Linear and four‐armed poly(l ‐lactide)‐block‐poly(d ‐lactide) (PLLA‐b‐PDLA) block copolymers are synthesized by ring‐opening polymerization of d ‐lactide on the end hydroxyl of linear and four‐armed PLLA prepolymers. DSC results indicate that the melting temperature and melting enthalpies of poly (lactide) stereocomplex in the copolymers are obviously lower than corresponding linear and four‐armed PLLA/PDLA blends. Compared with the four‐armed PLLA‐b‐PDLA copolymer, the similar linear PLLA‐b‐PDLA shows higher melting temperature (212.3 °C) and larger melting enthalpy (70.6 J g?1). After these copolymers blend with additional neat PLAs, DSC, and WAXD results show that the stereocomplex formation between free PLA molecular chain and enantiomeric PLA block is the major stereocomplex formation. In the linear copolymer/linear PLA blends, the stereocomplex crystallites (sc) as well as homochiral crystallites (hc) form in the copolymer/PLA cast films. However, in the four‐armed copolymer/linear PLA blends, both sc and hc develop in the four‐armed PLLA‐b‐PDLA/PDLA specimen, which means that the stereocomplexation mainly forms between free PDLA molecule and the inside PLLA block, and the outside PDLA block could form some microcrystallites. Although the melting enthalpies of stereocomplexes in the blends are smaller than that of neat copolymers, only two‐thirds of the molecular chains participate in the stereocomplex formation, and the crystallization efficiency strengthens. © 2014 Wiley Periodicals, Inc. The thermal properties, crystallization, and morphology of amphiphilic poly(D ‐lactide)‐b‐poly(N,N‐dimethylamino‐2‐ethyl methacrylate) (PDLA‐b‐PDMAEMA) and poly (L ‐lactide)‐b‐poly(N,N‐dimethylamino‐2‐ethyl methacrylate) (PLLA‐b‐PDMAEMA) copolymers were studied and compared to those of the corresponding poly(lactide) homopolymers. Additionally, stereocomplexation of these copolymers was studied. The crystallization kinetics of the PLA blocks was retarded by the presence of the PDMAEMA block. The studied copolymers were found to be miscible in the melt and the glassy state. The Avrami theory was able to predict the entire crystallization range of the PLA isothermal overall crystallization. The melting points of PLDA/PLLA and PLA/PLA‐b‐PDMAEMA stereocomplexes were higher than those formed by copolymer mixtures. This indicates that the PDMAEMA block is influencing the stability of the stereocomplex structures. For the low molecular weight samples, the stereocomplexes particles exhibited a conventional disk‐shape structure and, for high molecular weight samples, the particles displayed unusual star‐like shape morphology. © 2011 Wiley Periodicals, Inc. Poly(lactide)s [i.e. poly(lactic acid) (PLA)] and lactide copolymers are biodegradable, compostable, producible from renewable resources, and nontoxic to the human body and the environment. They have been used as biomedical materials for tissue regeneration, matrices for drug delivery systems, and alternatives for commercial polymeric materials to reduce the impact on the environment. Since stereocomplexation or stereocomplex formation between enantiomeric PLA, poly(L-lactide) [i.e. poly(L-lactic acid) (PLLA)] and poly(D-lactide) [i.e. poly(D-lactic acid) (PDLA)] was reported in 1987, numerous studies have been carried out with respect to the formation, structure, properties, degradation, and applications of the PLA stereocomplexes. Stereocomplexation enhances the mechanical properties, the thermal-resistance, and the hydrolysis-resistance of PLA-based materials. These improvements arise from a peculiarly strong interaction between L-lactyl unit sequences and D-lactyl unit sequences, and stereocomplexation opens a new way for the preparation of biomaterials such as hydrogels and particles for drug delivery systems. It was revealed that the crucial parameters affecting stereocomplexation are the mixing ratio and the molecular weight of L-lactyl and D-lactyl unit sequences. On the other hand, PDLA was found to form a stereocomplex with L-configured polypeptides in 2001. This kind of stereocomplexation is called "hetero-stereocomplexation" and differentiated from "homo-stereocomplexation" between L-lactyl and D-lactyl unit sequences. This paper reviews the methods for tracing PLA stereocomplexation, the methods for inducing PLA stereocompelxation, the parameters affecting PLA stereocomplexation, and the structure, properties, degradation, and applications of a variety of stereocomplexed PLA materials.  相似文献

