Incubation of deuteriated precursors in cultures of Flavobacterium produced specifically deuteriated carotenoids. Analysis of these led to several conclusions: i) Lycopene is a direct precursor of β,β‐carotene. ii) Its terminal Me groups retain their integrity during cyclization: there is a stereospecific folding of the 1,5‐diene. The Me(16,16′) groups of lycopene become the Me(16,16′) of β,β‐carotene. Consequently, the folding must follow the C2(E,E) mode. iii) Incorporation of deuterium was sufficiently extensive to permit CD measurements on the isolated β,β‐carotene, allowing its centers of chirality to be assigned as (1S,1′S). iv) The same chirality resulted from incorporation of [2H3]mevalonate into zeaxanthin. The syntheses of specifically deuteriated [2H3]GPP, [2H3]FPP, and [2H3]GG are described.  相似文献

The known solid‐state structure (Fig. 1, top) of cyclo(β‐HAla)4 was used to model the structure of the title compound 1 as a prospective somatostatin mimic (Fig. 1, bottom). The synthesis started with the N‐protected natural amino acids Boc‐Phe‐OH, Boc‐Trp‐OH, Boc‐Lys(2‐Cl‐Z)‐OH, and Boc‐Thr(OBn)‐OH, which were homologated to the corresponding β‐amino‐acid derivatives (Scheme 1) and coupled to the β‐tetrapeptide Boc‐β‐HTrp‐β‐HPhe‐β‐HThr(OBn)‐β‐HLys(2‐Cl‐Z)‐OMe ( 16 ); the (N‐Me)‐β‐HThr‐(N‐Me)‐β‐HPhe analog 17 was also prepared. C‐ and N‐terminal deprotection and cyclization through the pentafluorophenyl ester gave the insoluble β‐tetrapeptide with protected Thr and Lys side chains ( 18 ). Solubilization and debenzylation could only be effected in LiCl‐containing THF (ca. 10% yield; with ca. 55% recovery). HPLC Purification provided a sample of the title compound 1 , the structure of which, as determined by NMR‐spectroscopy (Fig. 2, left) was drastically different from the `theoretical' model (Fig. 1). There is a transannular H‐bond dividing the macrocyclic 16‐membered ring, thus forming a ten‐ and a twelve‐membered H‐bonded ring, the former mimicking, or actually being superimposable on, an α‐peptidic so‐called β‐turn. Still, the four side chains occupy equatorial positions on the ring, as planned, albeit with somewhat different geometry as compared to the `original'. The cycloβ‐tetrapeptide has micromolar affinities to the human somatostatin receptors (hsst 1 – 5). Thus, we have demonstrated for the first time that it is possible to mimic a natural peptide hormone with a small β‐peptide. Furthermore, we have discovered a simple way to construct the ubiquitous β‐turn motif with β‐peptides (which are known to be stable to mammalian peptidases).  相似文献

The importance of β‐peptides lies in their ability to mimic the conformational behavior of α‐peptides, even with a much shorter chain length, and in their resistance to proteases. To investigate the effect of substitution of β‐peptides on their dominant fold, we have carried out a molecular‐dynamics (MD) simulation study of two tetrapeptides, Ac‐(2R,3S)‐β2,3hVal(αMe)‐(2S)‐β2hPhe‐(R)‐β3hLys‐(2R,3S)‐β2,3‐Ala(αMe)‐NH2, differing in the substitution at the Cα of Phe2 (pepF with F, and pepH with H). Three simulations, unrestrained (UNRES), using 3J‐coupling biasing with local elevation in combination with either instantaneous (INS) or time‐averaging (AVE) NOE distance restraining, were carried out for each peptide. In the unrestrained simulations, we find three (pepF) and two (pepH) NOE distance bound violations of maximally 0.22 nm that involve the terminal residues. The restrained simulations match both the NOE distance bounds and 3J‐values derived from experiment. The fluorinated peptide shows a slightly larger conformational variability than the non‐fluorinated one.  相似文献

