A fluorimetric ion sensor based on fiber optics has been developed that employs Rhodamine 6G hydrophobically and electrostatically "trapped" on a Nafion film. The sensor is based on the measurement of quenching or enhancement of the Rhodamine 6G fluorescence by various ions. It was found that ions such as Co(2+), Cr(3+), Fe(2+), Fe(3+), Cu(2+), Ni(2+) and NH(+)(4) rapidly quench the Rhodamine 6G fluorescence at an initial rate that depends on the concentration of the ion. This quenching is then readily reversed by the addition of "reverser" ions such as H(+), Li(+), Na(+), K(+), Ba(2+), Ca(2+), Mn(2+), Zn(2+) and Mg(2+). Again, the initial rate for the attainment of the original fluorescence was found to depend on the concentration of the reverser ion. Therefore, by monitoring the quenching directly the concentration of quencher ions can be determined. In addition, by loading the film with quencher and monitoring the initial rate of return towards the original baseline signal, it is possible to determine non-quenching ions.  相似文献

The fluorescence of conjugated polyelectrolytes (CPEs) is quenched with very high efficiency by small molecule quenchers. This effect has been referred to as amplified quenching. In the present communication, we demonstrate that aggregation of a poly(phenylene ethynylene)-type CPE (PPE-CO2-) induced by Ca2+ has a pronounced effect on the amplified quenching of the polymer by the dication methyl viologen (MV2+). In particular, absorption and fluorescence spectroscopy of PPE-CO2- in methanol solution indicate that addition of a low concentration of Ca2+ induces aggregation of the polymer chains. The range of MV2+ concentrations within which linear Stern-Volmer quenching behavior is observed systematically decreases with increasing Ca2+ concentration to a point where superlinear quenching is observed immediately upon addition of MV2+. This finding is unequivocal evidence that the superlinear Stern-Volmer quenching behavior typically observed in CPE-quencher systems arises due to quencher-induced aggregation of the CPE chains.  相似文献

运用荧光淬灭技术,包括稳态荧光淬灭法(SSFQ)和时间分辨荧光淬灭法(TRFQ),研究了疏水缔合水溶性丙烯酰胺2苯氧基丙烯酸酯多嵌段共聚物[P(AM POEA)]在水溶液中自组装的聚集数.这类聚合物在水溶液中易形成胶束状聚集体,探针芘分子和淬灭剂二苯酮增溶于疏水微区,荧光测定结果很好地符合Poisson淬灭模型.实验结果表明聚合物链结构、聚合物浓度和无机盐对聚集体的尺寸具有重要影响.聚合物自组装聚集数NA随疏水单体含量的增加和疏水嵌段长度的减小而增大,同时也随聚合物浓度和NaCl浓度增加而增大.另外对聚合物链结构、聚集数和溶液粘度的相互关系进行了讨论.  相似文献

Aqueous solutions of anionic surfactants Cl(C3F6O)nCF2COOX, consisting of n = 2 and 3 perfluoroisopropoxy units and the counterion X = Na+ or NH4+, were studied by the method of fluorescence quenching with the use of (1-pyrenylbutyl)trimethylammonium bromide as a luminophore, and 1,1'-dimethyl-4,4'bipyridinium dichloride (methyl viologen) as a quencher. From the kinetics of fluorescence decay (time-resolved experiments) micellar aggregation numbers, N, and rate constants of the intramicellar quenching were determined for a wide range of surfactant concentrations, on the basis of the model developed by Infelta and Tachiya. The results are discussed in terms of the shape of the aggregates and the degree of counterion binding. The most important conclusions include: (i) a significant increase of N with increasing surfactant concentration suggests that spherical micelles formed at critical micellar concentration (CMC) transform into ellipsoidal aggregates, (ii) the degree of counterion binding to micelles is higher for NH4+ than for Na+, leading to higher N values in the case of the ammonium salt (n = 2), and (iii) at concentrations close to CMC the longer chain surfactant (n = 3) forms loose aggregates suggesting significant permeation with water molecules. An additional finding of this study is that the micelle-bound luminophore and quencher can form a ground-state complex, and for this reason the N values cannot be evaluated properly from the steady-state fluorescence intensity data using the equation proposed by Turro and Yekta.  相似文献

This study investigates the fluorescence quenching of a poly(p-phenyleneethynylene) (1) based polyelectrolyte by positively charged and neutral macromolecules. This work shows that the change in the fluorescence yield of 1 depends on a number of factors, including electrostatic, hydrophobic, and energy transfer interactions with the quencher and also changes in the solution conditions such as concentration and ionic strength. The fluorescence quenching is attributed to the formation of aggregates that form upon addition of different quenchers to a solution of 1 and/or the solution conditions. The extent of 1's aggregation is shown to depend on the type of interaction between the polymer and the quencher, the concentration of the polymer, and the ionic strength of the solution.  相似文献

