EDTA-catalyzed synthesis of 3,4-dihydroquinoxalin-2-amine derivatives by a three-component coupling of one-pot condensation reactions in an aqueous medium

This paper describes a simple and efficient one-pot synthetic approach for the preparation of biologically interesting 3,4-dihydroquinoxalin-2-amine derivatives using EDTA-catalyzed three-component reactions of o-phenylenediamines, carbonyl compounds, and isocyanides in an aqueous medium. This method is of great value because of its environmentally benign character, high yields, and ease of handling.     

A novel isocyanide-based three-component reaction: a facile synthesis of substituted 2H-pyran-3,4-dicarboxylates

An efficient method for synthesis of 2H-pyran-3,4-dicarboxylates using the three-component reaction of dithiocarbamates, dialkyl acetylenedicarboxylates, and isocyanides in solvent-free conditions is described. In these reactions, synthesis of dithiocarbamates is possibly based on the one-pot reaction of secondary amines, CS₂, and alkyl halides in solvent-free conditions without using a catalyst. The mild reaction conditions and high yields of the reaction exhibit the good synthetic advantage of these methods.     

PTSA catalyzed straightforward protocol for the synthesis of 2-(N-acyl)aminobenzimidazoles and 2-(N-acyl)aminobenzothiazoles in PEG

An efficient PTSA catalyzed synthesis of 2-(N-acyl)aminobenzimidazoles and 2-(N-acyl)aminobenzothiazoles has been described using S-ethylated-N-acylthioureas as substrates and polyethylene glycol as solvent.     

A novel three-component reaction of N-fluoropyridinium salts: a facile approach to imidazo[1,2-a]pyridines

The reaction of N-fluoropyridinium triflate with isonitriles in acetonitrile and propionitrile in the presence of NaBH(OAc)₃ led to the formation of the corresponding imidazo[1,2-a]pyridines in 44-73% yields. The proposed reaction mechanism involves the intermediate formation of a highly reactive carbene species and apparent reduction of the pyridinium intermediate with NaBH(OAc)₃ to yield the targeted heterocycles.     

Cascade reaction for 3-pyrrolines and pyrroles from nitroallylic acetates and N-mesyl 2-aminoethanones

Cascade reactions of nitroallylic acetates with methanesulfonyl 2-aminoethanones affords either the 3-pyrrolines or the pyrroles in one pot depending on reaction solvents and temperature. A possible mechanism for the entire sequence is proposed.     

InCl3-catalyzed four-component reaction: a novel synthesis of N-carbazolyl dihydropyridines

We have developed a simple, efficient, and environmentally benign microwave-assisted InCl₃-catalyzed synthesis of N-carbazolyldihydropyridines via a four-component reaction of 3-amino-9-ethylcarbazole, malononitrile, aromatic aldehydes, and acetylenic esters. The use of microwave heating allowed for reduced reaction times and resulted in higher yields. This four-component reaction is atom-efficient, high-yielding, and applicable to a wide variety of four-component reactions.     

Palladium-catalyzed cross-coupling reaction of aryldioxaborolane with 2-bromo-N,N-dimethylacetamide

A Suzuki-type cross-coupling of aryldioxaborolane with 2-bromo-N,N-dimethylacetamide in the presence of a catalytic amount of tricyclohexylphosphine as the ligand and hydroquinone as the free-radical scavenger has been demonstrated as a convenient and simple way for the synthesis of α-arylacetamide.     

Catalyst-free synthesis of functionalized dihydro-2-oxypyrroles by the reaction of enaminones and N,N′-bis(phenylmethylidene)phenylmethane

A catalyst-free and convenient approach for the preparation of substituted dihydro-2-oxypyrrole is described. This three-component reaction between primary amines, dialkyl acetylenedicarboxylate, and N,N′-bis(phenylmethylidene)phenylmethane proceeds in MeOH under reflux conditions in good to excellent yields.     

