Condensed tetrazolo-1,3,5-triazines. 4. Synthesis of salts of 5-aminotetrazolo[1,5-a]-1,3,5-triazin-7-one

2-Amino-4-azido-1,3,5-triazin-6(1H)-ones were synthesized by successive substitution of the trinitromethyl groups in 2-amino-4,6-bis(trinitromethyl)-1,3,5-triazines under the influence of azide and nitrite ions. Interaction of 2-amino-4-azido-1,3,5-triazin-6(1H)-ones with bases led to the azido-tetrazole tautomeric conversion give salts of 5-aminotetrazolo[1,5-a]-1,3,5-triazin-7-one. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 8, pp. 1211–1219, August, 2006.     

6-氨基-5-氰基-3-甲基-4-(3-硝基苯)-1-苯基吡唑[3,4-b]并吡啶与人血清白蛋白结合作用的光谱特征

采用紫外光谱法和荧光光谱法研究了6-氨基-5-氰基-3-甲基-4-(3-硝基苯)-1-苯基吡唑[3,4-b]并吡啶(6A)与人血清白蛋白(HSA)的结合作用,利用同步荧光法和三维荧光法研究了6A与HSA作用前后人血清白蛋白的构象变化。观测到生理pH7.4条件下6A使HSA的紫外吸收峰增强,特征荧光峰猝灭。Stern-Volmer曲线显示,6A对HSA的荧光猝灭可能是一个单一的静态猝灭过程,并且得出18℃和37℃时的结合位点数和结合常数。根据F rster非辐射转移理论可求出6A与HSA作用距离r=3.73 nm;根据基本热力学参数ΔH、ΔS和ΔG判断6A和HSA主要通过氢键和范德华力发生相互作用。     

Laser photolysis of 1-(4-nitrophenyl)-3-methylpyrazole and 1-ethyl-3-(4-nitrophenyl)-5-chloropyrazole

The spectral kinetic characteristics of the triplet states of 1-(4-nitrophenyl)-3-methylpyrazole (1) and 1-ethyl-3-(4-nitrophenyl)-5-chloropyrazole (2) were studied by the laser nanosecond photolysis technique in different solvents. The triplet lifetimes (τ_T) of molecules 1 and 2 were found to depend strongly on the solvent nature. An increase in τ_T by approximately two orders of magnitude on going from nonpolar and polar aprotic solvents (τ_T ≤ 15 ns) to aqueous-acetonitrile solutions (τ_T = 1100 ns for a volume acetonitrile to water ratio of 1 : 3) was analyzed, taking into account the influence of the medium on the relative contribution of the n,π*- and π,π*-configurations to the lowest triplet state. __________ Published in Russian in Izvestiya Akademii Nauk. D-脯氨酸催化的(3S,4S)-4-氨基-3-羟基-5-苯基戊酸衍生物的合成

报道了一条合成(3S,4S)-4-氨基-3-羟基-5-苯基戊酸(Ahppa)衍生物的新路线。以氨基保护的L-苯丙氨酸为起始原料,依次经Weinreb胺缩合、还原、aldol缩合及溴仿4步反应合成了3个Ahppa衍生物,总收率5.8%~6.7%,其结构经¹H NMR, ¹³C NMR和ESI-MS确证。对反应条件进行了探讨,结果表明：催化剂D-脯氨酸用量对反应收率影响不大,对立体选择性影响较大;氨基上保护基体积较大有利于提高反应立体选择性。     

One-step synthesis of 5-acetyl-2-amino-4-aryl-3-cyano-4H-pyrano[3,2-b]indoles. Molecular and crystal structure of 5-acetyl-2-amino-4-(4"-chloro-3"-nitrophenyl)-3-cyano-4H-pyrano[3,2-b]indole

A one-step procedure was developed for the synthesis of 5-acetyl-2-amino-4-aryl-3-cyano-4H-pyrano[3,2-b]indoles involving the three-component reaction of 1-acetylindol-3(2H)-one with aromatic aldehydes and malononitrile in ethanol in the presence of triethylamine as the catalyst. The structure of 5-acetyl-2-amino-4-(4"-chloro-3"-nitrophenyl)-3-cyano-4H-pyrano[3,2-b]indole was established by X-ray diffraction analysis.     

