It has been suggested that ultraviolet light induces free radical formation in skin, leading to photoaging and cancer. We have demonstrated by electron paramagnetic resonance that the ascorbate free radical is naturally present in unexposed skin at a very low steady state level. When a section of SKH-1 hairless mouse skin in an EPR cavity is exposed to UV light (4,500 J m−2−1, Xe lamp, 305 nm cutoff and IR filters), the ascorbate free radical signal intensity increases. These results indicate that UV light increases free radical oxidative stress, consistent with ascorbate's role as the terminal, small-molecule antioxidant. The initial radicals produced by UV light would have very short lifetimes at room temperature; thus, we have applied EPR spin trapping techniques to detect these radicals. Using α-[4-pyridyl 1-oxide]-N- tert -butyl nitrone (POBN), we have for the first time spin trapped a UV light-produced carbon-centered free radical from intact skin. The EPR spectra exhibited hyperfine splittings that are characteristic of POBN/alkyl radicals, aN= 15.56 G and aH= 2.70 G, possibly generated from membrane lipids as a result of β-scission of lipid alkoxyl radicals. Iron can act as a catalyst for free radical oxidative reactions; chronic exposure of skin to UV radiation causes increased iron deposition. Using our spin trapping system, we have shown that topical application of the iron-chelator, Desferal, to a section of skin reduces the UV light-induced POBN adduct radical signal. These results provide direct evidence for free radical generation and a role for iron in UV light-induced dermatopathology. We suggest that iron chelators can serve as photoprotective agents by preventing these oxidations.  相似文献

Unlike direct ESR, spin trap methodology depends on the absolute fidelity of the spin trap reaction. Two alternative reactions of 5,5-dimethyl-1-pyrroline N-oxide (DMPO) leading to radical adduct artifacts have been discovered and investigated: inverted spin trapping and the Forrester-Hepburn nucleophilic mechanism. These two alternate pathways to radical adducts are a combination of one-electron oxidation and nucleophilic addition, in either order. In biological systems, serious artifacts have been reported due to the Forrester-Hepburn mechanism, which is initiated by the addition of a nucleophile to DMPO. It has recently been demonstrated that (bi)sulfite (hydrated sulfur dioxide) can react with DMPO via a nonradical, nucleophilic reaction, and it has been further proposed that DMPO/(?)SO(3)(-) formation in biological systems is an artifact and not the result of spin trapping of sulfur trioxide anion radical ((?)SO(3)(-)). The one-electron oxidation of (bi)sulfite catalyzed by horseradish peroxidase (HRP)/hydrogen peroxide (H(2)O(2)) has been reinvestigated by ESR spin trapping with DMPO and oxygen uptake studies to obtain further evidence for the radical reaction mechanism. In the absence of DMPO, the initial rate of (bi)sulfite-dependent oxygen and H(2)O(2) consumption was determined to be half of the initial rate of DMPO/(?)SO(3)(-) radical adduct formation as determined by ESR, demonstrating that, under our experimental conditions, DMPO exclusively forms the radical adduct by trapping the (?)SO(3)(-).  相似文献

Electron spin resonance (ESR) techniques have been applied to a detailed study of batch and semicontinuous emulsion polymerization of methyl methacrylate; butyl acrylate and styrene have been briefly studied. Quenching of samples from the polymerization reactor followed by ESR analysis are useful, but we have developed continuous flow techniques which are preferable in many cases. ESR techniques can provide valuable information relating to the nature of free radical reactions, the concentration of propagating free radicals, and the kinetic processes in these reactions. Direct ESR analysis is most valuable but is not applicable to all systems. Spin trapping techniques can be useful for systems not accessible by direct analysis.  相似文献

The application of spin trapping to the detection of transient radicals generated in heterogeneous systems is discussed. The major part of this review focuses on the interaction of light with particles in dispersion and the resultant radicals which are produced. This includes photoactive pigments such as CdS, TiO2, ZnO and phthalocyanine in a variety of solvents as well as micelles, vesicles and microemulsions containing molecules such as phthalocyanine or chlorophyll. The utility of spin trapping in heterogeneous systems which are not a function of illumination (i.e. electrochemical reactions, catalysis and biological systems) is also discussed. It is demonstrated that reaction mechanisms can be better understood by the indirect detection of the radical intermediates which are involved in the various processes. This impacts studies in solar energy utilization, electrophotography, catalysis, electrochemistry, photosynthesis and the disease process.  相似文献

