The 6- and 7-(1-ethylbenzimidazolyl)-substituted 1-ethyl-1,4-dihydro-4-oxoquinoline-3-carboxylic acids were synthesized. Antibacterial activity was tested in vitro. Only one of the new compounds prepared showed slight antibacterial activity against one of the strains tested. So, they are of no interest as antibacterial agents.  相似文献

The synthesis and antimicrobial activity of 5-Hydroxy-l-ethyl-1,4-dihydrio-4-oxo-3-quinolinecarboxylic acids and their derivatives have been investigated. The 5-hydroxy-3-quinolinecarboxylic acids were prepared by thermal cyclization of 3-oxocyclohexenylaminomethylenemalonates followed by aromatiza-tion with iodine-ethanol, ethylation and hydrolysis of products. The 5-amino derivatives were prepared by several steps involving novel aromatization of isoxazole intermediates.  相似文献

Condensation of 2-bromoacetophenones with sodium succinimide gave N-phenacylsuccinimides ( 1 ) which were opened with sodium hydroxide to N-phenacylsuccinamic acids ( 2 ). The latter were cyclized to 5-aryl-2-oxazolepropionic acids ( 3 ) in sulfuric acid. Similar cyclization of N-phenacylphthalamic acid ( 5 ) and succinic acid 2-benzoylhydrazide ( 7 ) gave o-(5-phenyl-2-oxazolyl)benzoic acid ( 6 ) and 5-phenyl-1,3,4-oxadiazole-2-propionic acid ( 8 ). The succinamic acids 2 and the phthalamic acid 5 were observed to recyclize to the corresponding imides ( 1 and 4 ) on heating, and the succinic acid hydrazide 7 was similarly cyclized to N-benzamidosuccinimide ( 9 ) with acetic anhydride. Antiinflammatory screening data are reported for 3 , 6 and 8 .  相似文献

A series of 6-aroyl-4-oxohexanoic acids ( 2 ) was prepared for intermediate use by acid-catalyzed solvolysis of substituted 3-(2-furyl)acrylophenones ( 1 ). This reaction occasionally gave 5-aryl-2-furanpropionic acids ( 3 ) instead of, or in addition to, the desired diketones ( 2 ). Equilibrium between 2 and 3 was observed in the case where Ar is m-nitrophenyl. A rationalization for the formation of 3 is offered, and published reports with which our results differ are discussed. Diketones ( 2 ) were cyclized to furans ( 3 ) and antiinflammatory screening data are reported for the latter.  相似文献

The corresponding 5-bromo-6-alkoxydihydroorotic acid derivatives were obtained by reaction of orotic acid, its methyl ester, and amide with bromine in methanol and ethanol. The reaction of methyl and butyl orotates with methyl and ethyl hypobromites gives 5,5-dibromo-6-alkoxydihydroorotic acid esters. A series of 1-ethyl-1,4-dihydro-4-oxo-7-(4-thiazolyl)-3-quinolinecarboxylic acids and 1-ethyl-1,4-dihydro-4-oxo-7-(2-thiazolyl)-3-quinolinecarboxylic acids were prepared. Also prepared was 10-[2-(aminomethyl)-4-thiazolyl]-9-fluoro-2,3-dihydro-3-methyl-7-oxo-7H-pyrido[1,2,3-de][1,4]benzoxazine-6-carboxylic acid. Analogs with basic amine substituents on the thiazole moiety were found to have antibacterial activity.  相似文献

A general method for preparing 2-substituted-4-oxo-3-quinolinecarboxylic acids and 2-substituted-4-oxo-1,8-naphthyridine-3-carboxylic acids as new analogs in the quinolone class of antiinfectives has been developed. The reaction of a Grignard reagent in the presence of copper(I) iodide with the 4-oxo-3-quinolinecar-boxylic acid esters and 4-oxo-1,8-naphthyridine-3-carboxylic acid esters yields the desired 2-substituent. Re-introduction of the 2,3-double bond is effected by phenylselenation of the 3-position, oxidation to the selen-oxide, and in situ syn-elimisation. Depending on the degree of steric crowding between the 2-substituent and the 3-carboxylic acid group, hydrolysis of the ester to the carboxylic acid could be carried out under acidic or basic conditions.  相似文献

