[reaction: see text] An efficient methodology for the reductive alkylation of secondary amine with aldehyde and Et(3)SiH using an iridium complex as a catalyst has been developed. For example, treatment of dibutylamine with butyraldehyde and Et(3)SiH (a 1:1:1 molar amount of amine, aldehyde, and silane) in 1,4-dioxane at 75 degrees C under the influence of a catalytic amount of [IrCl(cod)](2) gave tributylamine in quantitative yield. In this reaction, no reduction of aldehyde took place. It was found that IrCl(3), which is a starting material for preparation of iridium complexes such as [IrCl(cod)](2), acts as an efficient catalyst for the present reductive alkylation of amine. In addition, a cheaper, easy-to-handle, and environmentally friendly reducing reagent such as polymethylhydrosiloxane (PMHS) in place of Et(3)SiH was also useful. Thus, a variety of secondary amines could be alkylated by allowing them to react with aldehydes and PMHS in the presence of an iridium catalyst to afford the corresponding tertiary amines in good to excellent yields. From the deuterium label experiments, it was revealed that silane and water, generated during the formation of enamine by the reaction of amine and aldehyde, seem to behave as a hydrogen source. Practical catalytic cross-coupling of secondary alkyl electrophiles with secondary alkyl nucleophiles under Cu catalysis has been realized. The use of TMEDA and LiOMe is critical for the success of the reaction. This cross-coupling reaction occurs via an S(N)2 mechanism with inversion of configuration and therefore provides a general approach for the stereocontrolled formation of C-C bonds between two tertiary carbons from chiral secondary alcohols.  相似文献

The enantioselective synthesis of optically active secondary amines via the asymmetric reduction of N-substituted ketimines with various chiral hydride reagents, such as Itsuno's reagent (1), Corey's reagent (2), K glucoride (3), Sharpless' reagent (4), and Mosher's reagent (5) has been investigated. Among the hydride reagents examined, 1 gave the best results in terms of asymmetric induction. Thus, the reduction of N-phenylimine derivatives of aromatic ketones with 1 provided the corresponding amines in 96–98% yields with high optical induction, such as 73 % ee for acetophenone N-phenylimine (6a), 87 % ee for propiophenone N-phenylimine (6b), 88 % ee for bulyrophenone N-phenylimine (6c), and 71 % ee for isobutyrophenone N-phenylimine (6d). In the case of N-alkyl ketimine derivatives, the reduction afforded somewhat lower optical inductions as compared to those of N-phenyl derivatives, giving 46 % ee for acetophenone N-benzylimine (6f), 52 % ee for acetophnone N-n-heptylimine (6g) and 43 % ee for acetophenone N-cyclohexylimine (6h). However, the substitution of a bulky alkyl group on nitrogen of the ketimines increases remarkably the optical induction of product amine, such as 80 % ee for acetophenone N-tert-butylimine (6e). The reduction of N-substituted aliphatic ketimines gave very low optical inductions (7.4 – 24 % ee). The catalytic effects of oxazaborolidines (1a and 2a) in the reduction of ketimines with 1 and 2 were also examined.  相似文献

Recently there has been considerable interest in using surface plasmon resonance (SPR) for the measurement of conformational changes of immobilized biomolecules that are induced by an exogenous analyte. While a number of studies have shown the analytical utility of such measurements, there has been no report which characterizes the specific secondary structure that actuates the change in SPR signal. The use of SPR to indicate the type of secondary structure present in two immobilized polypeptides, poly-L-lysine (PL) and poly-L-glutamic acid (PGA), and a globular protein, concanavalin A (Con A) is described in this report. The PL, PGA and Con A were modified with N-succinimidyl 3-(2-pyridyldithiol) propionate (SPDP) to introduce disulfide groups to facilitate the attachment onto gold-coated surfaces via self-assembly. Ethanol and 2,2,2-trifluoroethanol (TFE) were used to induce changes in the secondary structure of the immobilized polypeptides and the protein respectively. Using both circular dichroism (CD) and Fourier transform infrared (FTIR) spectroscopies, it has been demonstrated that it is possible to correlate the signal changes observed in SPR to the secondary conformation of the biomolecule. Both CD and FTIR showed that a decrease in SPR signal corresponded to a high content of beta, turn or unordered structures while an increase corresponded to a high alpha-helical content. The sensitivity of the SPR technique is comparable to that obtained in solution with CD and FTIR spectroscopies. These results are the first demonstration that SPR can be used to characterize secondary structures. There is potential, therefore, for SPR to be used as a technique to study secondary conformational changes of immobilized polypeptides and proteins.  相似文献

The effect of the analyte electron affinity on the liquid secondary ionization mass spectrometry beam-induced dehalogenation of simple bromoaromatic compounds in a glycerol matrix was investigated. The results show a definite trend of decreasing dehalogenation with increasing analyte electron affinity. At high analyte electron affinity (≥ 1.0 eV), no dehalogenation was observed. These results are consistent with electrochemical and pulse radiolysis studies where one electron reduction was shown to be responsible for dehalogenation. A chloroaromatic compound with high electron affinity, 4-(4-chloro-benzoyl)pyridine, exhibited reduction by hydrogen addition but not dehalogenation. The radiation chemistry of alcohols was used to elaborate a scheme of the reactive species generated in the glycerol matrix by kiloelectronvolt particle bombardment. The possible role of those species in reduction processes such as dehalogenation was evaluated. The observation that dehalogenation decreases with analyte electron affinity is mechanistically consistent with the proposition that secondary electron production is an intrinsic part of the bombardment process.  相似文献