The reactions of hydrated electron (eaq-) with various radicals have been studied in pulse radiolysis experiments. These radicals are hydroxyl radical (*OH), sulfite radical anion (*SO3-), carbonate radical anion (CO3*-), carbon dioxide radical anion (*CO2-), azidyl radical (*N3), dibromine radical anion (Br2*-), diiodine radical anion (I2*-), 2-hydroxy-2-propyl radical (*C(CH3)2OH), 2-hydroxy-2-methyl-1-propyl radical ((*CH2)(CH3)2COH), hydroxycyclohexadienyl radical (*C6H6OH), phenoxyl radical (C6H5O*), p-methylphenoxyl radical (p-(H3C)C6H4O*), p-benzosemiquinone radical anion (p-OC6H4O*-), and phenylthiyl radical (C6H5S*). The kinetics of eaq- was followed in the presence of the counter radicals in transient optical absorption measurements. The rate constants of the eaq- reactions with radicals have been determined over a temperature range of 5-75 degrees C from the kinetic analysis of systems of multiple second-order reactions. The observed high rate constants for all the eaq- + radical reactions have been analyzed with the Smoluchowski equation. This analysis suggests that many of the eaq- + radical reactions are diffusion-controlled with a spin factor of 1/4, while other reactions with *OH, *N3, Br2*-, I2*-, and C6H5S* have spin factors significantly larger than 1/4. Spin dynamics for the eaq-/radical pairs is discussed to explain the different spin factors. The reactions with *OH, *N3, Br2*-, and I2*- have also been found to have apparent activation energies less than that for diffusion control, and it is suggested that the spin factors for these reactions decrease with increasing temperature. Such a decrease in spin factor may reflect a changing competition between spin relaxation/conversion and diffusive escape from the radical pairs.  相似文献

The stability of membranes under the strong oxidizing conditions in fuel cells is one of the major challenges in the development of fuel cells based on proton exchange membranes (PEMs). This study is centered on the determination of the susceptibility to degradation of SPEEK membranes exposed to OH radicals, using both direct ESR and spin trapping with 5,5-dimethyl-1-pyrroline-1-oxide (DMPO). In order to achieve a complete picture on SPEEK degradation, two types of experiments were performed: 1. UV irradiation at 77 K of SPEEK membranes swollen by aqueous solutions of H2O2; 2. UV irradiation of SPEEK membranes swollen by aqueous solutions of H2O2 in the presence of DMPO as a spin trap. UV irradiation without oxygen of SPEEK at 77 K in acid or basic form in the presence of H2O2/H2O produced phenoxyl radicals as the predominant radicals detected by direct ESR or spin trapping methods. At pH 4, the oxygen radicals produced phenyl radicals as the predominant species detected by spin trapping methods. The hydroperoxyl radical, as DMPO/OOH adduct, was detected only when the DMPO/OH adduct was absent. The appearance of phenyl and phenoxyl radicals provides the evidence that OH radicals react with the aromatic ring of SPEEK or leading to the scission of its ether bridge.  相似文献

Using a competitive spin trapping method, relative spin trapping rates were quantified for various short-lived radicals (methyl, ethyl, and phenyl radicals). High static pressure was applied to the competitive spin-trapping system by employing high-pressure electron spin resonance (ESR) equipment. Under high pressure (490 bar), spin trapping rate constants for alkyl and phenyl radicals increased by 10 to 40%, and the increase was dependent on the structure of nitrone spin traps. A maximum increase was obtained when tert-butyl(4-pyridinylmethylene)amine N-oxide (4-POBN) was used as a spin trap. Activation volumes (DeltaDeltaV(double dagger)) for the two spin trapping reactions were calculated to be -17-(-9) cm(3) mol(-1) for the 4-POBN system.  相似文献

Here we describe the synthesis and characterisation of a new isoindole-based nitrone spin trap, 1,1,3-trimethylisoindole N-oxide (TMINO). This nitrone and its radical adducts (isoindoline nitroxides) exhibit enhanced stability with respect to other commonly used spin traps and their adducts. We also report EPR trapping studies of this new nitrone with some carbon- and oxygen-centred radicals including alkyl, aryl, hydroxyl and benzoyloxyl systems. The narrow EPR line-widths and stability of the resulting nitroxide spin adducts allowed the detection of the expected radicals as well as secondary and minor radical components in the reaction mixtures.  相似文献

