Aryl‐furyl substituted pyrazolines 2a–c and 4a–c were prepared by the reaction of α,β‐unsaturated carbonyl compounds with hydrazine or phenyl hydrazine. N‐chloroacetyl derivatives 3a–c were obtained by the N‐acetylation of 2a–c . The antibacterial activities of synthesized pyrazolines were examined by employing the disk‐diffusion technique. All synthesized compounds showed antibacterial effects in 1200 μg concentration. © 2003 Wiley Periodicals, Inc. Parallel and practical methods for the preparation of both (E)‐ and (Z)‐β‐aryl1‐β‐aryl2‐α,β‐unsaturated esters 1 and (E)‐ and (Z)‐α‐aryl1‐β‐aryl2‐α,β‐unsaturated esters 2 are described. These methods involve accessible, robust, stereocomplementary N‐methylimidazole (NMI)‐mediated enol tosylations (14 examples, 70–99 % yield), as well as stereoretentive Suzuki–Miyaura cross‐couplings (36 examples, 64–99 % yield). The highlighted feature of the present protocol is the use of parallel and stereocomplementary approaches to obtain highly (E)‐ and (Z)‐pure products 1 and 2 by utilizing sequential enol tosylations and cross‐coupling reactions. An expeditious and parallel synthesis of (E)‐ and (Z)‐zimelidine ( 3 ), which is a highly representative selective serotonin reuptake inhibitor (SSRI), was performed by utilizing the present methods.  相似文献

The sesquiterpene capnellene‐8β, 10α‐diol ( 1 ) was isolated from non‐polar extract of the soft coral Capnella sp. Ten acylation products of capnellene‐8β, 10α‐diol were prepared: 10α‐hydroxy‐8β‐O‐benzoylcapnellene ( 2 ), 10α‐hydroxy‐8β‐Op‐toluoylcapnellene ( 3 ), 10α‐hydroxy‐8β‐O‐4‐chlorobenzoyl‐capnellene ( 4 ), 10α‐hydroxy‐8β‐O‐2‐furoylcapnellene ( 5 ), 10α‐hydroxy‐8β‐O‐2‐thiophenoylcapnellene ( 6 ), 10α‐hydroxy‐8β‐O‐4‐fluorobenzoylcapnellene ( 7 ), 10α‐hydroxy‐8β‐O‐4‐propylbenzoylcapnellene ( 8 ), 10α‐hydroxy‐8β‐O‐cinnamoylcapnellene ( 9 ), 10α‐hydroxy‐8β‐O‐4‐nitrobenzoylcapnellene ( 10 ), and 10α‐hydroxy‐8β‐O‐4‐anisoylcapnellene ( 11 ). The structures of capnellene‐8β, 10α‐diol as well as its derivatives were established through standard spectroscopic analysis. The in vitro cytotoxic activities of the eleven compounds were evaluated against Hela, KB, Daoy, and WiDr human tumor cell lines.  相似文献

A novel series of pyrazoline and thiazole derivatives incorporating 2‐pyrazolin‐5‐one moiety were synthesized starting from α,β‐unsaturated ketones under the effect of hydrazine derivatives and thiosemicarbazide. The obtained pyrazolines 4a , 4b were treated with different reagents to afford N‐substituted pyrazolines 5a , 5b , 6a , 6b , 7a , 7b , 8a , 8b . N‐Thiocarbamoyl pyrazolines 12a , 12b were cyclized using phenacyl bromide, 2,3‐dichloroquinoxaline, and monochloroacetic acid afforded the novel pyrazolinyl thiazoles 13a , 13b , 14a , 14b , 15a , 15b , 16a , 16b , 16c , 16d , 16e , 16f . The newly synthesized compounds were characterized by analytical and spectral data.  相似文献

β‐Substituted chiral γ‐aminobutyric acids feature important biological activities and are valuable intermediates for the synthesis of pharmaceuticals. Herein, an efficient catalytic enantioselective approach for the synthesis of β‐substituted γ‐aminobutyric acid derivatives through visible‐light‐induced photocatalyst‐free asymmetric radical conjugate additions is reported. Various β‐substituted γ‐aminobutyric acid analogues, including previously inaccessible derivatives containing fluorinated quaternary stereocenters, were obtained in good yields (42–89 %) and with excellent enantioselectivity (90–97 % ee). Synthetically valuable applications were demonstrated by providing straightforward synthetic access to the pharmaceuticals or related bioactive compounds (S)‐pregabalin, (R)‐baclofen, (R)‐rolipram, and (S)‐nebracetam.  相似文献

