In this study, we propose a simple, cost-effective, and sensitive high-performance liquid chromatography with fluorescence detection (HPLC-FLD) for the simultaneous determination of seven bisphenols (bisphenol F (BPF), bisphenol E (BPE), bisphenol B (BPB), BADGE (bisphenol A diglycidyl ether), BADGE∙2H2O, BADGE∙H2O, BADGE∙2HCl) in human breast milk samples. The dispersive solid phase extraction (d-SPE) coupled with solid phase extraction (SPE) procedure performed well for the majority of the analytes with recoveries in the range 57–88% and relative standard deviations (RSD%) of less than 9.4%. During the d-SPE stage, no significant matrix effect was observed thanks to the application of different pairs of salts such as zirconium-dioxide-based sorbents (Z-Sep or Z-Sep +) and primary secondary amine (PSA) or QuEChERS Enhanced Matrix Removal-Lipid (EMR-Lipid) and PSA. The method limits of quantification (mLOQs) for all investigated analytes were set at satisfactory low values in the range 171.89–235.11 ng mL−1. Analyte concentrations were determined as the average value from human breast milk matrix samples. The results show that the d-SPE/SPE procedure, especially with the application of EMR-Lipid and PSA, could be used for further bisphenol analyses in human breast milk samples.  相似文献

建立了含油脂食品中17种邻苯二甲酸酯的分散固相萃取-气相色谱-质谱法检测方法。奶茶样品经乙腈-甲基叔丁基醚(9∶1,V/V)提取后,提取液用MAS-PAEC分散固相萃取管进行净化。调味包样品经乙腈(正己烷饱和)-甲基叔丁基醚(19∶1,V/V)提取2次后,提取液用CNW分散固相萃取管进行净化。采用基质匹配标准外标法进行定量分析。结果表明,奶茶中17种邻苯二甲酸酯的加标回收率为82.2%~125.4%;相对标准偏差小于16.5%;方法检出限为100~200μg/L。调味包中17种邻苯二甲酸酯的加标回收率为70.9%~115.5%;相对标准偏差小于9.8%;方法检出限为400~800μg/L。本方法快速、精确、简易、廉价、稳定,可应用于含油脂食品中17种邻苯二甲酸酯的实际检测分析。  相似文献

ABSTRACT

A study for the quantification method of neonicotinoid pesticides (Neos) in human urine that aims to develop the certified reference material (CRM) in the future was carried out. A modified Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) method based on dispersive solid phase extraction (d-SPE) with isotope dilution mass spectrometry (IDMS) was evaluated for the quantification of five Neos (acetamiprid, dinotefuran, imidacloprid, thiacloprid and thiamethoxam) in human urine control by optimisation of the extraction solvent, and the type and amount of d-SPE material. Two types of human urine controls (sample A and B) containing 5 μg/kg and/or 25 μg/kg of Neos were prepared and employed for this study. The results of spike test using sample A (2 mL) obtained by modified QuEChERS method with acetone extraction (10 mL) and d-SPE using a strong cation exchanger (silica gel modified with benzenesulphonic acid phase, SCX) 500 mg were as follows: 97.8%–103.1% for 5 μg/kg and 97.2%–102.7% for 25 μg/kg (described as percentage by the quantification results of IDMS relative to the spiked amount of pesticides). These results were comparable to those by SPE cartridge method using SCX functionalised hydrophilic styrene divinylbenzene (PCX). The repeatabilities of the analyses, represented as relative standard deviations, were also comparable for spike level 5 μg/kg and 25 μg/kg of Neos: 0.1%–7.2% for modified QuEChERS with IDMS and 0.2%–5.6% for PCX cartridge method. The results of spike test using sample B for a spike level 5 μg/kg were 96.0%–100.6%. These results indicate that modified QuEChERS method with IDMS can be used for the development of certified reference material and has the potential to use for the quantification of Neos in real human urine.  相似文献

A sensitive and reliable method was developed to determine methylene blue (MB) and its metabolite residues, including azure A (AZA), azure B (AZB), and azure C (AZC) in aquatic products by HPLC–MS/MS. The samples were extracted by acetonitrile and cleaned up by alumina-neutral (ALN) cartridges. The analytes were separated on a Sunfire C18 column (150 mm × 2.1 mm, 5 µm). The method was validated according to the European criteria of Commission Decision 2002/657/CE. Good linearity between 1–500 µg/L was obtained with correlation coefficients (R2) greater than 0.99. The limit of quantification (LOQ) was 1.0 µg/kg. The average recoveries at three levels of each compound (1, 5, and 10 µg/kg) were demonstrated to be in the range of 71.8–97.5%, with relative standard deviations (RSDs) from 1.05% to 8.63%. This method was suitable for the detection of methylene blue and its metabolite residues in aquatic products.  相似文献