The miktoarm star‐shaped poly(lactic acid) (PLA) copolymer, (PLLA)2‐core‐(PDLA)2, was synthesized via stepwise ring‐opening polymerization of lactide with dibromoneopentyl glycol as the starting material. 1H NMR and FTIR spectroscopy proved the feasibility of synthetic route and the successful preparation of star‐shaped PLA copolymers. The results of FTIR spectroscopy and XRD showed that the stereocomplex structure of the copolymer could be more perfect after solvent dissolution treatment. Effect of chain architectures on crystallization was investigated by studying the nonisothermal and isothermal crystallization of the miktoarm star‐shaped PLA copolymer and other stereocomplexes. Nonisothermal differential scanning calorimetry and polarizing optical microscopy tests indicated that (PLLA)2‐core‐(PDLA)2 exhibited the fastest formation of a stereocomplex in a dynamic test due to its special structure. In isothermal crystallization tests, the copolymer exhibited the fast crystal growth rate and the most perfect crystal morphology. The results reveal that the unique molecular structure has an important influence on the crystallization of the miktoarm star‐shaped PLA copolymer. © 2019 Wiley Periodicals, Inc. J. Polym. Poly(lactic acid) (PLA) stereocomplexes have high potential as renewable materials for advanced polymer applications, mainly due to their high melting temperature (Tm, typically 230–240°C). The properties of PLA stereocomplexes consisting of linear high molar mass homopolymers have been studied extensively in the past, but the available information about the possibilities to affect the thermal properties of the stereocomplex by varying the structure of the blend components has not been sufficient. Novel stereocomplexes containing linear or star‐shaped D ‐lactic acid (D ‐LA) oligomers and high molar mass poly(L ‐lactide) (L‐ PLA) were thus prepared. The Tm and melting enthalpy (ΔHm) of the racemic crystallites were found to depend strongly on both the blending ratio and the arm‐length of the D ‐lactic acid oligomer. The preparation method of the oligomers, i.e. step‐growth polymerization or ring‐opening polymerization (ROP), did not affect the Tm or ΔHm of the blends significantly. Slightly higher ΔHm values were, however, obtained, when linear oligomers were used. The results thus indicated that the Tm and ΔHm of PLA stereocomplexes could be optimized, simply by selecting a D ‐LA oligomer having a suitable arm‐length and structure as the other blend component. The possibility to adjust the melting behavior of the stereocomplex blend is a significant advantage and could make PLA suitable for a wider range of products used at elevated temperatures. Abstract

This paper gives a general overview of several approaches we have investigated for designing new PLA-based polymers with a broad range of properties and improved processability. These approaches include: copolymerization (block and stereoblock copolymers), microstructure and architecture control, and stereocomplexation. Multiblock copolymers with alternating “soft” and “hard” segments, synthesized over a broad range of chemical compositions, show properties ranging from hard plastics to elastomers. Stereoblock copolymers with alternating amorphous and semicrystalline PLA blocks combine the advantages of PLA homopolymers (crystallinity) and random copolymers (processability). Independent control of polymer architecture and microstructure allows for the synthesis of star polymers with various arm morphologies. A new method for stereocomplex formation between L-PLA and D-PLA, which combines in-situ polymerization with stereocomplexation, is also described. For the synthesis of these new materials we took advantage of: 1) chirality of lactide monomer, 2) retention of configuration during polymerization, 3) living nature of the ring-opening polymerization (ROP) of lactide in the presence of active hydrogen groups such as OH and NH2, and 4) control of the level of transesterification reactions.  相似文献

Two enantiomeric amphiphilic graft copolymers consisting of water soluble poly(2‐hydroxyethyl methacrylate) (HEMA) and biodegradable oligo(L ‐lactide) (OLLA) or oligo(D ‐lactide) (ODLA) were synthesized by free radical copolymerization. HEMA‐OL(D)LA macromonomers were synthesized by ring opening polymerization of L ‐ or D ‐lactide. Both HEMA‐OLA macromonomers and graft copolymers were characterized by NMR spectroscopy and gel permeation chromatography. Graft copolymers and their stereocomplexes were analyzed by wide angle X‐ray diffraction and differential scanning calorimetry (DSC). Due to the formation of stereocomplex crosslinks between poly(HEMA) main chains, amphiphilic, biodegradable hydrogels prepared by blending of two enantiomeric poly(HEMA‐g‐OLLA) and poly(HEMA‐g‐ODLA) degraded more slowly in phosphate buffered saline than individual optically pure poly‐(HEMA‐g‐OL(D)LA).  相似文献

