The determination of target analytes such as nonyl- and octylphenols and 17β-estradiol in fish homogenate require of solid–liquid extraction step. In this work microwave-assisted extraction (MAE) and focused-ultrasound liquid extraction (FUSLE) were studied as two different alternatives for extraction of the target compounds in zebrafish (Danio rerio) homogenate. In this work solid phase extraction (SPE) using 5-g and 10-g Florisil cartridges and gel permeation chromatography (GPC) were studied for the clean-up of the MAE and FUSLE extracts due to the non-selective extraction step. Although good recoveries were obtained both for SPE (106% and 126% range) and GPC (79% and 100% range) clean-up procedures, cleaner chromatograms were obtained after SPE and finally 5-g Florisil cartridges were tested since no improvement was observed when 10-g Florisil cartridges were used. Under optimized clean-up conditions, MAE and FUSLE provided comparable results for 4nOP and NP, while more accurate results were obtained for 4tOP and E2 after FUSLE. Finally, the method was applied to the determination of alkylphenols and 17β-estradiol in zebrafish homogenate that had been exposed to known concentrations of the target analytes. In the case of alkylphenols two different isomers of nonyl- and octylphenol (4-(3′,6′-dimethyl-3′-hepthyl)phenol, 363-NP, and 4-(3′-methyl-3′-hepthyl)phenol, 33-OP) were studied.  相似文献

A method to determine 21 organochlorine pesticides (OCPs) in tree leaves [chestnut (Castanea sativa), hazel (Corylus avellana), oak (Quercus robur) and walnut tree (Juglans regia)] based on microwave-assisted extraction (MAE) followed by solid-phase extraction (SPE) clean-up is described. After extraction with hexane:acetone (50:50), four different sorbents (Florisil, tandem Florisil + alumina, silica and ENVI-Carb) were assayed for the clean-up step. Pesticides were eluted with 5 mL of hexane:ethyl acetate (80:20) and determined by gas chromatography and electron capture detection (GC-ECD). Carbon was the sorbent, which provided colourless eluates and chromatograms with less interferent compounds. Analytical recoveries obtained were ca. 100% for all the studied pesticides with this sorbent.  相似文献

A fast, simple and inexpensive sample preparation method based on the matrix solid-phase extraction (SPE) technique is proposed for the isolation of cypermethrin and its metabolite residues from soils. Both the extraction and clean-up procedures were carried out in two steps and target compounds were determined by gas chromatography coupled with electron-impact mass spectrometry (GC-EI/MS). The characteristic ions and fragmentation mechanism of cypermethrin were evaluated by electron impact ionization mass spectrometry (EI/MS). After the optimization of different parameters, such as the extraction solvent, the pesticide was extracted from the matrix with methanol/acetone in a Soxhlet extractor, cleaned up on a Florisil column by elution with a mixture of 30% ethyl acetate in n-hexane and analyzed by gas chromatography-electron impact ionization mass spectrometry (GC-EI/MS) in the selected ion-monitoring mode (SIM) with permethrin as internal standard. Recovery was in the range 77–118% with relative standard deviations (RSD) between 2.5% and 10.2%. The limit of detection (LOD) was 6.5?µg/kg for cypermethrin. The developed method was linear in the injection range 6–30?ng, with correlation coefficients greater than 0.9957.  相似文献

A selective pressurized liquid extraction (SPLE) procedure capable of performing simultaneous extraction and clean-up has been demonstrated for multi-residue analysis of organochlorine pesticides (OCPs) in soil. The final method was performed at 100 degrees C for 3 x 10 min using acetone/n-heptane (1:1, v/v). Florisil was placed inside the extraction cell downstream the sample to remove interfering compounds. Extraction of two soil samples by SPLE gave a recovery which was over 80% for beta-endosulfan, endosulfan sulfate, p,p'-DDT and p,p'-DDE compared to PLE with off-line clean-up. The same trend was observed when applying the SPLE method to a certified reference soil sample (CRM 811-050) containing 13 OCPs, where the SPLE method gave 80-90% recovery vis-à-vis the PLE method with off-line clean-up. Feasibility of the SPLE method was further demonstrated by applying it to five real soil samples collected in Ethiopia.  相似文献

