Polycarbonyl heterocycles. Part III. Synthesis of 2,3-furanodione derivatives by ring transformations of thiazolidine-4,5-dione derivatives

The base catalyzed isomerization of 2-(aroyl-methylene)-3-aryl-thiazolidine-4,5-dione derivatives1 leads to salts of 1-aryl-3-aroyl-4-hydroxy-pyrroline-2-thio-5-one and aliphatic amines2. It was found that2 undergo further transformation to 4-thioanilido-5-arylo-2,3-furanodione derivatives4. This series of reaction provides a convenient synthetic route to 2,3-furanodione derivatives4.

Zusammenfassung Die basenkatalysierte Isomerisierung von 2-(Aroylmethylen)-3-aryl-thiazolidin-4,5-dionen1 führt zu Salzen von 1-Aryl-3-aroyl-4-hydroxy-pyrrolin-2-thio-5-on und aliphatischen Aminen2. Die letzteren gehen in 4-Thioanilido-5-arylo-2,3-furandion-Derivate4 über. Diese Reaktionsfolge stellte eine vorteilhafte synthetische Route zu 2,3-Furandionen dar.

     

New Dipyrrinonols Derived from 2,3-Dihydro-1H-pyrrol-3-ones

Summary. The reaction of 2-aminomethylene-2,3-dihydropyrrol-3(1H)-ones with ethyl bromoacetate, chloroacetonitrile, or dilute HCl gave rise to a series of methine dyes, namely 1-aryl-2-[3-hydroxy-1-aryl-4,5-diphenyl-1H-pyrrol-2-yl]methylene-4,5-diphenyl-2,3-dihydropyrrol-3(1H)-ones.     

2,3-Diferrocenylcyclopropenone in the reaction with organomagnesium compounds

The reactions of 2,3-diferrocenylcyclopropenone with ethyl- and benzylmagnesium chlorides afford 3,3-diethyl- and 3,3-dibenzyl-1,2-diferrocenylcyclopropenes along with products of nucleophilic opening of the three-membered ring: ,-unsaturated and saturated ketones (cis-1,2-diferrocenylpent-1-en-3-one and cis-1,2-diferrocenyl-4-phenylbut-1-en-3-one, 4,5-diferrocenylheptan-3-one, and 3,4-diferrocenyl-1,5-diphenylpentan-2-one). The products of insertion of intermediate diferrocenyl(vinyl)carbene at one of the -bonds of the starting 2,3-diferrocenylcyclopropenone were also isolated: 4-(2-oxo-1-ferrocenylbutyl)- and 4-(2-oxo-3-phenyl-1-ferrocenylpropyl)-2,3,4-triferrocenylcyclobutenones. 3,3-Dibenzyl-1,2-diferrocenylcyclopropene and one of the diastereomers of 4,5-diferrocenylheptan-3-one were studied by X-ray diffraction analysis.     

Novel Aspects of the Reaction of 3-(Benzimidazol-2-yl)-2-iminocoumarins with Aromatic Aldehydes

The reaction of 3-(benzimidazol-2-yl)-2-iminocoumarins with aromatic aldehydes has been studied. The condensation products 7-aryl-7H-benzo[4,5]imidazo[1,2-c]benzopyrano[3,2-e]pyrimidines or 3-(benzimidazol-2-yl)coumarins are formed depending on the nature of the substituent in the starting 2-iminocoumarin and aldehyde. In DMF medium, 7-aryl-7H-benzo[4,5]imidazo[1,2-c]benzopyrano[3,2-e]pyrimidines isomerize to the corresponding 7-aryl-14H-benzo[4,5]imidazo[1,2-c]benzopyrano[3,2-e]pyrimidines. The effect of the substituent on the isomerization process has been studied and the reaction mechanisms are discussed.

Crystal and molecular structure of (z)-isopropyl 2-hydroxy-4,5-dioxo-1-phenyl-3-[phenyl(phenylamino)-methylene]pyrrolidine-2-carboxylate

Reactions of isopropyl 2-(1-aryl-4,5-dioxo-2-phenyl-4,5-dihydro-1H-pyrrol-3-yl)-2-oxoacetates with aromatic amines involve a pyrroledione-pyrroledione recyclization to form isopropyl 1-aryl-2-hydroxy-4,5-dioxo-3-[phenyl(arylamino)methylene]pyrrolidine-2-carboxylates. The crystal and molecular structure of (Z)-isopropyl 2-hydroxy-4,5-dioxo-1-phenyl-3-[phenyl(phenylamino)methylene]pyrrolidine-2-carboxylate was proved by XRD analysis.     