The incorporation of the β‐amino acid residues into specific positions in the strands and β‐turn segments of peptide hairpins is being systematically explored. The presence of an additional torsion variable about the C(α) C(β) bond (θ) enhances the conformational repertoire in β‐residues. The conformational analysis of three designed peptide hairpins composed of α/β‐hybrid segments is described: Boc‐Leu‐Val‐Val‐DPro‐β Phe ‐Leu‐Val‐Val‐OMe ( 1 ), Boc‐Leu‐Val‐β Val ‐DPro‐Gly‐β Leu ‐Val‐Val‐OMe ( 2 ), and Boc‐Leu‐Val‐β Phe ‐Val‐DPro‐Gly‐Leu‐β Phe ‐Val‐Val‐OMe ( 3 ). 500‐MHz 1H‐NMR Analysis supports a preponderance of β‐hairpin conformation in solution for all three peptides, with critical cross‐strand NOEs providing evidence for the proposed structures. The crystal structure of peptide 2 reveals a β‐hairpin conformation with two β‐residues occupying facing, non‐H‐bonded positions in antiparallel β‐strands. Notably, βVal(3) adopts a gauche conformation about the C(α) C(β) bond (θ=+65°) without disturbing cross‐strand H‐bonding. The crystal structure of 2 , together with previously published crystal structures of peptides 3 and Boc‐β Phe ‐β Phe ‐DPro‐Gly‐β Phe ‐β Phe ‐OMe, provide an opportunity to visualize the packing of peptide sheets with local ‘polar segments' formed as a consequence of reversal peptide‐bond orientation. The available structural evidence for hairpins suggests that β‐residues can be accommodated into nucleating turn segments and into both the H‐bonding and non‐H‐bonding positions on the strands.  相似文献

13‐cis‐β,β‐Carotene, C40H56, crystallizes with a complete molecule in the asymmetric unit, whereas 15‐cis‐β,β‐carotene, also C40H56, has twofold symmetry about an axis through the central bond of the polyene chain. The polyene methyl groups are arranged on one side of the polyene chains for each molecule and the 6‐scisβ end groups, with the cyclohexene rings in half‐chair conformations, are twisted out of the planes of the polyene chains by angles ranging from 41.37 (17) to 52.2 (4)°. The molecules in each structure pack so that the arms of one occupy the cleft of the next, and there is significant π–π stacking of the almost‐parallel polyene chains of the 15‐cis isomer, which approach at distances of 3.319 (1)–3.591 (1) Å.  相似文献

Two polymorphs of 20‐desmethyl‐β‐carotene (systematic name: 20‐nor‐β,β‐carotene), C39H54, in monoclinic and triclinic space groups, were formed in the same vial by recrystallization from pyridine and water. Each polymorph crystallizes with the complete molecule as the asymmetric unit, and the two polymorphs show differing patterns of disorder. The β end rings of both polymorphs have the 6‐scis conformation, and are twisted out of the plane of the polyene chain by angles of −53.2 (8) and 47.3 (8)° for the monoclinic polymorph, and −43.6 (3) and 56.1 (3)° for the triclinic polymorph. The cyclohexene end groups are in the half‐chair conformation, but the triclinic polymorph shows disorder of one ring. Overlay of the molecules shows that they differ in the degree of nonplanarity of the polyene chains and the angles of twist of the end rings. The packing arrangements of the two polymorphs are quite different, with the monoclinic polymorph showing short intermolecular contacts of the disordered methyl groups with adjacent polyene chain atoms, and the triclinic polymorph showing π–π stacking interactions of the almost parallel polyene chains. The determination of the crystal structures of the two title polymorphs of 20‐desmethyl‐β‐carotene allows information to be gained regarding the structural effects on the polyene chain, as well as on the end groups, versus that of the parent compound β‐carotene. The absence of the methyl group is known to have an impact on various functions of the title compound.  相似文献

Oligomers of β‐substituted β‐amino acids (‘β3‐peptides') are known to adopt a helical secondary structure defined by 14‐membered ring hydrogen bonds ('14‐helix'). Here, we describe a deca‐β3‐peptide, 1 , that does not adopt the 14‐helical conformation and that may prefer an alternative secondary structure. β3‐Peptide 1 is composed exclusively of residues with side chains that are not branched adjacent to the β‐C‐atom (β3‐hLeu, β3‐hLys, and β3‐hTyr). In contrast, an analogous β‐peptide, 2 , containing β3‐hVal residues in place of the β3‐hLeu residues of 1 , adopts a 14‐helical conformation in MeOH, according to CD data. These results illustrate the importance of side‐chain branching in determining the conformational preferences of β3‐peptides.  相似文献

