The effect of Zn2+ on the O2- generation and change in intracellular Ca2+ concentration ([Ca2+]i) of rat peritoneal neutrophils was studied. Zymosan (serum-treated zymosan (STZ))-induced O2- generation was inhibited by Zn2+ at concentrations as low as 10 microM. A large amount of the inhibition was observed in the absence of extracellular Ca2+ but the inhibition could not be restored by increasing the extracellular Ca2+ concentration, indicating that Zn2+ does not necessarily inhibit the O2- generation competitively with extracellular Ca2+. In the absence of extracellular Ca2+, Zn2+ inhibited STZ-induced transient increase in [Ca2+]i in the concentration range that evoked a marked inhibition in the O2- generation. On the other hand, Zn2+ did not inhibit significantly STZ-induced uptake of 45Ca2+ from extracellular medium by the cells. From these results, it is suggested that Zn2+ inhibits STZ-induced release of Ca2+ from intracellular storage sites, resulting in the suppression of the activation mechanism of neutrophils.  相似文献

The effect of regucalcin, a calcium-binding protein isolated from rat liver cytosol, on cytosolic Ca2+/calmodulin-dependent protein kinase activity was investigated. The increase in cytosolic Ca2+/calmodulin-dependent protein kinase activity with passage of incubation time was clearly prevented by the presence of regucalcin (1.0 microM). An appreciable effect of regucalcin was seen at 0.5 microM. The cytosolic Ca2+/calmodulin-dependent protein kinase activity was fairly increased by increasing concentrations of added Ca2+ (0.25-1.0 mM). This increase was clearly blocked by the presence of regucalcin (1.0 microM). The inhibitory effect of regucalcin on the protein kinase activity was also seen with varying concentrations of calmodulin (2.5-15 micrograms). In the presence of regucalcin (1.0 microM), trifluoperazine (50 microM), an antagonist of calmodulin, significantly decreased the cytosolic Ca2+/calmodulin-dependent protein kinase activity. These results suggest that regucalcin can regulate the Ca2(+)-calmodulin effect in liver cytosol.  相似文献

The unicellular cyanobacterium Synechocystis sp. PCC 6803 (Syn6803) exhibits photomovement through gliding motility. For a better understanding of photomovement in Syn6803, we examined the effects of Ca2+ on photoorientation and motility using a computer-assisted videomicroscope motion analysis system. When calcium ion was chelated from the basic motility medium by adding 0.5 mM ethylene glycol-bis-(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA), the photoorientation was completely inhibited, whereas the gliding motility remained approximately 70% of the control. Photoorientation impaired by EGTA was nearly recovered within 30 min upon addition of 1 mM Ca2+. The recovery of photoorientation by Ca2+ was mimicked by either Mn2+ or Mg2+ but not by Ba2+ or Sr2+. Lanthanum ion at 10 microM completely inhibited both phototactic orientation and gliding motility of Syn6803. Furthermore, pimozide (voltage-gated L-type calcium channel inhibitor), orthovanadate (calcium efflux blocker) and A23187 (calcium ionophore) partially inhibited phototactic orientation and gliding motility. Interestingly, photoorientation was prevented with increasing concentrations of calmodulin antagonist such as trifluoperazine (TFP) and chlorpromazine, but gliding motility was inhibited in proportion to the concentration of TFP. The results we present strongly indicate that Ca2+ plays a significant role in regulating the photomovement of Syn6803.  相似文献

