Microalgae have a great potential for the production of healthy food and feed supplements. Their ability to convert carbon into high-value compounds and to be cultured in large scale without interfering with crop cultivation makes these photosynthetic microorganisms promising for the sustainable production of lipids. In particular, microalgae represent an alternative source of polyunsaturated fatty acids (PUFAs), whose consumption is related to various health benefits for humans and animals. In recent years, several strategies to improve PUFAs’ production in microalgae have been investigated. Such strategies include selecting the best performing species and strains and the optimization of culturing conditions, with special emphasis on the different cultivation systems and the effect of different abiotic factors on PUFAs’ accumulation in microalgae. Moreover, developments and results obtained through the most modern genetic and metabolic engineering techniques are described, focusing on the strategies that lead to an increased lipid production or an altered PUFAs’ profile. Additionally, we provide an overview of biotechnological applications of PUFAs derived from microalgae as safe and sustainable organisms, such as aquafeed and food ingredients, and of the main techniques (and their related issues) for PUFAs’ extraction and purification from microalgal biomass. 相似文献

Analysis of polyunsaturated fatty acids using high performance liquid chromatography–atmospheric pressure chemical ionization mass spectrometry is described. The standard fatty acid methyl esters from 16 to 22 carbons were analyzed by LC‐MS with APCI. The effect of orifice voltage and total carbon atoms versus number of double bonds in each homologue on the mass spectra is discussed. The correction coefficients for homologues from saturated fatty acids to hexaenoic acid are also mentioned. 相似文献

A high resolution approach to silver ion HPLC was studied for the separation of positional isomers of triacylglycerols (TAGs) containing long chain polyunsaturated fatty acids (PUFA) such as eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and docosapentaenoic acid (DPA) in enzymatically synthesized structured TAGs. Isopropanol was used as a novel modifier in a hexane-acetonitrile based mobile phase for silver ion HPLC. Peak identification was based on HPLC-mass spectroscopy and selectivities of lipases. Positional isomers of TAGs containing one molecule of EPA, DHA, or DPA with saturated fatty acids (FAs) such as caprylic acid and palmitic acid were separated within 13 min using a gradient of hexane-isopropanol-acetonitrile as mobile phase. TAGs containing two or more EPA, DHA, or DPA were also separated from each other within 25 min, but their positional isomers were unresolved. The retention characteristics of the TAG were found to be related to the number of carbon atoms in the FAs present in addition to the number of double bonds and their isomeric configuration. One isomer with an unsaturated FA in the sn-2 position eluted faster than the other with the unsaturated FA in the sn-1 or 3 position. Species with longer chain FAs attached to TAGs with the same degree of unsaturation eluted faster than those that have shorter chain FAs. For example, docosapentaenoylhexadecanoyloctanoin (DPA/C16/C8) was eluted faster than dioctanoyldocosapentaenoin (DPA/C8/C8). 相似文献

用超临界流体萃取 (SFE)及GC MS分析了新冷冻干燥及保存 1 5d ,30d ,45d ,60d ,75d ,90d后的鲜牡蛎粉中的 2 3种脂肪酸组分的质量分数。发现在存放过程中牡蛎脂肪酸的稳定性与其不饱和度有关 ;不饱和度越高 ,脂肪酸越易被氧化 ,其中多不饱和脂肪酸的氧化是逐渐进行的 ,没有特定的稳定期。放置 90d后 ,牡蛎脂肪酸中脑黄金EPA的质量分数由原来的 1 6 94%降至 5 43% ,DHA由 9 2 5%降至 2 86%。 相似文献

Cells respond to alterations in their membrane structure by activating hydrolytic enzymes. Thus, polyunsaturated fatty acids (PUFAs) are liberated. Free PUFAs react with molecular oxygen to give lipid hydroperoxide molecules (LOOHs). In case of severe cell injury, this physiological reaction switches to the generation of lipid peroxide radicals (LOO.). These radicals can attack nearly all biomolecules such as lipids, carbohydrates, proteins, nucleic acids and enzymes, impairing their biological functions. Identical cell responses are triggered by manipulation of food, for example, heating/grilling and particularly homogenization, representing cell injury. Cholesterol as well as diets rich in saturated fat have been postulated to accelerate the risk of atherosclerosis while food rich in unsaturated fatty acids has been claimed to lower this risk. However, the fact is that LOO. radicals generated from PUFAs can oxidize cholesterol to toxic cholesterol oxides, simulating a reduction in cholesterol level. In this review it is shown how active LOO. radicals interact with biomolecules at a speed transcending usual molecule–molecule reactions by several orders of magnitude. Here, it is explained how functional groups are fundamentally transformed by an attack of LOO. with an obliteration of essential biomolecules leading to pathological conditions. A serious reconsideration of the health and diet guidelines is required. 相似文献