A simple and rapid multiplug filtration cleanup method based on multiwalled carbon nanotubes was developed to determine 124 pesticide residues in rice, wheat, and corn, which could be done in a few seconds without conditioning and elution steps. Various combinations of sorbents were optimized for each matrix with a dispersive solid‐phase extraction procedure to get a satisfactory recovery and clean‐up performance. Good linearity was obtained for all pesticides with calibration curve coefficients larger than 0.9958. Most recoveries for the majority pesticides were between 70 and 120% (n = 5) with relative standard deviations below 20%. The limit of detection was 0.1–1.3 μg/kg, and the limit of quantification was 0.2–4.3 μg/kg for the pesticides in all matrices. The work suggests that the multiplug filtration cleanup method is better than the dispersive solid‐phase extraction method and it could be applied to routinely monitor pesticide residues in market samples.  相似文献

采用改进的QuEChERS(quick, easy, cheap, effective, rugged and safe)前处理法,结合气相色谱-串联质谱(GC-MS/MS)技术建立了检测烟草中49种农药残留的分析方法。样品用含0.1%乙酸的乙腈溶液提取,提取液被氮吹至干后,残渣用乙腈-乙酸乙酯(1:1, v/v)溶液溶解,溶解液经N-丙基乙二胺(PSA)吸附剂、无水MgSO4、C18吸附剂净化后,直接进行GC-MS/MS测定,内标法定量。实验结果表明,49种农药在低质量浓度(0.05 μg/L)的加标水平下的平均加标回收率为60.4%~104.8%,高质量浓度(5 μg/L)的平均加标回收率为70%~115%,相对标准偏差均小于15%;其中16种农药的方法检出限(LOD)分别为0.01~0.03 μg/kg,其余33种农药的LOD均小于0.01 μg/kg;相关系数都大于或等于0.991。该方法样品前处理简单、分析时间短、灵敏度和精密度均符合农药多残留痕量检测技术的要求,适用于烟草中多种农药残留的检测。  相似文献

建立了一种大豆和玉米中20种农药残留量的分散固相萃取气相色谱-负化学离子源质谱分析方法。样品经乙腈提取并浓缩后加入N-丙基乙二胺(PSA)、石墨化碳黑和C18 3种填料进行分散固相萃取净化,气相色谱-负化学离子源质谱分时段选择离子监测技术测定与确证,外标法定量。所有农药在20~400 μg/L范围内线性均良好;方法的定量限(LOQ)均不高于2 μg/kg;在5,10和20 μg/kg 3个添加水平下所有农药的平均回收率均处于70%~130%之间,相对标准偏差(RSD)低于17%;运用该方法检测大豆和玉米样品时没有干扰现象。  相似文献

Nanosheets of graphitic carbon nitride (GCN) were used as the dispersive solid-phase extraction (d-SPE) sorbent for the first time. GCN successfully purified complex matrices of soil, wheat, and wheat straw utilising a simple QuEChERS (quick, easy, cheap, effective, rugged, and safe) method with liquid chromatography tandem mass spectrometry. Pesticides recoveries in the range of 80–110%, small matrix effect, and decreased amounts guaranteed and distinguished this new sorbent. The application of a real formulation of pesticides under field conditions during 2015 in the wheat ecosystem illustrated the clean-up effect and application potential of GCN. It is a valuable and potential substitute for C18 according to the results comparison between two sorbents, including recoveries, matrix-matched calibration, and dissipation kinetics of pesticides. The new use of GCN also enriched the knowledge of d-SPE and sample preparation furthermore.  相似文献

A new method of sample preparation was developed and is reported for the first time. The approach combines in-vial filtration with dispersive solid-phase extraction (d-SPE) in a fast and convenient cleanup of QuEChERS (quick, easy, cheap, effective, rugged, and safe) extracts. The method was applied to simultaneous analysis of 42 diverse pesticides and 17 environmental contaminants, including polycyclic aromatic hydrocarbons, polychlorinated biphenyls (PCBs), and flame retardants, in shrimp as the sample matrix. Final extracts were analyzed by both low-pressure gas chromatography – triple quadrupole tandem mass spectrometry (LPGC-MS/MS), and high-performance liquid chromatography – triple quadrupole tandem mass spectrometry (HPLC-MS/MS) to provide a wide scope of analysis for targeted analytes. During method development, several different commercial sorbents for d-SPE were investigated and compared with respect to analyte recoveries. The method was validated at 10, 50, and 100 ng g−1 spiking levels (10-fold lower for PCBs), and the results for nearly all analytes were between 70 and 115% recoveries with ≤17% relative standard deviations. The method was shown to be simple, fast, and effective for multi-application analysis of chemical residues in the representative food and environmental marker matrix.  相似文献