The effect of regucalcin, a calcium-binding protein isolated from rat liver cytosol, on glucose-6-phosphatase in the microsomes of rat liver was investigated. Addition of Ca2+ up to 2.5 microM to the enzyme reaction mixture caused a significant increase of glucose-6-phosphatase activity in hepatic microsomes, while Ni2+, Zn2+, Cd2+, Cu2+, Mn2+ and Co2+ (20 microM) did not have an appreciable effect. Vanadate (V5+) markedly inhibited the enzyme activity; a significant inhibitory effect was seen at 10 microM V5+. The Ca2+-induced increase of glucose-6-phosphatase activity was reversed by the presence of regucalcin; the effect was complete at 1.0 microM of the protein. Regucalcium had no effect on the basal activity of the enzyme. Meanwhile, the inhibitory effect of V5+ (10-100 microM) on glucose-6-phosphatase was not appreciably blocked by the presence of regucalcin (up to 2.0 microM). The present data suggest that hepatic microsomal glucose-6-phosphatase is uniquely regulated by Ca2+ and V5+, of various metals, and that the Ca2+ effect is reversed by regucalcin. The present study supports the view that regucalcin plays an important role as a regulatory protein in liver cell function related to Ca2+.  相似文献

The uptake of Cu+ by rat liver mitochondria is rapid and extensive. Respiration is stimulated by 10 microM Cu+ then inhibited and the inhibition could not be relieved with uncoupling agents. Collapse of the membrane potential is induced by 5-10 microM Cu+. These effects are partially inhibited by radical scavengers indicating the involvement of radical production in these events. Reduction of the GSH content and production of peroxidation products by higher amounts of Cu+ was also demonstrated. Swelling of non-respiring rat liver and heart mitochondria in sodium or lithium acetate was used to study effects of Cu+ on the Na+/H+ exchanger. Swelling is stimulated by 5-100 microM Cu+. In the presence of a radical scavenger the swelling is reduced. In sodium nitrate media diltiazem-sensitive stimulated swelling is observed. Amiloride was found to inhibit Cu(+)-induced efflux of Ca2+. At high concentrations of Cu+, a general increase in permeability was the dominant feature.  相似文献

The effect of the calcium-binding protein regucalcin on the Ca2+ transport system in the liver microsomes from fed rats was investigated. Ca2+ transport was assayed by the method of Millipore filtration to estimate microsomal 45Ca2+ accumulation following addition of 10 mM adenosine triphosphate (ATP). 45Ca2+ uptake was retarded by the presence of regucalcin (1.0-4.0 microM). This retardation was remarkable at 1 min after regucalcin addition, while appreciable retardation was no longer seen at 5 min. Regucalcin (2.0 microM)-induced retardation of 45Ca2+ uptake was prevented by the presence of calmodulin (5 micrograms/ml). Calmodulin alone (1 and 5 micrograms/ml) caused a significant increase in 45Ca2+ uptake at 3 min after the start of incubation. Also, regucalcin (2.0 microM)-induced retardation of 45Ca2+ uptake was completely blocked by the presence of a Ca2(+)-trapping agent, oxalate (3 mM). On the other hand, 45Ca2+, which accumulated in microsomes during 5 min after ATP addition, was markedly released by the addition of regucalcin. This release was dose-dependent (0.5-4.0 microM). Guanosine triphosphate (GTP; 10-100 microM) caused a significant release of 45Ca2+ from the microsomes. The presence of regucalcin (2.0 microM) further enhanced the GTP effect. Regucalcin (2.0 microM)-induced release of 45Ca2+ was not blocked by the presence of the protein thiol-protecting agent dithiothreitol (0.1 mM). The presence of oxalate (3 mM) completely blocked the effect of regucalcin on 45Ca2+ release from the microsomes. These results indicate that regucalcin stimulates Ca2+ release from liver microsomes, and that the protein retards the microsomal Ca2+ uptake. The present study suggests that regucalcin can regulate the Ca2+ transport system in rat liver microsomes.  相似文献

The fluorescence spectra of Quin 2, (2-[(2-bis-[carboxymethyl] amino-5-methylphenoxy)methyl]-6-methoxy-8-bis [carboxymethyl] aminoquiniline), a Ca^2 probe, were investigated upon incubation with Ca^2 or La^3 . The results showed that binding of La^3 to Quin 2 resulted in different fluorescent spectrum from that of Ca^2 . Based on this observation, a fluorescent method was developed for simultaneously determination of the dissociation rates of Ca^2 and La^3 from a Ca-La- calmodulin complex (Ca2La2CaM).  相似文献

