Chiral recognition was observed in a biomembrane model. Micellar aggregates formed by enantiopure N-alkyl-N,N-dimethyl-N-(1-phenyl)ethylammonium bromide were in fact able to convert the racemic mixture of bilirubin-IXalpha into an enantiomerically enriched mixture. The stereochemical preference and the extent of enantiomeric enrichment depend on the length of the hydrophobic portion of the surfactant and on the concentration conditions, and changes in the stereochemical bias are reversible.  相似文献

Chiral recognition of the enantiomeric couples of ditryptophan and diphenylalanine was observed by (1)H NMR spectroscopy in micelles formed by sodium N-dodecanoyl-L-prolinate. Ditryptophan showed a selective association with the Z domains of the amidic aggregates, whereas diphenylalanine did not show any selectivity in the association. Partition coefficients between water and aggregates were evaluated by diffusion NMR experiments. Intramolecular distances of ditryptophan isomers associated with chiral aggregates were obtained by ROESY experiments and were used as constraints in molecular mechanics calculations. From these calculations, information on the conformation of the peptides in the chiral aggregates was obtained.  相似文献

A novel approach to the enzyme model for the synthesis of peptides has been established by using multi-functionalized chiral crown ethers as hosts. The new strategy consists of three key steps as follows. (1) Guest assembly: the host having one free thiol and one thioester withN-protected-amino acid or peptide proceeds via rapid intra-complex thiolysis of-amino acid ester salts to form the dithioester, and assembles two guests. (2) Amide formation: the intramolecular aminolysis occurs between the bound guests to form the amide bond. (3) Peptide chain elongation: as the thiol reactive group is regenerated, the above two reactions are repeated to elongate the peptide chain. In the present paper, we describe the multiple chiral recognition that could be achieved by the chiral crown ether in both the intra-complex thiolysis and the intramolecular aminolysis. For explanation of the chiral recognition, we propose a likely structure for the intermediate of the aminolysis.  相似文献

Examples of reverse turns as recognition motifs in biological systems can be found in high-resolution crystal structures of antibody-peptide complexes. Development of peptidomimetics is often based on replacing the amide backbone of peptides by sugar rings, steroids, benzodiazepines, or other hetero- and carbocycles. In this approach, the chemical scaffold of the peptide backbone can be replaced while retaining activity as long as the pharmacophoric groups of the peptide side chains stay in relatively the same place; in other words, similar functional groups must overlap in space for interaction with critical receptor sites. This study evaluates the potential of metal complexes of chiral pentaazacrowns (PAC) derived by reduction of cyclic pentapeptides as -turn mimetics. Due to the limited flexibility of the pendant chiral side groups in these metal complexes, one can potentially elicit information about the receptor-bound conformation from their binding affinities. 11 PAC crystal structures with different substitution patterns complexed with 3 different metals (Mn, Fe, Cd) as a prototypical database of potential side-chain orientations. Complexation with different metals induces subtle differences in the conformations of a particular azacrown scaffold. The lack of parameterization of transition metals for force field calculations precludes a thorough theoretical study. Thus, this study utilizes a simple geometrical comparison between the experimental data for crystalline PAC complexes and the side-chain orientations seen in classic -turns. The FOUNDATION program was used to overlap the C-C vectors of the corresponding ideal -turn side-chains to all possible leaving groups of the PAC complexes. When comparing the relative orientations of the chiral side chains, a strong overlap of the bonds (between about 0.1 Å to about 0.5 Å RMS for 3 residues and up to about 1 Å RMS for 4 residues) was observed for many of the molecules. Such metal complexes may lack complete peptidomimetic activity due to the lack of spatial overlap of all four side-chain residues, however, if only three peptide side chains are needed for receptor recognition and/or binding, the metal complexes should show biological activity.  相似文献

Thermoplastics have been increasingly used for fabricating microfluidic devices because of their low cost, mechanical/biocompatible attributes, and well-established manufacturing processes. However, there is sometimes a need to integrate such a device with components made from other materials such as polydimethylsiloxane (PDMS). Bonding thermoplastics with PDMS to produce hybrid devices is not straightforward. We have reported our method to modify the surface property of a cyclic olefin copolymer (COC) substrate by using corona discharge and grafting polymerization of 3-(trimethoxysilyl)propyl methacrylate; the modified surface enabled strong bonding of COC with PDMS. In this paper, we report our studies on the surface modification mechanism using attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS), atomic force microscopy (AFM) and contact angle measurement. Using this bonding method, we fabricated a three-layer (COC/PDMS/COC) hybrid device consisting of elastomer-based valve arrays. The microvalve operation was confirmed through the displacement of a dye solution in a fluidic channel when the elastomer membrane was pneumatically actuated. Valve-enabled microfluidic handling was demonstrated.  相似文献

The size and shape of the chiral cavity of a macrocyclic receptor were tuned by the alteration of the binaphthyl moiety to improve the chiral recognition/discrimination ability. For example, host 3 with the 3,5-bis(trifluoromethyl)phenyl group at the 3,3'-positions showed improved enantioselectivity for small molecules such as 2-chloropropionic acid and methyl lactate as evaluated by the binding constants. This host 3 also had an excellent ability as an NMR chiral solvating agent.  相似文献

