Kulokekahilide-2 is a potent cytotoxic depsipeptide isolated from the Hawaiian marine mollusk Philinopsis speciosa. The structure of kulokekahilide-2 was reported to be composed of five amino acids (l-Ala, l-Ile, MeGly, l-MePhe, d-Ala) and two hydroxy acids (d-Hica, 5S,6S,7S-Dtda); however, following its total synthesis, the 1H NMR spectrum of the synthetic compound was found to be different from that of the natural one, suggesting that the stereochemistry of the reported structure was incorrect. To determine the stereochemistry of the natural compound, arrays of analogues have been prepared using different sets of chiral amino acids, and the absolute stereochemistry of kulokekahilide-2 has been unambiguously confirmed to involve the combination 21-l-Ala, 24-d-MePhe, and 43-d-Ala.  相似文献

An attempt to carry out a total synthesis of kulokekahilide-2 (1) by macrolactonization of a seco acid prepared from a suitably protected hexapeptide and a dioxy acid moiety unexpectedly resulted in the formation of the 43-epimer (1a) of the cytotoxic depsipeptide, for which structure 1b has previously been proposed. A second attempt involving macrolactamization of the corresponding amino acid gave the target product, 1b, but the spectral data of the product did not match those of natural 1. Furthermore, neither 1a nor 1b showed any cytotoxicity, from which it is concluded that the structure of natural 1 is incorrect and should be re-examined.  相似文献

As part of our ongoing investigation of filamentous fungi for anticancer leads, an active fungal extract was identified from the Mycosynthetix library (MSX 55526; from the Order Sordariales). Bioactivity-directed fractionation yielded the known ergosterol peroxide (2) and 5α,8α-epidioxyergosta-6,9(11),22-trien-3β-ol (3), and a new benzoate trimer, termed thielavin B methyl ester (1). The structure elucidation of 1 was facilitated by the use of HRMS coupled to an APPI (atmospheric pressure photoionization) source. Compound 1 proved to be moderately active against a panel of three cancer cell lines.  相似文献

A series of novel 2-hydrazinylpyrido[2,3-b]pyrazin-3(4H)-one derivatives were synthesized and evaluated for their cytotoxic activities against A549,MDA-MB-231 and HT-29 cell lines in vitro.Pharmacological data indicated that compounds 5b,5c,10a and 10g possessed marked cytotoxicity,especially 10a(with IC50 values of 0.81,2.56 and 1.63μmol/L against A549,MDA-MB- 23 1 and HT29 cell lines,respectively),which had emerged as a lead compound.  相似文献

以硅胶为核,马来海松酸丙烯酸乙二醇酯和甲基丙烯酸为功能单体,采用涂覆悬浮聚合法合成了核壳型SiO2@松香基阳离子交换树脂(SiO2@RCER),研究了其对水中微量Cd2+的静态吸附性能。结果表明,在Cd2+溶液浓度为0.5 mg·L-1,pH=6.0,温度为303 K,吸附剂用量为10 g·L-1时,Cd2+去除率可达到100%;SiO2@RCER对Cd2+的吸附符合准二级动力学方程以及Langmuir吸附等温线,表明吸附过程为单分子层化学吸附。吸附前后材料的XPS图谱表明:SiO2@RCER对Cd2+吸附主要涉及Na+以及溶液中的Cd2+的阳离子交换。该吸附剂在对Cd2+、Pb2+浓度超标的矿区废水吸附处理后,高毒重金属浓度均达到国家生活饮用水水源水质一级标准(CJ3020-93),其他离子浓度也有所降低。  相似文献

Six homologous series of 4-(4′-alkoxy phenylazo) phenyl 4″-substituted benzoates (Ina-f) were prepared in which, within each homologous series, the length of the terminal alkoxy chain varies between 6, 8, 10, and 12 carbons, while the other terminal substituent, X, is a polar group that alternatively changed from CH3O, CH3, H, Br, NO2 and CN. Compounds prepared were characterised by spectroscopic methods, and their mesophase behaviour investigated by differential scanning calorimetry (DSC) and polarised optical microscopy (POM). The results were discussed in terms of mesomeric and polarisability effects. In each group of compounds, bearing the same alkoxy substituent, the nematic-to-isotropic transition temperatures (TC) were successfully correlated with the polarisability anisotropy of bonds to the substituent X. A comparative study was made between the investigated compounds and their previously prepared isomers, namely, 4-(4′-substituted phenylazo) phenyl 4″-alkoxybenzoates (IIna-f) in which the two terminal (alkoxy- and X) groups are exchanged.  相似文献

