 3‐[(2,2,3,3,3‐pentafluoropropoxy)methyl]pyridinium saccharinate, C9H9F5NO+·C7H4NO3S, (2), i.e. saccharinate (or 1,1‐dioxo‐1λ6,2‐benzothiazol‐3‐olate) salts of pyridinium with –CH2OCH2CF2CF2H and –CH2OCH2CF2CF3meta substituents, respectively, were investigated crystallographically in order to compare their fluorine‐related weak interactions in the solid state. Both salts demonstrate a stable synthon formed by the pyridinium cation and the saccharinate anion, in which a seven‐membered ring reveals a double hydrogen‐bonding pattern. The twist between the pyridinium plane and the saccharinate plane in (2) is 21.26 (8)° and that in (1) is 8.03 (6)°. Both salts also show stacks of alternating cation–anion π‐interactions. The layer distances, calculated from the centroid of the saccharinate plane to the neighbouring pyridinium planes, above and below, are 3.406 (2) and 3.517 (2) Å in (1), and 3.409 (3) and 3.458 (3) Å in (2).  相似文献

Enantiomerically pure triflones R1CH(R2)SO2CF3 have been synthesized starting from the corresponding chiral alcohols via thiols and trifluoromethylsulfanes. Key steps of the syntheses of the sulfanes are the photochemical trifluoromethylation of the thiols with CF3Hal (Hal=halide) or substitution of alkoxyphosphinediamines with CF3SSCF3. The deprotonation of RCH(Me)SO2CF3 (R=CH2Ph, iHex) with nBuLi with the formation of salts [RC(Me)? SO2CF3]Li and their electrophilic capture both occurred with high enantioselectivities. Displacement of the SO2CF3 group of (S)‐MeOCH2C(Me)(CH2Ph)SO2CF3 (95 % ee) by an ethyl group through the reaction with AlEt3 gave alkane MeOCH2C(Me)(CH2Ph)Et of 96 % ee. Racemization of salts [R1C(R2)SO2CF3]Li follows first‐order kinetics and is mainly an enthalpic process with small negative activation entropy as revealed by polarimetry and dynamic NMR (DNMR) spectroscopy. This is in accordance with a Cα? S bond rotation as the rate‐determining step. Lithium α‐(S)‐trifluoromethyl‐ and α‐(S)‐nonafluorobutylsulfonyl carbanion salts have a much higher racemization barrier than the corresponding α‐(S)‐tert‐butylsulfonyl carbanion salts. Whereas [PhCH2C(Me)SO2tBu]Li/DMPU (DMPU = dimethylpropylurea) has a half‐life of racemization at ?105 °C of 2.4 h, that of [PhCH2C(Me)SO2CF3]Li at ?78 °C is 30 d. DNMR spectroscopy of amides (PhCH2)2NSO2CF3 and (PhCH2)N(Ph)SO2CF3 gave N? S rotational barriers that seem to be distinctly higher than those of nonfluorinated sulfonamides. NMR spectroscopy of [PhCH2C(Ph)SO2R]M (M=Li, K, NBu4; R=CF3, tBu) shows for both salts a confinement of the negative charge mainly to the Cα atom and a significant benzylic stabilization that is weaker in the trifluoromethylsulfonyl carbanion. According to crystal structure analyses, the carbanions of salts {[PhCH2C(Ph)SO2CF3]Li? L }2 ( L =2 THF, tetramethylethylenediamine (TMEDA)) and [PhCH2C(Ph)SO2CF3]NBu4 have the typical chiral Cα? S conformation of α‐sulfonyl carbanions, planar Cα atoms, and short Cα? S bonds. Ab initio calculations of [MeC(Ph)SO2tBu]? and [MeC(Ph)SO2CF3]? showed for the fluorinated carbanion stronger nC→σ* and nO→σ* interactions and a weaker benzylic stabilization. According to natural bond orbital (NBO) calculations of [R1C(R2)SO2R]? (R=tBu, CF3) the nC→σ*S? R interaction is much stronger for R=CF3. Ab initio calculations gave for [MeC(Ph)SO2tBu]Li ? 2 Me2O an O,Li,Cα contact ion pair (CIP) and for [MeC(Ph)SO2CF3]Li ? 2 Me2O an O,Li,O CIP. According to cryoscopy, [PhCH2C(Ph)SO2CF3]Li, [iHexC(Me)SO2CF3]Li, and [PhCH2C(Ph)SO2CF3]NBu4 predominantly form monomers in tetrahydrofuran (THF) at ?108 °C. The NMR spectroscopic data of salts [R1(R2)SO2R3]Li (R3=tBu, CF3) indicate that the dominating monomeric CIPs are devoid of Cα? Li bonds.  相似文献