Dansyl-anthracene dyads 1 and 2 in CH(3)CN-H(2)O (7:3) selectively recognize Cu(2+) ions amongst alkali, alkaline earth and other heavy metal ions using both absorbance and fluorescence spectroscopy. In absorbance, the addition of Cu(2+) to the solution of dyads 1 or 2 results in appearance of broad absorption band from 200 nm to 725 nm for dyad 1 and from 200 nm to 520 nm for dyad 2. This is associated with color change from colorless to blue (for 1) and fluorescent green (for 2). This bathochromic shift of the spectrum could be assigned to internal charge transfer from sulfonamide nitrogen to anthracene moiety. In fluorescence, under similar conditions dyads 1 and 2 on addition of Cu(2+) selectively quench fluorescence due to dansyl moiety between 520-570 nm (for 1)/555-650 nm (for 2) with simultaneous fluorescence enhancement at 470 nm and 505 nm for dyads 1 and 2, respectively. Hence these dyads provide opportunity for ratiometric analysis of 1-50 μM Cu(2+). The other metal ions viz. Fe(3+), Co(2+), Ni(2+), Cd(2+), Zn(2+), Hg(2+), Ag(+), Pb(2+), Li(+), Na(+), K(+), Mg(2+), Ca(2+), Ba(2+) do not interfere in the estimation of Cu(2+) except Cr(3+) in case of dyad 1. The coordination of dimethylamino group of dansyl unit with Cu(2+) causes quenching of fluorescence due to dansyl moiety between 520-600 nm and also restricts the photoinduced electron transfer from dimethylamino to anthracene moiety to release fluorescence between 450-510 nm. This simultaneous quenching and release of fluorescence respectively due to dansyl and anthracene moieties emulates into Cu(2+) induced ratiometric change.  相似文献

Monitoring the levels of potentially toxic metal (PTM) ions (e.g., Hg(2+), Pb(2+), Cu(2+)) in aquatic ecosystems is important because these ions can have severe effects on human health and the environment. Gold (Au) nanomaterials are attractive sensing materials because of their unique size- and shape-dependent optical properties. This review focuses on optical assays for Hg(2+), Pb(2+), and Cu(2+) ions using functionalized Au nanomaterials. The syntheses of functionalized Au nanomaterials are discussed. We briefly review sensing approaches based on changes in absorbance resulting from metal ion-induced aggregation of Au nanoparticles (NPs) or direct deposition of metal ions onto Au NPs. The super-quenching properties of Au NPs allow them to be employed in 'turn on' and 'turn off' fluorescence approaches for the sensitive and selective detection of Hg(2+), Pb(2+), and Cu(2+) ions. We highlight approaches based on fluorescence quenching through analyte-induced aggregation or the formation of metallophilic complexes of Au nanodots (NDs). We discuss the roles of several factors affecting the selectivity and sensitivity of the nanosensors toward the analytes: the size of the Au nanomaterial, the length and sequence of the DNA or the nature of the thiol, the surface density of the recognition ligand, and the ionic strength and pH of the buffer solution. In addition, we emphasize the potential of using new nanomaterials (e.g., fluorescent silver nanoclusters) for the detection of PTM ions.  相似文献

The effects of several biologically important inorganic salts, including NaCl, NaI, NaBr, KCl, MgCl2, MgSO4 and CaCl2 on the electronic absorption and fluorescence spectra of Merocyanine 540 (MC-540) have been investigated in aqueous media at 25 degrees C. Depending on both the MC-540 concentration and the nature of salt, a new absorption band appears at about 515 nm, above the critical salt concentration (CSC), corresponding to salt-induced MC-540 aggregation. Several types of MC-540 fluorescence quenching by the salts are observed, according to their cationic charge and the nature of anion: in the case of monovalent ions (Na+, K+), a non-linear Stern-Volmer behaviour is observed, indicating variable contributions of dynamic and static quenching mechanisms, whereas for divalent alkaline-earth (Mg2+, Ca2+) ions, linear Stern-Volmer relationships are obtained. Using these results, an analytical quenchofluorimetric approach is proposed for the determination of magnesium ions.  相似文献