A new industrially viable process for the preparation of 1β‐(Ntert‐butyl carbamoyl)‐4‐aza‐5α‐androst‐1‐ene‐3‐one, also known by the generic name finasteride ( 6 ) from the new azaandrostane derivatives such as 1β‐(Ntert‐butyl carbamoyl)‐4‐benzoyl‐4‐aza‐5α‐androstane‐3‐one ( 4 ), 1β‐(Ntert‐butyl carbamoyl)‐4‐benzoyl‐4‐aza‐5α‐androst‐1‐ene‐3‐one ( 5 ) is reported. In this process, benzoyl group is demonstrated as a novel protecting group for lactamic NH group. The structures of newly prepared compounds were established on the basis of spectral data (IR, 1H‐NMR, and MS).  相似文献

Oximes of glucose, xylose, lactose, fructose, and mannose have been prepared. Nitrosation of the oximes of glucose, xylose, and lactose with NaNO2/HCl afforded 2‐(β‐glycopyranosyl)‐1‐hydroxydiazene‐2‐oxides, which were isolated as salts 13 , 22 , and 28 . Nitrosation of fructose oxime 29 furnished fructose, whereas nitrosation of mannose oxime 30 with NaNO2/HCl afforded the 1‐hydroxy‐2‐(β‐d‐ mannopyranosyl)diazene‐2‐oxide 32 , from which the p‐anisidinium salt 31 and the sodium salt 33 were prepared. However, nitrosation of 30 with isopentyl nitrite in aqueous solutions of CsOH or KOH resulted in the formation of the 2‐(α‐D ‐mannofuranosyl)‐1‐hydroxydiazene‐2‐oxide salts 34 and 35 , respectively. Methylation of the ammonium 2‐(β‐D ‐glucopyranosyl)‐1‐hydroxydiazene‐2‐oxide 13 yielded the 1‐methoxy compound, which was benzoylated to afford the tetra‐O‐benzoate 14 a , the structure of which was confirmed by X‐ray diffraction analysis. From the glucose O‐methyloximes 15 and 16 the N‐methoxy‐N‐nitroso‐2,3,4,6‐tetra‐O‐acetyl‐β‐D ‐glucopyranosylamine 18 was prepared. The structure of this compound was confirmed by X‐ray diffraction analysis. Treatment of acetobromoglucose with cupferron furnished the 1‐(2,3,4,6‐tetra‐O‐acetyl‐β‐D ‐glucopyranosyloxy)‐2‐phenyldiazene‐2‐oxide 20 .  相似文献

The human lectin galectin‐1 (hGal‐1) translates sugar signals, that is, β‐galactosides, into effects on the level of cells, for example, growth regulation, and has become a model for studying binding of biopharmaceutically relevant derivatives. Bound‐state conformations of Galβ‐C‐(1→3)‐Glcβ‐OMe ( 1 ) and its βGal‐(1→3)‐βGlc‐OMe disaccharide parent compound were studied by using NMR spectroscopy (transferred (TR)‐NOESY data), assisted by docking experiments and molecular dynamics (MD) simulations. The molecular recognition process involves a conformational selection event. Although free C‐glycoside access four distinct conformers in solution, hGal‐1 recognizes shape of a local minimum of compound 1 , the synΦ/synΨ conformer, not the structure at global minimum. MD simulations were run to explain, in structural terms, the observed geometry of the complex.  相似文献

A series of new N‐aryl‐α,β‐disubstituted amides (gem or E1; trans or E2) were synthesized in good yields by carbonylative addition of aniline derivatives 1a–f to aromatic alkynes 2a,b catalyzed by Pd(OAc)2 and 1,3‐bis(diphenylphosphino)propane. The catalytic synthesis of tertiary α,β‐unsaturated amides was also successfully achieved. Traces of products were observed in the absence of p‐toluenesulfonic acid used as an additive. A series of KF/Al2O3 catalyzed Michael-addition reactions between malononitrile and α,β-unsaturated cycloketones in DMF solution were studied. At room temperature, 2-cyano-3-aryl-3-(1,2,3,4-tetrahydronaphthalen-1-one-2-yl) propionitrile derivatives were synthesized by the reaction between 2-arylmethylidene-1,2,3,4-tetra-hydronaphthalen-1-one and malononitrile. However, if the temperature was increased to 80℃, 2-amino-3-cyano-4-aryl-4H-benzo[h]chromene derivatives were obtained in high yields. When the α,β-unsaturated ketones were replaced by 2,6-biarylmethylidenecyclohexanone or 2,5-biarylmethylidenecyclopentanone, another series of 2-amino-3-cyano-4H-pyran derivatives was isolated successfully. The structures of the products were confirmed by X-ray diffraction analysis.  相似文献