Synthesis and structures of 4,6-disubstituted 2-(5-methyl-1,2,4-oxadiazol-3-yl)-1,3,5-triazines

New 1,2,4-oxadiazolyl-1,3,5-triazines were synthesized from amidoximes derived from sym-triazine mononitriles. The use of aliphatic aldehydes for the synthesis of 4-alkyl-6-amino- 3,5-dicyanopyridine-2(1H)-thiones

The condensation of aliphatic aldehydes with malononitrile and cyanothioacetamide gives 4-alkyl-2,6-diamino-3,5-dicyano-4H-thiopyrans which recyclize into 4-alkyl-6-amino3,5-dicyanopyridine-2(IH)-thiones. 研究了1-（2-羟基-3,5-二硝基苯基）-3-[4（苯基偶氮）苯基]-三氮烯（HDNPAPT）试剂与汞离子的显色反应。在非离子表面活性剂TritonX-100存在下,pH10．3的NH3·H2O-NH4Cl介质中,汞与HDNPAPF形成1：4的红色络合物,表观摩尔吸光系数ε525＝1．63×105·mol-1·cm－1,汞离子浓度在0～0．64mg/L范围内符合比尔定律。方法应用于废水中微量汞的测定。     

Synthesis of 6-amino-4-aryl-5-cyano-3-(3-cyanopyridin-2-ylthiomethyl)-2,4-dihydropyrano[2,3-c]pyrazoles and their hydrogenated analogs. Molecular structure of 6-amino-5-cyano-3-(3-cyano-4,6-dimethylpyridin-2-ylthiomethyl)-4-(2-nitrophenyl)-2,4-dihydropyrano[2,3-c]pyrazole

Substituted 3-hydroxypyrazoles, which were prepared based on ethyl esters of substituted 4-(pyridin-2-ylthio)- or 4-(1,4-dihydropyridin-2-ylthio)acetoacetic acids and hydrazine hydrate, were used in the synthesis of 6-amino-4-aryl-5-cyano-3-(pyridin-2-ylthiomethyl)-2,4-dihydropyrano[2,3-c]pyrazoles. The molecular and crystal structure of 6-amino-5-cyano-3-(3-cyano-4,6-dimethylpyridin-2-ylthiomethyl)-4-(2-nitrophenyl)-2,4-dihydropyrano[2,3-c]pyrazole was established by X-ray diffraction analysis.     

Synthesis and transformations of 2-(5-amino-(mercapto)-1,3,4-thiadiazolylthio)-7-methyl-5-oxo-5H-1,3,4-thiadiazolo[3,2-a]pyrimidines

The reactions of 2-amino-5-mercapto-(or 2,5-dimercapto)-1,3,4-thiadiazoles with 2-bromo-7-methyl-5-oxo-5H-1, 3,4-thiadiazolo[3, 2-a]pyrimidine to give the corresponding sulfides have been studied. The possibility of S-alkylation and addition of quinone at the free mercapto group in the 1,3,4-thiadiazole ring has been shown. The reactions at the amino group with benzoyl chloride and chloroformates have been investigated. The conditions of cyciodehydration at the amino group with ethyl acetoacetate and bromination of the pyrimidine fragment of 7-methyl-5-oxo-5H-1,3,4-thiadiazolo[3,2-a]pyrimidine have been found.Translated fromIzvestiya Akademii Nauk. 1-(2-羟基-3,5-二硝基苯基)-3-[4-(苯基偶氮)苯基]-三氮烯-阴离子表面活性剂-季铵盐型阳离子表面活性剂显色反应及其应用

在三乙醇胺介质中,新试剂1-(2-羟基-3,5-二硝基苯基)-3-[4-(苯基偶氮)苯基]-三氮烯(HDNPAPT)与季铵盐型阳离子表面活性剂及阴离子表面活性剂(AS)反应显色,形成三元络合物.研究了显色体系的条件实验,建立了测定阴离子表面活性剂十二烷基苯磺酸钠(DBOSO_3Na)、十二烷基磺酸钠(DOSO_3Na)和十二烷基硫酸钠(DSO_4Na)的方法,表观摩尔吸光系数均达10~4以上.方法已应用于环境水样中阴离子表面活性剂的测定.     

