Citrus fruits are a valuable functional food and their peel is used in East Asian folk medicine. In this study, the polar components of the fruit tissues of Citrus grandis Osbeck were analyzed using high‐performance liquid chromatography–tandem mass spectrometry and compared with reported data. Among the 13 characterized compounds, eight flavonoids and one coumarin were identified for the first time in fruit tissues. The total amount of the identified components was the largest for the immature fruit peel, followed by mature fruit peel, mature fruit flesh, and immature fruit flesh. Naringin (2) and neohesperidin (3) were particularly rich in all samples. The antioxidant activity of the flavonoids extracted from fruit tissues increased in a dose‐dependent manner. Gas chromatography-mass spectrometry (GC-MS) and gas chromatography-olfactometry (GC-O) were used to determine the aromatic composition and aroma active compounds of fruit juice and peel oil of Jinchen sweet orange fruit. Totals of 49 and 32 compounds were identified in fruit juice and peel oil, respectively. GC-O was performed to study the aromatic profile of Jinchen fruit juice and peel oil. A total of 41 components appeared to contribute to the aroma of fruit juice and peel oil. Twelve components were the odorants perceived in both samples. The aromatic compositions of fruit juice were more complex than that of peel oil. Ethyl butanoate, beta-myrcene, octanal, linalool, alpha-pinene, and decanal were found to be responsible for the aromatic notes in fruit juice and peel oil. Nineteen components have been perceived only in the juice and ten compounds were described as aromatic components of only the peel oil by the panelists. These differences lead to the different overall aroma between fruit juice and peel oil. 相似文献

A mixture of polyphenol components was isolated from the fruits of C. annuum L. cv. Cupra, C. annuum L. cv. Orange glory, and C. annuum L. cv. ST4712 (CLST), via 70% methanol extraction followed by column chromatography over silica gel. The polyphenol components of the mixture were analyzed via HPLC-MS/MS and compared with the reported data. Three cinnamic acid derivatives and five flavonoid components in the fruits of the three varieties were identified for the first time in this study. The antioxidant activity and anticancer effect of the polyphenol mixtures of the three fruits were determined. The antioxidant and anticancer activities of CLST were substantially higher than those of C. annuum L. cv. Cupra and C. annuum L. cv. Orange glory. The high activities of CLST were attributed to the much higher concentration of quercetin derivatives in CLST. 相似文献

The skin of Vitis coignetiae Pulliat (meoru) grown wild in the Republic of Korea was analyzed for anthocyanins via HPLC coupled to ESI‐MS/MS in positive ion mode. Chromatographic separation was conducted via RP HPLC using a C18 column, with a 50‐min gradient from 0 to 80% methanol in water containing 0.5% formic acid. A total of 18 anthocyanins were identified. Among them, nine compounds were newly determined by comparing the retention time (tR) and mass fragmentation patterns with those of the previously reported anthocyanins for other grape varieties: malvidin hexose, peonidin 3‐galactoside, malvidin 3‐galactoside, cyanidin, petunidin, petunidin 3‐(6″‐coumaroyl)‐5‐diglucoside, peonidin, malvidin, and malvidin 3‐(6″‐coumaroyl)‐5‐diglucoside. The antioxidant activity of the V. coignetiae Pulliat anthocyanins was determined via 2,2‐diphenyl‐2‐picrylhydrazyl radical scavenging and 2,2′‐azinobis‐(3‐ethylbenzothiazoline‐6‐sulfonic acid) radical cation assays in a range of concentration from 25 to 500 mg/L. The capacity increased with concentration. The IC50 values, defined as the concentration of sample required to scavenge 50% of free radicals, were calculated as follows: 189.63±11.31 mg/L and 141.29±6.70 mg/L for 2,2‐diphenyl‐2‐picrylhydrazyl and 2,2′‐azinobis‐(3‐ethylbenzothiazoline‐6‐sulfonic acid) radical cation, respectively. The antioxidant activity of the V. coignetiae Pulliat anthocyanins is substantially higher than that of ascorbic acid and is similar to the effects of the extracts obtained from other grape varieties. 相似文献