The chiral compounds (R)‐ and (S)‐1‐benzoyl‐2,3,5,6‐tetrahydro‐3‐methyl‐2‐(1‐methylethyl)pyrimidin‐4(1H)‐one ((R)‐ and (S)‐ 1 ), derived from (R)‐ and (S)‐asparagine, respectively, were used as convenient starting materials for the preparation of the enantiomerically pure α‐alkylated (alkyl=Me, Et, Bn) α,β‐diamino acids (R)‐ and (S)‐ 11 – 13 . The chiral lithium enolates of (R)‐ and (S)‐ 1 were first alkylated, and the resulting diasteroisomeric products 5 – 7 were aminated with ‘di(tert‐butyl) azodicarboxylate’ (DBAD), giving rise to the diastereoisomerically pure (≥98%) compounds 8 – 10 . The target compounds (R)‐ and (S)‐ 11 – 13 could then be obtained in good yields and high purities by a hydrolysis/hydrogenolysis/hydrolysis sequence.  相似文献

Summary: Host‐guest complexes of α‐cyclodextrin (α‐CD) and methylated β‐cyclodextrin (Me‐β‐CD) with diacrylates and dimethacrylates of butan‐1,4‐diol and hexan‐1,6‐diol at varying stoichiometries were studied. The complexes were analyzed by means of 1H NMR, two‐dimensional ROESY spectroscopy and Job's curves, which clearly revealed the discriminating influence of the two hosts towards complex formation. The corresponding polymers were obtained using a redox initiator system in water. Thermal analysis and IR measurements of the polymers provided evidence for the existence of a polyrotaxane architecture.

Oxidations of 5α‐hydroxy‐B‐norcholestan‐3β‐yl acetate ( 8 ) with Pb(OAc)4 under thermal or photolytic conditions or in the presence of iodine afforded only complex mixtures of compounds. However, the HgO/I2 version of the hypoiodite reaction gave as the primary products the stereoisomeric (Z)‐ and (E)‐1(10)‐unsaturated 5,10‐seco B‐nor‐derivatives 10 and 11 , and the stereoisomeric (5R,10R)‐ and (5S,10S)‐acetals 14 and 15 (Scheme 4). Further reaction of these compounds under conditions of their formation afforded, in addition, the A‐nor 1,5‐cyclization products 13 and 16 (from 10 ) and 12 (from 11 ) (see also Scheme 6) and the 6‐iodo‐5,6‐secolactones 17 and 19 (from 14 and 15 , resp.) and 4‐iodo‐4,5‐secolactone 18 (from 15 ) (see also Scheme 7). Oxidations of 5β‐hydroxy‐B‐norcholestan‐3β‐yl acetate ( 9 ) with both hypoiodite‐forming reagents (Pb(OAc)4/I2 and HgO/I2) proceeded similarly to the HgO/I2 reaction of the corresponding 5α‐hydroxy analogue 8 . Photolytic Pb(OAc)4 oxidation of 9 afforded, in addition to the (Z)‐ and (E)‐5,10‐seco 1(10)‐unsaturated ketones 10 and 11 , their isomeric 5,10‐seco 10(19)‐unsaturated ketone 22 , the acetal 5‐acetate 21 , and 5β,19‐epoxy derivative 23 (Scheme 9). Exceptionally, in the thermal Pb(OAc)4 oxidation of 9 , the 5,10‐seco ketones 10, 11 , and 22 were not formed, the only reaction being the stereoselective formation of the 5,10‐ethers with the β‐oriented epoxy bridge, i.e. the (10R)‐enol ether 20 and (5S,10R)‐acetal 5‐acetate 21 (Scheme 8). Possible mechanistic interpretations of the above transformations are discussed.  相似文献