Heteropentapeptides containing the α‐ethylated α,α‐disubstituted amino acid (S)‐butylethylglycine and four dimethylglycine residues, i.e., CF3CO‐[(S)‐Beg]‐(Aib)4‐OEt ( 4 ) and CF3CO‐(Aib)2‐[(S)‐Beg]‐(Aib)2‐OEt ( 7 ), were synthesized by conventional solution methods. In the solid state, the preferred conformation of 4 was shown to be both a right‐handed (P) and a left‐handed (M) 310‐helical structure, and that of 7 was a right‐handed (P) 310‐helical structure. IR, CD, and 1H‐NMR spectra revealed that the dominant conformation of both 4 and 7 in solution was the 310‐helical structure. These conformations were also supported by molecular‐mechanics calculations.  相似文献

A series of novel α‐fluoroalkyl ammonium salts was obtained from the corresponding cyano compounds or nitriles by reaction with anhydrous HF. Room‐temperature stable trifluoromethyl ammonium salts were obtained in quantitative yield in a one‐step reaction at ambient temperature from the commercially available starting materials BrCN or ClCN. The novel cations [CF3CF2NH3]+, [HCF2CF2NH3]+, and [(NH3CF2)2]2+ were obtained from CF3CN, HCF2CN, and (CN)2, respectively, and anhydrous HF. The aforementioned fluorinated ammonium cations were isolated as room temperature stable [AsF6]? and/or [SbF6]? salts, and characterized by multi‐nuclear NMR and vibrational spectroscopy. The salts [HCF2NH3][AsF6] and [CF3NH3][Sb2F11] were characterized by their X‐ray crystal structure.  相似文献

An optically active (S)‐α‐ethylleucine ((S)‐αEtLeu) as a chiral α‐ethylated α,α‐disubstituted α‐amino acid was synthesized by means of a chiral acetal auxiliary of (R,R)‐cyclohexane‐1,2‐diol. The chiral α‐ethylated α,α‐disubstituted amino acid (S)‐αEtLeu was introduced into the peptides constructed from 2‐aminoisobutyric acid (=dimethylglycine, Aib), and also into the peptide prepared from diethylglycine (Deg). The X‐ray crystallographic analysis revealed that both right‐handed (P) and left‐handed (M) 310‐helical structures exist in the solid state of CF3CO‐(Aib)2‐[(S)‐αEtLeu]‐(Aib)2‐OEt ( 14 ) and CF3CO‐[(S)‐αEtLeu]‐(Deg)4‐OEt ( 18 ), respectively. The IR, CD, and 1H‐NMR spectra indicated that the dominant conformation of pentapeptides 14 and CF3CO‐[(S)‐αEtLeu]‐(Aib)4‐OEt ( 16 ) in solution is a 310‐helical structure, and that of 18 in solution is a planar C5 conformation. The conformation of peptides was also studied by molecular‐mechanics calculations.  相似文献

