Three complementary capillary electrophoresis (CE) methods were developed for the separation and quantification of ephedrine and pseudoephedrine stereoisomers. Either single or dual cyclodextrin-based chiral selector systems provided enantioselective separation of the compounds of interest. The three methods were applied to the analysis of a suite of five standard reference materials (SRMs) containing ephedra. Use of a high-sensitivity UV detection cell enhanced quantification of the analytes of interest over the wide range of concentrations encountered in the SRMs. Results for (-)-ephedrine ranged from 0.31 to 76.43 mg/g, and for (+)-pseudoephedrine ranged from 0.049 to 9.23 mg/g in the materials studied. Results from the three methods agreed well with each other and with the results from other methods of analysis. The addition of known amounts of specific enantiomers was used to confirm the enantiomeric identity of the analytes. The results obtained by the three CE methods were utilized for value assignment of the ephedrine alkaloid content of these five SRMs.  相似文献

In this work, simultaneous separation of eight stereoisomers of ephedrine and related compounds ((+/-)-ephedrine, (+/-)-pseudoephedrine, (+/-)-norephedrine and (+/-)-N-methylephedrine) was accomplished using a polymeric chiral surfactant, i.e. polysodium N-undecenoxycarbonyl-L-leucinate (poly-L-SUCL) by chiral (C)MEKC-ESI-MS. The conditions of CMEKC were first investigated. The baseline separation of all eight stereoisomers of ephedrine and related compounds was achieved under optimum CMEKC conditions (35 mM poly-L-SUCL, 15 mM NH(4)OAc, pH 6.0, 30% v/v ACN, 30 kV and 20 degrees C) in less than 30 min. Next, a central composite design for response surface modeling has been described to evaluate the electrospray chamber parameters and the sheath liquid conditions. Optimum mass abundance of stereoisomers of ephedrine and related compounds was observed using the spray chamber parameters, namely 250 degrees C drying gas temperature and 8 L/min drying gas flow rate at a nebulizer pressure of 4 psi. Furthermore, the experimental design indicates that the optimum mass abundance of the stereoisomers of ephedrine and related compounds can be obtained using a sheath liquid containing 80:20 v/v methanol-water, 5 mM NH(4)OAc at pH 8.5 delivered at 5 microL/min. Finally, compared to MEKC-UV, the use of poly-L-SUCL in MEKC-MS provided significantly higher sensitivity for stereoisomers of ephedrine and related compounds.  相似文献

A method was developed for determination of total iodine content in different standard reference materials (SRMs) and seafood products by inductively coupled plasma/mass spectrometry (ICP/MS). If iodine is present as iodide and nitric acid is used in the wet digestion system, the observed signal is not stable when iodine is measured by ICP/MS at m/z 127. To stabilize the iodine signal, 3% ammonia solution (1 + 1, v/v) was added to the digest. The limit of quantitation of the method, defined as 6 times the standard deviation in the blank solution (n = 20) was estimated to be 15 mg/kg (using 0.2 g dry mass and a dilution factor of 50). The precision, expressed as repeatability of the iodine concentration, varied between 3.2 and 12% in SRMs, with concentrations of 4.70-0.17 mg/kg dry matter. The described method was compared with a method using tetramethylammonium hydroxide extraction. Both methods showed good precision and trueness by analyses of SRMs. The 2 methods were used to determine iodine in seafood from the Barents Sea, the Norwegian Sea, and the North Sea. The results showed great variation between different fish species as well as between individuals within a species. The lowest values of iodine were recorded in muscle of ling (Molva molva) with a mean of 0.07 mg/kg fresh weight and a variation between 0.03 and 0.11 mg/kg fresh weight. The highest values were found in cod (Gadus morhua) from the Barents Sea, with a mean of 2.5 mg/kg and a variation between 0.7 and 12.7 mg/kg fresh weight.  相似文献

