A determination method has been optimized and validated for the simultaneous analysis of tetracycline (TC), oxytetracycline (OTC), chlortetracycline (CTC) and doxycycline (DC) in honey. Tetracyclines (TCs) were removed from honey samples by chelation with metal ions bound to small Chelating Sepharose Fast Flow columns and eluted with Na2EDTA-Mcllvaine pH 4.0 buffers. Extracts were further cleaned up by Oasis HLB solid-phase extraction (SPE), while other solid-phase extraction cartridges were compared. Chromatographic separation was achieved using a polar end-capped C 18 column with an isocratic mobile phase consisting of oxalic acid, acetonitrile and methanol. LC with ultraviolet absorbance at 355 nm resulted in the quantitation of all four tetracycline residues from honey samples fortified at 15, 50, and 100 ng/g, with liner ranges for tetracyclines of 0.05 to 2 μg/mL. Mean recoveries for tetracyclines were greater than 50% with R.S.D. values less than 10% (n= 18). Detection limits of 5, 5, 10, 10 ng/g for oxytetracycline, tetracycline, chlortetracycline and doxycycline, respectively and quantitation limits of 15 ng/g for all the four tetracyclines were determined. Direct confirmation of the four residues in honey (2-50 ng/g) was realized by liquid chromatography-tandem mass spectrometry (LC/MS/MS). The linear ranges of tetracyclines determined by LC/MS/MS were between 5 to 300 ng/mL, with the linear correlation coefficient r〉 0.995. The limits of detection of 1 to 2 ng/g were obtained for the analysis of the TCs in honey.  相似文献

A confirmatory method coupling liquid chromatography to tandem mass spectrometry (LC/MS/MS) is described for the determination of tetracycline, oxytetracycline, doxycycline and chlortetracycline in honey. Demeclocycline, another tetracycline molecule not reported for its usage in honey, was used as internal standard to quantify the four analytes. The sample preparation entails a clean-up on an Oasis HLB solid-phase extraction cartridge and analyses were realised by LC/MS/MS in selected reaction monitoring mode. The stability of tetracyclines was checked under various storage conditions at -20, +4 and +20 degrees C (both under dark and light exposures). Indeed, tetracyclines are not stable molecules and the epimerisation phenomenon was evaluated in this work. Appropriate correction factors of the MS/MS responses of each epimer were studied for each of the four tetracyclines to accurately quantify them. Moreover, the matrix effects encountered during the LC/MS/MS analyses were also studied in spiked experiments from blank honey samples of various geographical origins and different flower types.  相似文献

A sensitive, high performance liquid chromatography/tandem mass spectrometric (i.e. mass spectrometry/mass spectrometry; LC/MS/MS) method with on-line extraction and sample clean-up for the screening and confirmation of residues of sulfonamides in kidney is described. The sulfonamides are extracted from homogenized kidney with methanol. After centrifugation of the extract, an aliquot of the extract is directly injected on the LC/MS/MS system with further extraction and clean-up of the sample on-line. Detection of the analytes was achieved by positive electrospray ionization (ESI) followed by multiple reaction monitoring. For each sulfonamide the collisional decomposition of the protonated molecule to a common, abundant fragment ion was monitored. The method has been validated for sulfadimethoxine, sulfaquinoxaline, sulfamethazine, sulfamerazine, sulfathiazole, sulfamethoxazole, sulfadiazine and sulfapyridine. Calibration curves resulting from spiked blank kidney samples at the 10-200 microg/kg level showed good linear correlation. At the level of 50, 100 and 200 microg/kg both within- and between-day precision, as measured by relative standard deviation (RSD), were less than 16%. The limits of detection (LODs) ranged from 5 to 13.5 microg/kg. The recoveries ranged from 78 to 82%. The procedure provides a rapid, reliable and sensitive method for the determination of residues of sulfonamides in bovine kidney. The advantage of this method over existing methods is its decreased sample preparation and analysis time, which makes the method more suitable for routine analysis.  相似文献

