[reaction: see text] 8-Aza-5,7-dimethyl-2-trifluoromethylchromone reacts with alkyl mercaptoacetates to give pyrido derivatives of 2-oxa-7-thiabicyclo[3.2.1]octane, which undergo the reductive ring-opening to sulfanyl acetates. The latter compounds are useful CF(3)-containing building blocks for the preparation of a variety of 2-pyridone derivatives.  相似文献

[reaction: see text] A novel one-pot synthesis of N-difluoromethyl-2-pyridones is described. N-(Pyridin-2-yl)acetamide derivatives were excellent precursors for the preparation of N-difluoromethyl-2-pyridone derivatives. Difluoromethylation of 2-acetaminopyridine derivatives was achieved with sodium chlorodifluoroacetate as a difluorocarbene source in the presence of a catalytic amount of 18-crown-6. Subsequent in situ hydrolysis of resultant 1,2-dihydro-2-acetimino-1-difluoromethylpyridines proceeded under mild acidic conditions to afford the corresponding N-difluoromethyl-2-pyridones in moderate to good yields.  相似文献

Empirical information on the acidity of the propargylic proton from our previous work allowed us to develop novel synthetic transformations of readily available terminally trifluoromethylated propargylic alcohols 1 into the corresponding allenyl tosylates 3a, 1-tosyloxy- or 1-acyloxy-4,4,4-trifluorobutan-2-ones 4, and 2-(2,2,2-trifluoroethyl)prop-2-en-1-ones 5, which was enabled by such common bases as NaOH and tertiary amines for affecting ready abstraction of this proton.  相似文献

The nitration of 5,7-dimethyl-2-polyhaloalkylchromones affords either 5,7-dimethyl-6-nitro- or 5,7-dimethyl-6,8-dinitro-2-polyhaloalkylchromones, depending on the reaction conditions. Signals in the 1H and 13C NMR spectra of the sterically hindered chromones were completely assigned using the 2D NOESY, HETCOR, and COLOC spectra. The influence of nonplanar nitro groups on chemical shifts of carbon atoms was studied. Some spectral peculiarities of the peri-methyl group were revealed. The 1H-1H and 13C-1H spin-spin coupling constants, including the extreme six-bond long-range coupling between the protons of the Me(5) group and H(8), were determined.  相似文献

A series of benzimidazole bearing 2-pyridones 5a-k were synthesized and assessed in vitro for their activity as antimicrobial agents using the conventional broth dilution method. The results of the antimicrobial study revealed that compounds 5b, 5c, Sj and 5k exhibited substantial antibacterial activity while compound 5d emerged as amore potent antifungal agent compared to the standard drugs chloramphenicol and ketoconazole, respectively. It was observed that the presence of inductively electron withdrawing groups remarkably enhance the antibacterial activity of the newly synthesized compounds. Cytotoxicity studies suggested that none of the tested compounds exhibited any significant cytotoxic effects.  相似文献

A rapid and efficient synthesis of amino-substituted 2-pyridones was demonstrated by palladium-catalyzed amination reaction under microwave irradiation. This high-speed synthesis provided a number of amino-substituted 2-pyridones from the corresponding bromo-2-benzyloxypyridines via palladium-catalyzed amination followed by hydrogenolysis of benzyl ether.  相似文献

A new method for the synthesis of aminovinyl trifluoromethyl ketones was developed. The method is based on the reactions of 4-sulfonyl-1,1,1-trifluorobut-3-ene-2,2-diols with various alkyl-, aryl-, dialkyl-, and alkylarylamines. The stereochemistry of the compounds obtained was studied.  相似文献

Synthesis of 1,6-diamino-4-phenyl-3,5-dicyano-2-pyridone derivatives via a one-pot, three-component reaction of aryl aldehydes, malononitrile, and cyanoacetic hydrazide at room temperature using KF-Al2O3 as a recyclable catalyst have been developed. The reaction proceeds through the initial Knoevenagel condensation between aldehyde and malononitrile in the presence of KF-Al2O3 to form the benzylidene derivative which then undergoes Michael addition with cyanoacetic hydrazide followed by intramolecular cyclization of the resulting intermediate to produce the N-amino-2-pyridones in good to excellent yields. The structure of the synthesized compounds were characterized and established on the basis of their spectral data analysis and single-crystal XRD analysis.  相似文献

Phase-selective photochemical reaction of 2-pyridone derivatives was examined. Irradiation of 1 in benzene mainly gave rearrangement products 2. However, intermolecular [4 + 4] photocycloaddition proceeded quantitatively in the solid state, affording photodimers 3. An effective pi-pi stacking and dipole-dipole interaction between two pyridone moieties might play important roles in an effective arrangement of 1 for photodimerization in their crystal structures. Condensation of 6-earbethoxy-4-hydroxy-2-pyridone or a silyl derivative of 5-earbomethoxy-4-hydroxy-2-pyridone with 2′,3′,5′-tri-O-benzoyl-D-ribofuranosyl halide has provided the 3-deaza analogs of orotidine and uridine-5-carboxylic acid. The corresponding amides have also been prepared in view of their possible structural relationship to l-β-D-ribohiranosyl nicotinamide. Tri-O-benzoyl-3-deazauridine was treated with N-bromosuccinimide to give, after deblocking, 3-bromo-4-hydroxy-1-(β-D-ribofuranosyl)-2-pyridone. The anomeric configuration of these nuclcosides was confirmed by pmr spectroscopy.  相似文献

3-Dicyanomethylene-3-phenylpropionitrile coupled with diazotized aromatic amines (Ar-NH2) to afford hydrazono derivatives. The latter (Ar = C6H5, 4-CH3C6H4, 4-CH3OC6H4, and 4-ClC6H4) were readily cyclized upon reflux in aqueous NaOH to 3(2H) pyridazinimine derivatives, which were readily transformed into their corresponding pyridazinone derivatives on reflux in ethanolic HCl. The pyridazinimines undergo reductive cleavage and recyclization via loss of ammonia to afford the corresponding N-aryl pyrroles on reflux in glacial acetic acid with Zn dust. The pyridazinone derivatives, however, undergo the same reaction but with loss of the aromatic amine moiety to afford only one pyrrolone derivative. The hydrazono derivative (Ar = 2-NCC6H4) was cyclized into pyridazino[3,2-b]quinazolin-6-imine, which is easily converted into pyridazino[3,2-b]quinazolin-6-one on reflux in ethanolic HCl. The latter compound is also obtained from the hydrazo derivatives (Ar = 2-HOOCC6H4 and 2-MeOOCC6H4) by reflux in aqueous NaOH.  相似文献

Various aldehydes undergo smooth coupling with 5-(hydroxymethyl)-4,6,6-trimethylcyclohex-3-enol in the presence of p-TSA at ambient temperature to afford a novel series of hexahydro-8,8-dimethyl-1H-isochromen-7-ol derivatives in good yields with high selectivity. Heating the morpholinal of nitrosalicylic aldehyde with methyl propiolate in acetonitrile in the presence of Cul affords an equimolar mixture of 2-methoxycarbonylmethyl-3-morpholino-5-nitrobenzofuran and 3-methoxycarbonyl-2-morpholino-6-nitro-2H-chromene, the structures of which were determined by X-ray analysis. Reactions of acetylenic ketones (Ia-f) with methyl α-naphthylacetate in the presence of sodium methoxide gave the corresponding 6-aryl-3-α-naphthyl-4-phenyl-2-pyrones (IIa-f) which upon refluxing with hydrazine hydrate in ethanol gave the corresponding 1-amino-2-pyridone derivatives (VIa-f).
