A novel piezoelectric immunosensor has been developed for the detection of human complement C1-inhibitor. Anti-C1-inhibitor antibody was immobilized onto the gold electrodes of a 9 MHz AT-cut piezoelectric crystal. Coating the crystal with polyethyleneimine adhesion, followed by a glutaraldehyde cross-linking method to immobilize antibody showed better results than the physical adsorption method with respect to sensitivity and reproducibility. Under the optimized experimental conditions, the sensor showed good response to the C1-inhibitor in the range from 2.0 × 10–8 to 1.2 × 10–6 g. Other proteins in human serum did not remarkably interfere with the detection. The crystals could be regenerated 5 times, when bound materials on the crystal surface were eluted by strong acid and strong alkali solution and subsequently cleaned in an ultrasonic cleaner.  相似文献

A piezoelectric immunosensor has been developed for the detection of complement C4. Anti-C4 antibody was immobilized onto the gold electrodes of a 9 MHz AT-cut piezoelectric crystal. The coated crystal with the physical adsorption method to immobilize antibody showed the better results than the polyethyleneimine adhesion, glutaraldehyde cross-linking method with respect to sensitivity and reproducibility. The antibody-bound crystal with the physical adsorption method was successfully used for the detection of human complement C4 in the concentration range of 0.1-10 μg/mL for 40 min incubation time. The immunosensor system had good selectivity, and other materials in human serum did not interfere the detection remarkably. The crystal could be regenerated nearly 15 times when the bound materials on the crystal surface were eluted by strong acid and strong alkali solutions and subsequently cleaned in a ultrasonic cleaner.  相似文献

A polymer sorbent formed at the surface of gold-plated piezoelectric crystal by anodic oxidation of m-aminophenol was investigated by voltammetric, FTIR techniques for antibody coupling after activation by divinylsulphone. A novel film with increased capacity for immobilizing antibodies was obtained using phloroglucinol to modify poly-m-aminophenol via divinylsulphone. Compared with the dip-coating methods using polyethylenimine and (gamma-aminopropyl)trimethoxysilane, this new technique gave more reproducible results for the immobilization of antibody from sample to sample due to the improved homogeneity and reproducibility of the coating. With present modification method, a piezoelectric immunosensor was developed for the detection of Salmonella enteritidis. A detection limit of 1x10(5) cells ml(-1) and an assay time of 25 min were achieved.  相似文献

A new piezoelectric crystal impedance sensor for the determination of sulphur dioxide in aqueous solution is presented. It is realized using a series piezoelectric crystal device which is constructed by connecting an AT-cut piezoelectric crystal to a probe in series. The probe is filled with an internal electrolyte solution that is separated from sample solutions by a gas-permeable membrane. The present sensor exhibits a favourable frequency response to 1 x 10(-7)-1 x 10(-3) M sulphur dioxide. The detection limit is 1 x 10(-8) M. The effects of the sensor preparation are considered. Dynamic range, reproducibility, response time and selectivity of the sensor are also discussed. The proposed sensor has been used successfully for lamp sulphur determinations in petroleum samples.  相似文献

A selective, sensitive and simple ion-selective piezoelectric (ISP) sensor was developed for the direct determination of isoniazid (INH) in body fluids. Based on sensitive mass response of piezoelectric quartz crystal and selective adsorption/desorption across the modified film, the ISP sensor was fabricated by coating a PVC film containing activant on one electrode of a thickness-shear mode piezoelectric quartz crystal. The observed frequencies of ISP sensor were found to decrease with the increase of the INH concentration in a 0.1 M NaNO(3) solution. In this paper, three activants, INH-phosphotungstate (I), INH-silicotungstate (II), and INH-[BiI(4)](-) (III), were synthesized and investigated. Calibration graphs were linear from 6x10(-8) to 2x10(-3) M for I, 2x10(-7) to 2x10(-3) M for II and 2x10(-7) to 2x10(-3) M for III, with detection limits 6x10(-8) M for I, 2x10(-7) M for II and 2x10(-7) M for III, in a 0.1 M NaNO(3) solution at pH 7.0 and 37 degrees C. Recoveries were from 98% to 102% with R.S.D. up to 2%. Results for real samples obtained by the proposed method agreed well with those obtained by the conventional pharmacopeia method.  相似文献