Tenebrio molitor larvae (mealworm) is an edible insect and is considered a future food. Using liquid chromatography-tandem mass spectrometry (LC-MS/MS), a novel method for simultaneous analysis of 353 target analytes was developed and validated. Various sample preparation steps including “quick, easy, cheap, effective, rugged, and safe” (QuEChERS) extraction conditions, number of acetonitrile-hexane partitions, and dispersive-solid phase extraction (dSPE) sorbents were compared, and the optimal conditions were determined. In the established method, 5 g of homogenized mealworms was extracted with acetonitrile and treated with QuEChERS EN 15662 salts. The crude extract was subjected to three rounds of acetonitrile-hexane partitioning, and the acetonitrile layer was cleaned with C18 dSPE. The final solution was matrix-matched and injected into LC-MS/MS (2 μL). For target analytes, the limits of quantitation (LOQs) were ≤10 μg/kg, and the correlation coefficient (r2) of calibration was >0.990. In recovery tests, more than 90% of the pesticides showed an excellent recovery range (70–120%) with relative standard deviation (RSD) ≤20%. For more than 94% of pesticides, a negligible matrix effect (within ±20%) was observed. The analytical method was successfully applied and used for the detection of three urea pesticides in 4 of 11 mealworm samples.  相似文献

An innovative, rapid and stable method for simultaneous determination of three tetracycline (oxytetracycline, tetracycline and doxycycline) and two fluoroquinolone (ciprofloxacin and enrofloxacin) residues in poultry eggs by ultra-high performance liquid chromatography–fluorescence detection (UPLC-FLD) was established and optimized. The samples were homogenized and extracted with acetonitrile/ultrapure water (90:10, v/v) and then purified by solid-phase extraction (SPE). LC separation was achieved on an ACQUITY UPLC BEH C18 column (1.7 µm, 2.1 mm × 100 mm), and the mobile phase was composed of acetonitrile and a 0.1 mol/L malonic acid solution containing 50 mmol/L magnesium chloride (the pH was adjusted to 5.5 with ammonia). When the five target drugs were spiked at the limit of quantification, 0.5 times the maximum residue limit (MRL), 1.0 MRL and 2.0 MRL, the recoveries were above 83.5% and the precision ranged from 1.99% to 6.24%. These figures of merit complied with the parameter validation regulations of the EU and U.S. FDA. The limits of detection and quantifications of the targets were 0.1–13.4 µg/kg and 0.3–40.1 µg/kg, respectively. The proposed method was easily extended to quantitative analyses of target drug residues in 85 egg samples, thus demonstrating its reliability and applicability.  相似文献

Sludge generated after washing of fruits and vegetables during agro-food processes is a complex matrix and selective methods for the identification and quantification of pesticides’ residues are necessary in order to achieve a sustainable and effective management of the total sewage. The present work describes the development and validation of a reliable, simple and fast analytical method based on liquid chromatography-tandem mass spectrometry (LC-MS/MS) for the determination of 47 pesticides of different chemical classes, including organosphosphates, pyrethroids, neonicotinoids, triazoles and others, in sludge samples after QuEChERS sample preparation. The necessity of the individual steps of QuEChERS was investigated and the LC-ESI-MS/MS conditions were optimized to achieve maximum sensitivity of the target analytes. The method limits of detection (LODs) ranged between 0.0005 mg/kg (imidacloprid) and 0.05 mg/kg (beta cyfluthrin). The recoveries ranged between 71–120% and the repeatability of the method was ≤25% expressed as relative standard deviation. The method was applied to sludge samples generated after washing of fruits in an agro-fruit-packaging unit in Greece. The results showed the presence of 37 pesticides’ active substances with concentrations ranging from low ppbs, such as fludioxinil (5 μg/kg) up to low ppms such as beta cyfluthrin (3.5 mg/kg) and with their sum concentration reaching up to 19 mg/kg.  相似文献

