The binding stoichiometry, strength and structure of inclusion complexes formed between the neurotoxin tetramethylenedisulfotetramine (TETS) and both native and modified cyclodextrins (CyDs) were investigated using nuclear magnetic resonance (NMR) spectroscopy. Of all six examined cases, native β‐cyclodextrin (β‐CyD) and its chemically modified counterpart heptakis‐(2,3,6‐tris‐(2‐hydroxypropyl))‐β‐cyclodextrin (2HP‐β‐CyD) were found to associate most strongly with TETS as reflected in the magnitude of their binding constants (K = 537 ± 26 M?1 for β‐CyD and K = 514 ± 49 M?1 for 2HP‐β‐CyD). Two‐dimensional rotating‐frame Overhauser effect spectroscopy NMR experiments confirm close proximity of the TETS molecule to both β‐CyD and 2HP‐β‐CyD as intermolecular, through‐space interactions between the H3 and H5 protons located in the interior of the CyD cavity and the methylene protons of TETS were identified. Computationally and spectroscopically assisted analytical comparative investigation into the extraction of bisphenol A using three cyclodextrins, that is, α, β, and γ respectively, were performed. A simple, self-tailored μ-solid-phase extraction podium was used to extract bisphenol A from water samples, and high-performance liquid chromatography-ultraviolet was used for the qualitative and quantitative analysis of bisphenol A. Density functional theory first principle calculations, attenuated total reflectance Fourier-transform infrared spectroscopy and Fourier-transform Raman spectroscopy data supports the analytical selection of β-cyclodextrin as the adsorbent for bisphenol A extraction. Analytical optimization of various parameters including sample volume, sample pH, eluting solvent and its volume was performed to discover the most proper conditions for maximum extraction. Under the optimized conditions, a limit of detection value of 0.70 ng/ml and a limit of quantification value of 2.31 ng/ml was achieved with β-cyclodextrin, with recovery (%) values over 98.40–102.50 in real source water samples. Overall, well assisted by comprehensive computational and spectroscopic studies, a novel, simple, sensitive and economic analytical method was developed for the extraction of bisphenol A from source water using cyclodextrin. 相似文献

Hollow mesoporous SiO2 (mSiO2) nanostructures with movable nanoparticles (NPs) as cores, so‐called yolk‐shell nanocapsules (NCs), have attracted great research interest. However, a highly efficient, simple and general way to produce yolk‐mSiO2 shell NCs with tunable functional cores and shell compositions is still a great challenge. A facile, general and reproducible strategy has been developed for fabricating discrete, monodisperse and highly uniform yolk‐shell NCs under mild conditions, composed of mSiO2 shells and diverse functional NP cores with different compositions and shapes. These NPs can be Fe3O4 NPs, gold nanorods (GNRs), and rare‐earth upconversion NRs, endowing the yolk‐mSiO2 shell NCs with magnetic, plasmonic, and upconversion fluorescent properties. In addition, multifunctional yolk‐shell NCs with tunable interior hollow spaces and mSiO2 shell thickness can be precisely controlled. More importantly, fluorescent‐magnetic‐biotargeting multifunctional polyethyleneimine (PEI)‐modified fluorescent Fe3O4@mSiO2 yolk‐shell nanobioprobes as an example for simultaneous targeted fluorescence imaging and magnetically guided drug delivery to liver cancer cells is also demonstrated. This synthetic approach can be easily extended to the fabrication of multifunctional yolk@mSiO2 shell nanostructures that encapsulate various functional movable NP cores, which construct a potential platform for the simultaneous targeted delivery of drug/gene/DNA/siRNA and bio‐imaging. 相似文献

Amphiphilic fluorescent graft copolymer (PVP‐PyATAm) was successfully synthesized by the free radical copolymerizations of hydrophobic monomer N‐acryloyl‐thioureylene‐4‐(1‐pyrene)‐butyryl amide (PyATAm) with hydrophilic precursor polymers of vinyl‐functionalized poly (N‐vinylpyrrolidone) (Acryloyl‐PVP) in DMF. FT‐IR, 1H NMR, TEM, gel permeation chromatography‐multi‐angle laser light scattering, UV‐vis spectroscopy, viscometric measurement, and fluorescence spectroscopy were used to characterize this copolymer. The TEM observation showed that the copolymer PVP‐ PyATAm formed spherical micelles in an aqueous solution and the size of micelles was between 50 and 70 nm in diameter. The interaction of PVP‐PyATAm copolymer and plasmid DNA was examined by agarose gel electrophoresis and TEM. Results indicated that the copolymer–DNA complexes were self‐assembled and the size of complexes was between 90 and 120 nm in diameter. Cytotoxity studies using MTT colorimetric assays suggested good biocompatibility of PVP‐PyATAm in vitro. These results suggested the potential of this graft copolymer as gene delivery carrier. The synthesis, cytotoxicity, and behavior in cell culture of a new set of first- (G1) and second-generation (G2) dendrimers is reported. The surface functionality of these dendrimers has been varied to see whether structure/toxicity relations can be observed. The outermost functional groups are amines that are decorated either with protons, tert-butoxycarbonyl (Boc) or benzyloxycarbonyl (Cbz) protecting groups, Boc-protected or unprotected natural amino acid residues, ethylenediamine ligands, and/or dansyl fluorescence labels. The cytotoxicity was determined in vitro in concentration-dependent assays using the human MCF-7 breast cancer cell line. Cellular uptake and intracellular distribution was monitored by confocal fluorescence microscopy after internalization of the dansyl-labeled dendrimers by HeLa cells. 相似文献