将9MHz双面镀金石英晶体浸入蛋白A溶液中,在晶体电极表面形成一层均匀的蛋白A薄层,用于定向固定人体纤维蛋白抗体.在蛋白A层上形成一层有序致密的自组装抗体分子膜,研制成一种新型的用于人体纤维蛋白检测的压电免疫传感器.比较了3种固定抗体方法的效果,从传感器的灵敏度、稳定性、重现性等考虑,蛋白A吸附法优于聚乙烯亚胺及牛血清白蛋白固定抗体的方法.研究了蛋白A浓度、抗体效价以及抗原抗体反应时间等对传感器灵敏度的影响,考察了电极的选择性和再生能力.纤维蛋白在1×10-4~1×10-2g/L浓度范围内有良好响应.  相似文献

A novel method for monitoring, in real time, the formaldehyde (FA)-induced DNA-protein cross-linking process with the piezoelectric quartz crystal impedance (PQCI) technique is proposed. The method was used to monitor FA-induced DNA-lysozyme cross-link formation. Lysozyme was directly immobilized on the silver electrode surface of a piezoelectric quartz crystal by adsorption. The lysozyme-coated piezoelectric sensor was in contact with FA and DNA solutions. The time courses of the resonant frequency and equivalent circuit parameters of the sensor during the cross-linking were simultaneously obtained and are discussed in detail. On the basis of the feature of the multi-dimensional information provided by the PQCI technique, it was concluded that the observed frequency decrease could be mainly ascribed to the mass increase resulting from the cross-linking. According to the frequency decrease with time, the kinetics of the cross-linking process were quantitatively studied. A piezoelectric response model for the cross-linking was theoretically derived. Fitting the experimental data to the model, the kinetic parameters, such as the binding and dissociation rate constants (k(1) and k(-1)) and the cross-linking equilibrium constant (Ka), were determined. At 37 degrees C, the k(1), k(-1) and Ka values obtained were 7.0 (+/-0.1) x 10(-5) (microg ml(-1))(-1) s(-1), 6.6 (+/-0.1) x 10(-3) s(-1) and 1.06 (+/-0.02) x 10(-2) (microg ml(-1))(-1), respectively.  相似文献

Fullerene(C60)‐dibenzo‐16‐crown‐5‐oxyacetic acid (DBI6C5‐OCH2‐COOC60) was prepared and applied as the coating material on piezoelectric quartz crystals for detection of various metal ions and polar/nonpolar organic molecules. The C60‐crown ether‐coated piezoelectric crystal sensor with a home‐made computer interface for signal acquisition and data processing was applied as an ion chromatographic (IC) detector for various metal ions, e.g., alkali metal, alkaline earth metal and transition‐metal ions. The piezoelectric detector exhibited quite good sensitivity of 104 ~ 106 Hz/M and good detection limit of 10?3 ~ 10?4 M for these metal ions. The C60‐crown ether piezoelectric detector compared well with the commercial conductivity detector conventionally used for metal ions. The ionic size and ionic charge seemed to have significant effect on the frequency response of the piezoelectric detector. The C60‐crown ether coated piezoelectric crystal sensor was also employed as a high performance liquid chromatographic (HPLC) detector for various polar organic molecules with frequency responses in the order: amines > carboxylic acids > alcohols > ketones. Furthermore, nonpolar organic molecules, e.g., n‐hexane, 1‐hexene and 1‐hexyne, were also detected with this piezoelectric crystal detector. The frequency responses of the piezoelectric crystal detector for these nonpolar organic molecules were in the following order: alkynes > alkenes > alkanes. The effects of solvents and flow rate on the frequency responses of the piezoelectric crystal detector were investigated. The C60‐crown ether coated piezoelectric crystal detector also showed short response time (< 1 min.) and good reproducibility.  相似文献

