Rapid and sensitive indirect competitive enzyme‐linked immunosorbent assays (ic‐ELISA) and gold nanoparticle immunochromatographic strip tests were developed to detect thiamphenicol (TAP), florfenicol (FF) and chloramphenicol (CAP) in milk and honey samples. The generic monoclonal antibody for TAP, FF and CAP was prepared based on a hapten [D ‐threo‐1‐(4‐aminophenyl)‐2‐ dichloroacetylamino‐1,3‐propanediol], and the haptenwas linked to a carrier protein using the diazotization method. After the optimization of several parameters (coating, pH, sodium chloride content and methanol content), the ic‐ELISA was established. The quantitative working range for TAP was 0.11–1.36 ng/mL, with an IC50 of 0.39 ng/mL. The optimized ELISA showed cross‐reactivity to CAP (300%) and FF (15.6%), with IC50 values of 0.13 and 2.5 ng/mL, respectively. The analytical recovery of TAP, FF and CAP in milk and honey samples in the ic‐ELISA ranged from 81.2 to 112.9%. Based on this monoclonal antibody, a rapid and sensitive immunochromatographic test strip was also developed. This strip had a detection limit of 1 ng/mL for TAP, FF and CAP in milk and honey samples. Moreover, the test was completed within 10 min. Our results showed that the proposed ic‐ELISA and immunochromatographic test strip method are highly useful screening tools for TAP, FF and CAP detection in milk and honey samples. A polyclonal antiserum to streptomycin was generated in sheep using a streptomycin-bovine serum albumin conjugate as immunogen. Streptomycin was linked to the carrier protein with cyanuric chloride using a new two-step conjugation method. Plate coating antigen conjugates of streptomycin and gelatine were prepared using either cyanuric chloride (homologous bridge) or 1,4-butanediol diglycidyl ether to provide an heterologous complex. The reagents enabled the generation of a specific antiserum with a titre of 1/40,000 and the development of a sensitive ELISA method suitable for the measurement of streptomycin sulfate in milk, serum and water samples. A minimum detection value of 1 ng mL(-1) and a dynamic range of 1 to 200 ng mL(-1) were demonstrated in the three matrices. No detectable cross reactivity with any of the common aminoglycosides was found except the related dihydrostreptomycin which gave a 75% cross reaction value. The details of the preparation of the hapten-protein conjugates, characterisation of the antiserum and assay construction and assessment methods are presented. The introduction of new coupling methods and antibody assessment provide improved basic methodologies necessary for the advancement of immuno-analysis of streptomycin, one of the most widely used antimicrobial substances.  相似文献

Two rapid,sensitive and reliable immunoassay methods,namely competitive indirect enzyme-linked immunosorbent assay(CI- ELISA)and colloidal gold-based immunochromatographic assay(CGIA),were developed to detect ofloxacin(OFL).The linear range of the CI-ELISAwas from 0.5 to 128 ng/mL with a limit of detection(LOD)of 0.35 ng/mL.Good recoveries were obtained in analyzing simulated swine urine samples.The CGIA could accurately estimate OFL at concentrations as low as 10 ng/mL in less than 10 min,and test results were read visually without any instrument.  相似文献

We have developed a heterologous direct competitive enzyme-linked immunosorbent assay (ELISA) and a visual colloidal gold-based immunochromatographic assay (CGIA) for simultaneous determination of ofloxacin, marbofloxacin, and fleroxacin residues in milk using polyclonal antibodies. The half-maximum inhibition concentrations (IC50) of ofloxacin, marbofloxacin, fleroxacin, and limits of detection (LODs; calculated as IC15 values) are between 0.20 and 0.53?ng mL?1, and between 0.02 and 0.05?ng mL?1, respectively. The average recoveries range from of 78% to 113%, and the coefficients of variation of intra- and inter-assays are between 2 and 11%, and 3 to 19%, respectively. The LODs for ofloxacin, marbofloxacin, fleroxacin in milk are between 3.5 and 8.9?ng mL?1. The visual minimum detection limit of the optimized CGIA is 2?ng mL?1 for milk samples. The detection process can be completed within 10?min. The strips can be stored at 4?°C for 8?weeks without significant loss of activity. The results of the analysis of spiked samples showed that the CGIA can be applied to preliminary, fast, and on-site screening of milk samples. The ELISA and CGIA allow for a rapid, sensitive, and low-cost determination of (fluoro)quinolones residues in milk samples.