A simple and rapid microwave-assisted extraction (MAE) procedure was developed and optimized for the extraction of paclitaxel (Taxol) from the needles of yew trees Taxus baccata L. grown in Iranian habitats. The samples, immersed in a methanol-water mixture, were irradiated with microwaves in a closed-vessel system. The method was evaluated using a factorial design approach based on parameters such as extraction time, temperature, methanol concentration in water (v/v), and the ratio of grams of sample to 10 mL of solvent. Statistical treatment of the results revealed that the selected parameters were all significant except the extraction time. Optimum conditions would be 1.5 g samples in 10 mL solvent (90% methanol), an extraction temperature of 95 degrees C, and an extraction time of 7 min. The extracts has been analyzed by reverse-phase high-performance liquid chromatography with UV detection (LC/UV) at 227 nm for quantification. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used for confirmation. The main advantage of the proposed MAE method versus conventional solvent extraction (CSE) are the considerable reductions in time (7 min versus 16 h) and in solvent consumption (20 mL versus 150 mL). The MAE procedure yielded extracts that could be analyzed directly without any preliminary clean-up or solvent exchange steps. Both extraction methods show RSDs lower than 10% and lead to comparable recoveries of paclitaxel (87-92%).  相似文献

A procedure for the multiresidue determination of organochlorine pesticides and polychlorinated biphenyls in fish muscle samples has been developed. The method is based on the microwave-assisted extraction (MAE) of food samples from an acetonitrile-water (95 + 5, v/v) mixture followed by SPE cleanup of the extracts and analysis by GC with an electron capture detector. MAE operational parameters, such as the extraction solvent, temperature, and time, were optimized with respect to the extraction efficiency of the target compounds from food samples with 10-13% fat content. The chosen extraction technique allows reduction of the solvent consumption and extraction time when compared with methods already used. Acetonitrile is a good extraction solvent for low-fat matrixes (2-20% fat content), such as fish samples, because it does not significantly dissolve the highly polar proteins, salts, and sugars commonly found in food and gives high recoveries of a wide polarity range of analytes. For purification, SPE using LC-Florisil was shown to be sufficient for the removal of coextracted substances. Recoveries > 78% with RSD values < 15% were obtained for all compounds under the selected conditions. Method quantification limits were in the 5-10 microg/kg range. The method was applied to the analysis of samples of herring (Clupea harengus) purchased at the local fish market. The method is rapid and reliable for the determination of organochlorine analytes in fish muscle.  相似文献

Polychlorinated biphenyls (PCBs) in sludge are usually extracted by a technique such as Soxhlet with subsequent fractionation prior to long GC runs using GC-ECD or GC-HRMS. In this study, the extraction of selected chlorinated biphenyls (CBs) from a spiked sludge sample by three rapid techniques, i.e. ultrasonic (USE), pressurised-liquid (PLE), and microwave-assisted (MAE) extraction using a domestic microwave, was studied, with subsequent direct GC-ECD, GC-MS, or GC x GC-microECD analysis of the extracts. The main goal was to select an appropriate, and miniaturised, extraction method after only a brief optimisation and demonstrate the power of GC x GC analysis of dirty extracts. For PLE similar CB recoveries were found when extracting with either n-hexane or n-hexane/acetone (1/1). For USE and MAE, n-hexane/acetone (1/1) was the preferred extraction solvent. USE gave the best recoveries (80-95%; except 130% for CB 105). The only clean-up needed prior to GC-MS or GC x GC-gECD analysis was the removal of sulphur-containing compounds. GC-ECD was not suitable for these dirty extracts. The lowest LODs for the CBs (20 fg or 0.1 ng/g sludge) were found when combining USE and GC x GC-microECD, because of the powerful extraction, high separation power and excellent detectability provided by this technique.  相似文献