The thermal properties of two series of AB di-block copolymers of poly(ϵ-caprolactone) (A) and poly(lactide) (B) and their blends were studied. Each series contained poly(lactide) blocks of opposite chirality. The length of the poly(ϵ-caprolactone) blocks was not varied (DP = 70), whereas the poly(lactide) blocks were of varying length (DP = 5 − 80). Blends of polymers containing blocks of opposite chirality were prepared by mixing in solution. The melting temperature of the PLA phase was raised by approximately 55 °C in the blends due to stereocomplex formation. The melting temperatures of the crystalline PCL and PLA phases strongly depended on the composition of the block copolymers.  相似文献

The stereocomplex formation between enantioselective poly(lactide) (PLA) homopolymers is well understood. In this report an attempt is made to analyze the influence on the self‐assembling of the stereocomplex of enantiomorphic PLA‐PEG di‐ and tri‐blocks in different solvents. Powder diffraction studies showed the poly(ethylene glycol) (PEG) and the PLA blocks crystallize separately forming unique supra structures like rods, discs and coiled coils with dimensions in the micrometer scale in length and sub‐micrometer scale in diameter. The influence of the solvents on the crystal formation was shown in the formation of uniform structures. Discs emerged from equimolar mixtures of the D ‐ and L ‐configured di‐ and tri‐block copolymers, in dioxan and acetonitrile and in water the stereocomplexes crystallized mainly as rods. In some cases the rods were observed as coiled coils. The shape, the hydrophobic/hydrophilic content and the PEG coated surface of the discs give them a future potential as matrix for the controlled and targeted delivery of bioactive agents. A very important goal of researchers today is providing poly(L -lactide) (PLA)-based polymers with controllable degradation profiles, various (soft or elastic) mechanical properties, reactivity for chemical modification, and other functionalities while keeping the favorable characteristics of PLA. This article concerns the synthetic methods and properties of the following novel lactide copolymers: (1) random and block copolymers of depsipeptide and L -lactide with reactive (ionic) side-chain groups, (2) comb-type PLA and branched PLA, and (3) PLA-grafted polysaccharide and PLA with terminal saccharide residues. Poly(depsipeptide-random-L -lactide)s and polydepsipeptide-block-poly(L -lactide)s with reactive (ionic) side-chain groups should be useful for the preparation of matrices and microspheres with reactive surfaces because of their amphiphilic structures. Comb-type PLA and branched PLA show lower crystallinities than linear PLA. PLA-grafted polysaccharide should be useful for the preparation of matrices with various microstructures and mechanical and degradation properties through the introduction of hydrophilic segments. © 2003 Wiley Periodicals, Inc. In situ gel forming systems have been prepared by linking polylactic acid (PLA) to a water soluble and polyfunctional polymer, such as α,β-poly(N-2-hydroxyethyl)-d,l-aspartamide (PHEA). Three graft copolymers PHEA-PLA with a different derivatization degree in PLA, have been synthesized and characterized. PHEA-PLA graft copolymer with the highest amount in PLA has been used to prepare solutions in organic solvents able to give rise to gel-like matrices when injected into phosphate buffered saline solution. The chemical degradation of these gels has been evaluated and in vitro tests have been performed to evaluate the cell compatibility of the hydrolysis products. The possibility to use these gels for drug release has been investigated by incorporating leuprolide as a peptide model drug and by evaluating its in vitro release. To improve the drug release profile, PHEA-PLA graft copolymer has been derivatized with pendant carboxylic groups that are able to form an ion pair with the leuprolide thus reducing the burst effect and prolonging its release.  相似文献