The formation of radicals during the liquid-phase radiolysis of ethylamine, diethylamine, and triethylamine was studied by means of the spin trapping technique. The radicals produced in ion-molecule reactions and in the rearrangement and fragmentation reactions of the primary radical cations of the amines were identified. The structure and reactions of the primary radical cations were studied in a low-temperature CFCl3 freonic matrix in which amine radical cations were generated via charge transfer from matrix radical cations to amines during freon irradiation. The results of experiments in the liquid and solid phases are consistent with one another. The structure of neutral radicals and radical cations of the ethylamines was corroborated by quantum-chemical calculations.  相似文献

Free radicals produced either by γ or ultrasonic irradiation of poly(methyl methacrylate) (PMMA) in benzene solution were stabilized by spin trapping; they were identified by analysis of ESR spectra of the trapped radicals (the spin adducts). The radical species identified after γ-irradiation were methyl, ester (COOCH3), a pair of the chain scission radicals, ~CH2C(CH3)(COOCH3) and CH2C(CH3)(COOCH3)~, and phenyl radical originating from the solvent. The chain scission radicals were also detected by spin trapping after ultrasonic irradiation of the benzene solution. Taking account of the difference in the trapping rate for two spin trapping agents, 2,4,6-tri-t-butylnitrosobenzene (BNB) and penta-methyl-nitrosobenzene (PMNB) the radical species trapped by PMNB are assumed to be precursors of those trapped by BNB. Based on the radical species found by the spin trapping method, plausible degradation processes for PMMA in benzene solution are proposed.  相似文献

A new phosphorylated linear nitrone N-(4-hydroxybenzyliene)-1-diethoxyphosphoryl-1-methylethylamine N-oxide (4-HOPPN) was synthesized, and its X-ray structure was determined. The spin trapping ability of various kinds of free radicals by 4-HOPPN was evaluated. Kinetic study of decay of the superoxide spin adduct (4-HOPPN-OOH) shows the half-life time of 8.8 min. On the basis of the X-ray structural coordinates, theoretical analyses using density functional theory (DFT) calculations at the B3LYP/6-31+G(d,p)//B3LYP/6-31G(d) level were performed on spin-trapping reactions of superoxide radical with 4-HOPPN and PBN and three possible decay routes for their corresponding superoxide adducts. The comparative calculations on the spin-trapping reactions with superoxide radical predicted that both spin traps share an identical reaction type and have comparable potency when spin trapping superoxide radical. Analysis of the optimized geometries of 4-HOPPN-OOH and PBN-OOH reveals that an introduction of the phosphoryl group can efficiently stabilize the spin adduct through the intramolecular H-bonds, the intramolecular nonbonding attractive interactions, as well as the bulky steric protection. Examination of the decomposition thermodynamics of 4-HOPPN-OOH and PBN-OOH further supports the stabilizing role of the phosphoryl group to a linear phosphorylated spin adduct.  相似文献

Eight simple amides have been subjected to UV irradiation in the presence of either MNP or NO. In all cases radical species were generated: these were detected by means of EPR spectroscopy in the form of different nitroxides resulting from the trapping of the primary radicals. NO acted as a double spin trap, scavenging a radical to afford a diamagnetic nitroso derivative that in turn acted as trap towards another radical unit. As amido-groups are present in components of skin tissue and may be present in many therapeutic or cosmetic products used as skin sunscreen, and NO is a ubiquitous endogenous reactive species, the nitroxides detected in the present studies might participate in radical processes triggered by sun exposure and resulting in damages, even severe, of biological tissues.  相似文献

There is current interest in using high-intensity ultrasound to perform a range of chemical transformations, including polymerisation reactions. In this work, the technique of radical trapping, primarily using DPPH, has been used to measure radical production rates in a range of monomer and related systems when exposed to high intensity ultrasound. It has been shown that realistic rates of production can be obtained around room temperature equivalent to thermal decomposition rates > 100°C, making sonication a viable method for initiating polymerisation. Rates of initiation in a twophase organic in water system have also been measured. Some of the initiating species have been identified recording the ESR spectra of adducts with spin traps, although further analysis is needed before the complete range of radicals produced can be identified.  相似文献