N‐Acyl‐β2/β3‐dipeptide‐amide somatostatin analogs, 5 – 8 , with β2‐HTrp‐β3‐HLys ('natural' sequence) and β2‐HLys‐β3‐HTrp (retro‐sequence) have been synthesized (in solution). Depending on their relative configurations and on the nature of the terminal N‐acyl and terminal C‐amino group, the linear β‐dipeptide derivatives have affinities for the human receptor hsst 4, ranging from 250 to >10000 nanomolar (Fig. 3). Also, N‐Ac‐tetrapeptide amides 9 and 10 , which contain one α‐ and three β‐amino acid residues (NβαββC), have been prepared (solid‐phase synthesis), with the natural (Phe, Trp, Lys, Thr) and the retro‐sequence (Thr, Lys, Trp, Phe) of side chains and with two different configurations, each, of the two central amino acid residues. The novel ‘mixed', linear α/β‐peptides have affinities for the hsst 4 receptor ranging from 23 to >10000 nanomolar (Fig. 4), and, like ‘pure' β‐peptides, they are completely stable to a series of proteolytic enzymes. Thus, the peptidic turn of the cyclic tetradecapeptide somatostatin (Fig. 1) can be mimicked by simple linear di‐ and tetrapeptides. The tendency of β‐dipeptides for forming hydrogen‐bonded rings is confirmed by calculations at the B3LYP/6‐31G(d,p) level (Fig. 2). The reported results open new avenues for the design of low‐molecular‐weight peptidic drugs.  相似文献

The design and synthesis of β‐peptides from new C‐linked carbo‐β‐amino acids (β‐Caa) presented here, provides an opportunity to understand the impact of carbohydrate side chains on the formation and stability of helical structures. The β‐amino acids, Boc‐(S)‐β‐Caa(g)‐OMe 1 and Boc‐(R)‐β‐Caa(g)‐OMe 2 , having a D ‐galactopyranoside side chain were prepared from D ‐galactose. Similarly, the homo C‐linked carbo‐β‐amino acids (β‐hCaa); Boc‐(S)‐β‐hCaa(x)‐OMe 3 and Boc‐(R)‐β‐hCaa(x)‐OMe 4 , were prepared from D ‐glucose. The peptides derived from the above monomers were investigated by NMR, CD, and MD studies. The β‐peptides, especially the shorter ones obtained from the epimeric (at the amine stereocenter Cβ) 1 and 2 by the concept of alternating chirality, showed a much smaller propensity to form 10/12‐helices. This substantial destabilization of the helix could be attributed to the bulkier D ‐galactopyranoside side chain. Our efforts to prepare peptides with alternating 3 and 4 were unsuccessful. However, the β‐peptides derived from alternating geometrically heterochiral (at Cβ) 4 and Boc‐(R)‐β‐Caa(x)‐OMe 5 (D ‐xylose side chain) display robust right‐handed 10/12‐helices, while the mixed peptides with alternating 4 and Boc‐β‐hGly‐OMe 6 (β‐homoglycine), resulted in left‐handed β‐helices. These observations show a distinct influence of the side chains on helix formation as well as their stability.  相似文献

Synthesis of Diastereo- and Enantioselectively Deuterated β,ε-, β,β-, β,γ- and γ,γ-Carotenes We describe the synthesis of (1′R, 6′S)-[16′, 16′, 16′-2H3]-β, εcarotene, (1R, 1′R)-[16, 16, 16, 16′, 16′, 16′-2H6]-β, β-carotene, (1′R, 6′S)-[16′, 16′, 16′-2H3]-γ, γ-carotene and (1R, 1′R, 6S, 6′S)-[16, 16, 16, 16′, 16′, 16′-2H6]-γ, γ-carotene by a multistep degradation of (4R, 5S, 10S)-[18, 18, 18-2H3]-didehydroabietane to optically active deuterated β-, ε- and γ-C11-endgroups and subsequent building up according to schemes \documentclass{article}\pagestyle{empty}\begin{document}${\rm C}_{11} \to {\rm C}_{14}^{C_{\mathop {26}\limits_ \to }} \to {\rm C}_{40} $\end{document} and C11 → C14; C14+C12+C14→C40. NMR.- and chiroptical data allow the identification of the geminal methyl groups in all these compounds. The optical activity of all-(E)-[2H6]-β,β-carotene, which is solely due to the isotopically different substituent not directly attached to the chiral centres, is demonstrated by a significant CD.-effect at low temperature. Therefore, if an enzymatic cyclization of [17, 17, 17, 17′, 17′, 17′-2H6]lycopine can be achieved, the steric course of the cyclization step would be derivable from NMR.- and CD.-spectra with very small samples of the isolated cyclic carotenes. A general scheme for the possible course of the cyclization steps is presented.  相似文献