Methylated and pegylated poly(lactide)‐block‐poly(ε‐caprolactone)‐block‐poly(lactide) copolymers, PLA–P(CL‐co‐CLCH3)–PLA and PLA–P(CL‐co‐CLPEG)–PLA, were prepared in three steps: combining the formation of carbanion‐bearing dihydroxylated‐PCL, the coupling of iodomethane or bromoacetylated α‐hydroxyl‐ω‐methoxy‐poly(ethylene glycol) onto the carbanionic PCL, and finally the ring opening polymerization of DL ‐lactide initiated by the preformed grafted diOH‐PCL copolymers. The resulting block copolymers exhibited lower crystallinity, melting temperature, and hydrophobicity with respect to the original PCL. Degradation of the grafted copolymers was investigated in the presence of Pseudomonas cepacia lipase and compared with that of the triblock copolymer precursor. It is shown that the presence of the grafted substituents affected the enzymatic degradation of PCL segments. © 2005 Wiley Periodicals, Inc. A series of copolylactones was synthesized by ring‐opening copolymerization of glycolide, L ‐lactide and ?‐caprolactone, using stannous octoate as catalyst. The in vitro degradation behaviors of them were studied and data demonstrated different degradation rates which mainly depended on the compositions. Investigation of the 5‐fluorouracil (5‐Fu) release from these copolylactones revealed that the composition, degradation rate and the morphology of the polymeric matrix played an important role on the drug release kinetics. A sustained 5‐Fu release without initial time lag was obtained from random poly(lactide‐co‐glycolide‐co‐caprolactone) (r‐PGLC) drug carrier, and it differed from the cases of polylactide (PLA) or random poly(lactide‐co‐glycolide) (PLGA), which usually showed an initial time lag or biphasic drug release behavior. It was due to the low glass transition temperature (T g) of the r‐PGLC and the drug would diffuse faster in rubbery state under the experimental temperature. Furthermore, a significant change in the drug release behavior of r‐PGLC was observed when the temperatures were changed around the T g of the drug carrier, which implied that the drug release behavior could be regulated by adjusting the morphology of the drug carrier. Enantiopure poly(lactic acid) (PLA) can form stereocomplexes when enantiomeric PLA chains are mixed in equivalent amounts. Such materials provide interesting features that might be suitable for numerous applications. Despite several advantages, the main drawback of PLA is its narrow window of processing, thus limiting its use for industrial applications. Reported herein are achiral iron complexes, that are highly active, productive, and stereoselective under mild reaction conditions for the ring‐opening polymerization of lactide. The corresponding catalytic systems enable the production of stereoblock polymers with high molecular weights, allowing the formation of thermally stable and industrially relevant stereocomplexes.  相似文献

Nowadays, preparation of poly(lactide) (PLA) with high content of stereocomplex crystallites (SCs) is receiving more and more attentions. The stereocomplex formation between enantiomeric poly(l ‐lactide) and poly(d ‐lactide) can efficiently improve the corresponding mechanical properties and thermal stability. In the current work, using dynamic Monte Carlo simulations, the microscopic mechanism of SC formation in grafted polymers was investigated. The increase in grafting density can lead to the enhancement of SC formation. On one hand, the miscibility between the chains of different types can be improved due to the grafting. On the other hand, the increase in grafting density can result in the higher degree of chain extension and the change in crystal nucleation and growth modes. The changes facilitate the stereocomplex formation. These findings not only provide an effective way to prepare PLAs with high content of SCs, but also reveal the underlying mechanisms controlling the SC formation. © 2018 Wiley Periodicals, Inc. J. Environmental switches may be fabricated for the controlled release of pharmaceutical drug using a thermally responsive polymer with the intrinsic chemical and physical nature of stimuli‐sensitive smart materials. Particularly, much attention has been paid to the biomedical applications of poly(N‐isopropyl acrylamide) (PNIPAAm) because of its unique reversible transition at a specific lower critical solution temperature (LCST).Thermally sensitive block copolymers, poly(N‐isopropyl acrylamide‐b‐poly(L ‐lactide‐co‐glycolide) (PNIPAAm‐b‐PLGA), and polyethylene glycol‐poly (lactide‐co‐glycolide) (PEG‐PLGA) triblock copolymers with different compositions and length of PLGA block were synthesized via ring‐opening polymerization of lactide and glycolide in the presence of OH‐terminated PNIPAAm or PEG. The composition and structure of the polymer were determined by NMR and FTIR. The effect of important factors, such as ionic strength, pH, and polymer concentration on the phase transition behavior of temperature‐sensitive polymers, were investigated by cloud point measurements. The resulting thermosensitive polymers were used for the entrapment of a narcotic antagonist drug, naltrexone, as the model drug. The loading efficiency and drug release behavior of naltrexone‐loaded hydrogels were investigated. The naltrexone loaded thermosensitive polymers were able to sustain the release of naltrexone for different periods of time, depending on the polymer composition, and concentration. In vitro release studies showed that these thermosensitive polymers are able to deliver naltrexone in biologically active forms at a controlled rate for 3–8 weeks. Amphiphilic copolymers containing β-cyclodextrin (β-CD) core were synthesized successfully. Synthesis was initiated from tosylated cyclodextrin ((Tosyl)7-β-CD) which is containing two types of functional groups. Hydroxyl functional groups of (Tosyl)7-β-CD were used as initiator for ring opening polymerization of lactide. This step led to star polymers containing tosylated cyclodextrin core and polylactide arms (PLA-(Tosyl)7-β-CD). In the next step, tosyl groups of PLA-(Tosyl)7-β-CD were used as initiator for ring opening polymerization of 2-ethyl-2-oxazoline and amphiphilic copolymers containing cyclodextrin core, PLA and poly(2-ethyl-2-oxazoline) arms (PLA-β-CD-POX) were obtained. Loading of Congo red as guest molecule by amphiphilic copolymers was investigated. The release of guest molecule from chloroform solution of copolymers to water phase was also investigated.  相似文献