Due to the widespread use and potential toxicity of avermectins (AVMs), multi-residue monitoring of AVMs in edible tissues, especially in milk, has become increasingly important. With the aim of developing a broad-selective immunoassay for AVMs, a broad-specific monoclonal antibody (Mab) was raised. Based on this Mab, a homologous indirect enzyme-linked immunosorbent assay (ELISA) for the rapid detection of AVMs in milk was developed. Under the optimized conditions, the IC?? values in assay buffer were estimated to be 3.05 ng/mL for abamectin, 13.10 ng/mL for ivermectin, 38.96 ng/mL for eprinomectin, 61.00 ng/mL for doramectin, 14.38 ng/mL for emamectin benzoate. Detection capability (CCβ) of the ELISA was less than 5 ng/mL and 2 ng/mL in milk samples prepared by simple dilution and solvent extraction, respectively. The optimized ELISA was used to quantify AVMs in milk samples spiked at different amounts. The mean recovery and coefficient of variation (CV) were 95.90% and 15.42%, respectively. The Mab-based ELISA achieved a great improvement in AVMs detection. Results proved this broad-selective ELISA would be useful for the multi-residue determination of AVMs in milk without purification process.  相似文献

A new CE method with fluorescence detection is reported on the determination of ofloxacin in milk samples using graphene quantum dots (GQDs) for sensitivity enhancement. Injection of GQDs prior the standards/sample is crucial to increase the antibiotic fluorescence response. Clean‐up and preconcentration steps allowed for a good linear correlation in a concentration range between 50 and 1000 ng/mL for the ofloxacin, detection and quantification limits being 10.7 and 35.5 ng/mL, respectively. Optimal CE conditions for the seven‐fluoroquinolone separation method were assessed in terms of buffer type, pH, and voltage. The selective interaction of GQDs with ofloxacin as model analyte was subsequently studied finding a significant sensitivity improvement; therefore, the analytes would be detected at low concentrations by means of a commercial CE device equipped with a multi‐wavelength photoluminescence detector. Due to the different maximum emission wavelengths of the target compounds and the limitations shown by the single‐wavelength photoluminescence detector coupled to the CE system, we demonstrated the usefulness of the GQD‐assisted sensitivity‐enhanced CE method to determine ofloxacin in milk samples. This work opens an interesting possibility of using GQDs in separation techniques combined with photoluminescence detectors for lowering sensitivity levels typically provided by the mere device.  相似文献

Based on the enhancement of chemiluminescence (CL) of luminol-hydrogen peroxide-gold nanoparticles system by fluoroquinolones (FQs), a novel and rapid CL method is reported for the determination of FQs derivatives. Under the optimum conditions, the CL intensity is proportional to the concentration of FQs derivative in solution. The corresponding linear regression equations are established over the range of 0.08-1.28 μg/mL for norfloxacin, 0.013-1.32 μg/mL for ciprofloxacin, 0.014-1.4 μg/mL for lomefloxacin, 0.029-1.46 μg/mL for fleroxacin, 0.02-1.0 μg/mL for ofloxacin and 0.01-1.44 μg/mL for levofloxacin, respectively. The limits of detection (S/N = 3) are 3.2, 9.5, 7.0, 9.0, 8.0, and 8.0 ng/mL with the relative standard deviation (n = 11) 4.3, 1.5, 1.9, 1.3, 1.6 and 2.1% for norfloxacin, ciprofloxacin, lomefloxacin, fleroxacin, ofloxacin and levofloxacin, respectively. This proposed method has been applied to detect FQs derivatives in human urine successfully.  相似文献

采用微波辅助合成的荧光稀土二氧化硅纳米颗粒(BHHCT-Eu3+@SiO2)为标记物,建立了快速定量检测卡那霉素(Kana)残留的荧光免疫层析方法.实验结果表明,微波辅助合成的BHHCT-Eu3+@SiO2纳米颗粒呈球形,粒径约36 nm,具有良好的荧光发射性能,最大吸收波长和最大发射波长分别为343和615 nm.将BHHCT-Eu3+@SiO2与卡那霉素抗体(Kana-ab)通过醛基化葡聚糖交联,合成了荧光标记抗体Eu3+-Kana-ab,结合定量侧向层析读数仪,建立了牛奶中Kana残留的快速定量检测方法,对Kana的检出限(IC10)为0.85 ng/mL,半数抑制浓度(IC50)为12.76 ng/mL,检测范围(IC20-IC80)为3.0~76.0 ng/mL,牛奶中的Kana的加标回收率范围为93.7%~97.4%,RSD为3.1%~4.6%,与Kana类似物的交叉反应均<1%.牛奶中Kana残留的测定结果与ELISA方法相关性良好.  相似文献