A new and accurate HPLC method using sulfobutylether-beta-cyclodextrin (SBE-beta-CD) as chiral mobile phase additive (CMPA) was developed and validated for the determination of R-(+)pantoprazole in S-(-)pantoprazole. The influences of type and concentration of CD, ACN content and buffer pH of mobile phase on the resolution and retention of enantiomers were investigated. A baseline resolution of pantoprazole enantiomers was achieved on a Spherigel C18 column (150 mm x 4.6 mm, 5 microm) using ACN and 10 mM phosphate buffer (pH 2.5) containing 10 mM SBE-beta-CD (15:85 v/v) as mobile phase with a flow rate of 0.9 mL/min at 20 degrees C. The detection wavelength was set at 290 nm. The method was extensively validated in terms of accuracy, precision and linearity according to the International Conference on Harmonisation (ICH) guidelines and proved to be robust. The LOD and LOQ for R-(+)pantoprazole were 0.2 and 0.5 microg/mL, respectively, with 5 microL injection volume. A good linear relationship was obtained in the concentration range of 0.5-6.0 microg/mL with r(2) >0.999 for R-(+)pantoprazole. The percentage recovery of the R-(+)pantoprazole ranged from 92.1 to 101.2 in bulk drug of S-(-)pantoprazole. The method is capable of determining a minimum limit of 0.05% w/w of R-enantiomer in S-(-)pantoprazole bulk samples.  相似文献

A fast, economic, reproducible, accurate, effective, rugged and selective chiral-HPLC method was developed and validated for the enantiomeric resolution of nebivolol enantiomers [(+)-RRRS and (-)-SSSR)] in dosage formulation. The method was rapid as chiral separation occurred within only 12 min. The mobile phase used was n-heptane-ethanol-DEA (85:15:0.1, v/v) at 3.0 mL/min flow-rate with 225 nm detection. The column used was an amylase-based 3-AmyCoat (150 × 46 mm) [tris-(3,5-dimethylphenyl carbamate)]. The capacity factors of (+)-RRRS and (-)-SSSR enantiomers were 7.85 and 10.90 while the separation and resolution factors were 1.39 and 1.83, respectively. The limits of detection and quantitation for (+)-RRRS enantiomer were 4.5 and 10.00 μg/mL, while these values for (-)-SSSR enantiomer were 4.1 and 8.2 μg/mL, respectively. The linearity was observed in the concentrations range of 0.10-1.0 mg/mL for both enantiomers. The π-π interactions, hydrogen bonds, dipole-dipole interactions and steric effects control the chiral resolution of nebivolol enantiomers on the reported chiral column. The reported method can be used for the quality control of nebivolol in pharmaceutical preparations with good economy. In addition, this method can also be used for the analysis of (+)-RRRS and (-)-SSSR) enantiomers in biological and environmental samples.  相似文献

The preliminary validation of a high-performance liquid chromatography particle beam mass spectrometry method (HPLC-PB/MS) with electron impact ionization source for analysis of botanical extracts is presented. The LC-PB/MS system was evaluated for the analysis of ephedrine alkaloids using ephedra-containing National Institute of Standards and Technology dietary supplement standard reference materials (SRMs) 3241 Ephedra Sinica Stapf Native Extract and 3242 Ephedra Sinica Stapf Commercial Extract. The ephedrine alkaloids were separated by reversed-phase chromatography using a phenyl column at room temperature. A linear gradient method with a mobile phase composition varying from 5:95 [MeOH:0.1% trifluoroacetic acid (TFA) in water] to 20:80 (MeOH:0.1% TFA in water) at a flow rate of 1.0 ml/min, with an analysis time of less than 20 min, was used. The source block temperature was evaluated to determine the optimal operating conditions by monitoring the intensities and fragmentation patterns of the ephedrine alkaloids. Ephedrine and N-methylephedrine were taken as a representative of the test alkaloids. The LODs on the sub-nanogram level were achieved, with ephedrine, pseudoephedrine, and methylephedrine in the SRMs quantified by a standard addition method with recoveries of > or = 86% and RSDs of < or = 14% (n = 3).  相似文献