Coα‐(1H‐Imidazol‐1‐yl)‐Coβ‐methylcob(III)amide ( 4 ) was synthesized by methylation with methyl iodide of (1H‐imidazol‐1‐yl)cob(I)amide, obtained by electrochemical reduction of Coα‐(1H‐imidazol‐1‐yl)‐Coβ‐cyanocob(III)amide ( 5 ). The spectroscopic data and a single‐crystal X‐ray structure analysis indicated 4 to exhibit a base‐on constitution in solution and in the crystal. The crucial lengths of the axial Co−N and Co−CH3 bonds also emerged from the crystallographic data and were found to be smaller by 0.1 and 0.02 Å, respectively, than those in methylcob(III)alamin ( 2 ). The data of 4 support the view, that the `long' axial Co−N bonds as determined by X‐ray crystallography for the B12‐dependent methionine synthase, for methylmalonyl‐CoA mutase, and for glutamate mutase represent stretched Co−N bonds. The thermodynamic effect (the `trans influence') of the 1H‐imidazole base in 4 on the organometallic reactivity of this model for protein‐bound organometallic B12 cofactors was examined by studying Me‐group‐transfer equilibria in aqueous solution and using (5′,6′‐dimethyl‐1H‐benzimidazol‐1‐yl)cobamides (cobalamins) as reaction partners (Schemes 2 – 5, Table). In comparison with methylcob(III)alamin ( 2 ), 4 was found to be destabilized for an abstraction of the Co‐bound Me group by a CoIII electrophile. In contrast, the abstraction of the Co‐bound Me group by a radical(oid) CoII species was not significantly influenced thermodynamically by the exchange of the nucleotide base. Likewise, exploratory Me‐group‐transfer experiments with Me−CoIII and nucleophilic CoI corrinoids at pH 6.8 provided an apparent equilibrium constant near unity. However, this finding also was consistent with partial protonation of the imidazolylcob(I)amide at pH 6.8, suggesting an interesting pH dependence of the Megroup‐transfer equilibrium near neutral pH. Therefore, the replacement of the 5′,6′‐dimethyl‐1H‐benzimidazole base by an 1H‐imidazole moiety, as observed in methyl transferases and in C‐skeleton mutases, does not by itself strongly alter the inherent reactivity of the B12 cofactors in the crucial homolytic and nucleophilic‐heterolytic reactions involving the organometallic bond, but may help to enhance the control of the organometallic reactivity by protonation/deprotonation of the axial base.  相似文献

3′‐Epilutein (=(all‐E,3R,3′S,6′R)‐4′,5′‐didehydro‐5′,6′‐dihydro‐β,β‐carotene‐3,3′‐diol; 1 ), isolated from the flowers of Caltha palustris, was submitted to both thermal isomerization and I2‐catalyzed photoisomerization. The structures of the main products (9Z)‐ 1 , (9′Z)‐ 1 , (13Z)‐ 1 , (13′Z)‐ 1 , (15Z)‐ 1 , and (9Z,9′Z)‐ 1 were determined based on UV/VIS, CD, 1H‐NMR, and MS data.  相似文献

Inexpensive acryloyl chloride was converted in 91% overall yield to two derivatives of β‐alanine, (R,R,R)‐ 6 and (R,R,S)‐ 6 , containing two chiral auxiliaries. C‐Alkylation of (R,R,R)‐ and (R,R,S)‐ 6 via a dianion derivative, was performed by direct metallation with 2.2 equiv. of lithium hexamethyldisilazane (LHMDS) in THF at ?78°. C‐Alkylation of (R,R,S)‐ 6 ‐Li2 (‘matched' pair of chiral auxiliaries) afforded the mono‐alkylated products 8 – 11 in 29–96% yield and 54–95% stereoselectivity. Employment of LiCl as an additive generally increased stereoselectivities, whereas the effect of HMPA as a cosolvent was erratic. Chemical correlation of the major diastereoisomer from the alkylation reactions with (S)‐α‐alkyl‐β‐alanine ( 12 – 15 ) showed that addition of the electrophile preferentially takes place on the enolate's Si‐face. This conclusion is also supported by molecular‐modeling studies (ab initio HF/3‐21G), which indicate that the lowest‐energy conformation for (R,R,S)‐ 6 ‐Li2 presents the more sterically hindered Re‐face of the enolate. The theoretical studies also predict a determining role for N? Li? O chelation in (R,R,S)‐ 6 ‐Li2, giving rise to an interesting ‘ion‐triplet' configuration for the dilithium dianion.  相似文献

