A copper-catalyzed, three-component reaction between a sulfonyl azide, an alkyne, and a 2-aminothiophenol derived Schiff base is reported. This novel one-pot procedure involves a click reaction, a formal [2+2] cycloaddition, and S–S coupling, therefore, providing a unique method for the synthesis of novel disulfide-linked N-sulfonylazetidin-2-imines under mild conditions.  相似文献

A rapid protocol for the multicomponent microwave-assisted organocatalytic domino Knoevenagel/hetero Diels-Alder reaction (DKHDA) has been developed for the synthesis of 2,3-dihydropyran[2,3-c]pyrazoles. The reported procedure could be used for the fast generation of novel substituted 2,3-dihydropyran[2,3-c]pyrazoles with potential anti-tuberculosis activity.  相似文献

An easy, efficient, and simple one-pot approach for the synthesis of 1,3-thiazolidine-2-thiones via multicomponent reaction is reported. The reaction of a primary amine with carbon disulfide in the presence of dibenzoylacetylene or bis(4-methyl-1-benzoyl)acetylene in a mixture of CH2Cl2 and H2O after 5 h, afforded the title compound as alkene diastereomers.  相似文献

A one-pot, five-component strategy toward the synthesis of new indole substituted pyrrolidine and pyrrolizidine heterocycles through 1,3-dipolar cycloaddition reactions using ninhydrin, 1,2-phenylenediamine, amino acids, 3-cyanoacetyl indoles and aryl aldehydes is described. The features of this procedure were characterized by the mild reaction conditions, high yields, one-pot procedure, and operational simplicity.  相似文献

A general method for the synthesis of 1,2,3,4-tetrasubstituted pyrroles by [NMPH]CH3SO3 is reported. A full factorial design was performed in order to obtain a more robust and statistically correct optimum condition. The products were obtained in moderate to excellent yields. The developed methodology is very simple, cost-effective, and applicable for tri- and tetra-component reactions.  相似文献

A new domino process of three-component Povarov reaction and aerobic dehydrogenation was developed toward the synthesis of polysubstituted quinolines. 2,4-Disubstituted-8-nitroquinolines, which are important precursors for the synthesis of antimalarial primaquine drug-like molecules, were prepared conveniently by this method in moderate to good yields. Brönsted acid (HClO4)-modified montmorillonite was found to be a crucial catalyst in promoting the procedure.  相似文献

In an attempt to further exploit multicomponent reactions in the field of hybrid heterocyclic molecules, we describe a bidirectional approach for the synthesis of novel 1,3,5-thiadiazine-peptides molecules. The process relies on the execution of two Ugi reactions between dicarboxy-functionalized 1,3,5-thiadiazine with different amines and isocyanides. An alternative strategy relying on a sequential multicomponent macrocyclization procedure was also developed to afford thiadiazine peptide-like macrocycles. Both methods showed great chemical efficiency and versatility.  相似文献

One-pot synthesis of 2-(N-sulfonylimino)indolines has been developed. The procedure combines the copper-catalyzed three-component reaction of sulfonyl azides, o-bromophenylacetylenes, and amines and the copper-catalyzed intramolecular C-N coupling in one sequence, which afforded the products in moderate to good yields. The resulting 2-(N-sulfonylimino)indolines could be easily transformed to pharmaceutically valuable oxindoles (indolin-2-ones).  相似文献

Stereoselective 1,3-dipolar cycloadditions of exo-methylenesugars 1a-c to a sugar nitrone 2 were carried out with refluxing a toluene solution and afforded the corresponding cycloadducts, ketosyl spiro-isoxazolidine disaccharides 3a and 4a-c. Followed by reductive cleavage of the N-O bond of the isoxazolidine ring with the treatment of Zn-AcOH-Ac2O or with catalytic hydrogenation (Pd(OH)2/C), the cycloadduct 3a could be readily converted into a novel amino-C-disaccharide possessing a ketose form, providing an access to a novel amino-C-ketosyl disaccharides.  相似文献

Various tricyclic dienes were synthesized via enyne metathesis using the first generation Grubbs catalyst. The enyne metathesis proceeded smoothly in refluxing CH2Cl2 with a low catalyst loading (3.0 mol %), giving good yields (72-89%) of the tricyclic products 6 and 16. The resulting 1,3-dienes are suitable precursors of polycyclic structures via a Diels-Alder process. One-pot RCM/Diels-Alder reactions of the enyne products with dienophiles proceeded smoothly to afford polycyclic compounds as a single cycloadduct. The structures of the Diels-Alder adducts were determined by 1H NMR spectra and X-ray analysis. The cycloadducts were formed via the approach of the dienophiles towards the diene in endo mode.  相似文献

A practical synthesis of 2-substituted 1,3-cyclohexadienes by the cross enyne metathesis between alkynes and 1,5-hexadiene is reported. The isolation of the 1,3-cyclohexadienes has been hampered by the formation of an inseparable triene by-product. The use of a second consecutive cross alkene metathesis to give water-soluble products allowed removal of this by-product. Using this one-pot procedure, a synthesis of cyclohexadienes from simple starting materials was developed. The procedure was used in a three-step synthesis of a functionalized tetrahydroquinoline using Pd(II)-catalyzed chloroacetoxylation (Bäckvall reaction) for cyclohexadiene functionalization.  相似文献

The regioselective three-component condensation reaction of 2-hydroxy-1,4-naphthoquinone with isocyanides in the presence of a variety of aldehydes offers an easy one-pot access to linear naphtho[2,3-b]-furan-4,9-dione derivatives. This method has the advantage of good yields, high regioselectivity, and uses readily accessible substrates and reagents. Unsymmetrical diarylamines have been synthesized by a novel, unexpected multicomponent reaction between methoxyarylaldehydes, N-arylhydroxylamine and maleic anhydride. Using improved reaction protocols for three-component coupling reactions of aldehydes, dienophiles, and amides (AAD-reaction) or anhydrides (ANAD-reaction) or orthoesters (ALAD-reaction), a variety of functionalized hexahydroisoindolo derivatives were synthesized and fully characterized. Condensation of ubiquitous available aldehydes with unsaturated amides or anhydrides or orthoesters and subsequent Diels-Alder reactions with electron deficient dienophiles furnishes endo-selective enyne-reaction precursors in good to excellent yield.  相似文献