New transformation of α-acetylenic ketones under the action of 1,2-diaminoethane

A reaction of 3-aryl-1-(3,4,5-trimethoxyphenyl)prop-2-yn-1-ones with 1,2-diaminoethane in boiling dioxane leads to 2-aryl-4,5-dihydro-1H-imidazoles and 1-(3,4,5-trimethoxy-phenyl)ethanone.     

v-triazolines. XI . Rearrangement of 1-aryl-4,5-diamino-4,5-dihydro-v-triazoles

The reaction of arylazides with 1,2-diaminoethylenes ( 1a-b ) or α,β-diaminostyrenes ( 1c-f ) gave N-(1,2-diaminoethylideneanilines ( 2a-e ) and N-(1,2-diamino-2-phenylethylidene)anilines ( 2g-i ), respectively. These amidine derivatives are formed through the rearrangement of unstable 1-aryl-4,5-diamino-v-triazolines. The regiospecificity of the cycloaddition reaction has been elucidated on the basis of the products obtained.

1-Aryl-4,5-diaroyl-1H-pyrrole-2,3-diones reacted with benzyl- and phenylhydrazines to give N-aryl-2-(5-aryl-3-aroyl-1-benzyl(phenyl)-1H-pyrazol-4-yl)-2-oxoacetamides whose structure was proved by X-ray analysis.     

Synthesis of 2,3-Dihydroimidazo[1,2-a]pyridines from 1,3-Diketones

A series of 2-hydroxy-, 2-chloro-, and 2-(2-hydroxyethylamino)-6-aryl-4-polyfluoroalkyl-3-cyanopyridines has been synthesized. The latter react with phosphorus oxychloride to give fluorine containing 2,3-dihydroimidazo[1,2-a]pyridines.

Methyl 1-aryl-3-aroyl-4,5-dioxo-4,5-dihydro-1H-pyrrole-2-carboxylates reacted with 3-methyl-4-phenyl-1H-pyrazol-5-amine to give methyl 7-arylcarbamoyl-6-aroyl-2-methyl-3-phenylpyrazolo[1,5-a]-pyrimidine-5-carboxylates. The molecular and crystalline structures of methyl 6-benzoyl-7-(4-chlorophenylcarbamoyl)-2-methyl-3-phenylpyrazolo[1,5-a]pyrimidine-5-carboxylate were studied by X-ray analysis.     

Cyclisierungsreaktionen von Hydrazonen, 2. Mitt.: Eine neue Methode bei der Herstellung von 1-Oxo-2-aryl-1,2-dihydro-(as-triazino) [4,5-a]benzimidazol-4-carbonsäurenitrilen

Zusammenfassung Durch Kupplung von Diazoniumsalzen mit 2-Cyanmethylbenzimidazol wurden einige 2-Arylhydrazonocyanmethylbenzimidazole (1) hergestellt, durch deren Cyclisierung (Einwirkung von Chlorameisensäureäthylester in trockenem Pyridin) Nitrile der 1-Oxo-2-aryl-1,2-dihydro-(as-triazino) [4,5-a] benzimidazol-4-carbonsäuren (2) gewonnen wurden. Dieselben Verbindungen wurden auch aus 1-Carbäthoxy-2-cyanmethylbenzimidazol nach einer kürzlich¹ beschriebenen Methode gewonnen.

Cyclization reactions of hydrazones, II: New synthesis of 1-oxo-2-aryl-1,2-dihydro (as-triazino) [4,5-a]benzimidazole-4-carbonitriles

2-Arylhydrazonocyanomethylbenzimidazoles (1), obtained from diazonium salts and 2-cyanomethylbenzimidazole, cyclize by treatment with ethyl chloroformate in dry pyridine solution to give the corresponding 1-oxo-2-aryl-1,2-dihydro-(as-triazino)-[4,5-a]benzimidazole-4-carbonitriles (2). The same compounds were prepared from 1-carbethoxy-2-cyanomethylbenzimidazole according to the method recently described¹ too.

     

Interaction of 5-aryl-4-quinoxalinyl-2,3-dihydro-2,3-furandiones with mesitylamine and methyl(phenyl)hydrazine. Synthesis and crystal and molecular structure of 2-(1-mesityl-4,5-dioxo-2-phenyl-4,5-dihydro-3-pyrollyl)-3-phenylquinoxaline

The interaction of 5-aryl-4-quinoxaline-2,3-dihydro-2,3-furandiones with mesitylamine and methyl(phenyl) hydrazine results in the formation of 1-substituted 5-aryl-4-quinoxalinyl-2,3-dihydro-2,3-pyrrolediones. Interaction of 4,5-diphenyl-2,3-dihydro-2,3-pyrroledione with ethoxycarbonylmethylenetriphenylphosphorane: synthesis and crystal structure of 4,5-diphenyl-Z-2-ethoxycarbonylmethylene-2,3-dihydro-3-pyrrolone

1-Unsubstituted 4,5-diphenyl-2,3-dihydro-2,3-pyrroledione interacts with ethoxycarbonyl-methylenetriphenylphosphorane regioselectively to give 4,5-diphenyl-Z-2-ethoxycarbonyl-methylene-2,3-dihydro-3-pyrrolone. Reaction of 3-Benzoyl-2,3-dibromopropionic Acid with Substituted ortho-Phenylenediamines

3-Benzoyl-2,3-dibromopropionic acid reacts with 4-substituted o-phenylenediamines to give 3-aryl-2-carboxymethylene-1,2-dihydroquinoxalines.