In 1987, Berner and Landis reported that upon vacuum grinding of 80 million year old amber, a gas mixture was released which suggested an oxygen-rich prehistoric environment. Fundamental to their argument was the assumption that amber, an organic glass formed during the fossilization of tree sap, is a perfect sealant. Their assumption was challenged by three technical comments which collectively concluded that gases diffuse readily through amber. In order to defend their key assumption that gases are perfectly trapped in amber, Berner and Landis dismissed the data obtained through gravimetric sorption experiments as only a measure of surface adsorption rather than bulk absorption in and concomitant diffusion through the amber matrix. The validity of interpreting these gravimetric experiments as a measure of bulk diffusion is demonstrated by exploring the physical basis for interpreting gravimetric sorption data. Most importantly, new experimental gravimetric sorption data are presented which demonstrate an explicit separation of adsorption from diffusion-controlled absorption and also reveal that adsorption accounts for a very small fraction of the total sorption in amber. 相似文献

This study represents a systematic evaluation of protocols for protein extraction and cleanup for fruit proteomic analysis. Procedures were optimized using pooled lyophilized banana fruit pulp, which is known to be particularly tricky due to high concentrations of soluble polysaccharides, phenolics, and other substances that interfere with protein extraction and purification. A total of 18 combinations of three protein extraction procedures (SDS‐based, Triton X‐100‐based, and phenol‐based), three protein precipitating agents (ammonium acetate/methanol, TCA/acetone, and acetone), and two resolubilization buffers (classical Rabilloud and the so‐called R2D2) were compared for total protein yields and efficiency of recovery. The results demonstrate that while losses in total recovered protein are unavoidable, the degree of these losses depends on the method combinations used. Combinations based on buffer‐saturated phenol always gave the highest yields, and overall recovery and purity was highest when acetone was combined with the R2D2 buffer for protein purification and concentration. Comparative 2D‐PAGE analysis confirmed that this method combination produced high‐quality and reproducible gels and the largest numbers of spots per gel. The usefulness of this methodology was demonstrated on ripe fruits from several other species and shown to give excellent results. 相似文献

Summary Methods to determine the contribution of the chromatographic equipment to the total band broadening which involve replacing
the column by a union or a capillary tube are not suitable as they involve a fundamental change in the chromatographic system.
The linear extrapolation method, based on the estimation of the relative influence of the instrument variance on solutes with
different capacity factors, is a more attractive alternative method since the column remains in the chromatographic system.
This method is only valid when a number of conditions are satisfied. By meeting these conditions the error in the instrument
variance by using the linear extrapolation method was determined. At the same time, ways to minimise these errors were studied.
Use of the linear extrapolation method in combination with conventional columns of 4.6 mm i.d. appears to yield inaccurate
results.
In combination with microbore columns the method can be used, provided the columns have a maximum length of 5cm and contain
a packing material with a particle size of 2 or 3μm. The error in the determined instrument variance is then of the order

Agastache rugosa Kuntze (Korean mint) is used as a spice and in folk medicine in East Asia. The present study identified a total of 18 polyphenols from the flower, leaf, stem and roots of this plant using high‐performance liquid chromatography–tandem mass spectrometry. Fourteen of these compounds had not previously been identified in these plant tissues. Each polyphenol was validated in comparison with external calibration curves constructed using structurally related compounds, with determination coefficients >0.9993. The limits of detection and quantification were 0.092–0.650 and 0.307–2.167 mg/L, respectively. Recoveries of 61.92–116.44% were observed at two spiking levels, with 0.91–11% precision, expressed as relative standard deviation (except anthraquinone spiked at 10 mg/L). Hydroxycinnamic acid was the most abundant compound in the root, while the flowers showed the highest total flavonoid level. Antioxidant activities, determined in terms of reducing power, Fe2+ chelating activity and the radical scavenging activities using α,α‐diphenyl‐β‐picrylhydrazyl and 2‐2?‐azino‐bis‐3‐ethylbenzothiazoline‐6‐sulfonic acid, increased in a concentration‐dependent manner; the highest activity was identified in the stems, followed by leaves > flowers > roots. Evaluation of the antioxidant properties of litchi fruit phenolics in relation to pericarp browning prevention 总被引:1,自引:0,他引:1
Phenolics were extracted from litchi fruit pericarp (LFP) tissues, purified and their antioxidant properties analyzed. LFP phenolics strongly inhibited linoleic acid oxidation and exhibited a dose-dependent free-radical scavenging activity against alpha,alpha-diphenyl-beta-picrylhydrazyl (DPPH*) and hydroxyl radicals and superoxide anions. The degradation of deoxyribose by hydroxyl radicals was inhibited by phenolics acting mainly as iron ion chelators, rather than by directly scavenging the radicals. Phenolics from litchi fruit pericarp were found to display similar reducing power activity as ascorbic acid. The effect of phenolic compound treatment on pericarp browning and electrolyte leakage of litchi fruit was also evaluated and it was observed that application of exogenous litchi phenolics to harvested litchi fruit significantly prevented pericarp browning and delayed increases in electrolyte leakage. These results suggest that litchi pericarp tissue phenolics could be beneficial in scavenging free radicals, maintaining membrane integrity and, thereby inhibiting pericarp browning of litchi fruit. 相似文献