A novel chemiluminescence method for the determination of levofloxacin is presented, which is based on the inhibitory effect of levofloxacin on the chemiluminescence reaction between luminol and myoglobin in a flow-injection system. The decrement of chemiluminescence intensity is linear with the logarithm of levofloxacin concentration over the range from 0.07 to 100.0 ng/mL (r 2 = 0.9994), with the detection limit of 0.02 ng/mL (3σ). At a flow rate of 2.0 mL/min, a complete analytical process could be performed within 0.5 min, including sampling and washing, with a relative standard deviation of less than 3.0% (n = 5). The proposed procedure was applied successfully to the determination of levofloxacin in pharmaceutical preparations, human urine and serum without any pretreatment procedure.  相似文献

Abstract

Different ELISA techniques have been developed for the detemination of four widely used pesticides: 2,4-dichlorophenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), simazine and atrazine. Dependences between the assay scheme and the limiting detectable concentration of the pesticide were studied. The cases of preferential applying of the scheme with immobilized antibodies or one with immobilized pesticide-protein conjugate have been revealed. The following approaches resulting in lowering of ELISA sensitivity were proposed: preliminary incubation of the tested sample with antibodies, immobilization of antibodies via staphylococcal protein A, usage of monovalent fragments of antibodies instead of native ones and chemical modification of the pesticide molecules in the sample. Optimal combinations of these approaches permitted to lower the detection limit of the assays in about 5–30 times. The achieved sensitivities were 3 ng/mL for 2,4-D, 5 ng/mL for 2,4,5-T, 0.05 ng/mL for simazine, and 0.1 ng/mL for atrazine, being acceptable for purposes of ecological monitoring.  相似文献

Papaverine (1-(3,4-dimethoxybenzyl)-6,7-dimethoxyisoquinoline, PAP) is a member of the benzylisoquinoline sub-group of the opium alkaloids. It has been widely used for treating diseases like pulmonary arterial embolism and renal or biliary colic. In this paper, a specific conjugate of mono-demethylated papaverine-O-carboxylmethyl ether (MDMPAP-O-CME) and bovine serum albumin (BSA) was synthesized and used as the complete antigen (PAP-BSA), with which we successfully obtained a high-titer anti-PAP polyclonal antibody (pAb) by immunization of rabbits. The anti-PAP pAb showed high affinity to papaverine with an affinity constant (Kaff) of 7.3 × 107 L/mol. With this antibody, we established a sensitive immunochemical method for the determination of papaverine based on indirect competitive enzyme-linked immunosorbent assay (ELISA). The optimal concentrations of the coated antigen (PAP-OVA) and purified pAb used in the ELISA were 5 and 1.2 μg/mL, respectively. The cross reactivity of other benzylisoquinoline derived substances, including 1-(3,4-dihydroxybenzyl)-7-hydroxy-6-methoxy-isoquinoline (6-methoxy-papaveroline, MPAPO), morphine (MP) and codeine (CD) were all lower than 1%. The linear range of the calibration curve was 0.1-1000 ng/mL. Normal human serum samples were spiked with known amount of papaverine and measured by the ELISA. Recoveries were between 102% and 105%. Papaverine content in a commercial papaverine hydrochloride injection sample was also determined using the established ELISA. Compared with the results given by the control experiment of HPLC, the recoveries of ELISA to detect injection samples were 102-110%. The limits of detection for synthetic serum samples and injection samples of papaverine hydrochloride were 0.25 and 0.06 ng/mL, respectively.  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed to determine the presence of egg proteins in foods. The polyclonal antibodies developed were specific to whole egg proteins and did not cross-react with any of the 38 nuts, legumes, or other common food ingredients tested. The concentrations of egg proteins that will inhibit 50% of antibody-antigen binding, IC50, were 3-7 ng/mL, and the linear range was 0.5-62.5 ng/mL. The detection limit was 0.2 ppm for various foods. Recoveries ranged from 67 to 96%. The intra- and inter-assay coefficients of variation in this procedure were 10-13% for ice cream spiked at 0.8 and 1.6 ppm. The ELISA has been applied to ice creams, noodles, pasta, and breads. Egg proteins were identified in all declared egg products, and no false positives were found.  相似文献