Selective SPE of derivates of p-hydroxybenzoic acid (pHBA) from plant extract of Melissa officinalis is presented using a molecularly imprinted polymer (MIP) made with protocatechuic acid (PA) as template molecule. MIP was prepared with acrylamide as functional monomer, ethylene glycol dimethacrylate as crosslinking monomer and ACN as porogen. MIP was evaluated towards six phenolic acids: PA, gallic acid, pHBA, vanillic acid (VA), gentisic acid (GeA) and syringic acid (SyrA), and then steps of molecularly imprinted SPE (MISPE) procedure were optimized. The best specific binding capacity of MIP was obtained for PA in ACN (34.7 microg/g of MIP). Other tested acids were also bound on MIP if they were dissolved in this solvent. ACN was chosen as solvent for sample application. M. officinalis was extracted into methanol/water (4:1, v/v), the extract was then evaporated to dryness and dissolved in ACN before application on MIP. Water and ACN were used as washing solvents and elution of benzoic acids was performed by means of a mixture methanol/acetic acid (9:1, v/v). pHBA, GA, PA and VA were extracted with recoveries of 56.3-82.1% using this MISPE method. A method for simultaneous determination of eight adulterants including two appetite suppressants, two energy expenditure-enhancing drugs, one diuretic and three cathartics in slimming functional foods by high performance liquid chromatography with electrospray ionization-tandem mass spectrometry (HPLC-ESI-MS/MS) was established. After samples were ultrasonically extracted with 70% (v/v) methanol aqueous solution and centrifuged, the components of ephedrine, norpseudoephedrine, fenfluramine, sibutramine, clopamide, emodin, rhein, and chrysophanol in sample solution were separated by a Hypersil Gold column (2.1 mm × 150 mm, 5 μm) using a programmed gradient elution. A mobile phase consisting of 0.02% (v/v) formic acid-ammonium formate buffer solution (pH=3.50) and methanol was used for elution with a flow rate set at 250 μL/min and column temperature of 25 °C. Qualitative determination was based on characteristic ion pairs and retention time of the targeted compounds using SRM (selective reaction monitoring) mode. Clenbuterol and ibuprofen were internal standards in positive and negative ionization mode, respectively. The internal standard curves were used for quantification measurement. The average recoveries of three different concentrations were from 80.2% to 94.5%. The limits of detection (LODs) were from 0.03 to 0.66 mg/kg (except chrysophanol 1.6 mg/kg). The linear dynamic range covered from 1 to 500 μg/L (except chrysophanol 50-5000 μg/L) for the twelve samples analyzed. Adulterants in four different kinds of slimming functional foods were determined by this developed method, and satisfactory results were obtained. These experimental results showed that, adulteration of sibutramine or/and fenfluramine were the major adulterating components with contents varying from 6.1 to 1.3×10(3) mg/kg and 1.9 to 9.7×10(3) mg/kg, respectively. In addition, three cathartic compounds were detected in six of those tested samples, and ephedrine, norpseudoephedrine and clopamide were not detected in all samples.  相似文献

Micellar electrokinetic chromatography (MEKC) of highly hydrophobic compounds is generally difficult using sodium dodecyl sulfate micellar solutions. The polymeric surfactant, polysodium undecyl sulfate (poly-SUS) has been used to separate moderately to highly hydrophobic polychlorinated biphenyl (PCB) congeners by MEKC in the absence of cyclodextrins. Parameters such as concentration of acetonitrile (ACN), polymeric surfactant concentration, and the effect of pH were examined. Optimum MEKC conditions to get baseline resolution of nine PCBs was 7.5 mM borate in 40% (v/v) ACN fraction buffered at pH 9.2 using 0.5% (w/v) poly-SUS. The applied voltage was 30 kV and the temperature was maintained at 25 degrees C. Elution order for each PCB congener was found to be dependent on the degree of chlorination and hydrophobic character.  相似文献