Reactions of isopropyl 2-(1-aryl-4,5-dioxo-2-phenyl-4,5-dihydro-1H-pyrrol-3-yl)-2-oxoacetates with alkylamines involve pyrroledione-pyrroledione recyclization with formation of isopropyl 1-aryl-2-hydroxy-4,5-dioxo-3-[phenyl(alkylamino)methylidene]pyrrolidine-2-carboxylates.     

Synthesis and Properties of 6-Amino-7-hetaryl-5-R-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles

We have established that when 5-chloro-6-[cyano(2,3-dihydro-1-R-benzo[d]azol-2-yl)methyl]-2,3-pyrazinedicarbonitriles are reacted with nucleophilic reagents (aliphatic and aromatic amines, hydrogen sulfide), annelation of the five-membered ring occurs on the [b] face of the pyrazine with formation of 6-amino-7-hetaryl-5-R-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles and 6-amino-7-(1H-benzo[d]imidazol-2-yl)thieno[2,3-b]pyrazine-2,3-dicarbonitrile respectively. Further heating with excess of acylating reagent leads to formation of a novel heterocyclic system 1H-benzo[4,5]imidazo[1,2-c]pyrazino[2',3':4,5]pyrrolo[3,2-e]pyrimidine. Reaction of vicinal dinitriles with hydrazine hydrate leads to the novel system 1H-pyrrolo[2',3':5,6]pyrazino[2,3-d]pyridazine.     

Recyclization of Pyrrolediones with Arylaminoindenones. Synthesis of Indeno[1,2-<Emphasis Type="Italic">b</Emphasis>]pyridines

Dimethyl 1-aryl-4,5-dioxo-4,5-dihydro-1H-pyrrole-2,3-dicarboxylates reacted with 3-arylamino-1H-inden-1-ones to give the corresponding methyl 1-aryl-3-[2-(arylamino)-2-oxoacetyl]-2,5-dioxo-2,5-dihydro-1H-indeno[1,2-b]pyridine-4-carboxylates.     

Synthesis of 1-aryl-2-[2-(dimethylamino)ethyl]-1,2-dihydrochromeno[2,3-c]pyrrole-3,9-diones and their analogs

Methyl o-hydroxybenzoylpyruvate heated with N,N-dimethylethylenediamine and aromatic aldehydes affords in a high yield 5-aryl-3-hydroxy-4-(2-hydroxyphenyl)-1-[2-(dimethylamino)ethyl]-1,5-dihydro-2H-pyrrol-2-ones, which easily split off water at boiling in acetic acid and are converted into 1-aryl-2-[(2-dimethylamino)ethyl]-1,2-dihydrochromeno[2,3-c]pyrrol-3,9-diones. The developed route of synthesis provides a wide range of derivatives of 1-aryl-2-[ω-(dialkylamino)alkyl]-1,2-dihydrochromeno[2,3-c]pyrrole-3,9-diones.     

Pyrolysis and photolysis of 1-aroylamino-4,5-diaryl-1,2,3-triazoles: Generation and thermal transformations of 4,5-diaryl-1,2,3-triazolyl radicals

The pyrolysis of 1-aroylamino-4,5-diphenyl-1,2,3-triazoles 1 yields, pressumably via the 4,5-diphenyl-1,2,3-triazolyl radical ( 2a ), 2,3-diphenyl-2H-azirine ( 11a ) and 2-aryl-4,5-diphenylimidazoles 14 as the major products. Upon irradiation 1-benzoylamino-4,5-diphenyl-1,2,3-triazole ( 1a ) gives 4,5-diphenyl-1 (2)H-1,2,3-triazole ( 4a ) via the 1,2,3-triazolyl radical 2a , together with benzamide ( 5a ) and 1,2-bisbenzoylhydrazine ( 6a ). Products 5a and 6a result from the benzoylamino radical 3a by hydrogen atom abstraction and dimerization respectively.     

Experimental and Theoretical DFT Investigations in the [2,3]-Wittig-Type Rearrangement of Propargyl/Allyl-Oxy-Pyrazolones

Here we report the synthesis of interesting 3-alkyl-4-hydroxy-1-aryl-4-(propa-1,2-dienyl)1H-pyrazol-5(4H)-ones and 9-alkyl-7-aryl-1-oxa-7,8-diazaspiro[4.4]nona-3,8-dien-6-ones, starting from 1,2-diaza-1,3-dienes (DDs) and propargyl alcohol. The reaction proceeds through a sequence Michael-type nucleophilic attack/cyclization/[2,3]-Wittig rearrangement. In the same way, the reaction between the aforementioned DDs and allyl alcohol furnished 4-allyl-4-hydroxy-3-alkyl-1-aryl-1H-pyrazol-5(4H)-ones. A DFT study was also carried out, in order to have decisive clarifications about the mechanism.