The quenching of the fluorescence of HPTS (1-hydroxypyrene-3,6,8-trisulfonate) by Cu(2+), Ni(2+), Co(2+), I(-), and CP(+) (cetylpyridinium cation) has been studied in the w/o microemulsion medium formed with water, AOT [sodium salt of bis (2-ethylhexyl) sulfosuccinic acid], and heptane as components at two [H(2)O]/[AOT] ratios (omega), 6 and 20. The quenching process has been found to be dynamic in nature. The lifetimes of HPTS in the microemulsion medium in the absence and in the presence of quencher have been determined. The analysis of the results has been performed in terms of the Stern-Volmer equation and the quenching sphere of action model. The Poisson distribution equation has been also used in the analysis of the probability of quencher distribution in the microemulsion compartment. The quenching of HPTS has been found to be much lower in microemulsion than in bulk water.  相似文献

To mimic and measure calcium ion-mediated carbohydrate-carbohydrate interactions, four lactose derivatives have been synthesized for assembly on gold nanoparticles. The series of lactose derivatives varied by the length of the thiolated ethylene glycol anchor chain [O(CH2CH2O)(m)CH2CH2SH; where m = 0, 1, 2, and 3] used to self-assemble the carbohydrates to the preformed gold nanoparticles of ca. 16 nm diameter. Upon addition of calcium ions to the lactose-stabilized nanoparticles, rapid carbohydrate-carbohydrate interactions were visualized and subsequently measured using UV-visible spectrometry and transmission electron microscopy (TEM). The nanoparticle aggregates formed via metal-mediated carbohydrate-carbohydrate interactions could be readily redispersed through the addition of EDTA. Multiple reaggregation and redispersion cycles were achieved, confirming that the aggregation process was due to metal ion-mediated carbohydrate interactions rather than calcium chelation by residual citrate ions on the particle surface. The essential involvement of the lactose moiety in Ca2+ complexation was shown by control measurements on related D-glucose-derivatized nanoparticles, where a significantly reduced aggregation response was obtained only at high ion concentrations. Other group 2 metal ions with radii larger than that of calcium, viz., barium and strontium, were also shown to mediate the aggregation of the lactose-stabilized nanoparticles. The induced aggregation of the lactose nanoparticles was determined to be quantitatively dependent upon the calcium ion concentration. Furthermore, the analytical sensitivity of the calcium-induced aggregation and the linear dynamic range were dependent on the length of the ethylene glycol anchor chain. The shortest ethylene glycol chain (m = 0) gave the most sensitive response with the optimum limit of detection (0.8 mM Ca2+), whereas the longest ethylene glycol chain (m = 3) provides a measurement of calcium ion concentration over the largest linear dynamic range (10-35 mM Ca2+). This work has shown that the self-assembled deposition of lactose derivatives on gold nanoparticles provides multivalent carbohydrate surfaces that can be used as mimics for the measurement of biologically relevant carbohydrate-carbohydrate interactions. Additionally, this study has highlighted the importance of the structure and length of the ligand that anchors the carbohydrate sugar to the gold particle surface to facilitate such carbohydrate interactions and for "tuning" the analytical characteristics of bioassays developed using metal nanoparticle technology.  相似文献

The dynamics of fluorescence quenching of a conjugated polyelectrolyte by a cyanine dye are investigated by femtosecond fluorescence up-conversion and polarization resolved transient absorption. The data are analyzed with a model based on the random walk of the exciton within the polymer chain and a long-range direct energy transfer between polymer and dye. We find that rapid intrachain energy migration toward complex sites with the dye leads to the highly efficient energy transfer, whereas the contribution from direct, long-range energy transfer is negligible. We determine the actual density of complexes with the dye along the polymer chain. A clear deviation from calculations based on a constant complex association constant is found and explained by a reduced effective polymer concentration due to aggregation. Altogether, the quenching efficiency is found to be limited by (i) the energetic disorder within the polymer chain and (ii) the formation of loose polymer aggregates.  相似文献

New amphiphilic gelators that contained both Schiff base and L-glutamide moieties, abbreviated as o-SLG and p-SLG, were synthesized and their self-assembly in various organic solvents in the absence and presence of metal ions was investigated. Gelation test revealed that o-SLG formed a thermotropic gel in many organic solvents, whilst p-SLG did not. When metal ions, such as Cu(2+), Zn(2+), Mg(2+), Ni(2+), were added, different behaviors were observed. The addition of Cu(2+) induced p-SLG to from an organogel. In the case of o-SLG, the addition of Cu(2+) and Mg(2+) ions maintained the gelating ability of the compound, whilst Zn(2+) and Ni(2+) ions destroyed the gel. In addition, the introduction of Cu(2+) ions caused the nanofiber gel to perform a chiral twist, whilst the Mg(2+) ions enhanced the fluorescence of the gel. More interestingly, the Mg(2+)-ion-mediated organogel showed differences in the fluorescence quenching by D- and L-tartaric acid, thus showing a chiral recognition ability.  相似文献