Two-pot synthesis of N,N-disubstituted 4H-3,1-benzothiazin-2-amines from aryl(2-isothiocyanatophenyl)methanones and secondary amines

A convenient synthesis of N,N-disubstituted 4H-3,1-benzothiazin-2-amines from aryl(2-isothiocyanatophenyl)methanones using a two-pot procedure has been developed. Thus, treatment of these isothiocyanato ketones with secondary amines gave the corresponding keto thioureas, which were allowed to react with sodium borohydride or methylmagnesium bromide to afford 1,1-dialkyl-3-{2-[aryl(hydroxy)methyl]phenyl}thioureas or 1,1-dialkyl-3-[2-(1-aryl-1-hydroxyethyl)phenyl]thioureas, respectively, in one pot. Hydrobromic acid-mediated cyclization of these hydroxy thiourea precursors provided the desired 4H-3,1-benzothiazin-2-amines.     

A novel one-pot synthesis of hydroximoyl chlorides and 2-isoxazolines using N-tert-butyl-N-chlorocyanamide

Treatment of aldoximes with N-tert-butyl-N-chlorocyanamide gave hydroximoyl chlorides in quantitative yields in less than a minute, which on dehydrohalogenation in the presence of triethylamine gave the corresponding nitrile oxides. The nitrile oxides underwent 1,3-dipolar addition to dipolarophiles and gave 2-isoxazolines in excellent yields under mild conditions.     

A facile procedure for synthesis of 3-[2-(N,N-dialkylamino)ethyl]-3-fluorooxindoles by direct fluorination of N,N-dialkyltryptamines

A practical procedure for the synthesis of 3-fluorooxindole derivatives having basic amine moieties was developed, which involves Selectfluor™-mediated oxidative fluorination of N,N-dialkyltryptamines in the presence of Lewis acid. This procedure was applied to an antimigraine drug, rizatriptan, to afford the corresponding 3-fluorooxindole, which is a potential fluorine-containing drug candidate.     

A one-pot four-component synthesis of N-arylidene-2-aryl-imidazo[1,2-a]azin-3-amines

N-Arylidene-2-aryl-imidazo[1,2-a]azin-3-amines were synthesized via a one-pot, four-component condensation reaction using a 2-aminoazine, toluene-4-sulfonylmethyl isocyanide (TsCH₂NC), and two equivalents of readily available aromatic aldehydes. The reaction was performed in diethyl ether as the solvent, with p-toluenesulfonic acid (p-TSA) as the catalyst at reflux temperature.     

A novel, one-pot, three-component synthesis of 4H-pyrido[1,2-a]pyrimidines

A novel, one-pot, three-component synthesis of 4H-pyrido[1,2-a]pyrimidines is described. The reactive 1:1 zwitterionic intermediate, formed by the addition of isocyanides to dialkyl acetylenedicarboxylates, was trapped by N-(2-pyridyl)amides to produce the title compounds under mild reaction conditions in good yields.     

Radiation synthesis and characterization of poly(N,N-dimethylaminoethyl methacrylate-co-N-vinyl 2-pyrrolidone) hydrogels

In this study, radiation synthesis and characterization of swelling behavior and network structure of poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA), and poly(N,N-dimethylaminoethyl methacrylate-co-N-vinyl 2-pyrrolidone) (P(DMAEMA-co-VP)), hydrogels were investigated. PDMAEMA and P(DMAEMA-co-VP) hydrogels in the rod forms were prepared by irradiating the ternary mixtures of DMAEMA/VP/cross-linking agent, ethyleneglycol dimethacrylate (EGDMA), by gamma rays at ambient temperature. In composition ranges where the three components were completely miscible, water was also added to the ternary mixture to provide the formation of homogeneous polymerization and gelation. The influence of irradiation dose, comonomer, VP, and cross-linking agent, EGDMA, content on the total percentage gelation and monomer conversion were investigated. The effect of pH and temperature on the swelling behavior of hydrogels have also been examined. Hydrogels showed typical pH response and temperature responses, such as low-pH and low temperature swelling and high-pH and high temperature deswelling. Polymer-solvent interaction parameter (χ) and enthalpy and entropy changes appearing in the χ parameter for the P(DMAEMA-co-VP)-water system were determined by using Flory-Rehner theory of swelling equilibrium. The negative values for ΔH and ΔS indicate that prepared pure PDMAEMA and P(DMAEMA-co-VP) hydrogels have lower critical solution temperature (LCST) and Flory-Rehner theory of swelling equilibrium provides a satisfactory agreement to the experimental swelling data of the hydrogels.     