A liquid chromatographic/tandem mass spectrometric (LC/MS/MS) multiresidue method for the simultaneous quantitative determination of oxytetracycline, 4-epi-oxytetracycline, tetracycline, 4-epi-tetracycline, chlortetracycline, 4-epi-chlortetracycline and doxycycline in milk has been developed. An extraction procedure consisting of a liquid extraction of the milk samples with trichloroacetic acid was performed. The extract was centrifuged and the supernatant was filtered. Solid-phase extraction (SPE) with an OASIS HLB SPE column was used to clean up the sample extracts. The samples were analysed by LC/MS/MS. The LC separation was performed on a reversed-phase C18 column using gradient elution with a mobile phase consisting of water and a mixture of methanol/acetonitrile. The tetracycline analytes were detected with a quadrupole mass spectrometer using positive ion electrospray ionisation. The confirmatory method has acceptable detection limits and the different tetracyclines can be detected at a residue concentration between 5 and 20 microg/L. The method is validated according to the European requirements for veterinary drug residues and all determined parameters were found to conform to the criteria. The recovery values ranged from 90.4 to 101.2% with relative standard deviations (RSDs) no larger than 9.7%. The overall or between-day precision of the analytical assay determined as repeatability at several residue concentrations and expressed as RSD ranged from 3.3 to 10%. This analytical assay is a useful tool within the Belgian monitoring programme for confirmation of samples which have been positively screened for residues of tetracyclines in raw farm cow's milk.  相似文献

建立了分散液液微萃取(DLLME)技术进行样品前处理,高效液相色谱(HPLC)法测定猪肾脏中土霉素(OTC)、四环素(TC)、金霉素(CTC)3种四环素类抗生素(TCs)残留量的方法。考察了分散剂种类、离子液体用量、分散剂用量、样品溶液p H值、萃取时间、盐效应等因素对萃取效率的影响。优化后的实验条件为:以丙酮为分散剂,离子液体([BMIM]PF6)用量为50μL,分散剂用量为140μL,样品溶液p H值为3.0,萃取时间为15 min,不添加盐。该方法在0.1~10.0 mg/L范围内线性关系良好(r2≥0.999 5),土霉素、四环素和金霉素的相对标准偏差(RSD)为2.2%~3.1%,检出限(LOD)为54~93μg/L,富集倍数为7.0~27.8,且样品的加标回收率达99.5%~101.1%。该法准确度和精密度均满足分析方法的要求,实现了对猪肾脏中土霉素、四环素、金霉素3种四环素类抗生素残留量的快速、绿色、灵敏和准确检测。  相似文献

The separation of tetracyclines (TCs) using reversed-phase liquid chromatography (LC) is proposed. The use of an amide-based stationary phase prevents the interaction of tetracyclines with the residual silanol groups and thus avoids the appearance of tailed peaks. Detection was based on using an UV spectrophotometer and gradient elution with acetonitrile-oxalic acid as mobile phase permitted good separation of all the peaks. Specificity was demonstrated by the retention characteristics, UV spectra and peak purity index. Linearity, precision, recovery and sensitivity were satisfactory. The procedure was applied to the analysis of tetracycline residues (tetracycline, oxytetracycline (OTC), chlortetracycline (CTC), doxycycline (DC), minocycline (MINO) and methacycline (MTC)) in honey of different types. Extraction involved using a mild acidic solvent containing EDTA to release protein-bound or sugar-bound tetracyclines. For the clean-up step, solid phase extraction using phenyl cartridges was applied. Detection limits in the honey using the proposed procedure are between 15 and 30 ng g(-1), depending on the tetracycline.  相似文献

The potential use of a class-specific enzyme-linked immunosorbent assay (ELISA) in studying the occurrence and fate of tetracyclines in the environment was evaluated. Several manure samples collected from hog lagoons and cattle feedlots were screened for the presence of tetracycline residues using ELISA. The levels varied from less than the detection limit (0.5 parts per billion) to 200 parts per million. The degradation of tetracyclines in soil-applied manure was followed using ELISA to measure the decline in tetracycline concentrations. Low levels of tetracyclines remained detectable in soil for up to 28 days. The ELISA procedure also proved useful in determining the leaching potential of tetracyclines in undisturbed soil columns and in the analysis of total tetracyclines in manure, soil, and water. Based on the cross-reactivity of the antibodies employed, this ELISA method can be an important screening tool for the presence of other tetracycline compounds, such as chlortetracycline and oxytetracycline. The ELISA method also detects the epimers of tetracyclines and the corresponding dehydration by-products, anhydrotetracyclines. Analysis of selected manure extracts by liquid chromatography with mass spectrometry (LC-MS) showed lower concentrations of total tetracyclines compared to the values obtained by ELISA, indicating the presence of other structurally related compounds or transformation products of tetracyclines being detected by ELISA in the samples. Because analysis of manure and soil samples by LC-MS requires extensive clean-up procedures, ELISA provides an alternative method for conducting environmental fate and transport studies of antibiotics.  相似文献