The influence of metal cations Li(+), Na(+), K(+), Cs(+), Mg(2+), Ca(2+), Sr(2+), Ba(2+), Co(2+), Ni(2+), Cu(2+), Zn(2+), Pb(2+) and Al(3+) on the spectroscopic properties of the dansyl (1-dimethylaminonaphthalene-5-sulfonyl) group covalently linked to monoaza crown ethers 1-aza-15-crown-5 (1,4,7,10,-tetraoxa-13-azacyclopentadecane) (A15C5) and 1-aza-crown-6 (1,4,7,10,13-pentaoxa-16-azacyclooctadecane) (A18C6) was investigated by means of absorption and emission spectrophotometry. Interaction of the alkali metal ions with both fluoroionophores is weak, while alkaline earth metal ions interact strongly causing 50 and 85% quenching of dansyl fluorescence of N-(5-dimethylamine-1-naphthalenesulfonylo)-1,4,7,10,-tetraoxa-13-azacyclopentadecane (A15C5-Dns) and N-(5-dimethylamine-1-naphthalenesulfonylo)-1,4,7,10,13-pentaoxa-16-azacyclooctadecane (A18C6-Dns), respectively. The Cu(2+), Pb(2+) and Al(3+) cations interact very strongly with dansyl chromophore, causing a major change in absorption spectrum of the chromophore and forming non-fluorescent complexes. The Co(2+), Ni(2+), Zn(2+), Mg(2+) cations interact moderately with both fluoroionophores causing quenching of dansyl fluorescence by several percent only.  相似文献

A rhodamine B derivative 4 containing a highly electron-rich S atom has been synthesized as a fluorescence turn-on chemodosimeter for Cu(2+). Following Cu(2+)-promoted ring-opening, redox and hydrolysis reactions, comparable amplifications of absorption and fluorescence signals were observed upon addition of Cu(2+); this suggests that chemodosimeter 4 effectively avoided the fluorescence quenching caused by the paramagnetic nature of Cu(2+). Importantly, 4 can selectively recognize Cu(2+) in aqueous media in the presence of other trace metal ions in organisms (such as Fe(3+), Fe(2+), Cu(+), Zn(2+), Cr(3+), Mn(2+), Co(2+), and Ni(2+)), abundant cellular cations (such as Na(+), K(+), Mg(2+), and Ca(2+)), and the prevalent toxic metal ions in the environment (such as Pb(2+) and Cd(2+)) with high sensitivity (detection limit < or =10 ppb) and a rapid response time (< or =1 min). Moreover, by virtue of the chemodosimeter as fluorescent probe for Cu(2+), confocal and two-photon microscopy experiments revealed a significant increase of intracellular Cu(2+) concentration and the subcellular distribution of Cu(2+), which was internalized into the living HeLa cells upon incubation in growth medium supplemented with 50 muM CuCl(2) for 20 h.  相似文献

Novel fluorescent probes have been developed for the ultratrace detection of heavy metal ions by capillary electrophoresis using laser-induced fluorescence detection. Based on a molecular design, the probes are composed of an octadentate chelating moiety, a macrocyclic DOTA (tetraazacyclododecanetetraacetic acid) and an acyclic DTPA (diethylenetriaminepentaacetic acid) frame, a spacer and a fluorophore (fluorescein). These were chosen on the basis of their ability to form kinetically inert and highly emissive complexes, and to prevent a quenching effect even with heavy and paramagnetic metal ions. Addition of a cationic polymer, polybrene, in the separation buffer provided high resolution and simultaneous detection of Ca(2+), Mg(2+), Cu(2+), Zn(2+), Ni(2+), Co(2+), Mn(2+), Cd(2+) and Pb(2+). The direct fluorescence detection of these metal ions with high sensitivity at lower ppt levels, typically 2-7 × 10(-11) M (potentially sub-ppt), was successfully achieved. While separation of anionic compounds using a counter cation ("Ion Association (IA)" mode) is typically controlled by the ion association equilibrium constants, K(ass), it was found that differences in the mobilities, μ(ep(IAC)), of the ion association complexes formed between the probe complexes and counter cations are the driving forces for separation in this new method. This suggests that each of the polybrene-probe complexes has different chemical structures among metal ions, which were able to be determined by CD spectra in this investigation. This novel separation mode was termed the "Ion Association Complex (IAC)" mode, distinct from the IA mode.  相似文献