A concise and convergent total synthesis of the highly cytotoxic marine natural product apratoxin A is accomplished by an 18‐step linear sequence. The high sensitivity of the thiazoline, bearing an adjacent β‐hydroxyl group at the C35‐position, results in the assembly process requiring the inclusion of appropriate protecting groups and the careful optimization of all individual transformations. In the synthesis of 3,7‐dihydroxy‐2,5,8,8‐tetramethylnonanoic acid (Dtena), the three reagent‐controlled asymmetric reactions enables us to introduce four chiral carbon centers in a dihydroxylated fatty acid moiety. Formation of the hindered ester and sterically‐unfavorable N‐methylamide bonds were successfully demonstrated. The thiazoline in apratoxin A was constructed by Tf2O and Ph3PO‐mediated dehydrative cyclization, and final macrocyclization was achieved between N‐methylisoleucine and proline residues. Moreover, an oxazoline analogue and a C34 epimer of apratoxin A have also been elaborated in a similar approach. This synthetic route would enable assembly of other analogues differing in stereocenters of Dtena and their amino acids.  相似文献

Anion exchange behavior of 20 anionic chemical species was studied focusing on hydrochloric acid solutions. Elution characteristics for some of these anions were followed by using a basic anion exchange resin. Estimates of distribution coefficients in 10−3-9 mol/l acid were obtained by analytical determinations of these elements in column effluent solutions. The adsorption data observed in this medium suggests many applications in analytical separations.  相似文献

以离子交换树脂(D001)为载体,四乙烯五胺(TEPA)为改性剂,采用三种不同的方法制备了一系列固态胺吸附剂。采用N2吸附-脱附、傅里叶变换红外光谱(FT-IR)、热重分析(TGA)等手段对吸附剂进行表征。在固定床反应器中考察了TEPA负载量、吸附温度、进气流量和CO2分压等因素对CO2吸附性能的影响。结果表明,配位法制得的固态胺吸附剂分散性和稳定性较好,且在TEPA负载量为40%,吸附温度为65℃,进气流量为40 mL/min时有最大CO2吸附量达4 mmol/g。经过10次吸附-脱附循环实验后,CO2吸附量下降3.98%。热力学、动力学研究结果表明,CO2吸附是物理吸附和化学吸附的结果。  相似文献

Three new penta- and hexacoordinated tin compounds (1-3) were prepared from PhSnCl3, Ph2SnCl2 and Ph3SnOH and 3-methyl-2-hydroxy-2-cyclopenten-1-one (L). Compounds 1-3 were characterized by IR, mass spectra, elemental analysis, 1H, 13C, and 119Sn NMR. The ligand acts as a bidentate giving the tin esters and coordinating the tin by the carbonyl group. Compound 1 (PhSnCl2L · EtOH) has an hexacoordinated tin atom, with an octahedral distorted geometry, which is a stereogenic center. Compounds 2 (Ph2SnClL) and 3 (Ph3SnL) have pentacoordinated tin atoms. The structures were determined by X-ray diffraction analyses. In the solid state 1 presents a racemic pair, linked by strong hydrogen bonds and 2 and 3 “Berry exchange coordinate” geometry.  相似文献

Propellanes are polycyclic compounds in which tricyclic systems share one carbon–carbon single bond. Propellane frameworks that consist of larger sized rings are found in a variety of natural products. As an approach to the stereoselective synthesis of the propellane framework, one of the efficient methods is forming several rings in a single operation. Lapidilectine B (1) is composed of a propellane framework and was synthesized through the oxidative cyclization of trisubstituted alkenes. When the alkene with an ester moiety was treated with N-iodosuccinimide (NIS), iodocyclization proceeded to give the cyclic carbamate. On the other hand, when PhI(OAc)2 was allowed to react in the carboxyl form, a furoindolin-2-one structure corresponding to the A-B-C ring of lapidilectine B (1) was produced. Furthermore, when Pd(OAc)2 catalyst was used for cyclization under oxidative conditions, the product yield was improved.  相似文献