建立了磁性固相萃取-高效液相色谱-串联质谱同时测定环境水样中四环素类抗生素的方法。以6种四环素类抗生素(差向四环素、土霉素、四环素、去甲金霉素、金霉素和脱水四环素)为目标化合物,考察并优化了吸附和解吸条件,确定了最佳萃取条件。萃取后的目标化合物经ZORBAX Eclipse Plus C18柱分离,用高效液相色谱-串联质谱在多反应监测(MRM)模式下进行检测。在优化的条件下,6种四环素在1~100 μg/L范围内线性关系良好,线性相关系数为0.9967~0.9993,检出限为2.44~25.21 ng/L,样品加标回收率为80.6%~90.0%,日内相对标准偏差(RSDs)为0.6%~2.5%,日间RSDs为1.1%~7.1%。该方法灵敏度高、背景干扰低,适用于环境水样中6种痕量四环素类抗生素的同时检测。  相似文献

A rapid, sensitive, and robust CE-MS method has been developed for the determination of tetracycline, oxytetracycline, and chlortetracycline in milk. Field-amplified sample stacking with electromigration injection (FASS-EMI) was used for the online concentration of tetracyclines. The conditions of separation, MS detection, and stacking were systematically optimized. The optimum buffer composition was 35 mM Tris, 1.1% formic acid, 5% methanol, and 15% ACN. By using the online concentration method of field-enhanced sample stacking (FESI)-EMI stacking, the sensitivity was increased six- to seven-fold. The RSDs (n=6) of the relative migration time of tetracyclines were 1.1-1.4% for intraday and 2.4-2.9% for interday. The RSDs (n=6) of the relative peak area of tetracyclines were 3.2-4.6% for intraday and 4.7-6.1% for interday. The LODs (S/N=3) were 7.14 ng/mL for tetracycline, 11.4 ng/mL for oxytetracycline, and 14.9 ng/mL for chlortetracycline. The method has been successfully used to analyze tetracyclines residues in bovine milk.  相似文献

Efficient methods are needed for analysis of veterinary drug residues in food. A number of methods are available for single analytes. Multiresidue methods are now increasingly available. It is still rare, however, to find methods not involving mass spectrometry which allow for analysis of more than one class of drug residue. An efficient multiresidue method for the simultaneous determination of fluoroquinolones (FQs) and tetracyclines (TCs) in catfish muscle has now been developed. This method involves an extraction of the analytes with a mixture of acetonitrile and citrate buffer containing magnesium chloride. After centrifugation and evaporation of the supernatants, the residues are determined using high performance liquid chromatography with fluorescence detection. With this method, five fluoroquinolones and three tetracyclines were determined in fortified catfish muscle at levels of 20, 50, and 100 ng g(-1). Average recoveries for ciprofloxacin (CIP), sarafloxacin (SAR), danofloxacin (DANO), enrofloxacin (ENRO), difloxacin (DIF), oxytetracycline (OTC), tetracycline (TC), and chlortetracycline (CTC) were in the range of 60-92% with good relative standard deviations. The limits of quantitation ranged from 0.15 to 1.5 ng g(-1). Utilization of the method to successfully analyze catfish muscle samples incurred with enrofloxacin and with oxytetracycline is described.  相似文献

An ultra-performance liquid chromatography coupled with tandem mass spectrometry (UPLC/MS/MS) method was developed to screen and confirm multi-class veterinary drug residues in pig tissues including pig kidney, liver and meat. Twenty-one drugs of two different classes including seven tetracyclines and four types of quinolones (quinoline, naphthyridine, pyridopyrimidine and cinoline) were determined simultaneously in a single run. The homogenized sample tissues were extracted with EDTA-McIlvaine buffer solution and further purified using a polymer-based Oasis HLB solid-phase extraction (SPE) cartridge. An ACQUITY UPLC BEH C18 column was used to separate the analytes followed by tandem mass spectrometry using an electrospray ionization source. MS data acquisition was performed in the positive ion multiple reaction monitoring mode, selecting two ion transitions for each target compound. Recovery studies were performed at different fortification levels. The overall average recoveries from pig muscle, kidney, and liver fortified with quinolones and tetracyclines at three levels ranged from 80.2 to 117.8% based on the use of matrix-fortified calibration with the coefficients of variation ranging from 2.1 to 17.8% (n=6). The limits of quantitation (LOQs) of quinolones and tetracyclines in different tissues ranged from 0.03-4.50 microg/kg and 0.16-10.00 microg/kg, respectively. The effects of the extraction solvent, SPE cartridge, elution solvent and sample matrix on the analyte recovery as well as the effects of the mobile phase composition and column temperature on the chromatographic behavior were also studied.  相似文献