The effect of various metals on uridine diphosphate (UDP)-glucuronyltransferase and beta-glucuronidase activities in rat liver microsomes was investigated. The presence of Mn2+, Cd2+, Zn2+, V5+, Ni2+, Co2+, Cu+ or Ca2+ (20 microM) in the enzyme reaction mixture did not cause a significant alteration of UDP-glucuronyltransferase activity in hepatic microsomes. Of these metals, Zn2+ and Cd2+ (20 microM) caused a remarkable increase in hepatic microsomal beta-glucuronidase activity. Appreciable effects of Zn2+ and Cd2+ on beta-glucuronidase activity were seen at 5.0 microM, and the effects were saturated at 50 microM. Ca2+ (5.0-50 microM) and/or the Ca2(+)-binding protein regucalcin (2.0 microM) did not have an appreciable effect on UDP-glucuronyltransferase and beta-glucuronidase activities in hepatic microsomes. Thus, Zn2+ and Cd2+ uniquely increased beta-glucuronidase activity. The Zn2(+)- and Cd2(+)-induced increase in beta-glucuronidase activity was completely reversed by the presence of an SH group-protecting reagent (dithiothreitol). The response of the microsomal enzyme to Zn2+ and Cd2+ (20 microM) was no longer seen after treatment with 0.2% Triton X-100 [polyoxyethylene(10)octylphenyl ether], indicating that the stimulation by these metals is dependent on membrane association. The present study suggests that, of various metals tested, Zn2+ and Cd2+ can uniquely increase hepatic microsomal beta-glucuronidase activity and that their effect is based on binding to membranous SH groups, beside the enzyme protein.  相似文献

The effect of regucalcin, a calcium-binding protein isolated from rat liver cytosol, on Ca2+-adenosine triphosphatase (ATPase) activity in hepatic microsomes was investigated. Mg2+-ATPase activity was clearly increased by the presence of 50 microM Ca2+. Regucalcin (1.0-4.0 microM) caused a remarkable elevation (about 3-fold) of Ca2+-ATPase activity. Also, Mg2+-ATPase activity was increased (about 1.6-fold) by the presence of regucalcin (2.0 and 4.0 microM). Guanosine-5'-O-(3-thiotriphosphate) (GTPrs; 10(-5) and 10(-4) M) and nicotinamide adenine dinucleotide phosphate oxidized form (NADP+; 10(-5) to 10(-3) M) or reduced form (NADPH; 10(-4) and 10(-3) M) significantly increased Ca2+-ATPase activity. These increases were not enhanced by the presence of regucalcin (2.0 microM). Of various metal ions, a comparatively low concentration of V5+ (10(-5) M) or Cd2+ (10(-6) M) significantly increased Ca2+-ATPase activity, while Hg2+, Zn2+, Cu2+ and Mn2+ did not have such an effect. Regucalcin (2.0 microM) did not enhance the effect of V5+ and Cd2+ on Ca2+-ATPase activity. The present finding, that regucalcin activates hepatic microsomal Ca2+-ATPase, suggests a cell physiological role of regucalcin as an activator in the microsomal Ca2+-pump activity. This action of regucalcin may not be influenced by other regulators.  相似文献