The effect of crystallization of a hydrophobic poly(lactide) block on the self-organization of biocompatible and biodegradable amphiphilic poly(lactide)-block-poly(ethylene oxide) (PLA-b-PEO) copolymers in a dilute aqueous solution has been investigated. It was demonstrated that the co-crystallization of poly(L,L-lactide) [P(L,L)LA] and poly(d,d-lactide) [P(d,d)LA] chains under equimolar mixing of P(L,L)LA46-b-PEO113 and P(d,d)LA56-b-PEO113 copolymers resulted in the formation of stable and spontaneously water-redispersible stereocomplex micelles with semicrystalline P(L,L)LA/P(d,d)LA cores. It was shown that the P(L,L)LA46 / P(d,d)LA56-b-PEO113 stereo-complex micelles produced by dialysis can be potential vehicles for the anticancer agent oxaliplatin  相似文献

Diblock and multiblock copolymers composed of a poly(D,L-lactide) (PLA) or poly(trimethylene carbonate) (PTMC) core with a hydrophilic chain of poly(ethylene glycol) (PEG) were prepared. These copolymers, in which the core is connected to PEG through a polyfunctional molecule such as citric, mucic, or tartaric acid, may be used to form nanoparticles for drug delivery applications. Branched copolymers were prepared by direct amidation between the polyfunctional acid and methoxy PEGamine, followed by ring-opening polymerization of lactide or trimethyl carbonate to form the PLA and PTMC block copolymers. In addition, a complex multiblock copolymer of biotin-PEG-poly[lactic-co-(glycolic acid)] (PLGA) for application in an avidin-biotin system was prepared for possible design of nanospheres with targeting properties. Studies of drug release from polymeric systems containing multiblock copolymers and studies of polymer degradation were also performed.  相似文献

Graft copolymers of poly(tulipalin A) (PT) and poly(DL‐lactide) (PDLLA) (PT‐g‐PDLLA) having various graft lengths and ratios were synthesized by free‐radical copolymerization of α‐methylene‐γ‐butyrolactone (MBL) and PDLLA macromonomers (HEMA‐PDLLA) terminated by 2‐hydroxyethyl methacrylate (HEMA)‐terminated. HEMA‐PDLLA were synthesized by ring opening polymerization (ROP) of DL‐lactide in the presence of HEMA. Both HEMA‐PDLLA and the copolymers were characterized by NMR spectroscopy and gel permeation chromatography (GPC). The thermal properties of the graft copolymers were found to depend on the graft length and the ratio. The copolymers consisting of PDLLA side chains of Mn = 500 Da showed a single Tg between Tgs of the two component polymers, suggesting a miscible state of PT and PDLLA. In contrast, the copolymers consisting of PDLLA side chains of Mn = 1100, 2000, and 7000 Da showed two isolated Tg, suggesting two segregated domains. The AFM phase images of the copolymers supported the single and phase‐separated morphologies for the former and latter systems, respectively. © 2011 Wiley Periodicals, Inc.