A simple, rapid and precise analytical method for the residual tetracyclines in honey has been established using a tandem cartridge clean-up system (prepacked reversed-phase and ion-exchange cartridges) followed by high-performance liquid chromatography. The recoveries of oxytetracycline (OTC), tetracycline (TC), chlortetracycline (CTC) and doxycycline (DC) from honey spiked at a level of 1.0 ppm are 87.1, 85.3, 98.0 and 99.0%, respectively, with coefficients of variation of 1.1-3.9%. The detection limits in honey are 0.02 ppm for OTC and TC, and 0.05 ppm for CTC and DC, respectively. The time required for the analysis of four samples is only 1 h.  相似文献

The increased production of ethanol in the US has resulted in large amounts of distillers grains (DG) which is an excellent feed supplement for livestock. However, the use of antimicrobials during ethanol fermentation has led to a growing concern over the possibility of their residues remaining in DG. To enable the detection of antimicrobial residues, a robust LC–MS/MS method was developed that included 13 antibiotics of diverse chemistries, ampicillin, penicillin G, tetracycline, oxytetracycline, chlortetracycline, bacitracin A, virginiamycin M1, chloramphenicol, erythromycin A, clarithromycin, tylosin A, monensin A and streptomycin. The residues were extracted with an aqueous solution of EDTA and trichloroacetic acid followed by methanol. The combined extract was subjected to a two-track cleanup and concentration on either hydrophilic polymeric or weak cation exchange solid phase extraction cartridges. The extracts are analyzed by LC/ion trap tandem mass spectrometry. The method was validated in dry DG matrix. Absolute recoveries of the analytes ranged from 50 to 100%. Accuracy ranged from 89 to 111% based on calibration by processed standards. The limits of detection and relative standard deviation are satisfactory to support future surveillance studies. The method was subsequently tested in three different end-products of DG: distillers dry grains, distillers wet grains and distillers grains solubles.  相似文献

An on-line simple and rapid method for the simultaneous determination of tetracycline (TC), oxytetracycline (OTC), chlortetracycline (CTC) and doxycycline (DC) residues in fish muscle was developed by coupling in-tube solid-phase microextraction (SPME) to high-performance liquid chromatography (HPLC) with a photodiode array detector. Biocompatible poly (methacrylic acid-ethylene glycol dimethacrylate) monolithic capillary was selected as the extraction medium, and no precipitating protein and removing fat steps were required prior to extraction. In order to optimize the extraction of these compounds, several in-tube SPME parameters were investigated. Simply performed by extracting with 0.01 M EDTA-MacIlvaine buffer solution (pH 4.0) and centrifugation, the sample then could be directly injected into the device for extraction. The limits of detection of tetracycline, oxytetracycline, chlortetracycline and doxycycline were calculated to be 22, 16, 30 and 21 ng/g, respectively. The calibration curves showed linearity in the range of 100-10,000 ng/g with a linear coefficient R2 value above 0.9980. Excellent method reproducibility was found by intra- and inter-day precisions, yielding the R.S.D.s less than 4.22% and 5.71%, respectively.  相似文献

A magnetic solid phase extraction method coupled to capillary electrophoresis is proposed for the determination of tetracycline, oxytetracycline, chlortetracycline and doxycycline in milk samples. Five different magnetic phenyl silica adsorbents covered with magnetite were synthesized by varying the molar ratio of phenyltrimethylsilane and tetramethylorthosilicate; these adsorbents were evaluated in terms of their pH and degree of hydrophobicity for tetracycline retention. The optimal, selected combination of conditions was a pH of 10.0 and a magnetic sorbent ratio of 4:1; under these conditions, the retention capacity ranged from 99.7% to 101.2% for the four tetracyclines analyzed. The elution conditions and initial sample volume of the proposed extraction method were also optimized, and the best results were obtained with 1×10(-3) M acetic acid in methanol as eluent and a 200 ml of sample volume. Under optimal conditions, average recoveries ranged from 94.2% to 99.8% and the limits of detection ranged from 2 to 9 μg l(-1) for the four tetracyclines. After the proposed method was optimized and validated, 25 milk samples of different brands were analyzed, oxytetracycline residues were detected in five samples, in concentrations ranging from 98 to 213 μg l(-1). Subsequent analysis of positive samples by SPE-CE and magnetic solid phase extraction-HPLC revealed than no significant differences were found from results obtained by the proposed methodology. Thus, the developed magnetic extraction is a robust pre-concentration technique that can be coupled to other analytical methods for the quantitative determination of tetracyclines.  相似文献