The lanthanide complexes LnL 2 (NO 3 ) 3 ( 3a-g ) are obtained where Ln is La, Sm, Yb, Er, Ce, Eu, Gd, and L is the diethyl ester of 2-dimethylamino-2-oxoethylphosphonic acid [(C 2 H 5 O) 2 P(O)CH 2 CON(CH 3 ) 2 ] 1 . They are characterized by elemental analysis, i.r. and NMR spectroscopy. The crystal structure of 3a is determined by single crystal X-ray diffraction. The complex is found to crystallize in the triclinic space group P 1 with a = 8.4220(17) Å, b = 11.123(2) Å, c = 17.560(4) Å, f = 87.20(3);, g = 82.27(3);, n = 76.89(3);, V = 1587.3(5) Å 3 , Z = 2, calcd = 1.614 mg/m 3 , R = 0.047, R w = 0.107, S = 1.034 for 5762 reflections with I > 2 (I). The structure contains monomeric units of the complex with the lanthanum atom coordinated by 10 oxygen atoms, six of them from the three bidentate nitrate ions and four from the two phosphonate ligands. The coordination is realized by both phosphoryl and amide-carbonyl oxygen atoms. The stereochemistry of the starting ligand 1 is investigated by NMR spectroscopy.  相似文献

Ag2CO3是一种典型的银基半导体,可在可见光照射下降解各种有机染料,但制备成本高,光腐蚀严重,稳定性差,难以循环利用等,因而限制了它的实际应用.针对这些问题,目前多数的改进措施是构建异质结,有效的分离光生电子与空穴来提高Ag2CO3的光催化性能.比如典型的异质结光催化剂有TiO2/Ag2CO3,Ag2CO3/ZnO,Ag2O/Ag2CO3和AgX/Ag2CO3等.也有在表面化学沉积,光化学还原Ag等贵金属形成等离子体等方式提高其光催化性能,但是很少通过特殊形貌控制以提高Ag2CO3的光催化性能.最近的研究表明,由于多尺度微球结构催化剂具有高效的光捕能力,同时具有比表面积大、易沉降,良好的物质传输能力和表面的渗透性,因而在液相光催化反应中具有明显的优势.因此,我们期望制备出一个多尺度微球结构Ag2CO3光催化剂.CaMg(CO3)2是一种具有微球结构的半导体,它与Ag2CO3有相同的阴离子结构,但是两者在水溶液中的溶解度相差较大,利用这个特性理论上可以将两个不同的半导体结合在一起,得到一种新型的复合微球.本文以CaMg(CO3)2微球为硬模板,通过简单的离子交换成功制备了粒径约为10μm的CaMg(CO3)2@Ag2CO3微球.利用X射线衍射、N2物理吸附、扫描电镜、傅里叶变换红外光谱和紫外-可见漫反射吸收光谱、光电流等手段对在不同反应时间与温度下制得的CaMg(CO3)2与Ag2CO3的复合物进行了表征.结果表明,在40°C下Ag+与Ca2+、Mg2+离子交换4 h后,得到了一种多尺度CaMg(CO3)2@Ag2CO3复合微球.此时,微球中Ag2CO3的含量约为2.56%.结果表明,这种具有多尺度结构的复合微球能够增强可见光的吸收.电化学阻抗测试和光电流测试表明,CaMg(CO3)2核的存在可以降低光生载流子的迁移阻力,进而促进光生电子与空穴的分离.在光降解酸性橙II的测试中,核壳结构的CaMg(CO3)2@Ag2CO3复合微球表现出了更高的催化活性,而且具有更好的循环使用性能.同时,相对于纯Ag2CO3光催化剂来说,CaMg(CO3)2@Ag2CO3复合微球制备的成本大幅度降低.ESR测试证明了·OH为CaMg(CO3)2@Ag2CO3复合微球光催化过程中的主要活性物质.  相似文献

正A series of 2,6-disubstituted-4-morpholinothieno[3,2-d]pyrimidine derivatives were synthesized and their cytotoxic activity against H460,HT-29,MDA-MB-231,U87MG and H1975 cancer cell lines were evaluated in vitro.Most of the target compounds exhibited moderate to excellent activity to the tested cell lines.The most promising compound 23(0.84μmol/L,0.23μmol/L, 2.52μmol/L,1.80μmol/L) was 1.0,2.9,29.3 and 4.3 times more active than GDC-0941(0.87μmol/L,0.66μmol/L,73.8μmol/L, 7.77μmol/L) against H460,HT-29,MDA-MB-231 and U87MG cell lines,respectively.  相似文献