Compounds including the free or coordinated gas‐phase cations [Ag(η2‐C2H4)n]+ (n=1–3) were stabilized with very weakly coordinating anions [A]? (A=Al{OC(CH3)(CF3)2}4, n=1 ( 1 ); Al{OC(H)(CF3)2}4, n=2 ( 3 ); Al{OC(CF3)3}4, n=3 ( 5 ); {(F3C)3CO}3Al‐F‐Al{OC(CF3)3}3, n=3 ( 6 )). They were prepared by reaction of the respective silver(I) salts with stoichiometric amounts of ethene in CH2Cl2 solution. As a reference we also prepared the isobutene complex [(Me2C?CH2)Ag(Al{OC(CH3)(CF3)2}4)] ( 2 ). The compounds were characterized by multinuclear solution‐NMR, solid‐state MAS‐NMR, IR and Raman spectroscopy as well as by their single crystal X‐ray structures. MAS‐NMR spectroscopy shows that the [Ag(η2‐C2H4)3]+ cation in its [Al{OC(CF3)3}4]? salt exhibits time‐averaged D3h‐symmetry and freely rotates around its principal z‐axis in the solid state. All routine X‐ray structures (2θmax.<55°) converged within the 3σ limit at C?C double bond lengths that were shorter or similar to that of free ethene. In contrast, the respective Raman active C?C stretching modes indicated red‐shifts of 38 to 45 cm?1, suggesting a slight C?C bond elongation. This mismatch is owed to residual librational motion at 100 K, the temperature of the data collection, as well as the lack of high angular data owing to the anisotropic electron distribution in the ethene molecule. Therefore, a method for the extraction of the C?C distance in [M(C2H4)] complexes from experimental Raman data was developed and meaningful C?C distances were obtained. These spectroscopic C?C distances compare well to newly collected X‐ray data obtained at high resolution (2θmax.=100°) and low temperature (100 K). To complement the experimental data as well as to obtain further insight into bond formation, the complexes with up to three ligands were studied theoretically. The calculations were performed with DFT (BP86/TZVPP, PBE0/TZVPP), MP2/TZVPP and partly CCSD(T)/AUG‐cc‐pVTZ methods. In most cases several isomers were considered. Additionally, [M(C2H4)3] (M=Cu+, Ag+, Au+, Ni0, Pd0, Pt0, Na+) were investigated with AIM theory to substantiate the preference for a planar conformation and to estimate the importance of σ donation and π back donation. Comparing the group 10 and 11 analogues, we find that the lack of π back bonding in the group 11 cations is almost compensated by increased σ donation.  相似文献

The preparation of three new N‐Fmoc‐protected (Fmoc=[(9H‐fluoren‐9‐yl)methoxy]carbonyl) β2‐homoamino acids with proteinogenic side chains (from Ile, Tyr, and Met) is described, the key step being a diastereoselective amidomethylation of the corresponding Ti‐enolates of 3‐acyl‐4‐isopropyl‐5,5‐diphenyloxazolidin‐2‐ones with CbzNHCH2OMe/TiCl4 (Cbz=(benzyloxy)carbonyl) in yields of 60–70% and with diastereoselectivities of >90%. Removal of the chiral auxiliary with LiOH or NaOH gives the N‐Cbz‐protected β‐amino acids, which were subjected to an N‐Cbz/N‐Fmoc (Fmoc=[(9H‐fluoren‐9‐yl)methoxy]carbonyl) protective‐group exchange. The method is suitable for large‐scale preparation of Fmoc‐β2hXaa‐OH for solid‐phase syntheses of β‐peptides. The Fmoc‐amino acids and all compounds leading to them have been fully characterized by melting points, optical rotations, IR, 1H‐ and 13C‐NMR, and mass spectra, as well as by elemental analyses.  相似文献

The difluoromethyl group (CF2H) is considered to be a lipophilic and metabolically stable bioisostere of an amino (NH2) group. Therefore, methods that can rapidly convert an NH2 group into a CF2H group would be of great value to medicinal chemistry. We report herein an efficient Cu‐catalyzed approach for the conversion of alkyl pyridinium salts, which can be readily synthesized from the corresponding alkyl amines, to their alkyl difluoromethane analogues. This method tolerates a broad range of functional groups and can be applied to the late‐stage modification of complex amino‐containing pharmaceuticals.  相似文献

A detailed investigation of the reactions of PhSO2CF2H and PhSO2CH2F with (E)‐chalcone (=(E)‐1,3‐diphenylprop‐2‐en‐1‐one) at low temperatures revealed that these two reactions were kinetically controlled, and the ratios of 1,2‐ vs. 1,4‐adducts, which did not change much over time at these temperatures, reflect the relative rates of the two reaction pathways. The controlled experiments of converting the PhSO2CF2‐ and PhSO2CHF‐substituted 1,2‐adducts to 1,4‐adducts showed that these isomerizations are not favored due to the low stability and hard‐soft nature of PhSO2CF and PhSO2CHF? anions. Moreover, taking advantage of the remarkable stability and softness of (PhSO2)2CF? anion, an efficient thermodynamically controlled isomerization of (PhSO2)2CF‐substituted 1,2‐adduct to 1,4‐adduct was achieved for the first time.  相似文献