The effect of lead (Pb) on the sorption of phenol onto montmorillonite-based sorbents was studied using a batch equilibration method when phenol and Pb were sorbed simultaneously and either Pb or phenol was previously sorbed. The sorbents were Na(+)-, Ca(2+)-, hexadecyl trimethylammonium (HDTMA)-Na(+)-, and HDTMA-Ca(2+)-montmorillonites. Pb diminished the sorption of phenol largely onto Na(+)- or Ca(2+)-montmorillonites, while phenol had little effect on the sorption of Pb onto all sorbents. Pb had no effect on the sorption of phenol onto HDTMA-Na(+)- or HDTMA-Ca(2+)-montmorillonites either. The sorption capacity of phenol followed the order HDTMA-Na(+)- > HDTMA-Ca(2+)- > Na(+)- > Ca(2+)-montmorillonites. The pseudo-second-order equation described the kinetics of phenol sorption well. Sorption isotherms of phenol followed the Freundlich equation. Phenol sorption on HDTMA-Na(+)- and HDTMA-Ca(2+)-montmorillonites was linear, while that on Na(+)- and Ca(2+)-montmorillonites was nonlinear.  相似文献

Two Ca2(+)-dependent membrane-binding proteins with apparent molecular weights of 70000 (calelectrin70) and 32000 (calelectrin32) were isolated from bovine liver using phenyl-Sepharose affinity chromatography followed by diethylaminoethyl (DEAE)-cellulose and Ultrogel AcA44 chromatographies. Limited proteolysis and immunological analyses indicated that calelectrin32 was not a digested product from calelectrin70. Both calelectrins bound to phosphatidylserine and to calmodulin in a Ca2(+)-dependent manner. Circular dichroism studies showed that the apparent alpha-helical contents of calelectrin70 and calelectrin32 were 25 and 40%, respectively and they underwent Ca2(+)-dependent conformational changes. When the calelectrins were incubated with a brain microtubule preparation, they were phosphorylated by endogenous kinase(s) and phosphorylation occurred on serine residues. Moreover, calelectrin70 showed an inhibitory action on endogenous kinase activity in the presence of Ca2+.  相似文献

The membrane potential (Um), the main factor of the excitation-contraction coupling, of human allantochorial placental vascular smooth muscle cells (VSMCs) has been previously shown to depend on voltage-sensitive K+ channels. These channels were blocked by high external K+. To characterize other channels which regulated Um, various constrictor or/and vasodilators and channel blockers were used. Serotonin depolarized VSMCs, in normal medium, but induced a more marked depolarization in VSMCs predepolarized by high external K+. This depolarization was inhibited by nifedipine, a blocker of voltage-gated Ca2+ channels. Acetylcholine, sodium nitroprusside (without effect on Um in normal medium), hyperpolarized the predepolarized-high K+ medium VSMCs. This hyperpolarization was inhibited after addition of charybotoxin (a blocker of Ca2+-activated K+ channels) or/and glibenclamide (a blocker of ATP-sensitive K+ channels). A similar effect was obtained with isoproterenol. These results indicated that membrane potential of human placental allantochorial VSMCs was regulated by voltage-gated, Ca2+- and ATP-sensitive K+ channels and by voltage-dependent Ca2+ channels.  相似文献

The effect of heparin on the calcium-binding protein regucalcin-stimulated Ca2+ release from rat liver microsomes was investigated. Ca2+ release was assayed by the method of Millipore filtration to estimate microsomal 45Ca2+ accumulation following the addition of 10 mM adenosine triphosphate. The addition of regucalcin (1.0 microM) or inositol 1,4,5-trisphosphate [Ins(1,4,5)P3; 1.0 microM] stimulated 45Ca2+ release from rat liver microsomes. These effects were completely inhibited by the presence of heparin (10.0 micrograms/ml). Regucalcin did not enhance the effect of Ins(1,4,5)P3. These results suggest that regucalcin affects 45Ca2+ release involved in Ins(1,4,5)P3 action in rat liver microsomes.  相似文献

The extent of H/D exchange of the protein calmodulin in solution was monitored by mass spectrometry following electrospray ionization (ESI) of the protein. In the absence of Ca2+, approximately 115 protons are exchanged for deuteriums after 60 min. As the calmodulin is titrated with Ca2+, the extent of exchange decreases significantly (i.e., by 24 protons), indicating Ca(2+)-induced folding of the protein to a tighter, less solvent-accessible form. The extent of H/D exchange ceases to decrease when the amount of added Ca2+ is sufficient to convert greater than 80% of the calmodulin to a form bound by four calcium ions. Lysozyme, a protein of similar molecular weight, does not show a significant decrease in the extent of H/D exchange as it binds to Ca2+, indicating that the changes in H/D exchange for calmodulin reflect tertiary structural change that occur upon binding with Ca2+.  相似文献