Organogold compounds arising from the trapping of carbon-centered radicals by gold chlorides (AuCl, AuCl3, and HAuCl4) and colloidal gold have been characterized by different spectroscopic techniques. Laser flash photolysis has provided kinetic evidence of the occurrence of radical trapping in the microsecond time scale, while theoretical calculations give the energetics of the elementary steps. The trapping of carbon radicals by gold species observed here may explain why the presence of radical initiators enhance dramatically the activity of gold catalysts for some reactions occurring through radical intermediates.  相似文献

Modeling of the addition of various radicals to C60 fullerene is currently an active research area. However, the radicals considered are not able to adequately model polymeric radicals. In this work, we have performed a theoretical study of the possible reactions of C60 fullerene with 1‐n‐phenylpropyl radicals, which are used to model polystyrene radicals. Several possible ways of subsequent addition of up to four 1‐phenylpropyl radicals to C60 have been analyzed, the structures of the intermediates have been defined and thermal properties, such as the activation enthalpies of the corresponding reactions, have been calculated using density functional theory with the approximation of PBE/3z. It is shown that the topology of the spin density distribution on the fullerenyl radical causes regioselectivity for further radical addition. According to the energetic characteristics of the reactions, we assume the possibility of formation of products of one‐, two‐, three‐, and four‐ addition of the growth radical to the fullerene core in radical polymerization of styrene in the presence of C60 fullerene. © 2016 Wiley Periodicals, Inc.  相似文献

The study of the influence of free radicals in the biological process depends primarily on the capacity to detect these reactive species. In this work we have studied the application of mass spectrometry to the identification of hydroxyl radical species. The detection and identification by collisional activation mass-analyzed ion kinetic energy spectrometry (CA-MIKES) of a spin adduct of DMPO with the hydroxyl radical [(DMPO + O) + H]+ (m/z 130) has demonstrated that mass spectrometry can be a powerful tool in the detection and identification of spin adducts of DMPO with hydroxyl radical species. We were also able to detect the capture of secondary free radicals using ethanol by detecting and identifying the corresponding adduct [(DMPO + ethanol) + H]+. Other spin adducts have also been detected and identified. We consider that the use of mass spectrometry is a relevant technique for the detection of free hydroxyl radicals, especially in complex mixtures, since mass spectrometry is able to discriminate these adducts in such situations. Moreover, using this approach, it was possible to identify new spin adducts.  相似文献

Given that spin trapping/electron paramagnetic resonance (EPR) spectroscopy has become the primary technique to identify important biologically generated free radicals, such as superoxide (O(2)(*-)), in vitro and in vivo models, evaluation of the efficiency of specific spin traps to identify this free radical is paramount. Recently, a family of ester-containing nitrones has been prepared, which appears to have distinct advantages for spin trapping O(2)(*-) compared to the well-studied spin traps 5,5-dimethyl-1-pyrroline N-oxide 1 and 5-(diethoxyphosphoryl)-5-methyl-1-pyrroline N-oxide 2. An important determinant in the selection of a spin trap is the rate constant (k(app)) for its reaction with O(2)(*-), and several different methods have been employed in estimating this k(app). In this paper, the two most frequently used scavengers of O(2)(*-), ferricytochrome c and Cu/Zn-SOD, were evaluated as competitive inhibitors for spin trapping this free radical. Data presented herein demonstrate that SOD is the preferred compound when determining the k(app) for the reaction of O(2)(*-) with spin traps. Using this model, the k(app) for the reaction of nitrone 1, 5-tert-butoxycarbonyl-5-methyl-1-pyrroline N-oxide 3, and 5-methoxycarbonyl-5-methyl-1-pyrroline N-oxide 4 with O(2)(*)(-) was estimated to be 24.6 +/- 3.1, 73.0 +/- 12, and 89.4 +/- 1.0 M(-1) s(-1) at pH 7.0, respectively. Several other comparative studies between known spin traps were also undertaken.  相似文献

EPR spectroscopy in conjunction with spin trapping has been employed extensively and successfully to detect and identify low-molecular-weight reactive radicals in biological systems. In contrast, this technique has been relatively infrequently used to investigate high-molecular-weight species generated as a result of radical-induced damage to biological macromolecules, such as DNA, RNA, proteins and carbohydrates, despite the fact that the destruction or alteration of these materials is known to play a key role in a large number of cellular injuries and diseases. Recent progress in the detection and identification of these macromolecular radicals by EPR spin trapping is reviewed, with particular emphasis on supplementary techniques which often yield valuable information as to the nature and identity of the species detected.  相似文献