建立了在线凝胶渗透色谱-气相色谱/质谱(GPC-GC/MS)分析茶叶中153种农药残留的方法。样品用乙腈超声提取,提取液经石墨化炭黑固相萃取柱净化后,经GPC-GC/MS在线净化、分离和检测。方法的加标回收率为73.32%~117.05%,相对标准偏差为0.76%~13.18%。方法的检出限和定量限范围分别为0.0003~0.006 mg/kg和0.001~0.02 mg/kg。该方法样品前处理简单、分析时间短,灵敏度和精密度均符合农药多残留检测技术的要求,适用于茶叶中多种农药残留的检测。  相似文献

采用多壁碳纳米管固相萃取-超高效液相色谱-串联质谱技术同时测定蜂蜜中的磺胺类、喹诺酮类、硝基咪唑类和四环素类等52种兽药残留. 样品用Na2EDTA-Mcllvaine提取,经改性的多壁碳纳米管固相萃取小柱净化,通过Waters C18色谱柱分离,以乙腈和0.1%(体积分数)的甲酸水溶液为流动相进行梯度洗脱,采用电喷雾-正离子多反应监测的质谱模式,以基质外标法进行定量分析. 实验结果表明,52种兽药在相应的浓度范围内线性相关系数均大于0.99,该方法的定量限为1.5 ~7.5 μg/kg. 在7.5,25和50 μg/kg 3个浓度添加水平下,各种兽药的回收率为67.3% ~117.8%,相对标准偏差为1.9% ~17.4%. 结果表明,多壁碳纳米管固相萃取材料具有较好的净化效果,适用于蜂蜜中多类兽药残留的同时检测.  相似文献

A multiclass/multiresidue method has been developed and validated for the determination of 21 veterinary drug residues in shrimp, including sulfonamides (sulfadiazine, sulfamerazine, sulfamethazine, sulfachloropyridazine, sulfadimethoxine, and sulfaquinoxaline); tetracyclines (oxytetracycline, tetracycline, and chlortetracycline); (fluoro)quinolones (norfloxacin, ciprofloxacin, enrofloxacin, sarafloxacin, difloxacin, flumequine, oxolinic acid, and nalidixic acid); and cationic dyes (malachite green, gentian violet, leucomalachite green, and leucogentian violet), using HPLC/MS/MS. All drugs were quantifiable over a no less than 10-fold range with matrix-matched standards for linear external calibration, except for oxytetracycline, tetracycline, norfloxacin, and ciprofloxacin, for which norfloxacin-d5 was used as an internal standard. Two grams of preground shrimp sample was extracted twice with extractant at two different pH values. The combined supernatant was further diluted with an aqueous internal standard solution, and 50 microL extract was injected into the HPLC instrument. An online SPE system was set up for automated sample cleanup. A triple quadrupole mass spectrometer equipped with an electrospray ionization source was operated in the multiple-reaction-monitoring mode to acquire data. The method has been validated at three levels within the designated linear ranges for each drug, with accuracies between 77 and 115%, and most CV values below 15%.  相似文献

A graphene-based solid-phase extraction (SPE) column was prepared for the isolation of tetracyclines from milk followed by determination by high-performance liquid chromatography. Graphene provided better separation for tetracyclines than amine-modified graphene and carboxyl-modified graphene. The optimized graphene-based SPE column showed high absorption capacities (greater than 4,660?ng) and high recoveries (exceeding 92%) for tetracycline, oxytetracycline, chlortetracycline, and doxycycline and was successfully reused at least fifty times. The limits of detection in milk were from 10 to 20?ng/mL, with recoveries between 82.3 and 103.6%. Furthermore, the system showed superior performance than two commercial SPE cartridges with respect to recovery, purification, and reusability. Therefore, this approach is suitable for the determination of tetracyclines in milk.  相似文献