Annonalide (3β,20‐epoxy‐3α,16‐dihydroxy‐15‐oxo‐7‐pimaren‐19,6β‐olide, C20H26O6, 1 ) is the major (9βH)‐pimarane diterpene isolated from tubers of Cassimirella ampla, and it exhibits cytotoxic properties upon interaction with ctDNA. We have prepared new derivatives of 1 by modification of the (9βH)‐pimarane backbone and report here the semisynthesis and absolute configuration of a novel rearranged 19,20‐δ‐lactone (9βH)‐pimarane. Our approach was the reduction of the carbonyl groups of 1 with sodium borohydride, at positions C15 (no stereoselectivity) and C3 (stereoselective reduction), followed by rearrangement of the 6,19‐γ‐lactone ring into the six‐membered 19,20‐δ‐lactone ring in 4a (3β,6β,16‐trihydroxy‐7‐pimaren‐19,20β‐olide monohydrate, C20H30O6·H2O). The absolute structure of the new compound, 4a , was determined unambiguously with a Flack parameter x of −0.01 (11), supporting the stereochemistry assignment of 1 redetermined here. Besides the changes in the pattern of covalent bonds caused by reduction and lactone rearrangement, the conformation of one of the three fused cyclohexane rings is profoundly different in 4a , adopting a chair conformation instead of the boat shape found in 1 . Furthermore, the intramolecular hydrogen bond present in 1 is lost in new compound 4a , due to hydrogen bonding between the 3‐OH group and the solvent water molecule.  相似文献

The synthesis of a novel series of twelve 4‐(trihalomethyl)dipyrimidin‐2‐ylamines, from the cyclo‐condensation reaction of 4‐(trichloromethyl)‐2‐guanidinopyrimidine, with β‐alkoxyvinyl trihalomethyl ketones, of general formula: X3C‐C(O)‐C(R2)=C(R1)‐OR, where: X = F, Cl; R = Me, Et, ‐(CH2)2‐, ‐(CH2)3‐; R1 = H, Me; R2 = H, Me, ‐(CH2)2‐, ‐(CH2)3‐, is reported. The reactions were carried out in acetonitrile under reflux for 16 hours, leading to the dipyrimidin‐2‐ylamines in 65‐90% yield. Depending on the substituents of the vinyl ketone, tetrahydropyrimidines or aromatic pyrimidine rings were obtained from the cyclization reaction. When X = Cl, elimination of the trichloromethyl group was observed during the cyclization step. The structure of 4‐(trihalomethyl)dipyrimidin‐2‐ylamines was studied in detail by 1H‐, 13C‐ and 2D‐nmr spectroscopy.  相似文献

The conversion of 2α,3α‐dioxy‐substituted phyllocladane derivatives into the corresponding 3‐ketone proceeds in an unexpected manner: Depending on the reaction conditions, the corresponding 3β‐hydroxy‐substituted compound is formed almost quantitatively, or the desired ketone can be isolated directly (see preceding paper). The reaction mechanism is now disclosed to be a stereospecific C(3)→C(2)‐hydride shift by investigating the reactions of the synthesized (±)‐trans‐decalin‐type (trans‐1,5,5‐trimethylbicyclo[4.4.0]decanes) and (±)‐podocarpane‐type (trans‐1,2,3,4,4a,9,10,10a‐octahydro‐1,1,4a‐trimethylphenanthrenes) model compounds 25 and 35 and of their D‐labeled isomers 25′ and 35′ (Scheme 6). The latter afforded the corresponding 3β‐hydroxy (2β‐D)‐derivatives 38 and 39 as well as the (2β‐D)‐3‐ketones of the general type 5b′ (e.g., 36′ ), thus evidencing a suprafacial (C3)→C(2)‐deuteride shift. This reaction mechanism seems to be a general feature of such 3α,4α‐dioxy‐substituted 1,5,5‐trimethylbicyclo[4.4.0]decane congeners.  相似文献