A new, single-step extraction and purification method based on matrix solid-phase dispersion (MSPD) was developed to determine 17 polycyclic aromatic hydrocarbons (PAHs) in sewage sludge samples. The MSPD method consists of sample homogenisation, exhaustive extraction and clean-up by a single process. The different operational parameters of the method, such as the type of dispersant, type and amount of additives, clean-up co-sorbent and extractive solvent were evaluated. Reversed-phase (C18) and polymeric (Oasis HLB and Oasis MAX) materials, as well as normal phase sorbents (Florisil, silica, neutral alumina) and an inert support (sand) were tested to assess the sorbents effect on the yield and selectivity of the MSPD process. Analysis of extracts was performed by high performance liquid chromatography (HPLC) coupled with fluorescence detection.Quantification limits obtained for all of these considered compounds (between 0.0001 and 0.005 μg g−1 dry mass) were well below of the limits recommended in the EU. The extraction yields for the different compounds obtained by MSPD ranged from 76.3% to 103.6%. On the other hand, the extraction efficiency of the optimised method is compared with that achieved by microwave-assisted extraction and the method was applied to the analysis of real sewage sludge samples. A certified reference material (sewage sludge (BCR 088)) and a reference material (sewage sludge (RTC-CNS312-04)) were used to validate the proposed method.  相似文献

建立了一种同时测定大米中有机氯、有机磷、氨基甲酸酯和拟除虫菊酯等4类农药残留量的分析方法。通过比较二氯甲烷、三氯甲烷、乙腈、乙酸乙酯和不同比例的己烷-丙酮混合溶剂等8种溶剂的提取效果,选择以二氯甲烷为提取溶剂;以Florisil固相萃取小柱净化,通过以不同比例的己烷-丙酮作洗脱溶剂,发现体积比为4∶1的己烷-丙酮的洗脱效果最佳,在选定的洗脱条件下,样品的净化效果良好;用气相色谱-质谱测定,以保留时间、选择离子及其相对丰度定性,以外标法定量。以低限加标样品的3倍信噪比确定方法的检出限(LODs),以两个添加水平测定样品的回收率和相对标准偏差(RSD)。该方法的检出限达到μg/kg水平;除敌敌畏、乐果、pp′-DDT等几种农药外,大多数农药的加标回收率在75%和120%之间,RSD均低于10.4%,r≥0.992。该方法简便、快速、灵敏,能够满足同时测定大米中多种类残留农药的要求,可以作为大米中农药多残留的例行分析和确证分析的方法。  相似文献

Three extraction methodologies (Soxhlet, ultrasonic and pressurized liquid extraction) and several clean-up procedures (Florisil, silica and alumina in cartridges or glass column format) were tested and compared to extract 16 US Environmental Protection Agency (EPA) polycyclic aromatic hydrocarbons (PAHs) from Pinus pinea L. needles. Quantification was done by gas chromatography with mass spectrometry, by internal standard method using five deuterated PAH surrogate standards. Among the several extraction and clean-up procedures tested, ultrasonic extraction followed by alumina cartridge clean-up was the preferred method, yielding recoveries between 72 and 100% and limits of detection between 0.22 and 0.71 ng/g dry weight. The performance of the method was tested to determine PAHs in naturally contaminated samples.  相似文献

A multi-residue method developed for the analysis of triazine herbicides, simazine, atrazine, propazine and prometryne, in sheep liver is presented. The method is based on microwave-assisted extraction (MAE) of sheep liver using methanol as extractant and analysis of extracts by high performance liquid chromatography (HPLC) and ultraviolet detection. MAE operational parameters, the solvent type and volume, extraction temperature and time, were optimized in detail with respect to extraction efficiency of the target compounds from sheep liver. The recoveries of the method at two different spiked levels were assessed by analyzing spiked liver samples and were found to be in the range from 90 to 102% with good precision (<11%).  相似文献