Novel Approaches to Synthesis of 4-Alkyl-6-amino-5-cyano-3-methyl(propyl, phenyl)-2H,4H-pyrano[2,3-c]pyrazoles

We have obtained 4-alkyl-6-amino-5-cyano-3-methyl(propyl, phenyl)-2H,4H-pyrano[2,3-c]pyrazoles by reaction of 4-alkylmethylene-3-substituted 5-pyrazolones with malononitrile or cyanothioacetamide. We have used X-ray diffraction to study the structure of 6-amino-5-cyano-4-isopropyl(hexyl)-3-phenyl-2H,4H-pyrano[2,3-c]pyrazoles.     

Asymmetric synthesis of organoelemental analogs of natural compounds. 7. (2R,3S)-2-amino-3-hydroxy-5-phosphonovaleric acid

Synthesis of 4-amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-triazole-3-thione and its reactions with C-electrophiles

4-Amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-trazole-3-thione is formed from the reaction of 4,6-diphenylpyrimidinecarboxylic acid or its ethyl ester with thiocarbonyl hydrazide. Alkylation of the product leads to S-alkyl derivaties or 6-substituted 3-(4,6-diphenyl-2-pyriimidinyl)-7H-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazine. Acetylation of 4-amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-triazole-3-thione gave under different conditions monoacetyl-, diacetyl, and triacetyl derivatives at the amino group and the N₍₂₎ atom, whereas benzoylation gave a benzoyl group at the amino group and 3-(4,6-diphenyl-2-pyrimidinyl)-6-phenyl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, 1088–1094, July, 2007.     

Reaction of 6-methoxybenzo[b]furan-3(2H)-one and benzo[b]thiophen-3(2H)-one with 2-aryl-1,1-dicyanoethylenes as a convenient synthetic route to substituted 2-amino-4-aryl-1,3-dicyano-7-methoxydibenzo[b,d]furans and 2-amino-4-aryl-3-cyano-4H-benzothieno[3,2-b]pyrans

The reactions of 6-methoxybenzo[b]furan-3(2H)-one with 2-aryl-1,1-dicyanoethylenes and malononitrile or with aromatic aldehyde and two moles of malononitrile afford 2-amino-4-aryl-1,3-dicyano-7-methoxydibenzo[b,d]furans. The reactions of benzo[b]thiophen-3(2H)-one with 2-aryl-1,1-dicyanoethylenes or with aromatic aldehyde and one mole of malononitrile produce 2-amino-4-aryl-3-cyano-4H-benzothieno[3,2-b]pyrans.     

Synthesis and Anti-HIV-1 Activity of 2-[2-(3,5-Dimethylphenoxy)ethylthio]pyrimidin-4(3H)-ones

New 2-[2-(3,5-dimethylphenoxy)ethyl]thio derivatives of pyrimidin-4(3H)-one containing various substituents at positions 5 and 6 of the pyrimidine ring were synthesized. It was shown that alkylation of 2-thiouracils with 1-bromo-2-(3,5-dimethylphenoxy)ethane in DMF takes place exclusively at the sulfur atom. The obtained 6-benzyl and 6-(2,6-difluorobenzyl) derivatives have clearly defined virus-inhibiting properties with respect to type 1 human immunodeficiency virus in vitro and suppress its reproduction by 50% at concentrations of 1.3 and 11.2 mM respectively.     

Synthesis of (2-hydroxy-5-nitrophenyl)di[1-(2-hydroxynaphthyl)]methane and 12-[1-(2-hydroxynaphthyl)]-10-nitro-12H-benzo[α]xanthene from of 5-nitrosalicylaldehyde derivatives and β-naphthol

The reaction of the morpholinal or anil of 5-nitrosalicylaldehyde with β-naphthol yielded (2-hydroxy-5-nitrophenyl)di[1-(2-hydroxynaphthyl)]methane. Condensation of the latter compound in boiling nitromethane gave 12-[1-(2-hydroxynaphthyl)]-10-nitro-12H-benzo[α]xanthene. Translated fromIzvestiya Akademii Nauk. We have synthesized a series of (1R,2R,4S,5S,8S)-2,8-diaryl-4-(4-nitrophenyl)-1-aza-3,7-dioxabicyclo[3.3.0]octanes as a result of reaction of (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediol with aromatic aldehydes.
