A simple synthesis of tetrasubstituted purines is disclosed based on the solution phase elaboration of 4,6-dichloro-2-methylthio-5-nitropyrimidine. One-pot sequential C4 and C6 chloride substitution by secondary and primary amines yields 4,6-diamino-2-methylthio-5-nitropyrimidines. mCPBA-mediated oxidation of the methylthio moiety to the corresponding sulfone allows facile substitution at the 2-position. CrCl2 assisted reduction of the nitro group, followed by acid catalyzed orthoester cyclization, then provides novel tetrasubstituted purines not accessible by other methods.  相似文献

Purine analogues exhibiting a wide range of pharmacological activities have been considered a privileged structure in medicinal chemistry. In addition, the purine core consisting of four points of structural diversity is a well-sought scaffold in combinatorial chemistry. Although most of the efforts have been focused on 2,6,9-, 6,8,9-, or 2,8,9-trisubstituted purines, syntheses of 2,6,8,9-tetrasubstituted purines are rare. This paper presents a parallel solution phase approach for the synthesis of fully substituted purines via a 6-sulfur-substituted pyrimidine as the key intermediate. This strategy combining construction and modification of the purine ring thus increases the structural diversity of the final products. Sequential substitution of chlorines in 4,6-dichloro-2-methyl-5-nitropyrimidine with primary amine and benzylmercaptan afforded the 4-(substituted)amino-6-benzylthio-5-nitropyrimidine, which was readily converted to its diaminopyrimidine analogue by reduction of the nitro group. The diaminopyrimidine intermediate was cyclized to construct the purine ring with a C-8 substituent. Eventual oxidation of sulfur to sulfone and subsequent displacement by a primary or secondary amine provided the desired 2,6,8,9-tetrasubstituted purine analogues. This synthetic methodology was validated with the synthesis of a 216-member purine library.  相似文献

2-Amino-4,6-dichloro-5-nitropyrimidine is an intermediate required for the preparation of nitropyrimidines as inactivators of the DNA repairing protein MGMT. When attempting its synthesis, 2-amino-4,5,6-trichloropyrimidine is obtained instead, via unusual aromatic substitution of the nitro group in 2-amino-4-hydroxy-5-nitropyrimidin-6-one by chloride. The synthesis, the reactivity of 4,5,6-trichloropyrimidine and the efficient preparation of 2-amino-4,6-dichloro-5-nitropyrimidine are presented.  相似文献

The acid catalyzed reaction of 4,6-di-O-acetyl-3-deoxy-3-(ethoxycarbonylamino)-D-glucal and 6-chloropurine in nitrometliane solution gave 6-ehloro-9-(4′,6′-di-O-acetyl-2′,3′-dideoxy-3′-ethoxy-carbonylamino-α- and β-D-arafemohexopyranosyl)purine. These were converted to the corresponding deblocked 6-dimetliylaminopurine nucleosides by treatment with ethanolic dimethylamine; acetylation of these gave the respective 4′,6′-di-O-acetyl derivatives. The anomeric assignments for the nucleosides were based on their nmr spectral data.  相似文献

A highly efficient synthesis of the vitamin D(3) system on solid support is described. Two synthetic strategies for the solid-phase synthesis of vitamin D(3) were developed. One is for 11-hydroxy analogues, and the other is for most other synthetic analogues. In the latter strategy, the sulfonate-linked CD-ring 58 was initially immobilized on PS-DES resin to give solid-supported CD-ring 63 (Scheme 10). Similarly, solid-supported CD-ring 63 was prepared by attachment of the CD-ring 10 to the chlorosulfonate resin 64. The vitamin D(3) system was synthesized by Horner-Wadsworth-Emmons reaction of the A-ring phosphine oxide to a solid-supported CD-ring, followed by simultaneous introduction of the side chain and cleavage from resin with a Cu(I)-catalyzed Grignard reagent. Parallel synthesis of the vitamin D(3) analogues was accomplished by a split and pool methodology utilizing radio frequency encoded combinatorial chemistry, and a manual parallel synthesizer for side chain diversification and deprotection. Additionally, we demonstrated the synthesis of various A-rings in a similar protocol for efficient preparation of building blocks.  相似文献