A microwave-assisted extraction (MAE) method has been developed and optimized for the extraction of six tricyclic antidepressants (TCAD; nordoxepin, nortriptyline, imipramine, amitriptyline, doxepin, dezypramine) from human serum. Optimal parameters of MAE (solvent and extraction temperature) for water solution of these drugs were defined. The microwave-assisted procedure developed was validated by extraction of serum samples at two concentration levels and then successfully applied to the analysis of reference material. Limit of quantification, precision and recovery were found for the studied compounds in the ranges 0.04-0.15 microg/mL, 1.57-4.34% (RSD) and 94-105%, respectively.  相似文献

This paper describes a new extraction method for the determination of aliphatic hydrocarbons (AHs) in soil and sediment samples, using continuous microwave-assisted extraction (MAE) combined with liquid-liquid extraction, for clean-up purposes. Analytical determinations were carried out by gas chromatography coupled with impact ionization mass spectrometry. The influence of the experimental conditions was tested using an agricultural soil spiked with standards (stored at 4 degrees C for 1 month) as reference soil. Maximum extraction efficiencies (80-90%) were achieved using 0.1-1.0g of sample, 60microl of water and 3ml of n-hexane (extractant) and 5min of extraction time; less than 70% of the most volatile hydrocarbons (C(9)-C(12)) were recovered since many evaporated during the drying step of the sample. MAE was compared with a conventional extraction method such as Soxhlet and a good agreement in the results was obtained (average recovery percentage value of 105% by comparing MAE against Soxhlet). Quality parameters such as linear range (0.5-800microg/g), limits of detection (LODs) (0.1-0.2microg/g) and precision (RSD, 4-6%) were determined using spiked soil samples. This method was successfully applied to the analysis of aliphatic hydrocarbons (C(9)-C(27) including pristane and phytane) in contaminated real samples.  相似文献

A selective pressurized liquid extraction (SPLE) method, followed by gas chromatography–mass spectrometry (GC–MS), for the simultaneous extraction and clean-up of estrone (E1), 17β-estradiol (E2), 17α-ethynylestradiol (EE2), estriol (E3) and bisphenol A (BPA) from soil samples is described. The on-line clean-up of soil by SPLE was achieved using different organic matter retainers, including silica, alumina and Florisil, the most effective being silica. Thus, different amounts of silica, in conjunction with different extraction solvents (acetone, ethyl acetate, isohexane and dichloromethane), either alone or in combination, were used to extract the target chemicals from spiked soil samples. It was shown that 3 g silica resulted in satisfactory rates of recovery of target compounds and acetone:dichloromethane (1:3, v/v) was efficient in extracting and eluting estrogenic compounds for SPLE. Variables affecting the SPLE efficiency, including temperature and pressure were studied; the optimum parameters were 60 °C and 1500 psi, respectively. The limits of detection (LODs) of the proposed method were 0.02–0.37 ng g−1 for the different estrogenic chemicals studied. The outputs using the proposed method were linear over the range from 0.1 to 120 ng g−1 for E1, E2, EE2, 0.2–120 ng g−1 for E3, and 0.5–120 ng g−1 for BPA. The optimized method was further verified by performing spiking experiments in natural soil matrices; good rates of recovery and reproducibility were achieved for all selected compounds and the method was successfully applied to soil samples from Northeast Scotland, for the determination of the target compounds.  相似文献