The adsorption mechanism of nortryptiline on C18-bonded Discovery   总被引:1,自引:0,他引:1  
The adsorption isotherms of an ionizable compound, nortriptyline, were accurately measured by frontal analysis (FA) on a C(18)-Discovery column, first without buffer (in an aqueous solution of acetonitrile at 15%, v/v of ACN), then with a buffer (in 28%, v/v ACN solution). The buffers were aqueous solutions containing 20 mM of formic acid or a phosphate buffer at pH 2.70. The linear range of the isotherm could not be reached with the non-buffered mobile phase using a dynamic range larger than 40,000 (from 1.2 x 10(-3) g/L to 50 g/L). With a 20 mM buffer in the liquid phase, the isotherm is linear for concentrations of nortriptyline inferior to 10(-3) g/L (or 3 micromol/L). The adsorption energy distribution (AED) was calculated to determine the heterogeneity of the adsorption process. AED and FA were consistent and lead to a trimodal distribution. A tri-Moreau and a tri-Langmuir isotherm models accounted the best for the adsorption of nortriptyline without and with buffer, respectively. The nature of the buffer affects significantly the middle-energy sites while the properties of the lowest and highest of the three types of energy sites are almost unchanged. The desorption profiles of nortriptyline show some anomalies in relation with the formation of a complex multilayer adsorbed phase of acetonitrile whose excess isotherm was measured by the minor disturbance method. The C(18)-Discovery column has about the same total saturation capacity, around 200 g of nortriptyline per liter of adsorbent (or 116 mg/g), with or without buffer. About 98-99% of the available surface consists in low energy sites. The coexistence of these different types of sites on the surface solves the McCalley's enigma, that the column efficiency begins to drop rapidly when the analyte concentration reaches values that are almost one hundred times lower than those that could be predicted from the isotherm data acquired under the same experimental conditions. Due to the presence of some relatively rare high energy sites, the largest part of the saturation capacity is not practically useful.  相似文献

A fast liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the analysis of acetylcholine (ACh) in cultured cells. [(2)H(4)]Acetylcholine (ACh-d(4)) was used as an internal standard for calibration. ACh was extracted from the cell lysate with acetonitrile (ACN)/water (80/20, v/v) and the crude extract was analyzed without further purification. Isocratic hydrophilic interaction chromatography (HILIC) with (10 mM) ammonium formate/ACN (35/75, v/v) as mobile phase was used for separation. ACh was eluted within 5 min and detected using electrospray-MS/MS in the positive ion mode. The limit of detection (LOD) was found to be 1.5f mol (0.3 nmol/L) ACh with a S/N ratio of 3:1. The approach was used for the measurement of ACh in undifferentiated SN56 cells and the ACh content was determined to be 1272+/-109 pmol/mg protein.  相似文献

A simple and inexpensive CE method was developed for the determination of peimine and peiminine. Because of the lack of an UV chromophore of peimine and peiminine, the detection method chosen was indirect UV detection, with N‐(1‐naphthyl)ethylenediamine dihydrochloride (NED) as the UV absorbing probe. It was thought that NED, a chromophoric ion, may form hydrogen bonding pairs with the analytes to cause significant changes in separation selectivity. Additionally, the hydrophobic interactions between analytes and the probe also play a crucial role in achieving a resolution between the two analytes. The analyses were carried out with a background electrolyte composed of 66% MeOH–ACN (1:1, v/v), 34% aqueous buffer containing 15 mM NaH2PO4, 2.5 mM NED, 4 mM H3PO4. MeOH–ACN mixtures used as organic modifiers can not only reduce the adsorption of NED to the capillary wall, but also decrease the baseline noise and drift. The method provided a linear response ranging from 5 to 200 μg/mL. The limits of detection (LODs) for peimine and peiminine were 3.9 and 4.1 μg/mL, respectively. The repeatabilities (n = 3) reached relative standard deviation values (RSDs) of 3.4 and 4.1% for the peak areas, 4.0 and 4.4% for the peak heights, and 0.29 and 0.30% for the migration time of peimine and peiminine, respectively. Regression equations revealed linear relationships (r = 0.9995–0.9996) between the peak area of each analyte and the concentration. The method developed was successfully applied to quantify peimine and peiminine in chloroform extracts of the ground Bulbus Fritillariae Thunbergii.  相似文献