A direct and convenient method has been developed for the synthesis of optically active pyrrolidines bearing a quaternary stereogenic center containing a CF3 group at the C‐3 position of the pyrrolidine ring. The synthesis system, CuI/Si‐FOXAP‐catalyzed exo‐selective 1,3‐dipolar cycloaddition of azomethine ylides with β‐CF3‐β,β‐disubstituted nitroalkenes, provides pyrrolidines with high diastereoselectivities (up to >98:2 d.r.) and excellent enantioselectivities (up to >99.9 ee) and performs well for a broad scope of substrates under mild conditions.  相似文献

To enable a comparison between a C—H…X hydrogen bond and a halogen bond, the structures of two fluorous‐substituted pyridinium iodide salts have been determined. 4‐[(2,2‐Difluoroethoxy)methyl]pyridinium iodide, C8H10F2NO+·I, (1), has a –CH2OCH2CF2H substituent at the para position of the pyridinium ring and 4‐[(3‐chloro‐2,2,3,3‐tetrafluoropropoxy)methyl]pyridinium iodide, C9H9ClF4NO+·I, (2), has a –CH2OCH2CF2CF2Cl substituent at the para position of the pyridinium ring. In salt (1), the iodide anion is involved in one N—H…I and three C—H…I hydrogen bonds, which, together with C—H…F hydrogen bonds, link the cations and anions into a three‐dimensional network. For salt (2), the iodide anion is involved in one N—H…I hydrogen bond, two C—H…I hydrogen bonds and one C—Cl…I halogen bond; additional C—H…F and C—F…F interactions link the cations and anions into a three‐dimensional arrangement.  相似文献

The Ser, Cys, and His side chains play decisive roles in the syntheses, structures, and functions of proteins and enzymes. For our structural and biomedical investigations of β‐peptides consisting of amino acids with proteinogenic side chains, we needed to have reliable preparative access to the title compounds. The two β3‐homoamino acid derivatives were obtained by Arndt–Eistert methodology from Boc‐His(Ts)‐OH and Fmoc‐Cys(PMB)‐OH (Schemes 2–4), with the side‐chain functional groups' reactivities requiring special precautions. The β2‐homoamino acids were prepared with the help of the chiral oxazolidinone auxiliary DIOZ by diastereoselective aldol additions of suitable Ti‐enolates to formaldehyde (generated in situ from trioxane) and subsequent functional‐group manipulations. These include OH→OtBu etherification (for β2hSer; Schemes 5 and 6), OH→STrt replacement (for β2hCys; Scheme 7), and CH2OH→CH2N3→CH2NH2 transformations (for β2hHis; Schemes 9–11). Including protection/deprotection/re‐protection reactions, it takes up to ten steps to obtain the enantiomerically pure target compounds from commercial precursors. Unsuccessful approaches, pitfalls, and optimization procedures are also discussed. The final products and the intermediate compounds are fully characterized by retention times (tR), melting points, optical rotations, HPLC on chiral columns, IR, 1H‐ and 13C‐NMR spectroscopy, mass spectrometry, elemental analyses, and (in some cases) by X‐ray crystal‐structure analysis.  相似文献

Due to its unique physical and electrophilic properties, the difluoromethyl group (−CF2H) has been playing an irreplaceable role in the field of pharmaceutical and agrochemical industry. Methods that could efficiently incorporate the difluoromethyl group into the target molecules are increasing in the recent years. Developing a stable and efficient difluoromethylating reagent is thus highly attractive. In this review, we describe the development of a nucleophilic difluoromethylation reagent [(SIPr)Ag(CF2H)], including its elemental reaction, difluoromethylation reaction with different types of electrophiles, and its application in the synthesis of a nucleophilic and an electrophilic difluoromethylthiolating reagent.  相似文献