以 2 ( 3,5 二氯 2 吡啶偶氮 ) 5 二甲氨基苯胺作为分光光度法测定微量钌 (Ⅱ )的新显色剂。在pH 4 0~ 6 0的HAc NaAc缓冲溶液中 ,钌 (Ⅱ )可与试剂形成稳定的配合物。在乙醇存在下 ,于 0 0 6~ 0 9mol/LHCl或 0 0 3~ 1 5mol/LH2 SO4介质中可转变为稳定的绿蓝色配合物 ,其最大吸收波长位于 636nm ,表观摩尔吸光系数为 1 0 7× 1 0 5L·mol- 1 ·cm- 1 。钌 (Ⅱ )浓度在 0~ 0 9mg/L范围内符合比尔定律。利用EDTA作掩蔽剂 ,5 0倍量的Fe3+、Cu2 +、Co2 +、Ni2 +等对测定无干扰 ,所拟方法已用于贵金属精矿中钌的测定  相似文献

A new simple, very sensitive, selective and accurate procedure for the determination of trace amounts of iron(II) by solid-phase spectrophotometry (SPS) has been developed. The procedure is based on fixation of iron(II) as 2,3-dichloro-6-(3-carboxy-2-hydroxy-1-naphthylazo)quinoxaline on a styrene-divinylbenzene anion-exchange resin. The absorbance of resin sorbed iron(II) complex is measured directly at 743 and 830nm. Iron(III) was determined by difference measurements after reduction of iron(III) to iron(II) with hydroxylamine hydrochloride. Calibration is linear over the range 1.0-20 microgL(-1) of Fe(II) with relative standard deviation (R.S.D.) of 1.65% (n=10.0). The detection and quantification limits for 100mL sample system are 280 and 950 ngL(-1) using 0.5 g of the exchanger. The molar absorptivity and Sandell sensitivity are also calculated and found to be 2.86 x 10(6)Lmol(-1)cm(-1) and 0.0196 ngcm(-2), respectively. The proposed procedure has been successfully applied to determine iron(II) and iron(III) in tap, mineral and well water samples.  相似文献

The thermal behavior of 4,6‐bis‐(5‐amino‐3‐nitro‐1,2,4‐triazol‐1‐yl)‐5‐nitropyrimidine (BANTNP) was studied under a non‐isothermal condition by DSC, PDSC and TG/DTG methods. The kinetic parameters (Ea and A) of the exothermic decomposition reaction are 304.52 kJ·mol?1 and 1024.47 s?1 at 0.1 MPa, 272.52 kJ·mol?1 and 1021.76 s?1 at 5.0 MPa, respectively. The kinetic equation at 0.1 MPa can be expressed as: dα/dT=1025.3(1?α)3/4exp(?3.8044×104/T)/β The critical temperature of thermal explosion is 588.28 K. The specific heat capacity of BANTNP was determined with a Micro‐DSC method, and the standard molar specific heat capacity is 397.54 J·mol?1·K?1 at 298.15 K. The adiabatic time‐to‐explosion of BANTNP was calculated to be 11.75 s.  相似文献

4-(1-Alkynyl)-3-bromo- and 4-(1-alkynyl)-3-chloro-2(5H)-furanones have been regioselectively synthesized in moderate to good yields by a new version of the Pd/Cu-catalyzed Sonogashira reaction involving treatment of 1-alkynes with 3,4-dibromo- and 3,4-dichloro-2(5H)-furanone, respectively, in the presence of KF as a base. 4-(1-Alkynyl)-3-bromo-2(5H)-furanones have been found to be able to undergo Stille-type and Suzuki-type reactions with aryl(tributyl)tins and arylboronic acids, respectively, to give 4-(1-alkynyl)-3-aryl-2(5H)-furanones in modest to satisfactory yields. Some 4-(1-alkynyl)-substituted 2(5H)-furanones so prepared have been found to exhibit significant cytotoxic activities, especially against human leukemia cell lines.  相似文献

Cyclization of unsymmetrical thioureas affords 3-(heteroaryl)-iminothiazolidin-4-ones with excellent levels of regiocontrol. In the absence of base, 2-(pyridylmethyl) and 2-(aminomethyl)benzimidazolyl substituents on the thiourea scavenge acid that is generated upon sulfur alkylation with bromoesters. The resulting conjugate acid plays an important role in influencing the regiochemical outcome and overall rate of the reaction.  相似文献