建立了QuEChERS-液相色谱-串联质谱同时测定棕榈原油中敌草快、乐果、倍硫磷、倍硫磷亚砜、倍硫磷砜、氟吡甲禾灵残留量的方法。以回收率及基质效应为考察指标,评估了乙二胺-N-丙基硅烷(PSA)/C18混合物、C18键合锆胶(Z-Sep+)/C18混合物、Z-Sep+、C18 4种分散吸附剂的净化效果,结果表明Z-Sep+/C18的净化效果最佳。样品采用含1%(v/v)乙酸的甲醇溶液提取,Z-Sep+/C18分散固相萃取净化;液相色谱以Atlantis T3色谱柱梯度洗脱分离,质谱分析采用电喷雾正离子电离、多反应监测模式,以基质匹配校准曲线外标法进行定量。6种农药在3个添加水平(10、20、250 μg/kg)下的回收率范围为70.0%~97.9%,相对标准偏差范围为2.6%~10.2%; 6种农药的定量限(S/N≥10)为0.1~2.5 μg/kg。该方法简便、灵敏、准确、环保,适用于棕榈原油中上述6种农药残留的定性、定量分析。  相似文献

建立了基于QuEChERS方法的液相色谱-串联质谱同时测定甜玉米及毛豆中32种磺酰脲类除草剂残留的方法。以提取液净化前后蒸发残渣质量及回收率为考察指标,评估了C18、氧化锆包覆硅胶(Z-Sep)/C18混合物、C18键合锆胶(Z-Sep+) 3种分散吸附剂对甜玉米的净化效果,结果Z-Sep/C18吸附剂的净化效果最佳。样品采用乙腈提取、无水硫酸镁及氯化钠盐析、Z-Sep/C18分散固相萃取净化;液相色谱以CSH C18色谱柱梯度洗脱分离,质谱分析采用电喷雾负离子电离、按时间表的多反应监测模式(Scheduled MRM),以基质匹配校准曲线外标法进行定量。32种农药在甜玉米及毛豆中3个添加水平(10、20、100 μg/kg)的回收率为80.0%~108.2%,相对标准偏差为1.2%~13.0%, 32种农药的定量限(S/N≥10)为0.2~5.0 μg/kg。方法简便、灵敏、环保,适用于甜玉米及毛豆中32种磺酰类除草剂残留的定性定量分析。  相似文献

In this work, for the first time, Ultra-High-Performance Liquid Chromatography–Tandem Mass Spectrometry (UHPLC–MS/MS) method was developed for qualitative and quantitative analysis of veterinary antibiotics (cephalosporins, diaminopyrimidines, fluoro(quinolones), lincosamides, macrolides, penicillins, pleuromutilins, sulfonamides, tetracyclines, and sulfones) in hen eggshells. The sample preparation method is based on a liquid–liquid extraction with a mixture of metaphosphoric acid, ascorbic acid, EDTA disodium salt dihydrate, and acetonitrile. The chromatographic separation was performed on Luna® Omega Polar C18 10 column in gradient elution mode and quantitated in an 8 min run. Validation such as linearity, selectivity, precision, recovery, matrix effect, limit of quantification (LOQ), and limit of detection (LOD) was found to be within the acceptance criteria of the validation guidelines of the Commission Decision 2002/657/EC and EUR 28099 EN. Average recoveries ranged from 81–120%. The calculated LOQ values ranged from 1 to 10 µg/kg, the LOD values ranged from 0.3 to 4.0 µg/kg, depending on analyte. The developed method has been successfully applied to the determination of antibacterial compounds in hen eggshell samples obtained from different sources. The results revealed that enrofloxacin, lincomycin, doxycycline, and oxytetracycline were detected in hen eggshell samples.  相似文献