Capillary ElectroChromatography (CEC) on a fused silica capillary column (40 cm L × 100 μm i.d.) packed with 3 μm octadecylsilica (ODS) was evaluated for the analysis of the triglycerides and their fatty acids in fish oils, and more especially in the oil of Moroccan Sardinia pilchardus. The very high complexity of the lipids in fish oil is well illustrated by CEC with a nonaqueous mobile phase consisting of acetonitrile/isopropanol/n-hexane in the ratio 57/38/5 to which 50 mM ammonium acetate has been added. In order to unravel the complexity, the oil was hydrolyzed and the free fatty acids (FFAs), the methyl esters (FAMEs), and the phenacyl esters (FAPEs) were analyzed by CEC on the same column used for the analysis of the triglycerides. Isocratic elution was achieved with the mobile phase acetonitrile/50 mM MES pH 6 in ratio 9/1. The migration characteristics of FFAs, FAMEs, and FAPEs are compared. The analysis of FAPE derivatives has the advantage that quantification applying UV detection is possible and moreover that the number of double bonds in the fatty acid chains can be elucidated by measuring the UV abundance ratio 240/210 nm. 相似文献

The decomposition of unsaturated fatty acids and tocopherols in 10 commercial edible oils during deep frying was investigated. The dominant tocopherol in oils rich in polyunsaturated fatty acids (PUFAs) was γ-tocopherol, except for natural perilla oil (δ-tocopherol dominant), and the main tocopherol in oleic acid-rich oils was α-tocopherol. The PUFA-rich oils had higher tocopherol contents than the oleic acid-rich oils. Both the reduction rate of total unsaturated fatty acid (TUFA) and total tocopherol (TToc) were linear with frying time (t). The decomposition rate of TToc is faster than that of TUFA since the slope values obtained from fitting equations (Y?=?k t) kTToc (1.520–14.483) were obviously larger than for kTUFA (0.155–0.270). By establishing a dynamic decomposition index, unsaturated fatty acids and tocopherol in oils showed dynamic decomposition over multiple frying cycles. The obtained results showed that decomposition characteristics of oils are related to their fatty acid compositions. 相似文献

PUFA content in the glyceride mixture (including TG, DG, and MG) increased as hydrolysis progressed. The effects of operational
parameters (pH, temperature, time, and enzyme concentration) on the extent of hydrolysis were investigated. Based on these
results, optimal reaction conditions were established. At optimal conditions (pH 4.0, 35°C, 3 h, and enzyme concentration
of 500 U/g oil), the level of PUFA in the glyceride mixture was raised to 40%. The individual TG and DG fractions contained

Abstract Poly (crown ether) was employed as a catalyst of derivatization of lower fatty acids to the p-bromophenacyl esters for gas chromatographic determination of them. The esterification reaction proceeds quantitatively under mild conditions, i.e. at room temperature and within 30 min. The poly (crown ether) did not interfere with the gas chromatogram of the esters unlike monomeric crown ethers. 丙二醛(MDA)是生物分子氧化的典型羰基中间体, 是脂质过氧化的典型终末产物. 这类活性羰基中间体引起组织蛋白的老化与恶化及DNA损伤. 牛磺酸在人体内具有非常广泛的生理功能, 而g-氨基丁酸(GABA)是神经系统的一种重要抑制性神经递质, 因此二者是两种很重要的非蛋白氨基酸. 研究了在生理条件下牛磺酸或GABA是否直接诱捕MDA从而保护组织蛋白不受伤害. 将牛磺酸或GABA与MDA在生理条件下温浴48 h, 经高效液相、质谱等分析手段分析, 发现牛磺酸或GABA与MDA直接反应, 通过HPLC分离, 都生成两种主产物, 一种是没有荧光的紫外吸收峰为274~278 nm 的产物, 一种是有荧光的产物, 这种类似脂褐素(Ex. 392~395 nm/Em. 456~364 nm)的荧光产物是1,4-二氢吡啶衍生物. 牛磺酸或GABA与MDA的反应表明它们在生理条件下具有清除活性羰基的功能, 而这种毒性羰基与很多羰基紧张相关的疾病及衰老有关. 相似文献