The reactions of 1,2-dichloroethane with 2-(ethylamino)ethanol or diethylamine have been investigated in several solvents from 51 to 80°C. A reaction mechanism has been proposed where 1,2-dichloroethane reacts with the secondary amines in both bimolecular substitution (SN 2) and elimination (E2) reactions; the substitution product is rapidly converted in an aziridinium ion and undergoes a consecutive reaction with the starting amine to give a tetrasubstituted ethylenediamine. The use of commercially available (SIPr)Pd(cinnamyl)Cl (SIPr = 1,3-bis(2,6-diisopropylphenyl)-4,5-dihydroimidazol-2-ylidene) as a precatalyst for the anaerobic oxidation of secondary alcohols is described. The use of this complex allows for a drastic reduction in the reaction times and catalyst loading when compared to the unsaturated counterpart. This catalytic system is compatible with the use of microwave dielectric heating, decreasing even further catalyst loading and reaction times. Domino Pd-catalyzed oxidation-arylation reactions of secondary alcohols are also presented.  相似文献

Novel catalytic reductions of tertiary and secondary phosphine oxides to phosphines have been developed. Using tetramethyldisiloxane (TMDS) as a mild reducing agent in the presence of copper complexes, PO bonds are selectively reduced in the presence of other reducible functional groups (FGs) such as ketones, esters, and olefins. Based on this transformation, an efficient one pot reduction/phosphination domino sequence allows for the synthesis of a variety of functionalized aromatic and aliphatic phosphines in good yields.  相似文献

A novel and highly diastereoselective samarium-catalyzed tandem rearrangement/reduction of secondary alpha-hydroxy epoxides, which involves a C1 to C3 carbon migration rearrangement and a very interesting hetero-Tishchenko reduction of the intermediate aldehyde and the reductant aldehyde, has been reported. This reaction could be developed to provide a facile and stereoselective construction of 2-quarternary 1,3-diol units with an hydroxymethyl moiety attached to the diastereogenic quaternary carbon center. Detailed investigations have been carried out concerning the screening of the aldehydes as a reductant, the optimization of reaction conditions, and the substrate scope of this tandem reaction. A catalytic cycle for this reaction, the electronic and steric effects of the reductant aldehydes, and the mechanism for the acyl migration of 1,3-diol monoesters are proposed.  相似文献

An enantioselective asymmetric reduction of phenyl ring-containing prochiral ketones to yield the corresponding optically active secondary alcohols was achieved with W110A secondary alcohol dehydrogenase from Thermoanaerobacter ethanolicus (W110A TESADH) in Tris buffer using 2-propanol (30%, v/v) as cosolvent and cosubstrate. This concentration of 2-propanol was crucial not only to enhance the solubility of hydrophobic phenyl ring-containing substrates in the aqueous reaction medium, but also to shift the equilibrium in the reduction direction. The resulting alcohols have S-configuration, in agreement with Prelog's rule, in which the nicotinamide-adenine dinucleotide phosphate (NADPH) cofactor transfers its pro-R hydride to the re face of the ketone. A series of phenyl ring-containing ketones, such as 4-phenyl-2-butanone (1a) and 1-phenyl-1,3-butadione (2a), were reduced with good to excellent yields and high enantioselectivities. On the other hand, 1-phenyl-2-propanone (7a) was reduced with lower ee than 2-butanone derivatives. (R)-Alcohols, the anti-Prelog products, were obtained by enantiospecific oxidation of (S)-alcohols through oxidative kinetic resolution of the rac-alcohols using W110A TESADH in Tris buffer/acetone (90:10, v/v).  相似文献

In this article, a facile, efficient and practical method for Ni-catalyzed direct C-H amination of benzoxazole with secondary amines has been developed. This procedure requires Ni(OAc)(2)·4H(2)O as catalyst, TBHP as oxidant and acid as the additive. A variety of substituted benzoxazol-2-amines were synthesized in moderate to good yields.  相似文献

Pseudo-first-order rate constants (k(obs)) have been measured spectrophotometrically for reactions of O-4-nitrophenyl thionobenzoate (2) with a series of primary and acyclic secondary amines. The plots of k(obs) vs amine concentration are linear for the reaction of 2 with primary amines. The slope of the Br?nsted-type plot for the reaction of 2 with primary amines decreases from 0.77 to 0.17 as the amine basicity increases, indicating that the reaction proceeds through a zwitterionic addition intermediate in which the rate-determining step changes from the breakdown of the intermediate to the reaction products to the formation of the intermediate as the amine basicity increases. On the other hand, for reactions with all the acyclic secondary amines studied, the plot of k(obs) vs amine concentration exhibits an upward curvature, suggesting that the reaction proceeds through two intermediates, e.g., a zwitterionic addition intermediate and an anionic intermediate. The microscopic rate constants (k(1), k(-)(1), k(2), and k(3) where available) have been determined for the reactions of 2 with all the primary and secondary amines studied. The k(1) value is larger for the reaction with the primary amine than for the reaction with the isobasic acyclic secondary amines, while the k(-)(1) value is much larger for the latter reaction than for the former reaction. The k(3) value for the reaction with secondary amine is independent of the amine basicity. The small k(2)/k(-)(1) ratio is proposed to be responsible for the deprotonation process observed in aminolyses of carbonyl or thiocarbonyl derivatives.  相似文献