Condensation of appropriate β-keto esters with phenylhydrazine gave 5-oxo-1-phenyl-2-pyrazoline-3-and 4-alkanoic esters 1 and 3 which were saponified to the corresponding alkanoic acids 2 and 4 . Analogous condensation of the same β-keto esters with hydrazobenzene gave 5-oxo-1,2-diphenyl-3-pyrazoline-3-and 4-alkanoic esters 5 and 7 which were similarly converted to acids 6 and 8 . The structures of the oxopyrazolines as revealed by their infrared absorption are discussed, and results of their antiinflammatory screening are reported.  相似文献

A novel preparative method for 1-alkyl-1,4-dihydro-4-oxo-3-quinolinecarboxylic acids was developed. The key process is the cyclization of N-alkylanilinomethylenemalonales, which was effected successfully in the presence of polyphosphoric acid, polyphosphate ester, boron tri-fluoride or a mixture of acetic anhydride and sulfuric acid. With phosphorus oxychloride, N-alkylanilinomethylenemalonates yielded 1-alkyl-4-chloro-3-carbethoxyquinolinium salts which were hydrolyzed readily to ethyl 1-alkyl-1,4-dihydro-4-oxo-3-quinolineearboxylates or their acids. By means of this novel method several new 1-alkyl-1,4-dihydro-4-oxo-3-quinolineearboxylic acids were prepared.  相似文献

A series of 2-anilino-1,6-dihydro-6-oxo-5-pyrimidinecarboxylic acids with various substituents was synthesized and evaluated in the rat passive cutaneous anaphylaxis test for antiallergic activity. High activity by intraperitoneal and oral administrations was observed for the 3-trifluoromethyl and 2-alkoxyanilino derivatives (64, 79, 81, 82 and 85). Malaria chemotherapy is greatly threatened by the recent emergence and spread of resistance in the Plasmodium falciparum parasite against artemisinins and their partner drugs. Therefore, it is an urgent priority to develop new antimalarials. Plasmepsin V (PMV) is regarded as a superior drug target for its essential role in protein export. In this study, we performed virtual screening based on homology modeling of PMV structure, molecular docking and pharmacophore model analysis against a library with 1,535,478 compounds, which yielded 233 hits. Their antimalarial activities were assessed amongst four non-peptidomimetic compounds that demonstrated the promising inhibition of parasite growth, with mean IC50 values of 6.67 μM, 5.10 μM, 12.55 μM and 8.31 μM. No significant affection to the viability of L929 cells was detected in these candidates. These four compounds displayed strong binding activities with the PfPMV model through H-bond, hydrophobic, halogen bond or π-π interactions in molecular docking, with binding scores under −9.0 kcal/mol. The experimental validation of molecule-protein interaction identified the binding of four compounds with multiple plasmepsins; however, only compound 47 showed interaction with plasmepsin V, which exhibited the potential to be developed as an active PfPMV inhibitor.  相似文献

The cyclization of m-substituted anilinomethylenemalonates ( 1 ) in the presence of polyphosphate ester and some other cyclizing agents gave mixtures of the isomeric ethyl 5- ( 2 ) and 7-substituted 4-hydroxy-3-quinolinecarboxylates ( 3 ), which led to mixtures of the corresponding quinolinecarboxylic acids ( 4 and 5 ) by hydrolysis. The proportions of 4 and 5 in the mixtures were determined on the basis of their nmr spectra. Novel 5-chloro- ( 8a ), 5-methyl- ( 8b ) and 5-nitro-1-ethyl-1,4-dihydro-4-oxo-3-quinolinecarboxylic acids were prepared and evaluated for antimicrobial activities. The rapid emergence of drug resistance to the current antimalarial agents has led to the urgent need for the discovery of new and effective compounds. In this work, a series of 5-phenoxy primaquine analogs with 8-aminoquinoline core (7a–7h) was synthesized and investigated for their antimalarial activity against Plasmodium falciparum. Most analogs showed improved blood antimalarial activity compared to the original primaquine. To further explore a drug hybrid strategy, a conjugate compound between tetraoxane and the representative 5-phenoxy-primaquine analog 7a was synthesized. In our work, the hybrid compound 12 exhibited almost a 30-fold increase in the blood antimalarial activity (IC50 = 0.38 ± 0.11 μM) compared to that of primaquine, with relatively low toxicity against mammalian cells (SI = 45.61). Furthermore, we found that these 5-phenoxy primaquine analogs and the hybrid exhibit significant heme polymerization inhibition, an activity similar to that of chloroquine, which could contribute to their improved antimalarial activity. The 5-phenoxy primaquine analogs and the tetraoxane hybrid could serve as promising candidates for the further development of antimalarial agents.  相似文献