A rapid and sensitive biosensor immunoassay was developed for determination of ivermectin residues in bovine milk. A detection limit of 16.2 ng/mL was achieved. A Biacore optical biosensor based on surface plasmon resonance was used, and a range of extraction techniques was investigated. In the final assay procedure, ivermectin was extracted with acetonitrile followed by C8 solid-phase extraction cleanup. It was proven experimentally that 2 methods of milk storage, freezing or addition of mercury-containing compounds as preservatives, could be used without considerable change in detected concentrations (samples were fortified with ivermectin after storage). The average values for milk samples spiked at 100 and 50 ng/mL concentrations were 102.6 and 51.5 ng/mL, respectively. Extraction and analysis of 20 milk samples were performed within a single working day.  相似文献

Diethylstilbestrol (DES) is a synthetic estrogen that has ever been used worldwide. Polyclonal antibodies (PAbs) were used in immunoassay for detection of DES residues in environmental and agricultural samples in previous paper. In this paper, an indirect competitive enzyme-linked immunosorbent assay (icELISA) was developed based on monoclonal antibody (MAb) for the determination of diethylstilbestrol. Mono-o-carboxypropyldiethylstilbestrol (DES-CP) and mono-o-carboxymethyldiethylstilbestrol (DES-CME) were synthesized to be haptens. DES-CP was coupled to bovine serum albumin (BSA) to be an immunogen in BALB/c female mouse for MAb production. The MAb was characterized for specificity and affinity to DES in icELISA. Under the optimum condition, the icELISA showed an ICs0 of 9.8 ng/mL, the limit of detection (IC20) of 2.3 ng/mL and a working range of 2-42 ng/mL. Hexestrol and dienestrol exhibited cross-reactivity values were 44% and 27%, respectively. Cross-reactivity of natural estrogen 17β-estradiol was less than 0.1%. The influences of some factors such as salt concentration, pH and organic solvent concentration on the assay were evaluated. The concentrations of DES in the fortified water samples determined by the assay were correlated well with the fortification levels. The results were conf'm'ned with analysis by HPLC.  相似文献

A competitive enzyme-linked immunosorbent assay (ELISA) was developed for the quantitative detection of the diethylstilbesterol (DES). Polyclonal rabbit antisera, raised against protein conjugate diethylstilbesterol-mono-caroxyl-propyl-ethyl-bovine-serum-albumin (DES-MCPE-BSA), were utilized in immobilized antibody-based and competitive immunoassays. Assay conditions, including concentrations of antisera and horseradish peroxidase, (HRP)-DES, were optimized. The effects of incubation time, surfactant concentration, ionic strength and pH of the medium were also investigated. The typical calibration curve gave an average IC(50) value of 2.4 ng/mL, calibration range from 0.2 to 30.5 ng/mL and a detection limit of 0.07 ng/mL. The specificity of the assay was tested against DES structurally related compounds, and the assay proved highly selective for DES. Assay performance was validated using spiked chicken meat and liver tissue samples. Moreover, it was compared with liquid chromatography-tandem mass spectrometry. The ion pair for quantification of DES was m/z 267.4/251.4, and the linear equation of DES was y = 0.1033x + 0.0126 (r = 0.9960). The two analytical methods can be applied to monitor DES and other steroid residues in foods.  相似文献