A poly(methacrylic acid-ethylene glycol dimethacrylate, MAA-EGDMA) monolithic capillary was used for the direct and on-line extraction of telmisartan from Sprague-Dawley rat tissue (heart, kidney, and liver) homogenates. Under optimized conditions, the tissue homogenates were simply diluted with a mixture of phosphate buffer (pH 2)/ACN (90:8 v/v), and then injected for extraction only after centrifugation and filtration. Coupled to HPLC with fluorescence detection, the method was linear over the range of 1.25-1500 ng/g for telmisartan in heart and kidney, 12.5-15 000 ng/g in liver with correlation coefficients over 0.9992. The detection limits were found to be in the range from 0.24 to 1.8 ng/g. RSDs for intra- and inter-day ranged from 1.2 to 8.1%. The determination of telmisartan in treated rat tissues was achieved by using the proposed method.  相似文献

为建立混合物中羟基封端聚丁二烯(HTPB)的临界点色谱定量方法,以C18为固定相,考察了HTPB在四氢呋喃-乙腈和四氢呋喃-水两种流动相体系下的临界点色谱条件。结果表明,四氢呋喃-乙腈体积比为70.7:29.3以及四氢呋喃-水体积比为92:8时,HTPB的保留值与其相对分子质量无关。将该临界点色谱方法用于胶黏剂混合组分中HTPB的测定,结果表明,HTPB在46.7~216.4 mg/L范围内呈良好线性关系,相关系数(r)>0.99,检出限为4.2 mg/L。自配HTPB和矿物油双组分样品的加标回收率为89.2%~101.1%,相对标准偏差小于0.66%(n=6)。应用该方法对市售聚氨酯胶黏剂进行检测,测定结果为HTPB成分占26.6%。该方法快速、准确,能满足聚合物混合产品的生产质量控制和失效分析的要求。  相似文献

A molecularly imprinted polymer prepared using 1-allylpiperazine (1-ALPP) as the functional monomer, trimethyltrimethacrylate (TRIM) as the crosslinker and the zearalenone (ZON)-mimicking template cyclododecanyl-2,4-dihydroxybenzoate (CDHB) has been applied to the clean-up and preconcentration of this mycotoxin (zearalenone) and a related metabolite, alpha-zearalenol (alpha-ZOL), from cereal and swine feed sample extracts. The extraction of ZON and alpha-ZOL from the food samples was accomplished using pressurized liquid extraction (PLE) with MeOH/ACN (50:50, v/v) as the extraction solvent, at 50 degrees C and 1500 psi. The extracted samples were cleaned up and preconcentrated through the MIP cartridge and analyzed using HPLC with fluorescence detection (lambda (exc)=271/ lambda (em)=452 nm). The stationary phase was a polar endcapped C18 column, and ACN/MeOH/water 10/55/35 (v/v/v, 15 mM ammonium acetate) at a flow rate of 1.0 mL min(-1) was used as the mobile phase. The method was applied to the analysis of ZON and alpha-ZOL in wheat, corn, barley, rye, rice and swine feed samples fortified with 50, 100 and 400 ng g(-1) of both mycotoxins, and it gave recoveries of between 85 and 97% (RSD 2.1-6.7%, n=3) and 87-97% (RSD 2.3-5.6%, n=3) for alpha-ZOL and ZON, respectively. In this work, NACE with UV detection is combined with SPE using multi-walled carbon nanotubes (MWCNT) as stationary phase to determine a group of seven pesticides (pirimicarb, pyrifenox, penconazol, carbendazim, cyromazine, pyrimethanil and cyprodinil) in mineral water samples using ametryn as internal standard. The optimized BGE, consisting of a mixture of MeOH and ACN (1:2 v/v) with 90 mM SDS and 20.5 mM HClO(4), was satisfactory to get a good resolution of the seven compounds in less than 13 min. On-line preconcentration was carried out by electrokinetic injection of the sample dissolved in 78:22 v/v MeOH/ACN, 1.11 mM HClO(4). Repeatability was studied for the same day (n=4), for nine different days (n=36) and for four different capillaries. RSD values were appropriate in all cases, i.e. in the range 4.3-9.4% between different capillaries. MWCNT of 10-15 nm od, 2-6 nm id and 0.1-10 mum length were used as SPE materials for the preconcentration of these pesticides from water samples. SPE parameters influencing the enrichment were optimized and the most favorable conditions were as follows: the amount of stationary phase, eluent, sample pH and sample volume were 40 mg MWCNT, 10 mL ACN and 10 mL dichloromethane containing 5% v/v formic acid, pH 8.0, and 750 mL, respectively. Mean recovery values ranged between 53 and 94% for Milli-Q water and between 47 and 93% for mineral waters (RSD values were in the range 2-16%). The method allowed the determination of these pesticides at concentrations below the maximum residue limits established by the European Union legislation (LOD in the range 27-58 ng/L). When the cost, amount and type of the carbon nanotubes used in this work are compared with those carbon nanotubes previously used in the literature it is clear that the proposed materials can be used as economical stationary phases, even cheaper than conventional SPE cartridges.  相似文献