Reaction of GeCl4 and CyPH2 leads to a range of products from which crystals of [CyPH3]+[GeCl3]- have been obtained. The major intermediate in this reaction, Cy(H)PGeCl3, can be obtained as the dominant product only when an excess of GeCl4 is used in the preparation. Similarly, crystals of [Ph2PH2]+[GeCl3]- have been obtained from reaction of Ph2PH and GeCl4(1 : 1). The structures of both primary and secondary phosphonium cations are reported. Aerosol-assisted and low-pressure chemical vapour deposition experiments using Cy(H)PGeCl3 as precursor lead to the deposition of thin films containing both germanium and phosphorus, contaminated with large amounts of oxygen. Singlet oxygen (1O?), produced during photodynamic therapy, deactivates during its interaction with tissues by producing reactive oxygen species (ROS) and peroxides as well as other degradation products. Here we investigated the role of parameters of light delivery, O(2), and temperature on the ROS and peroxides production, secondary to 1O?. A series of simple in vitro experiments has been performed with Rose Bengal (RB) as a 1O? producer, human serum (HS) as a target and dichlorofluorescein (DCFH) as a nonspecific marker, becoming fluorescent when oxidized. The overall secondary production of ROS and peroxides in HS had also been compared to fetal calf serum (FCS) or mice sera. Increasing power but with a same delivered energy decreased secondary ROS and peroxides when increasing power with a same duration for light delivery increased them. Increasing delivered energy increased linearly secondary ROS or peroxides. Delivering O? by bubbling before light delivery increased secondary ROS or peroxides, when Ar decreased them. Delivering gases after light delivery had no influence on secondary ROS or peroxides production. Increasing temperature from 20 to 40 °C increased secondary ROS or peroxides production but freezing after light delivery either before or after measurement had only a mild influence. Secondary ROS or peroxides production was 2 or 4 times lower in HS than in FCS or nude mice sera respectively. PDT seems to consist of two subsequent phases, both linked but developing independently. The intensity of photo-reactions varied with the model, human sera producing less secondary ROS than fetal calf or mouse sera. Search for new sensitizers should consider secondary ROS-induced pathways in addition to 1O? production.  相似文献

A novel one-pot reaction has been developed for the reduction of aldehydes, ketones and primary, secondary and tertiary alcohols into their corresponding alkyl function. This is also the first reported method which can efficiently reduce primary, secondary, or tertiary alcohols, without affecting carbon-carbon double bonds, into their corresponding alkyl function in high yields. The impact of in-line coagulation pre-treatment of secondary effluent on the operation of an immersed hollow-fibre ultrafiltration membrane pilot was evaluated as part of a larger study on optimising phosphorus removal. The efficacy of alum and ferric chloride was investigated, with an emphasis on alum use. Both coagulants were found to shift the particle-size distribution of organic matter in the feed towards larger fractions, with a notable reduction in colloidal matter. This was reflected in a reduction of both average daily transmembrane pressure increases, as well as a reduction of transmembrane pressure increases within backpulse intervals. Fouling reduction was observed with both lower and higher membrane packing density modules (membrane surface areas of 55.7 and 62.7 m2/module). The results of one-way analysis of variance (ANOVA) testing indicate that for this pilot system, chemical pre-treatment and solids concentrations in the feed water played a statistically significant role in determining transmembrane pressure variations. Membrane packing density and membrane production method did not exhibit a statistically significant effect on transmembrane pressure under the conditions of this study.  相似文献

Ozonization reaction of ethene in neat film at 77 K was performed. Separation of ethene secondary ozonide from the other products of the reaction was performed by continuous pumping of the reactor. Only the products, which evaporated from the walls of the reactor at 185 K, were transferred to the gas cell. The high-resolution infrared absorption spectrum of gaseous ethene secondary ozonide (C(2)H(4)O(3)) in a static gas long-path absorption cell has been recorded in the 900-1100 cm(-1) spectral region at 185 K. The spectral resolution was 0.003 cm(-1). Analyses of the nu(7)(A) band at 1037.0 cm(-1), the nu(8)(A) band at 956.1 cm(-1), and the nu(18)(B) band at 1082.1 cm(-1) have been performed using the Watson Hamiltonian model (A, reduction; III(r), representation). A set of ground-state rotational and quartic centrifugal distortion constants have been obtained, and upper state spectroscopic constants have been determined for the bands investigated. A local resonance observed in nu(18) is explained as c-Coriolis interaction with nu(10) + nu(11).  相似文献
