Positively charged α‐chymotrypsin (ChT) formed a 1:1 complex with negatively charged 5,10,15,20‐tetrakis(4‐sulfonatophenyl)porphyrinato iron(III) (FeTPPS) in phosphate buffer at pH 7.4 through electrostatic interaction. In spite of the large binding constant (K=4.8×105 M ?1), FeTPPS could not completely inhibit the catalysis of ChT in the hydrolysis of the model substrate, N‐succinyl‐L ‐phenylalanine p‐nitroanilide (SPNA). The degree of inhibition (60 %) was saturated at 1.6 equivalents of FeTPPS, which indicates that covering of the active site of ChT by FeTPPS was insufficient. The enzymatic activity lowered by FeTPPS was entirely recovered for the freshly prepared sample when the porphyrin on the protein surface was detached by per‐O‐methylated β‐cyclodextrin (TMe‐β‐CD), which formed a stable 1:2 inclusion complex with FeTPPS (K1=1.26×106 M ?1, K2=6.3×104 M ?1). FeTPPS gradually induced irreversible denaturation of ChT, and the denatured ChT further lost its catalytic ability. No repairing effect of TMe‐β‐CD was observed with irreversibly denatured ChT. A new reversible inhibitor, 5,10,15,20‐tetrakis[4‐(3,5‐dicarboxyphenylmethoxy)phenyl]porphyrinato iron(III) (FeP8M), was then designed, and its inhibitory behavior was examined. FeP8M formed very stable 1:1 and 1:2 FeP8M/ChT complexes with ChT, the K1 and K2 values being 2.0×108 and 1.0×106 M ?1, respectively. FeP8M effectively inhibited the ChT‐catalyzed hydrolysis of SPNA (maximum degree of inhibition=85 %), and the activity of ChT was recovered by per‐O‐methylated γ‐cyclodextrin. No irreversible denaturation of ChT occurred upon binding with FeP8M. The kinetic data support the observation that, for nonincubated samples, both inhibitors did not cause significant conformational change in ChT and inhibited the ChT activity by covering the active site of the enzyme. 相似文献

As easy as 1, 2, 3 : A palladium‐catalyzed three‐component coupling generates α,β‐unsaturated γ‐amino acids in a single step (see scheme). The reaction is believed to involve migration of a vinyl substituent to a highly electrophilic palladium carbene. Unlike previous synthetic approaches, this synthesis provides access to γ‐amino acids with non‐natural side chains.

The α‐chymotrypsin activity was tested in aqueous media with the presence of novel cationic amine–based gemini surfactant, with different spacer chain lengths and head group size, and also compared with the cationic cetyltrimethylammonium bromide (CTAB) and cetyltriphenylphosphonium bromide (CTPB) surfactants and aqueous buffer only. The p‐nitrophenyl acetate (PNPA) hydrolysis rate was monitored in the presence of the surfactant concentration at 30°C. Most of these gemini surfactants gave higher catalytic activity as compared to cationic CTAB and CTPB. The highest superactivity was measured in the presence of gemini 16‐12‐16, [dodecanediyl‐1,12‐bis(cetyldimethylammonium bromide)] surfactant at pH 7.5. The catalytic reaction follows the Michaelis–Menten mechanism. The catalytic rate constants, kcat, show the same profile that the catalytic affinity; KM being enhanced with increasing space chain length. The results are favorable for considering that the amine‐based gemini surfactant influences more than both the aqueous and cationic micellar media. 相似文献

A concise synthesis of α‐benzylidene‐γ‐methyl‐γ‐butyrolactones 5a – g from substituted benzaldehydes is described. Compounds 1a – g on reaction with phosphorane 2 , provide the pentenoates 3a – g , which can be hydrolyzed to the acids 4a – g . The latter are cyclized to the corresponding butyrolactones 5a – g in excellent yields. The pentenoates 3a – g , on acid catalyzed cyclization, also provide 5a – g in very high yields. 相似文献

Fully stereodivergent dual‐catalytic α‐allylation of protected α‐amino‐ and α‐hydroxyacetaldehydes is achieved through iridium‐ and amine‐catalyzed substitution of racemic allylic alcohols with chiral enamines generated in situ. The operationally simple method furnishes useful aldehyde building blocks in good yields, more than 99 % ee, and with d.r. values greater than 20:1 in some cases. Additionally, the γ,δ‐unsaturated products can be further functionalized in a stereodivergent fashion with high selectivity and with preservation of stereochemical integrity at the Cα position. 相似文献