A new method was developed for the determination of six fluoroquinolone antibiotics including fleroxacin, levofloxacin, ciprofloxacin, lomfloxacin, enrofloxacin, and sparfloxacin in chicken breast muscle, in which the extraction and clean-up were performed in one step by microwave irradiation. The mixture of ACN containing 0.3% v/v phosphoric acid/water pH 3 (70:30, v/v) was used as the extraction solution and hexane was used as the clean-up solution. The extract was analyzed by liquid chromatography-tandem mass spectrometry system. The RSDs of intra- and inter-day obtained are in the range of 1.0-10.4 and 3.8-13.6%, respectively. In the three fortified levels of chicken breast muscle (20, 100, and 500 ng/g), the recoveries of fluoroquinolone antibiotics ranging from 66.0 to 97.2% are obtained. The LODs are in the range of 2.7-6.7 ng/g. This method simplifies the process of the sample preparation and reduces the operation errors.  相似文献

An ion chromatographic method with conductivity detection for the simultaneous determination of ephedrine, pseudoephedrine and norephedrine was developed. A mixture of 2.0 mmol/L HNO3 and 2% (v/v) acetonitrile was used as eluent. The three ephedrine-like compounds were separated and determined within 20 min. The linear ranges were 0.08-50 microg/mL for ephedrine, 0.08-40 microg/mL for pseudoephedrine and 0.06-40 microg/mL for norephedrine. The detection limits were 0.03 microg/mL for ephedrine and pseudoephedrine, and 0.02 microg/mL for norephedrine. The method has been applied successfully to the determination of these sympathomimetics in pharmaceutical preparations and in Ephedra herbs.  相似文献

(+)‐Pinoresinol 4,4′‐di‐O‐βD ‐glucopyranoside ((+)‐PDG) is one of the major lignans with various pharmacological activities which could be isolated from Duzhong and other plant species. In this study, a diastereomeric impurity, (?)‐pinoresinol 4,4′‐di‐O‐βD ‐glucopyranoside ((?)‐PDG), the main impurity was identified in (+)‐PDG chemical reference substance (CRS) and a reliable chromatographic method for rapid purity determination of (+)‐PDG CRS was firstly developed. The optimal chromatographic condition was found to be using ACN/1,4‐dioxane–water (2.5:6:91.5, v/v/v) as mobile phase on a Waters Acquity UPLC HSS T3 column (2.1 mm×100 mm, 1.8 μm) with column temperature of 37°C. The method was validated and applied to determine the chromatographic purity of five (+)‐PDG CRS samples. The content of (?)‐PDG in four commercial (+)‐PDG CRS was 8.47–20.30%, whereas no (?)‐PDG was detected in our in‐house prepared (+)‐PDG CRS in which purity was confirmed to be 99.80%. The above results confirmed that this method is fast and highly efficient for purity determination of the (+)‐PDG CRS.  相似文献

A high-performance liquid chromatographic method was optimized and validated for the determination of atenolol and chlorthalidone (CT) in human breast milk. The milk samples were extracted and purified using ACN and phosphoric acid for precipitation of proteins followed by removal of ACN and milk fats by extraction with methylene chloride. The samples were applied, after an extraction procedure, to a cyanide column using a mobile phase consisting of ACN/water (35:65 v/v) and buffered at pH 4.0 with flow rate of 1.0 mL/min. Quantitation was achieved with UV detection at 225 nm using guaifenesin as the internal standard. The effectiveness of protein precipitation and clean up procedure were investigated.