A series of gas-phase reactions involving molecular Ca-containing ions was studied by the pulsed laser ablation of a calcite target to produce Ca(+) in a fast flow of He, followed by the addition of reagents downstream and detection of ions by quadrupole mass spectrometry. Most of the reactions that were studied are important for describing the chemistry of meteor-ablated calcium in the earth's upper atmosphere. The following rate coefficients were measured: k(CaO(+) + O --> Ca(+) + O(2)) = (4.2 +/- 2.8) x 10(-11) at 197 K and (6.3 +/- 3.0) x 10(-11) at 294 K; k(CaO(+) + CO --> Ca(+) + CO(2), 294 K) = (2.8 +/- 1.5) x 10(-10); k(Ca(+).CO(2) + O(2) --> CaO(2)(+) + CO(2), 294 K) = (1.2 +/- 0.5) x10(-10); k(Ca(+).CO(2) + H(2)O --> Ca(+).H(2)O + CO(2)) = (13.0 +/- 4.0) x 10(-10); and k(Ca(+).H(2)O + O(2) --> CaO(2)(+) + H(2)O, 294 K) = (4.0 +/- 2.5) x 10(-10) cm(3) molecule(-1) s(-1). The quoted uncertainties are a combination of the 1sigma standard errors in the kinetic data and the systematic errors in the models used to extract the rate coefficients. Rate coefficients were also obtained for the following recombination (also termed association) reactions in He bath gas: k(Ca(+).CO(2) + CO(2) --> Ca(+).(CO(2))(2), 294 K) = (2.6 +/- 1.0) x 10(-29); k(Ca(+).H(2)O + H(2)O --> Ca(+).(H(2)O)(2)) = (1.6 +/- 1.1) x 10(-27); and k(CaO(2)(+) + O(2) --> CaO(2)(+).O(2)) < 1 x 10(-31) cm(6) molecule(-2) s(-1). These recombination rate coefficients, as well as those for the ligand-switching reactions listed above, were then interpreted using a combination of high level quantum chemistry calculations and RRKM theory using an inverse Laplace transform solution of the master equation. The surprisingly slow reaction between CaO(+) and O was explained using quantum chemistry calculations on the lowest (2)A', (2)A' and (4)A' potential energy surfaces. These calculations indicate that reaction mostly occurs on the (2)A' surface, leading to production of Ca(+)((2)S) + O(2)((1)Delta(g)). The importance of this reaction for controlling the lifetime of Ca(+) in the upper mesosphere and lower thermosphere is then discussed.  相似文献

通过固相反应制备了系列Ca掺杂的Ba2Al2Si10N14O4∶Eu2+绿色荧光粉,发现当半径较大的Ba被Ca取代后导致了晶格的收缩,通过X射线衍射(XRD)测量和Unitcell软件计算发现Ca的最大掺杂量在20%。Ca掺入Eu0.4Ba1.6Al2Si10N14O4荧光粉后,可有效地提高光转换性能,并使激发光谱发生一定程度的红移和宽化,从而被近紫外宽波段光有效激发,与近紫外LED的发射光谱匹配。同时Ca的掺杂也使发射光谱发生了可控的红移,可以由520 nm的绿光红移至548 nm的黄光区域。进一步发现Eu2+的淬灭浓度随着20%Ca的掺入而降低,这是由于Ca掺入导致的晶格收缩使Eu2+离子间距离减小。最后在CIE色度图中对不同Ca,Eu浓度的荧光粉的色坐标位置进行比较,发现可通过Ca,Eu浓度的变化在很大范围内调制荧光粉的发光性能。  相似文献