We report a novel 1:1 cocrystal of β‐alanine with dl ‐tartaric acid, C3H7NO2·C4H6O6, (II), and three new molecular salts of dl ‐tartaric acid with β‐alanine {3‐azaniumylpropanoic acid–3‐azaniumylpropanoate dl ‐tartaric acid–dl ‐tartrate, [H(C3H7NO2)2]+·[H(C4H5O6)2], (III)}, γ‐aminobutyric acid [3‐carboxypropanaminium dl ‐tartrate, C4H10NO2+·C4H5O6, (IV)] and dl ‐α‐aminobutyric acid {dl ‐2‐azaniumylbutanoic acid–dl ‐2‐azaniumylbutanoate dl ‐tartaric acid–dl ‐tartrate, [H(C4H9NO2)2]+·[H(C4H5O6)2], (V)}. The crystal structures of binary crystals of dl ‐tartaric acid with glycine, (I), β‐alanine, (II) and (III), GABA, (IV), and dl ‐AABA, (V), have similar molecular packing and crystallographic motifs. The shortest amino acid (i.e. glycine) forms a cocrystal, (I), with dl ‐tartaric acid, whereas the larger amino acids form molecular salts, viz. (IV) and (V). β‐Alanine is the only amino acid capable of forming both a cocrystal [i.e. (II)] and a molecular salt [i.e. (III)] with dl ‐tartaric acid. The cocrystals of glycine and β‐alanine with dl ‐tartaric acid, i.e. (I) and (II), respectively, contain chains of amino acid zwitterions, similar to the structure of pure glycine. In the structures of the molecular salts of amino acids, the amino acid cations form isolated dimers [of β‐alanine in (III), GABA in (IV) and dl ‐AABA in (V)], which are linked by strong O—H…O hydrogen bonds. Moreover, the three crystal structures comprise different types of dimeric cations, i.e. (AA)+ in (III) and (V), and A+A+ in (IV). Molecular salts (IV) and (V) are the first examples of molecular salts of GABA and dl ‐AABA that contain dimers of amino acid cations. The geometry of each investigated amino acid (except dl ‐AABA) correlates with the melting point of its mixed crystal.  相似文献

A series of heteroligated (salicylaldiminato)(β‐enaminoketonato)titanium complexes [3‐But‐2‐OC6H3CH = N(C6F5)] [PhN = C(R1)CHC(R2)O]TiCl2 [ 3a : R1 = CF3, R2 = tBu; 3b : R1 = Me, R2 = CF3; 3c : R1 = CF3, R2 = Ph; 3d : R1 = CF3, R2 = C6H4Ph(p ); 3e : R1 = CF3, R2 = C6H4Ph(o ); 3f : R = CF3, R2 = C6H4Cl(p ); 3g : R1 = CF3; R2 = C6H3Cl2(2,5); 3h : R1 = CF3, R2 = C6H4Me(p )] were investigated as catalysts for ethylene (co)polymerization. In the presence of modified methylaluminoxane as a cocatalyst, these complexes showed activities about 50%–1000% and 10%–100% higher than their corresponding bis(β‐enaminoketonato) titanium complexes for ethylene homo‐ and ethylene/1‐hexene copolymerization, respectively. They produced high or moderate molecular weight copolymers with 1‐hexene incorporations about 10%–200% higher than their homoligated counterpart pentafluorinated FI‐Ti complex. Among them, complex 3b displayed the highest activity [2.06 × 106 g/molTi?h], affording copolymers with the highest 1‐hexene incorporations of 34.8 mol% under mild conditions. Moreover, catalyst 3h with electron‐donating group not only exhibited much higher 1‐hexene incorporations (9.0 mol% vs. 3.2 mol%) than pentafluorinated FI‐Ti complex but also generated copolymers with similar narrow molecular weight distributions (M w/M n = 1.20–1.26). When the 1‐hexene concentration in the feed was about 2.0 mol/L and the hexene incorporation of resultant polymer was about 9.0 mol%, a quasi‐living copolymerization behavior could be achieved. 1H and 13C NMR spectroscopic analysis of their resulting copolymers demonstrated the possible copolymerization mechanism, which was related with the chain initiation, monomer insertion style, chain transfer and termination during the polymerization process. © 2017 Wiley Periodicals, Inc.