Beeswax is a complex mixture of lipophilic compounds and other components such as aliphatic alcohols and carotenoids. Then, extraction and clean-up for pesticide analysis in beeswax is a challenge. In this work, a multiresidue method for the analysis of dichlorvos (DCV), diazinon, malathion, methyl parathion and coumaphos (CMF) in beeswax was developed. The proposed approach is based on matrix solid-phase dispersion extraction. The adsorbent for sample clean-up was studied and a simplex-centroid cubic statistical design was applied to evaluate pure solvents and their binary and ternary mixtures to elute the analytes. Finally, Florisil and ethyl acetate were chosen as solid support and eluting solvent, respectively. After extraction, pesticides were separated and detected by gas chromatography/mass spectrometry. The method achieved acceptable recoveries (70–85%; except for DCV, 24–38%) with relative standard deviations below 5%. The repeatability of the method was lower than 8% and interday variability was below 12%. The limit of detection (LOD) for the analytes varies between 0.2 and 2.6 µg?kg?1 and limit of quantification from 0.93 to 8.8 µg?kg?1. LOD reached for CMF was below the maximum residue limit allowed by the legislation of the United States and Canada.  相似文献

A novel sample preparation method for auxin analysis in plant samples was developed by vacuum microwave-assisted extraction (VMAE) followed by molecularly imprinted clean-up procedure. The method was based on two steps. In the first one, conventional solvent extraction was replaced by VMAE for extraction of auxins from plant tissues. This step provided efficient extraction of 3-indole acetic acid (IAA) from plant with dramatically decreased extraction time, furthermore prevented auxins from degradation by creating a reduced oxygen environment under vacuum condition. In the second step, the raw extract of VMAE was further subjected to a clean-up procedure by magnetic molecularly imprinted polymer (MIP) beads. Owing to the high molecular recognition ability of the magnetic MIP beads for IAA and 3-indole-butyric acid (IBA), the two target auxins in plants can be selectively enriched and the interfering substance can be eliminated by dealing with a magnetic separation procedure. Both the VMAE and the molecularly imprinted clean-up conditions were investigated. The proposed sample preparation method was coupled with high-performance liquid chromatogram and fluorescence detection for determination of IAA and IBA in peas and rice. The detection limits obtained for IAA and IBA were 0.47 and 1.6 ng/mL and the relative standard deviation were 2.3% and 2.1%, respectively. The IAA contents in pea seeds, pea embryo, pea roots and rice seeds were determined. The recoveries were ranged from 70.0% to 85.6%. The proposed method was also applied to investigate the developmental profiles of IAA concentration in pea seeds and rice seeds during seed germination.  相似文献

Two methods for extracting organochlorine (OCs) and organophosphorus (OPs) pesticides from animal liver have been developed. The determination was carried out by gas chromatography with electron impact ionization tandem mass spectrometry (GC-(EI-)MS/MS) using a triple quadrupole (QqQ) analyzer. First, a liquid-solid extraction performed with a high-speed homogenizer (Polytron) using ethyl acetate as solvent, and a subsequent clean-up by gel permeation chromatography (GPC) was applied, determining 34 pesticides. Secondly, a matrix solid phase dispersion (MSPD) extraction with octadecylsilyl (C(18)) sorbent combined with a Florisil clean-up and ethyl acetate elution was performed, analyzing 25 compounds. These methodologies have been tested and compared in the sample pre-treatment due to the fatty nature of the matrix. The GPC method was finally selected and validated, yielding recoveries in the range 70-115%, with precision values expressed as relative standard deviation (RSD) lower or equal to 20%, at the spiking levels of 25 and 50 microg kg(-1), and limits of quantification (LOQs) lower than the maximum residue levels (MRLs) set by the European Union in animal products, except for isofenphos. Linearity was also studied ranging between 5 and 300 microg kg(-1) for most of pesticides. This method was applied to the analysis of real liver samples of chicken, pork and lamb.  相似文献