The detection and quantification of nut allergens remains a major challenge. The liquid chroma-tography tandem mass spectrometry (LC-MS/MS) is emerging as one of the most widely used methods, but sample preparation prior to the analysis is still a key issue. The objective of this work was to establish optimized protocols for extraction, tryptic digestion and LC-MS analysis of almond, cashew, hazelnut, peanut, pistachio and walnut samples. Ammonium bicar-bonate/urea extraction (Ambi/urea), SDS buffer extraction (SDS), polyvinylpolypyrroli-done (PVPP) extraction, trichloroacetic acid/acetone extraction (TCA/acetone) and chloro-form/methanol/sodium chloride precipitation (CM/NaCl) as well as the performances of con-ventional tryptic digestion and microwave-assisted breakdown were investigated. Overall, the protein extraction yields ranged from 14.9 ± 0.5 (almond extract from CM/NaCl) to 76.5 ± 1.3% (hazelnut extract from Ambi/urea). Electrophoretic profiling showed that the SDS extraction method clearly presented a high amount of extracted proteins in the range of 0–15 kDa, 15–35 kDa, 35–70 kDa and 70–250 kDa compared to the other methods. The linearity of the LC-MS methods in the range of 0 to 0.4 µg equivalent defatted nut flour was assessed and recovery of internal standards GWGG and DPLNV(d8)LKPR ranged from 80 to 120%. The identified bi-omarkers peptides were used to relatively quantifier selected allergenic protein form the inves-tigated nut samples. Considering the overall results, it can be concluded that SDS buffer allows a better protein extraction from almond, peanut and walnut samples while PVPP buffer is more appropriate for cashew, pistachio and hazelnut samples. It was also found that conventional overnight digestion is indicated for cashew, pistachio and hazelnut samples, while microwave assisted tryptic digestion is recommended for almond, hazelnut and peanut extracts.  相似文献

A high‐throughput, rapid, and efficient modified QuEChERS (quick, easy, cheap, effective, rugged, and safe) method with a simple cleanup procedure has been developed for simultaneously determining 227 pesticides in pepper samples by liquid chromatography with tandem mass spectrometry (running time: 10 min). Pesticide residues were extracted/partitioned with an acetonitrile/DisQuE QuEChERS pouch, and the resulting samples were cleaned up with different methods: dispersive solid‐phase extraction with primary secondary amines or multiwalled carbon nanotubes and graphitized carbon solid mini cartridge column. The results indicated that multiwalled carbon nanotubes dispersive sorbents achieved the best recoveries and had less matrix interference. The numbers of pesticides with a recovery in the range of 70–120% were 199 at a spiked level of 40 μg/kg. The correlation coefficients (r2) for 227 pesticides were above 0.99, while the limits of quantitation of pesticides in pepper samples ranged from 0.13 to 13.51 μg/kg (S/N = 10), and the limits of detection ranged from 0.04 to 4.05 μg/kg (S/N = 3). The relative standard deviations of approximately 197 pesticides were below 20% at spiked levels of 40 μg/kg. Based on these results, the proposed method was chosen as the most suitable cleanup procedure for the determination of multiresidue pesticides in pepper samples.  相似文献

An improved analytical method was developed for the simultaneous quantification of several plant growth regulators and fungicides (carbendazim, pyrimethanil, metalaxyl, triadimefon, paclobutrazol, thiophanate, prochloraz, dimethomorph, difenoconazole, (4‐chlorophenoxy)‐acetic acid, (2,4‐dichlorophenoxy)‐acetic acid, thiadiazuron, forchlorfenuron and gibberellins) in fruits followed by ultra high performance liquid chromatography with tandem mass spectrometry. Samples were extracted and purified using a modified QuEChERS method. Different extraction solvents and sorbents in the QuEChERS method were compared. Optimum results were followed by the addition of 1% acetic acid in acetonitrile; C18 sorbent was added due to the acidic nature of several pesticides. The recoveries of the pesticides were in the range 73.7–118.4%, with relative standard deviations lower than 16.63%. Limits of detection ranged from 0.1–1.0 μg/kg. The method presented here is simple, rapid, sensitive and can be applied to large‐scale monitoring programs to screen the presences of pesticides in fruits.  相似文献

A novel method for detecting pesticide multi-residue in grass forage (alfalfa and oat) was established based on the one-step automatic extraction and purification technology of quick, easy, cheap, effective, rugged, and safe combined with ultrahigh-performance liquid chromatography quadrupole Orbitrap high-resolution mass spectrometry. The crushed sample was extracted with acetonitrile with 1% acetate, followed by a cleanup step with a primary-secondary amine, octadecylsilane, and graphitized carbon black. The extraction and purification were carried out using the one-step automatic pretreatment equipment. The target pesticides were acquired in positive ion electrospray ionization mode and full scan/data dependent secondary scan mode. The calibration curve shows good linearity over the corresponding concentration range, with the coefficient of determination greater than 0.99. The screening detection limits were 0.5–50 μg/kg, and the limit of quantification for the 206 pesticides was set at 1–50 μg/kg. At the spiking levels of one, two, and 10 times of limit of quantification, more than 95% of pesticides had recovery between 70–120%, with a relative standard deviation ≤20%. The method was proved to be simple, rapid, high-sensitivity, and could be routinely used for the high throughput screening and quantitative analysis of pesticide residues in alfalfa and oat.  相似文献