The method describes the use of a piezoelectric quartz crystal (PQC) as a substitute for ion-selective electrodes. The approach is feasible when the membrane materials are electrically non-conductive and membrane potential measurements are consequently not possible. An ion-selective piezoelectric sensor sensitive to atropine sulfate was constructed by coating a PVC membrane containing activant on one the side of a PQC. On the basis of selective adsorption of atropine ions across the modified film and the sensitive mass response of PQC, the method exhibits a sensitive, rapid response and is easy to operate without pretreatment of the sample. The logarithm of the frequency shift gave a linear relationship with the logarithm of atropine sulfate concentration in the 1.0 x 10(-8)-1.0 x 10(-3) M range with a detection limit of 5.0 x 10(-9) M at pH 7.0. Recoveries were from 98.7-102.2%. Two activants, atropine tetraphenylborate and atropine dipicrylaminate, were synthesized and investigated. Influencing factors were also examined and optimized. The results for real samples obtained by the proposed method agreed with those obtained by conventional methods.  相似文献

The construction and general performance characteristics of three piezoelectric quartz crystal sensors responsive to the pentoxyverine are described here. This kind of non-potentiometric sensing method is based on use of ion-pair complexes of the pentoxyverine cation with three counter anions, namely, tungstophosphate, tetraphenylborate and picrolonate. The complexes were embedded in a PVC matrix. Adsorption of the pentoxyverine ion on the complex caused a frequency decrease of the crystal. The frequency decrease was proportional to the amount of adsorbed analyte. The influencing factors were investigated in detail, and then optimized. The proposed sensors exhibit reasonable selectivity and a higher sensitivity than the potentiometric sensors. For a sensor modified with pentoxyverine-phosphotungstate, the calibration graph was linear over concentration of 1.0 x 10(-7) - 5.0 x 10(-5) M with a detection limit of 6 x 10(-8) M at pH 5.4.  相似文献

A method using HPLC-CL linkage was developed for simultaneous determination of N,N-dimethylaniline and phenol in wastewater, based on the strong sensitive chemiluminescence of the luminol-K3Fe(CN)6 systems in alkaline medium. The separation was carried out on a Hypersil ODS column with a mobile phase of ethanol-0.01% triethylamine (2:1, v/v). The linear ranges for N,N-dimethylaniline determinations were 2.0 x 10(-7) - 2.5 x 10(-5) g/mL and 4.0 x 10(-5) - 1.5 x 10(-4) g/mL with a detection limit (3sigma) of 1.20 x 10(-8) g/mL; the relative standard deviation (3sigma) for 5.0 x 10(-6) g/mL N,N-dimethylaniline was 1.4% (n = 6). The range for phenol was from 5.1 x 10(-7) to 1.3 x 10(-4) g/mL, and a detection limit (3sigma) of 2.5 x 10(-8) g/mL could be obtained. The method can be useful for the determination of N,N-dimethylaniline and phenol in some environmental samples.  相似文献

A piezoelectric immunosensor was developed for detecting Mycobacteria Tuberculosis (M. TB). Protein A was modified onto a silver-coated piezoelectric quartz-crystal sensor. Antibodies could be bond onto the crystal surface in an ordered orientation through protein A. An impendence analyzer was employed to record the admittance-frequency curve. The experimental results showed a successful and less-rigid bonding of protein A and antibodies. Compared with other traditional detection methods of TB, the method suggested in this paper was sensitive, selective and effective. The nonspecific response was limited by using control antibody-rabbit anti-honey bee venom (anti-HBV). A glycine-HCI buffer solution (pH = 2.4) was used to release antibodies from a crystal coated with protein A, and 0.5 mg/mL anti-TB was used to remove TB from a crystal that was bonded by an antibody. Good reusability was exhibited. Spike samples of sputum and saliva from normal people with and without adding M. tuberculosis were diagnosed using the proposed method. A novel piezoelectric immunosensor has been developed for the determination of beta-indole acetic acid (IAA) in dilute solutions. The detection is based on competitive immunoreaction between a hapten (IAA) and an antigen (IAA-BSA, hapten-protein conjugation) bound to an anti-IAA antibody, immobilized on a quartz crystal microbalance (QCM). The frequency change (y) of the sensor caused by antigen is linearly related to the logarithm of the concentration of IAA (x) in the range of 0.5 ng/ml - 5 microg/ml with a regression equation of the form y = -23x + 151 (r = 0.9937).  相似文献

Abstract

A piezoelectric crystal immunosensor has been developed for the detection and determination of cortisol. Cortisol antibody was layered onto the gold electrodes of a 10 MHz piezoelectric crystal which was precoated with either protein A or gluteraldehyde. Crystals pre-coated with protein A showed the best results with respect to stability and sensitivity.