The reactions of (20R)‐3β‐acetoxy‐21‐hydroxymethylpregn‐5‐en‐20‐ol ( 2 ) and (20R)‐3β‐acetoxypregn‐5‐ene‐20,21‐diol ( 11 ) with phenylphosphonic dichloride 3 and aryl dichlorophosphates 4–6 afforded novel types of P‐heterocyclic androst‐5‐ene derivatives 7–10 and 12 as epimeric pairs. The diastereomers were separated by column chromatography and were characterized by NMR spectroscopy. Estimation of the stereostructures of the corresponding epimers by B3LYP/631G(d) DFT ab initio calculations suggested that the six‐membered hetero ring in compounds 7b and 8a–10a adopts predominantly a chair conformation, with the P‐substituents in their preferred orientation. The cyclic phosphonate moiety in 7a or 8b–10b , however, seems to exist as an equilibrium mixture of chair–distorted‐boat or chairchair forms. The theoretical calculations indicate that the conformational equilibrium is shifted toward the distorted‐boat conformer for 7a , with a pseudoequatorial P‐phenyl substituent, whereas for 8b–10b the chair conformer with an equatorial P‐phenoxy group predominates. © 2008 Wiley Periodicals, Inc. Alkyl (Z)‐2‐[(E)‐2‐ethoxycarbonyl‐2‐(2‐pyridinyl)ethenyl]amino‐3‐dimethylaminopropenoates 7 and 8 were prepared from ethyl 2‐pyridinylacetate (1) in two steps. Substitution of the dimethylamino group with alkyl‐, aryl‐, or heteroarylamines afforded the corresponding β‐alkyl‐ 22–24 , β‐aryl‐ 25–35 , and β‐herteroaryl‐amino‐α,β‐didehydro‐α‐amino acid 36 and 37 derivatives, intermediates for further preparation of various heterocyclic systems. The orientation around both double bonds were determined by various nmr techniques.  相似文献

The development of efficient methods for stereocontrolled synthesis of polyol derivatives has been of continuing interest for the synthetic community. We describe herein tandem olefin cross‐metathesis/hemiacetalization/intramolecular oxa‐Michael addition of allylic/homoallylic alcohols, α,β‐unsaturated ketones, and aldehydes, which enabled the synthesis of syn‐1,2‐ and syn‐1,3‐diol derivatives in a step‐economical manner. A series of differentially protected polyol derivatives could be obtained in subsequent transformations via chemoselective/regioselective cleavage of the acetal moiety of the tandem reaction products.  相似文献

A series of α,β‐unsaturated ketones containing quinolone moieties 2 , 3 , 4 , 5 , 6 were synthesized by condensation of 7‐methoxyquinoline‐2,4(1H,3H)‐dione ( 1 ) with different aryl aldehydes. Pyrazole derivatives 8 , 9 , 10 , 11 were also synthesized via refluxing of α,β‐unsaturated ketones 2 , 3 , 4 , 5 , 6 with hydrazine derivatives. Newly synthesized compounds were characterized by elemental analyses, spectral data, and screened for their antioxidant and antitumor activities. Geometrical optimizations of the molecular structures for different synthesized compounds were studied.  相似文献

New asymmetric conjugate reduction of β,β‐disubstituted α,β‐unsaturated ketones and esters was accomplished with alkoxylhydrosilanes in the presence of chiral rhodium(2,6‐bisoxazolinylphenyl) complexes in high yields and high enantioselectivity. (E)‐4‐Phenyl‐3‐penten‐2‐one and (E)‐4‐phenyl‐4‐isopropyl‐3‐penten‐2‐one were readily reduced at 60 °C in 95 % ee and 98 % ee, respectively, by 1 mol % of catalyst loading. (EtO)2MeSiH proved to be the best hydrogen donor of choice. tert‐Butyl (E)‐β‐methylcinnamate and β‐isopropylcinnamate could also be reduced to the corresponding dihydrocinnamate derivatives up to 98 % ee.  相似文献

The structures of methyl 3β‐acetoxy‐12‐oxo‐18β‐olean‐28‐oate [C33H52O5, (I)] and methyl 3β‐acetoxy‐12,19‐dioxoolean‐9(11),13(18)‐dien‐28‐oate [C33H46O6, (II)] are described. In (I), all rings are in the chair conformation, rings D and E are cis and the other rings trans‐fused. In compound (II), only rings A and E are in the chair conformation, ring B has a distorted chair conformation, ring C a distorted half‐boat and ring D an insignificantly distorted half‐chair conformation.  相似文献