The yolk protein precursor vitellogenin (Vtg) in plasma has proved to be a simple and sensitive biomarker for assessing exposure of fish to environmental estrogens. Within international bodies such as the Organization for Economic Cooperation and Development (OECD) work is ongoing to develop screening and testing programmes for endocrine disrupting effects of new chemicals, and in the focus of this development are the fish test species common carp (Cyprinus carpio), fathead minnow (Pimephales promelas), zebrafish (Danio rerio) and Japanese medaka (Oryzias latipes). In this study we have developed quantitative enzyme linked immunosorbent assays (ELISAs) for Vtg in common carp/fathead minnow, zebrafish and Japanese medaka. The assays were developed using a combination of monoclonal and polyclonal fish Vtg antibodies in a sandwich format, using stabilized Vtg from the test species as a standard. The carp Vtg ELISA has a working range of 1–63 ng/mL, a minimal detection limit of 0.6 ng/mL, and may also be used for quantification of Vtg in fathead minnow. In fathead minnow whole-body homogenate samples, the practical detection limit is 400 ng/mL due to the matrix effect. The zebrafish Vtg ELISA has a working range of 0.5–63 ng/mL, a minimal detection limit of 0.4 ng/mL, and a practical detection limit of 200 ng/mL in whole-body homogenate samples. The medaka Vtg ELISA has a working range of 0.25–16 ng/mL, a minimal detection limit of 0.1 ng/mL, and a practical detection limit of 125 ng/mL in whole-body homogenate samples. The intra- and inter-assay variations were below 20% for all assays. The assays were evaluated with sets of representative samples spanning the wide dynamic range of Vtg-levels found in fish exposed to environmental estrogens, and all three assays are currently undergoing international inter-laboratory validation.  相似文献

Snake venom contains bioactive materials for drug development, diagnosis, and treatment. After separating and purifying the kallikrein-like enzyme (AHP-Ka) from Agkistrodon halys pallas venom for the first time, a monoclonal antibody against AHP-Ka was prepared and characterized. An indirect sandwich enzyme-linked immunosorbent assay (ELISA) based on the monoclonal antibody was developed and validated for the pharmacokinetic analysis of AHP-Ka in rat plasma. The method was calibrated using rat plasma and 1:100 dilution of plasma was selected to prepare a calibration curve to validate the precision, accuracy, and stability of the ELISA method. A good linear relationship was obtained in a working range from 3.9 ng/mL to 62.5 ng/mL with a limit of detection of 2.94 ng/mL. Intra- and inter-batch precision were less than 10%. The average recovery ranged from 94.6% to 104.4% in rat plasma at the concentrations of 5 ng/mL, 15 ng/mL, and 45 ng/mL, respectively. The ELISA method was successfully used for the pharmacokinetic study of AHP-Ka in Sprague-Dawley rat plasma after intravenous administration. The work is expected to contribute to future preclinical development of AHP-Ka.  相似文献

Four ELISA formats, antigen-coated indirect, antigen-coated direct, antibody-coated, and the secondary antibody-coated, were developed using monoclonal antibody to determine cephalexin. Results showed that the secondary antibody-coated method of ELISA had a better performance in the establishment of standard curves. The optimized secondary antibody-coated ELISA was used to determine cephalexin spiked in pig muscle, pig kidney, pig liver, chicken muscle, chicken liver, and cow's milk. The limits of detection were 0.09 ng/g, 0.15 ng/g, 0.26 ng/g, 0.13 ng/g, 0.19 ng/g, and 0.08 ng/mL in pig muscle, pig kidney, pig liver, chicken muscle chicken liver, and cow's milk, respectively. A mean recovery of 77.2–128.5% and coefficient of variation of 2.6–14.7% were obtained. The results given by the ELISA method were in agreement with those of the LC-MS/MS method, which confirmed the potential of the ELISA method for the monitoring of cephalexin in milk and animal tissues.  相似文献

A rapid CE coupled with potential gradient detection (PGD) for the separation and detection of four quinolones, namely, enoxacin, ofloxacin (OFL), fleroxacin, and pazufloxacin, was described. Separation was performed in a fused-silica capillary (75 microm x 8.5 cm) using a buffer of 30 mM Tris and 4 mM phosphoric acid at pH 8.9. Under the separation voltage of 3 kV, the quinolones were separated within 2.8 min with good linearity (r(2) >or= 0.985). The method was successfully applied in determining OFL in a pharmaceutical formulation. Also, a liquid-liquid extraction (LLE) method was developed and coupled to CE-PGD in determining quinolones that spiked in milk samples. With dichloromethane and hexane for enrichment and purification, the LLE recoveries of the four quinolones were in the range of 77-106%. The detection limits of the quinolones with LLE-CE-PGD were from 23 to 65 ng/mL. The proposed CE-PGD method was validated with an HPLC method, and the results indicated consistency between the two methods.  相似文献