The sensor was successfully used for the determination of cortisol in standard solutions from 36–3628 μg/L (part per billion). The advantages of the proposed sensor include simplicity, short analysis time, cost effectiveness and selectivity. The results demonstrate the feasibility of cortisol assay in clinical testing and in drug monitoring.  相似文献

Various fullerene C60‐proteins such as C60‐myoglobin (C60‐Mb), C60‐hemoglobin (C60‐Hb) and C60‐gliadin, coated piezoelectric quartz crystals were prepared and applied in piezoelectric quartz crystal immunosensors for protein‐antibodies such as anti‐myoglobin (Anti‐Mb), anti‐hemoglobin (Anti‐Hb) and anti‐gliadin respectively. The immobilizations of myoglobin, hemoglobin and gliadin onto Fullerene C60 were studied with a C60‐coated piezoelectric crystal detection system, respectively. The partially irreversible frequency responses for theses proteins were observed by a desorption study, implying that C60 can strongly adsorb these proteins. Thus, immobilized C60‐Mb, C60‐Hb and C60‐gliadin coating materials were successfully prepared and identified with FTIR spectrometry. The C60‐Mb, C60‐Hb and C60‐gliadin coated piezoelectric (PZ) quartz crystal immunosensors with homemade computer interfaces for signal acquisition and data processing were developed and applied for detection of Anti‐Mb, Anti‐Hb and anti‐gliadin respectively. The C60‐protein coated PZ immunosensors for Anti‐Mb, Anti‐Hb and antigliadin exhibited linear frequency responses to the concentrations of theses anti‐proteins with sensitivities of 1.43 × 103, 2.59 × 103 and 8.05 × 103 Hz/(mg/mL) respectively. The detection limits of these PZ‐immunosensors were 4.36 × 10?3, 3.23 × 10?3 and 1.98 × 10?3 mg/mL for Anti‐Mb, Anti‐Hb and anti‐gliadin respectively. Effects of pH and temperature on the frequency responses of the anti‐protein PZ‐immunosensors were also investigated. The optimum pH of these anti‐proteins and the optimum temperature for the PZ‐immunosensors were observed at pH = 7 and around 30 °C respectively. The interferences of various common species in human blood, e.g., cysteine, tyrosine, urea, glucose, ascorbic acid and metal ions, to these anti‐protein PZ‐immunosensors were also investigated respectively. These species showed nearly no interference or quite small interference with the anti‐protein PZ‐immunosensors. The reproducibility and lifetime of these immobilized C60‐protein coated PZ crystal immunosensors were also investigated and discussed.  相似文献

A new method of monitoring drug binding to DNA, in real-time, by means of the piezoelectric quartz-crystal-impedance (PQCI) technique, is proposed. The method was used to monitor the binding of an anticancer drug, mitoxantrone (MX), to double-stranded calf-thymus DNA covalently immobilized on the thioglycollic acid-modified gold electrode surface of a quartz crystal. Optimum experimental conditions for the immobilization were established. The DNA-anchored piezoelectric sensor was in contact with MX solution. The time courses of the resonant frequency and the equivalent circuit characteristics of the sensor were also obtained during the study of DNA-drug binding. On the basis of the analysis of the multidimensional information provided by the PQCI technique the observed frequency decrease was mainly ascribed to the increase in the mass of the sensor surface resulting from the binding. The kinetics of the binding process were studied quantitatively by monitoring the frequency change with time and a piezoelectric response model for the binding was derived theoretically. The experimental data were fitted to the model and the binding and dissociation rate constants and the binding equilibrium constant were estimated to be 66.0+/-0.1 mol(-1) L s(-1), 1.4+/-0.1x10(-4) s(-1), and 4.71+/-0.07x10(5) mol(-1) L, respectively, at 37 degrees C.  相似文献