The two double‐bond isomers 3‐iodo‐2,6,6‐trimethylbicyclo[3.1.1]hept‐2‐ene ( 6b ) and 3‐iodo‐4,6,6‐trimethylbicyclo[3.1.1]hept‐2‐ene ( 11 ) were synthesized by reacting 2,6,6‐trimethylbicyclo[3.1.1]heptan‐3‐one ( 9 ) with hydrazine, followed by treatment with I2 in the presence of Et3N. Treatment of 11 with t‐BuOK as base in diglyme at 220° resulted in the formation of 9 and 6,6‐dimethyl‐4‐methylidenebicyclo[3.1.1]hept‐2‐ene ( 12 ). For the formation of 9 , the cyclic allene 7 is proposed as an intermediate. Treatment of the second isomer, 6b , with t‐BuOK at 170° gave rise to the diene 12 and the dimerization product 17 . The underlying mechanism of this transformation is discussed. On the basis of density‐functional‐theory (DFT) calculations on the allene 7 and the alkyne 15 , the formation of the latter as the intermediate was excluded. 相似文献

The title compounds, both C23H34O5, are the 5α and 5β configurations of two diacetate epimers. The 5β‐diacetate crystallizes in an hexagonal structure, unusual for steroid molecules. The unit cell has an accessible solvent volume of 358 Å3, responsible for clathrate behaviour. The 5β‐epimer also features some shorter than average bond lengths in the 3α,4β‐acetoxy groups. The conformations of the molecules of both epimers are compared with those obtained through abinitio quantum chemistry calculations. Cohesion of the crystals can be attributed to van der Waals and weak molecular C—H⋯O interactions. 相似文献

Engineering biomaterials with integrin‐binding activity is a very powerful approach to promote cell adhesion, modulate cell behavior, and induce specific biological responses at the surface level. The aim of this Review is to illustrate the evolution of surface‐coating molecules in this field: from peptides and proteins with relatively low integrin‐binding activity and receptor selectivity to highly active and selective peptidomimetic ligands. In particular, we will bring into focus the difficult challenge of achieving selectivity between the two closely related integrin subtypes αvβ3 and α5β1. The functionalization of surfaces with such peptidomimetics opens the way for a new generation of highly specific cell‐instructive surfaces to dissect the biological role of integrin subtypes and for application in tissue engineering and regenerative medicine. 相似文献

SF6 was applied as pentafluorosulfanylation reagent to prepare ethers with a vicinal SF5 substituent through a one‐step method involving photoredox catalysis. This method shows a broad substrate scope with respect to applicable alcohols for the conversion of α‐methyl and α‐phenyl styrenes. The products bear a new structural motif with two functional groups installed in one step. The alkoxy group allows elimination and azidation as further transformations into valuable pentafluorosulfanylated compounds. These results confirm that non‐toxic SF6 is a useful SF5 transfer reagent if properly activated by photoredox catalysis, and toxic reagents are completely avoided. In combination with light as an energy source, a high level of sustainability is achieved. Through this method, the proposed potential of the SF5 substituent in medicinal chemistry, agrochemistry, and materials chemistry may be exploited in the future. 相似文献

The synthesis of all 20 common natural proteinogenic and 4 otherα‐amino acid‐isosteric α‐amino tetrazoles has been accomplished, whereby the carboxyl group is replaced by the isosteric 5‐tetrazolyl group. The short process involves the use of the key Ugi tetrazole reaction followed by deprotection chemistries. The tetrazole group is bioisosteric to the carboxylic acid and is widely used in medicinal chemistry and drug design. Surprisingly, several of the common α‐amino acid‐isosteric α‐amino tetrazoles are unknown up to now. Therefore a rapid synthetic access to this compound class and non‐natural derivatives is of high interest to advance the field. 相似文献

In the title compound, C21H31N3O4, a potential inhibitor of aromatase, all rings are fused trans. Rings A, B and C have chair conformations which are slightly flattened. Ring D has a 14α‐envelope conformation. The steroid nucleus has a small twist, as shown by the C19—C10⋯C13—C18 torsion angle of 6.6 (2)°. Ab initio calculations of the equilibrium geometry of the molecule reproduce this small twist, which appears to be due to the steric effect of the 6β‐azide substituent rather than to packing effects. 相似文献