N-(Diethoxyphosphoryl)-O-benzylhydroxylamine—a convenient substrate for the synthesis of N-substituted O-benzylhydroxylamines

Easily available N-(diethoxyphosphoryl)-O-benzylhydroxylamine was shown to be convenient, orthogonally protected substrate for regioselective N-alkylation by means of diverse halides under basic conditions (sodium hydride/tetrabutylammonium bromide). An efficient procedure for dephosphorylation of N-substituted N-(diethoxyphosphoryl)-O-benzylhydroxylamine to provide N-substituted O-benzylhydroxylamines was also established.     

N,N-Dichlorobis(2,4,6-trichlorophenyl)urea (CC-2) as a new reagent for the synthesis of pyrimidone and pyrimidine derivatives via Biginelli reaction

A simple and efficient method for the synthesis of 3,4-dihydropyrimidin-2-(1H)one and benzo[4,5]imidazo/thioazo[1,2-a]pyrimidine derivatives has been described using N,N′-dichlorobis(2,4,6-trichlorophenyl)urea (CC-2) as a new reagent. This method is found to be efficient and convenient for the synthesis of pyrimidone and pyrimidine derivatives.     

l-Proline-catalyzed three-component domino reactions for the synthesis of highly functionalized pyrazolo[3,4-b]pyridines

A series of novel N,3-dimethyl-5-nitro-1-aryl-1H-pyrazolo[3,4-b]pyridin-6-amines was obtained in good yields from the domino reactions of N-methyl-1-(methylthio)-2-nitroprop-1-en-1-amine, aromatic aldehydes, and 3-methyl-1-aryl-1H-pyrazol-5-amines in ethanol in the presence of green catalyst, l-proline. This transformation involves the formation of two C–C and one C–N bonds leading to the creation of a six-membered ring in a one-pot operation.     

A novel and facile synthesis of 2-(cyclohexylamino)-6,7-dihydro-3-aryl-1H-indole-4(5H)-ones via a one-pot multi-component reaction

A simple and efficient synthesis of 2-(cyclohexylamino)-6,7-dihydro-3-aryl-1H-indole-4(5H)-ones was achieved via a one-pot multi-component reaction of cyclohexyl isocyanide, an aldehyde, a 1,3-dicarbonyl compound, and ammonium acetate in the presence of a catalytic amount of KHSO₄ in acetonitrile.     

Rational use of substituted N-allyl and N,N-diallylanilines in the stereoselective synthesis of novel 2-alkenyltetrahydro-1-benzazepines

Two new series of 1,4-epoxy-2-exo-vinyl(isopropenyl)tetrahydro-1-benzazepines and cis-2-vinyl(isopropenyl)-4-hydroxytetrahydro-1-benzazepines were prepared by an efficient three/four-step route from available substituted N,N-diallylanilines and mono N-allylanilines. The amino-Claisen rearrangement and the sequential oxidation/intramolecular 1,3-dipolar cycloaddition reactions were used as the key steps in this synthesis. All the synthesized compounds were fully characterized by IR, GC-MS and NMR techniques.     

N,N′-Diisopropyl- and N,N′-dicyclohexylthiourea cadmium(II) complexes as precursors for the synthesis of CdS nanoparticles

Crystals of the cadmium(II) complexes of N,N-diisopropylthiourea and N,N-dicyclohexylthiourea were obtained and their X-ray single crystal structures determined. These complexes are air-stable, easy to prepare and inexpensive and decompose cleanly to give good quality crystalline CdS. The nanoparticles of CdS thus obtained showed quantum confinement effects in their optical spectra, with close to band-edge emission in luminescence experiments. The broad diffraction patterns observed are typical of nanodimensional particles. The variation of concentration of precursor-to-HDA ratio change the isolated materials from spheres to rod-shaped. TEM images showed agglomerates of needle-like plate of particles.