A detailed study on the photophysical and complexing properties of calixarenes bearing two and four dansyl derivatives (Calix-DANS2 and Calix-DANS4) in a CH3CN/H2O mixture (60:40 v/v) is reported. Calix-DANS2 shows a high selectivity towards Hg2+ over interfering cations (Na+, K+, Ca2+, Cu2+, Zn2+, Cd2+ and Pb2+) and a sensitivity in the 10(-7) mol L(-1) concentration range. The complexation of mercury ion induces a strong fluorescence quenching due to a well-defined electron transfer process from the fluorophore to the metal center. Calix-DANS4 exhibits an extremely high affinity for Pb2+ with a high selectivity over various competing ions. The unprecedented detection limit (4 microg L(-1)) is fully compatible with the level defined by the World Health Organisation. The affinity of Calix-DANS4 for Pb2+ can be rationalized by the activation of the inert pair of electrons on Pb2+. The number of fluorophores involved in the complexation can be determined from a careful time-resolved fluorescence characterization.  相似文献

Present study demonstrated that fibrillar beta-amyloid peptide (fAbeta1-42) induced ATP release, which in turn activated NADPH oxidase via the P2X7 receptor (P2X7R). Reactive oxygen species (ROS) production in fAbeta1-42- treated microglia appeared to require Ca2+ influx from extracellular sources, because ROS generation was abolished to control levels in the absence of extracellular Ca2+. Considering previous observation of superoxide generation by Ca2+ influx through P2X7R in microglia, we hypothesized that ROS production in fAbeta-stimulated microglia might be mediated by ATP released from the microglia. We therefore examined whether fAbeta1-42-induced Ca2+ influx was mediated through P2X7R activation. In serial experiments, we found that microglial pretreatment with the P2X7R antagonists Pyridoxal-phosphate-6-azophenyl-2',4'- disulfonate (100 microM) or oxidized ATP (100 microM) inhibited fAbeta-induced Ca2+ influx and reduced ROS generation to basal levels. Furthermore, ATP efflux from fAbeta1-42- stimulated microglia was observed, and apyrase treatment decreased the generation of ROS. These findings provide conclusive evidence that fAbeta-stimulated ROS generation in microglial cells is regulated by ATP released from the microglia in an autocrine manner.  相似文献

Heavy metal cations Cd2+, Pb2+, and Hg2+ were added to substitute for Ca2+ in culture media to study their effect on the relationship between CaM and the proliferation of cultured rabbit aortic smooth muscle cells (ASMC). It was found that all the heavy metal cations studied stimulated the proliferation of ASMC in varying degrees, increased the CaM content in cells at late G1 stage and decreased the activity of cAMP PDE. These results suggest that the adverse effect of heavy metals may be related to the pathogenesis of atherosclerosis and hypertensive disease.  相似文献

The electrophoretic properties of purified calmodulin were investigated. High performance capillary electrophoresis of this Ca2+(-)binding protein in free solution at pH 2.5 resulted in an elution of a single peak with a retention time of approximately 4.7 min. Addition of [ethylene-bis(oxyethylenenitrilo]) N,N,N',N'-tetraacetic acid (EGTA) to the protein prior to capillary electrophoresis completely abolished this electrophoretic profile. Polyacrylamide gel electrophoresis of calmodulin under denaturing and nondenaturing conditions also revealed a single polypeptide band. However, the relative electrophoretic mobilities of this protein could vary, depending on the presence or absence of Ca2+. The pI of calmodulin was estimated to be 3.7 by using isoelectric focusing techniques. Analysis of this acidic protein by high performance capillary electrophoresis at pH 8.0 revealed that it could be resolved into two major and one minor polypeptide peaks, regardless of the presence or absence of Ca2+. These findings suggest that capillary electrophoresis at near physiological pH may differentiate the microheterogeneity of calmodulin.  相似文献