用碱催化法将青海湖裸鲤鱼油甲酯化,以气相色谱/质谱法分析鱼油中的脂肪酸。青海湖裸鲤可食用部分中鱼油含量为25.13%。从鱼油中共鉴定出47种脂肪酸,包括直链、单支链、多支链饱和脂肪酸,单不饱和、多不饱和脂肪酸,环丙烷基、呋喃基脂肪酸等。不饱和脂肪酸含量为73.6%,其中多不饱和脂肪酸含量为25.4%,以C18∶2(4.9%),C18∶3(3.1%),C20∶4(1.3%),C20∶5(二十碳五烯酸(EPA), 9.4%)和C22∶6(二十二碳六烯酸(DHA), 6.7%)为主。单不饱和脂肪酸含量为48.2%,主要由C16∶1(20.3%),C18∶1(25.9%)构成。饱和脂肪酸含量为25.7%,主要有C14∶0(3.4%),C16∶0 (19.4%)和C18∶0(1.1%)。青海湖裸鲤鱼油中还存在不常见的环丙烷基和呋喃基脂肪酸及多种奇数碳链和支链脂肪酸。因此,青海湖裸鲤是功能性脂肪酸的重要膳食来源。 相似文献

A balanced ratio of fatty acids n-6 to n-3 in chicken eggs is important for health and to help prevent and manage obesity and other diseases. Traditionally, fish oil or flax seed has been utilized as feed additives to decrease the ratio of n-6 to n-3(n-6:n-3) fatty acids in eggs. The hull of spina date seed(HSDS) is a common agricultural waste product in China, from which wood vinegar(HSDSWV) may be derived. This study evaluated HSDSWV as a supplement in hen feeds to improve the quality of eggs and decrease the ratio of fatty acids n-6:n-3. HSDSWV was obtained via carbonization, and refined. Six concentrations(nil to 0.5%) of HSDSWV were prepared and fed to 6 hen groups, respectively, for 50 d. The fatty acids of the hen's egg yolks were analyzed by gas chromatography/electron ionization-mass spectrometry(GC/EI-MS) in the selected ion monitoring(SIM) mode. The 0.2% HSDSWV resulted in the best egg yolk quality, with a lower percentage of linoleic acid(C18:2n6) and higher percentages of cis-5,8,11,14,17-eicosapentaenoic acid(C20:5n3) and cis-4,7,10,13,16,19-docosahexaenoic acid(C22:6n3), and thus a lower n-6:n-3 ratio compared with the other HSDSWV concentrations. In addition, the eggs contained higher levels of yolk fat and egg yolk than the controls did. In conclusion, to modify the fatty acid composition of hens' eggs and obtain a balanced ratio of n-6:n-3, 0.2% HSDSWV may be considered suitable as a dietary supplement in hens' feed. 相似文献

二十碳五烯酸和二十二碳六烯酸是两种非常重要的ω-3多不饱和脂肪酸,广泛用于膳食补充剂和药品,同时它们的生理作用并不完全相同,因此分离制备它们的高纯度单体十分必要。首先以聚苯乙烯/二乙烯基苯(PS/DVB)聚合物为固定相,在液相色谱上分离二十碳五烯酸乙酯(EPA-EE)和二十二碳六烯酸乙酯(DHA-EE),考察了流动相、填料粒径、温度对分离的影响。然后采用粒径20 μ m、孔径10 nm的PS/DVB填料装填了8根150 mm×10 mm的半制备色谱柱,测定了半制备柱装填的均一性。最后尝试在模拟移动床(SMB)色谱上分离EPA-EE和DHA-EE的混合物,探究了Ⅱ区和Ⅲ区的流量、进料流量、进料浓度对分离的影响,结果表明SMB制备的EPA-EE和DHA-EE的相对纯度分别为91.6%和93.6%,回收率分别为97.0%和91.6%,固定相生产率为5.97 g/(L\5h),溶剂消耗为1.52 L/g。SMB制备EPA-EE和DHA-EE具有较大的应用潜力。 相似文献