A chiral stationary phase (CSP 1) derived from an (S)-N-3,5-dinitrobenzoyl-1-naphthylglycine showed excellent enantiomeric separation for amino acid derivatives with a fluorogenic reagent, 4-fluoro-7-nitro-2,1,3-benzoxadiazole (NBD-F), in high-performance liquid chromatography (HPLC). We compared elution profiles (separation factor and elution order) of NBD-amino acids and their analogs on HPLC, to determine the diastereomeric complex between the chiral moiety of CSP 1 and NBD-amino acid, which is responsible for the chiral recognition. (1)H-NMR studies of a mixture of model compound of CSP 1 and NBD-Ala suggest that the diastereomeric complex is composed of two hydrogen bonding sites at the amino proton and oxygen atom, and a pi-pi interaction by the benzofurazan structure (2,1,3-benzoxadiazole) of NBD-amino acid. Furthermore CSP 1 was able to separate esters, amides and alpha-methyl amino acids derivatized with NBD-F.  相似文献

The purpose of this paper was to study the enantioseparation mechanism of triadimenol compounds by carboxymethylated (CM)‐β‐CD mediated CE. All the enantiomers were separated under the same experimental conditions to study the chiral recognition mechanism using a 30 mM sodium dihydrogen phosphate buffer at pH 2.2 adjusted by phosphoric acid. The inclusion courses between CM‐β‐CD and enantiomers were investigated by the means of molecular docking technique. It was found that there were at least three points (one hydrophobic bond and two hydrogen bonds) involved in the interaction of each enantiomer with the chiral selectors. A new mathematic model has been built up based on the results of molecular mechanics calculations, which could analyze the relationship between the resolution of enantioseparation and the interaction energy in the docking area. Comparing the results of the separation by CE, the established mathematic model demonstrated good capability to predict chiral separation of triadimenol enantiomers using CM‐β‐CD mediated CE.  相似文献

The chemical characteristics of the polar parts of phospholipids as the main components of biological membranes were investigated by using infrared (IR) spectroscopy and theoretical calculations with water as a probe molecule. The logical key molecule used in this study is methylphosphocholine (MePC) as it is not only a representative model for a polar lipid headgroup but itself has biological significance. Isolated MePC forms a compact (folded) structure which is essentially stabilized by two intramolecular C-H...O type hydrogen bonds. At lower hydration, considerable wavenumber shifts were revealed by IR spectroscopy: the frequencies of the (O-P-O)- stretches were strongly redshifted, whereas methyl and methylene C-H and O-P-O stretches shifted surprisingly to blue. The origin of both red- and blueshifts was rationalized, on the basis of molecular-dynamics and quantum-chemistry calculations. In more detail, the hydration-induced blueshifts of C-H stretches could be shown to arise from several origins: disruption of the intramolecular C-H...O hydrogen bonds, formation of intermolecular C-H...O(water) H-bonds. The stepwise disruption of the intramolecular hydrogen bonds appeared to be the main feature that causes partial unfolding of the compact structure. However, the transition from a folded to extended MePC structure was completed only at high hydration. One might hypothesize that the mechanism of hydration-driven conformational changes as described here for MePC could be transferred to other zwitterions with relevant internal C-H...O hydrogen bonds.  相似文献

The dirhodium complex Rh(+II)2[(+)-MTPA]4 is introduced as an NMR auxiliary for enantiodifferentiation. As a soft Lewis acid, it is particularly useful for the recognition of soft Lewis base molecules. The donor properties of ligands can be divided into several categories and are exemplified by various classes of phosphorus-containing functionalities. In addition, the thermodynamics of the underlying ligand-complex equilibria are discussed and stereochemical implications demonstrated. The extension of the dirhodium method to absolute configuration determination is evaluated. The outlook for future applications with further non-phosphorus ligands is presented.  相似文献

Terpyridine macrocycle 1 is shown to be a strong chelating agent for organic ammonium salts and also a useful chromophore in fluorescent sensing. The pi-bonding of the beta-diketonate ligands in C2-symmetric (dike)2TiX2 complexes engenders a significant electronic dissymmetry, with the LUMO of the complex resembling a "tipped" dz2 orbital whose tilt with respect to the plane containing the metal and the two X groups is determined by the configuration at titanium. The ability of this electronic dissymmetry to promote chiral recognition has been probed in 1,1'-bi-2-naphtholate complexes (RCOCHCOR)2Ti(BINOL) (R = CH3, Ph, or tBu). The complexes show exclusively one diastereomer by NMR, which has been established as the one predicted on electronic grounds by crystallography of the dibenzoylmethane and 2,2,6,6-tetramethylheptanedione complexes. Confirmation that the selectivity is due to electronic rather than steric factors is given by the behavior of the tin analogues, which are structurally similar but show much smaller diastereoselectivities.  相似文献

We report quantum dynamical simulations for the laser controlled isomerization of 1-(2-cis-fluoroethenyl)-2-fluorobenzene based on one-dimensional electronic ground and excited state potentials obtained from (TD)DFT calculations. 1-(2-cis-fluoroethenyl)-2-fluorobenzene supports two chiral and one achiral atropisomers, the latter being the most stable isomer at room temperature. Using a linearly polarized IR laser pulse the molecule is excited to an internal rotation around its chiral axis, i.e. around the C-C single bond between phenyl ring and ethenyl group, changing the molecular chirality. A second linearly polarized laser pulse stops the torsion to prepare the desired enantiomeric form of the molecule. This laser control allows the selective switching between the achiral and either the left- or right-handed form of the molecule. Once the chirality is "switched on" linearly polarized UV laser pulses allow the selective change of the chirality using the electronic excited state as intermediate state.  相似文献