Well-defined α,ω-methacryloyl poly-ε-caprolactone (PCL) and poly(d,l )-lactide P(D,L)LA dimacromonomers have been synthesized by living ring-opening polymerization of the parent monomers initiated by diethylaluminum 2-hydroxyethylmethacrylate (Et2Al O (CH2)2 O C(O) C(CH3)CH2) and terminated by reaction of the propagating Al alkoxide groups with methacryloyl chloride. These dimacromonomers have been copolymerized with a hydrophilic comonomer, i.e., 2-hydroxyethylmethacrylate, in bulk at 65°C by using benzoyl peroxide as a free-radical initiator. The swelling ability of the amphiphilic PHEMA/PCL or P(D,L)LA networks has been investigated in both aqueous and organic media. Effect of network composition and molecular weight of the dimacromonomer on the swelling kinetics and the equilibrium solvent uptake has been studied. Lipophilic dexamethasone acetate and the hydrophilic sodium phosphate counterpart have been incorporated into the amphiphilic gels and their release has been studied in relation to the gel characteristics. © 1999 John Wiley & Sons, Inc. Solvent-driven aggregation of a series of porphyrin derivatives was studied by UV/Vis and circular dichroism spectroscopy. The porphyrins are characterised by the presence in the meso positions of steroidal moieties further conjugated with glucosyl groups. The presence of these groups makes the investigated macrocycles amphiphilic and soluble in aqueous solvent, namely, dimethyl acetamide/water. Aggregation of the macrocycles is triggered by a change in bulk solvent composition leading to formation of large architectures that express supramolecular chirality, steered by the presence of the stereogenic centres on the periphery of the macrocycles. The aggregation behaviour and chiroptical features of the aggregates are strongly dependent on the number of moieties decorating the periphery of the porphyrin framework. In particular, experimental evidence indicates that the structure of the steroid linker dictates the overall chirality of the supramolecular architectures. Moreover, the porphyrin concentration strongly affects the aggregation mechanism and the CD intensities of the spectra. Notably, AFM investigations reveal strong differences in aggregate morphology that are dependent on the nature of the appended functional groups, and closely in line with the changes in aggregation mechanism. The suprastructures formed at lower concentration show a network of long fibrous structures spanning over tens of micrometres, whereas the aggregates formed at higher concentration have smaller rod-shaped structures that can be recognised as the result of coalescence of smaller globular structures. The fully steroid substituted derivative forms globular structures over the whole concentration range explored. Finally, a rationale for the aggregation phenomena was given by semiempirical calculations at the PM6 level. 相似文献

1,2-Naphthalene-ring-expanded tetraazachlorins (TACs), tetraazabacteriochlorins (TABCs), and tetraazaisobacteriochlorins (TAiBCs) have been synthesized. Procedures for the synthesis of the starting materials, that is, derivatives of 1,2-naphthalenedicarboxylic acid, have been reinvestigated and improved. Nine possible derivatives, including four, two, and three structural isomers of TACs, TABCs, and TAiBCs, respectively, were separated by using thin-layer chromatography (TLC) or high-performance liquid chromatography (HPLC), and the structure of each isomer was determined by (1)H NMR spectroscopy combined with the NOE technique. The formation ratio of each isomer was rationalized in terms of the intramolecular steric repulsion effect, which was predicted by geometry optimizations at the DFT level. The derived compounds were characterized by using IR, electronic, and magnetic circular dichroism (MCD) spectroscopy, and by electrochemical methods. Frequency calculations at the DFT level correctly reproduced the experimental IR spectra and, in particular, distinguished between the three isomers of the TAiBCs. In the electronic absorption and MCD spectra of the TAC and TABC species, the Q-band splits into two intense components similarly to the 2,3-naphthalene-fused derivatives described in our preceding paper, although no significant spectral differences were observed from species to species. On the other hand, the spectra of the TAiBCs showed moderate differences depending on the structure of the isomer. The spectroscopic properties as well as the electrochemical behavior of these chlorins resemble those of the corresponding benzene-fused derivatives rather than the 2,3-naphthalene-fused derivatives. Molecular-orbital and configuration-interaction calculations within the framework of the ZINDO/S method were helpful in the discussions of the above observations. 相似文献

Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxy-butyrate)-poly(ethyl ethylene
phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation
chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher
content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers
self-assemble into biodegradable nanoparticles with a core-shell micellar structure in aqueous solution, verified by the probe-based
fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate)
(PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and
size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles
as a model drug and a sustained drug release from the micelles is observed. MTT assay also demonstrates that the block copolymers
are biocompatible, rendering these copolymers attractive for drug delivery.

Oral delivery remains a challenge for a number of drug candidates with low absorption profile (poor membrane permeability, lack of stability, solubility issues or susceptibility to enzymatic degradation) and various methodologies have been investigated to overcome it. The approach presented here consists of associating, by ion-pairing, a hydrophilic, ionizable model drug to a series of synthetic counter-ionic entities with a controlled degree of lipophilicity in order to enhance its penetration of biomembranes and offer some protection against in situ degradation, while preserving its biologically active chemical structure. We report herein the synthesis of a series of positively charged potential penetration enhancers designed from d-glucose, 2-aminododecanoic acid, and additional lipophilic amino acids, and converted afterward into quaternary ammoniums in an optimized, innovative one-step reaction. Each liposaccharide derivative synthesized was fully characterized and their ability to generate micelles in solution was assessed by isothermal titration calorimetry. 相似文献