A new method for determining endocrine disrupter compounds (EDCs) in sewage sludge is described in this paper. EDCs studied were bisphenol A (BPA) and alkylphenols (APs). In order to obtain a fast and simple method, selective pressurised liquid extraction (SPLE) and focused ultrasound solid-liquid extraction (FUSLE) were tested. Best results for SPLE were obtained using Florisil as clean-up sorbent and dichloromethane as extraction solvent, while temperature was the only significant variable. Analyte extraction by SPLE was completed in only one extraction cycle of 1 min at 130 °C. FUSLE was carried out in one step of 20 s at 75% power (0.5 cycles) and with 8 mL of ethyl acetate. Although the optimised FUSLE process was faster, simpler and cheaper, SPLE provided higher recovery values (ranging from 81 to 105%) and therefore SPLE-based method was selected and validated. The SPLE and GC-MS method showed an LOD of 10.7 ng/g for BPA and LODs between 1.2 and 41.6 ng/g for APs. Relative standard deviation values lower than 6% were obtained for all analytes. As a result, an efficient, fast and simple method based on SPLE and GC-MS for the determination of BPA and APs in sewage sludge is proposed.  相似文献

An improved method was developed for the simultaneous determination of eight steroid and phenolic endocrine disrupting chemicals, such as oestrone (E1), 17β-oestradiol (E2), oestriol (E3), 17α-ethynylestradoil (EE2), 4-nonylphenol (4-NP), bisphenol A (BPA), 4-tert-octylphenol (4-t-OP) and 4-cumylphenol (4-CP), in biological samples. The optimal extraction and clean-up procedures were investigated using microwave-assisted extraction (MAE), automated gel permeation chromatography (GPC) and solid phase extraction (SPE). As a consequence, the most efficient extraction was achieved by using MAE with methanol as solvent at an extraction temperature of 110°C for 20?min. The clean-up of extracts was carried out by GPC on a Biobeads S-X3 column with cyclohexane/ethyl acetate (1:1, v/v) as mobile phase. Target compounds were eluted in the fraction from 7–14?min retention time. Moreover, the cleanest extracts were obtained by solid phase extraction with C-18 cartridges after the elution with 15?mL ethyl acetate. The final sample extracts were derivatised using N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) (1% trimethylchlorosilane, TMCS) as derivatisation reagent with pyridine as the solvent. Quantification was performed by gas chromatography-mass spectrometry (GC-MS) with electron ionisation (EI) and selected ion monitoring (SIM) mode. The method was validated by spiked samples which showed good recovery and reproducibility. The overall recoveries ranged between 55.1% and 100.6%, with relative standard deviations (RSD) of 2.3–12.7% for the entire procedure. Method detection limits (MDL) ranged from 0.3 to 0.7?ng?g?1 dry weight (dw). Performance of the method was demonstrated by its application on tissues from fish exposed to high concentration of EDCs in the laboratory. The developed method is a promising approach for the analysis of steroid and phenolic endocrine disrupting chemicals in various biological samples.  相似文献

A fast and simple method for the analysis of polybrominated diphenyl ethers (PBDEs) in fish samples was developed using a one-step extraction and clean-up by means of pressurized liquid extraction (PLE) combined with gas chromatography-ion trap tandem mass spectrometry (GC-ITMS-MS). The selective PLE method provided to obtain ready-to-analyse extracts without any additional clean-up step, using a sorbent as fat retainer inside the PLE cell. Several PLE operating conditions, such as solvent type, extraction temperature and time, number of cycles and type of fat retainer, were studied. Using Florisil as fat retainer, maximum recoveries of PBDEs (83-108%) with minimum presence of matrix-interfering compounds were obtained using a mixture of n-hexane:dichloromethane 90:10 (v/v) as solvent, an extraction temperature of 100 °C and a static extraction time of 5 min in combination with three static cycles. Quality parameters of the method were established using standards and fish samples. Limits of detection and quantification ranged from 10 to 34 pg g−1 wet weight and between 34 and 68 pg g−1 wet weight, respectively. In addition, good linearity (between 1 and 500 ng ml−1) and high precision (RSD % < 15%) were achieved. The method was validated using the standard reference material SRM-1945 (whale blubber) and was then applied to the analysis of PBDEs in fish samples.  相似文献