Background
Hylocereus polyrhizus and Hylocereus undatus are two varieties of the commonly called pitaya fruits, and pitaya fruits have gained popularity in many countries all over the world. However, studies on chemical composition and the nutritional quality of pitaya flesh peel are limited.Results
Extracts of pitaya (H. polyrhizus and H. undatus) peel were extracted by supercritical carbon dioxide extraction, and analyzed by gas chromatography–mass spectrometry analysis. Their cytotoxic and antioxidant activities were investigated. The main components of H. polyrhizus extract were β-amyrin (15.87%), α-amyrin (13.90%), octacosane (12.2%), γ-sitosterol (9.35%), octadecane (6.27%), 1-tetracosanol (5.19%), stigmast-4-en-3-one (4.65%), and campesterol (4.16%), whereas H. undatus were β-amyrin (23.39%), γ-sitosterol (19.32%), and octadecane (9.25%), heptacosane (5.52%), campesterol (5.27%), nonacosane (5.02%), and trichloroacetic acid, hexadecyl ester (5.21%). Both of the two extracts possessed good cytotoxic activities against PC3, Bcap-37, and MGC-803 cells (IC50 values ranging from 0.61 to 0.73 mg/mL), and the activities of their main components were also studied. Furthermore, these extracts also presented some radical scavenging activities, with IC50 values of 0.83 and 0.91 mg/mL, respectively.Conclusion
This paper provides evidence for studying the chemical composition of supercritical carbon dioxide extracts of pitaya peel and their biological activity. 相似文献15.

AimThe purpose of the study is to research the effect of quercetin, which has a good antioxidant characteristic in rats exposed to sodium fluoride (NaF) toxication, on oxidant and antioxidant activity in liver tissue and erythrocyte.Material and method40 Swiss albino male rats with a weight of approximately 20–25 gr were used in the study. The rats were separated into 4 equal groups (n = 10). The first group, the control group, was given normal drinking water; the second group was given 12 mg/kg/day NaF; the third group was given 40 mg/kg/day quercetin; the fourth group was given 12 mg/kg/day NaF+40 mg/kg/day quercetin orally for 30 days. As a result of the experiment, plasma AST and ALT activities were found. Total oxidant capacity (TOC) and total antioxidant capacity (TAC) analyses were made in liver tissue and erythrocyte hemogenisate. Histopathological analyses of the liver tissue were conducted.ResultsNo changes were found in plasma ALT activities between groups. Increases were found in AST activity of all groups. While erythrocyte TAC level was found to increase in fluorine and fluorine + quercetin group when compared with the control group, erythrocyte TOC level was found to increase in all groups when compared with the control group. No significant change was found in TOC and TAC levels in the liver tissue. In liver tissue histopathology, while hydropic degeneration and cellular necrosis were found in vena centralis area in fluorine group, hydropic degeneration was found in heptocytes in centrilobular area in the quercetin group. In the fourth group, hydropic degeneration and centrilobular necrosis were more severe. It was found that in rats which are given NaF experimentally, fluorine and quercetin caused histopathological degenerations in the liver tissue during this time.ConclusionWhile no changes were found in TOC and TAC levels of liver tissue, antioxidant capacity was found to be stimulated in fluorine groups in erythrocyte. 相似文献