The structurally characterized lower rim 1,3-di{4-antipyrine}amide conjugate of calix[4]arene (L) exhibits high selectivity toward Hg(2+) among other biologically important metal ions, viz., Na(+), K(+), Ca(2+), Mg(2+), Mn(2+), Fe(2+), Co(2+), Ni(2+), Cu(2+), Zn(2+), Cd(2+), Hg(2+), Pb(2+), and Ag(+) as studied by fluorescence, absorption, and ESI MS. L acts as a sensor for Hg(2+) by switch-off fluorescence and exhibits a lowest detectable concentration of 1.87 ± 0.1 ppm. The complex formed between L and Hg(2+) is found to be 1:1 on the basis of absorption and fluorescence titrations and was confirmed by ESI MS. The coordination features of the mercury complex of L were derived on the basis of DFT computations and found that the Hg(2+) is bound through an N(2)O(2) extending from both the arms to result in a distorted octahedral geometry with two vacant sites. The nanostructural features such as shape and size obtained using AFM and TEM distinguishes L from its Hg(2+) complex and were different from those of the simple mercuric perchlorate. L is also suited to sense pyrimidine bases by fluorescence quenching with a minimum detection limit of 1.15 ± 0.1 ppm in the case of cytosine. The nature of interaction of pyrimidine bases with L has been further studied by DFT computational calculations and found to have interactions through a hydrogen bonding and NH-π interaction between the host and the guest.  相似文献

In this study, an assay to quantify the presence of aluminum ions using a salicylimine-based receptor was developed utilizing turn-on fluorescence enhancement. Upon treatment with aluminum ions, the fluorescence of the sensor was enhanced at 510 nm due to formation of a 1:1 complex between the chemosensor and the aluminum ions at room temperature. As the concentration of Al(3+) was increased, the fluorescence gradually increased. Other metal ions, such as Na(+), Ag(+), K(+), Ca(2+), Mg(2+), Hg(2+), Mn(2+), Co(2+), Ni(2+), Cu(2+), Zn(2+), Cd(2+), Pb(2+), Cr(3+), Fe(3+), and In(3+), had no such significant effect on the fluorescence. In addition, we show that the probe could be used to map intracellular Al(3+) distribution in live cells by confocal microscopy.  相似文献

The interaction between salvianic acid A sodium (SAS) and bovine serum albumin (BSA) was investigated using fluorescence and ultraviolet spectroscopy at different temperatures under imitated physiological conditions. The experimental results showed that the fluorescence of BSA was quenched by SAS through a static quenching procedure. The binding constants of SAS with BSA were 2.03, 1.17 and 0.71×10(5) L mol(-1) at 291, 298 and 305 K, respectively. Negative values of ΔG, ΔH, and ΔS indicate that the interaction between SAS and BSA is driven by hydrogen bonds and van der Waals forces. According to F?rster non-radiation energy transfer theory, the binding distance between BSA and SAS was calculated to be about 2.92 nm. The effect of SAS on the conformation of BSA was analyzed using synchronous fluorescence spectroscopy. In addition, the effect of some metal ions Cu(2+), Ca(2+), Mg(2+), and Zn(2+) on the binding constant between SAS and BSA was examined.  相似文献

We demonstrate highly sensitive and selective potassium ion detection against excess sodium ions in water, by modulating the interaction between the G-quadruplex-forming molecular beacon aptamer (MBA) and cationic conjugated polyelectrolyte (CPE). The K(+)-specific aptamer sequence in MBA is used as the molecular recognition element, and the high binding specificity of MBA for potassium ions offers selectivity against a range of metal ions. The hairpin-type MBA labeled with a fluorophore and quencher at both termini undergoes a conformational change (by complexation with CPEs) to either an open-chain form or a G-quadruplex in the absence or presence of K(+) ions. Conformational changes of MBA as well as fluorescence (of the fluorophore in MBA) quenching or amplification via fluorescence resonance energy transfer from CPEs provide clear signal turn-off and -on in the presence or absence of K(+). The detection limit of the K(+) assays is determined to be ~1.5 nM in the presence of 100 mM Na(+) ions, which is ~3 orders of magnitude lower than those reported previously. The successful detection of 5'-adenosine triphosphate (ATP) with the MBA containing an ATP-specific aptamer sequence is also demonstrated using the same sensor scheme. The scheme reported herein is applicable to the detection of other kinds of G-rich aptamer-binding chemicals and biomolecules.  相似文献