Within the concepts of structurally-thermodynamic characteristics of solvation and pseudo-chemical potential, the sample collection of the most authentic experimental data on solubility of gaseous He, Ne, Ar, Kr, Xe, and Rn in H2O and D2O is analysed at ≈0.1 MPa and T = 278–318 K. The conclusion is drawn that at deuteration of water molecules and also with increasing molar mass of noble gas, the relative contribution of effect of its hydrophobic hydration decreases. However in case of pass from lightweight noble gases (He, Ne, Ar) to heavy ones (Kr, Xe, Rn), structural transformations in their aqueous solutions become more expressed as a whole due to strengthening interaction between dissolved substance and solvent. 相似文献

We report on the application of a simple and versatile antioxidant capacity assay for dietary polyphenols, vitamin C and vitamin
E utilizing the copper(II)-neocuproine (Cu(II)-Nc) reagent as the chromogenic oxidant, which we term the CUPRAC (cupric reducing
antioxidant capacity) method. It involves mixing the antioxidant solution (directly or after acid hydrolysis) with solutions
of CuCl2, neocuproine, and ammonium acetate at pH 7, and measuring the absorbance at 450 nm after 30 min. Slowly reacting antioxidants
required an incubation at 50 °C for 20 min for color development. The flavonoid glycosides were hydrolyzed to their corresponding
aglycones by refluxing in 1.2 M HCl-containing 50% MeOH for fully exhibiting their antioxidant potencies. Certain compounds
also needed incubation after acid hydrolysis for color development. The CUPRAC absorbances of mixture constituents were additive,
indicating lack of chemical deviations from Beer’s law. The CUPRAC antioxidant capacities of a wide range of polyphenolics
are reported in this work and compared to those found by ABTS/persulfate and Folin assays. The trolox-equivalent capacities
of the antioxidants were linearly correlated (r = 0.8) to those found by ABTS but not to those of Folin. The highest antioxidant capacities in the CUPRAC method were observed
for epicatechin gallate, epigallocatechin gallate, quercetin, fisetin, epigallocatechin, catechin, caffeic acid, epicatechin,
gallic acid, rutin, and chlorogenic acid in this order, in accordance with theoretical expectations. The experiences of other
CUPRAC users also are summarized.

The flavonoid content of several methanolic extract fractions of Navel orange peel (flavedo and albedo of Citrus sinensis) cultivated in Crete (Greece) was first analysed phytochemically and then assessed for its antioxidant activity in vitro. The chemical structures of the constituents fractionated were originally determined by comparing their retention times and the obtained UV spectral data with the available bibliographic data and further verified by detailed LC-DAD-MS (ESI+) analysis. The main flavonoid groups found within the fractions examined were polymethoxylated flavones, O-glycosylated flavones, C-glycosylated flavones, O-glycosylated flavonols, O-glycosylated flavanones and phenolic acids along with their ester derivatives. In addition, the quantitative HPLC analysis confirmed that hesperidin is the major flavonoid glycoside found in the orange peel. Interestingly enough, its quantity at 48 mg/g of dry peel permits the commercial use of orange peel as a source for the production of hesperidin. The antioxidant activity of the orange peel methanolic extract fractions was evaluated by applying two complementary methodologies, DPPH(*) assay and the Co(II)/EDTA-induced luminol chemiluminescence approach. Overall, the results have shown that orange peel methanolic extracts possess moderate antioxidant activity as compared with the activity seen in tests where the corresponding aglycones, diosmetin and hesperetin were assessed in different ratios. 相似文献

We have performed CCSD(T), MP2, and DF-LMP2 calculations of the interaction energy of CO on the MgF(2)(110) surface by applying the method of increments and an embedded cluster model. In addition, we performed periodic HF, B3LYP, and DF-LMP2 calculations and compare them to the cluster results. The incremental CCSD(T) calculations predict an interaction energy of E(int) = -0.37?eV with a C-down orientation of CO above a Mg(2+) ion at the surface with a basis set of VTZ quality. We find that electron correlation constitutes about 50% of the binding energy and a detailed evaluation of the increments shows that the largest contribution to the correlation energy originates from the CO interaction with the closest F ions on the second layer. 相似文献
