The bioaffinity of receptor-ligand interactions is investigated by determining the binding constant (association constant or dissociation constant) of the resulting complex utilizing affinity capillary electrophoresis (ACE). The ACE binding assay was established with a potent immunosuppressant, deoxyspergualin (DSG), that binds specifically to Hsc70, a constitutive or cognate member of heat shock protein 70 (Hsp70) family. Quantitative determination of binding constants under different running buffer systems provide comparative results. The association constants for the interaction between Hsc70 protein and DSG were found to be 5.7·104 M−1 in a buffer with pH 6.95 and 6.3·104 M−1 in a buffer with pH 5.30 (or corresponding dissociation constants, 18 and 16 μM, respectively) based on Scatchard analyses. Binding of DSG to a synthetic peptide, SINPDEAVAYGAAV-QAAILSGDK, one of the DSG-binding fragments found from tryptic digest of Hsc70 protein, provides further detailed information for the understanding of Hsc70 binding domain. The applicability of using coated capillaries was also evaluated for probing Hsc70-DSG interaction.  相似文献

用量子化学方法研究了丙酸与多乙烯多胺缩合反应机理。采用AM1方法全优化计算了丙酸多胺盐及其质子化盐的几何构型、电子结构以及酸催化下的亲核加成和消除反应的机能曲线,求得该两步反应的活化能分别为6.258kJ/mol和206.15kJ/mol。消除反应是速度控制步骤。发现质子化后丙酸羰基碳原子上的净电荷增大,前线分子轨道能级差减小,前线分子轨道间相互作用增强,表明酸催化大大增强反应活性。  相似文献

The stabilities of 3,4-dihydroxybenzylamine (DHBA), dopamine, 3-methoxytyramine, normetanephrine and metanephrine standards under acid, base and enzymatic hydrolysis conditions were studied. Basic incubation media were not suitable for 3,4-dihydroxy compounds, but acid and enzymatic hydrolysis conditions were applicable to all the compounds. The results of acid and enzymatic hydrolysis were comparable and the enzymatic hydrolysis was applied to a urine matrix. A method including solid-phase extraction (SPE) with a copolymer sorbent was developed for purification of the urine samples. Due to poor recovery of DHBA, the most frequently used internal standard in catecholamine analysis, this compound was replaced with the 3-O-methoxy structure. The recoveries of the compounds in spiked urine samples in SPE were between 96.4 and 124.4%. The repeatability of the combination of enzymatic hydrolysis and SPE pretreatment was good for all the compounds, except for dopamine and 3-methoxytyramine due to some matrix compounds still interfering with the separation. The analyses were performed with capillary electrophoresis in an ammonium acetate buffer with UV detection. The validation data for the compounds including limit of detection, limit of quantification, linearity and repeatability of the method are presented.  相似文献

A UV spectrometric method was developed to determine the molar absorptivity (C) and formation constant (Kc) of the association complex of unsubstituted chalcone in cyclohexane, in the concentration range from 4.00·10−4 to 2.00·10−2 mol dm−3. The thermodynamic and spectroscopic magnitudes such as Kc and C contribute to the understanding of the physicochemical behavior of several ,β-unsaturated carbonylic compounds, of low solubility in water, as it is the case of numerous flavonoids of chemical and biological importance. The studied association complex, formed by two chalcone molecules, is characterized by the constants C (300.8 nm)=4.98·104 dm3 mol−1 cm−1 and Kc=5.58·103. The method proposed is convenient for the study of solute–solute molecular associations particularly those due to dipole–dipole interactions.  相似文献

The emissive properties of a bichromophor molecule (1) are reported. This contains an anilino group as an electron donor (D) and a 1-cyanonaphthalene group as an electron acceptor (A) interconnected by a saturated hydrocarbon bridge of limited flexibility, which holds D and A far apart in the electronic ground state. The emission spectrum of 1, both in solution and in the gas phase, indicates that quantitative formation of an intramolecular exciplex between D and A occurs. This exciplex formation was studied as a function of excitation energy in molecules of 1 isolated in a supersonic free jet. A barrier of 1700 ± 200 cm−1 was found between the Franck-Condon excited conformation and the conformation of the exciplex. Although this value is significantly higher than that reported earlier for exciplex formation between chromophores connected by a simple polymethylene chain (≈ 900 cm−1) it is much lower than the barrier predicted for folding the bridge in 1 sufficiently to bring D and A in close contact. A tentative explanation of this discrepancy is given.  相似文献

In this paper, a modified nearest-neighbor regression method (kNN) is proposed to model a process with incomplete information of the measurements. This technique is based on the variation of the coefficients used to weight the distances of the instances. The case study selected for testing this algorithm was the photocatalytic degradation of Reactive Red 184 (RR184), a dye belonging to the group of azo compounds, which is widely used in manufacturing paint paper, leather and fabrics. The process is conducted with TiO2 as catalyst (an inexpensive semiconductor material, completely inert chemically and biologically), in the presence of H2O2 (with the role of increasing the rate of photo-oxidation), at different pH values. The final concentration of RR184 is predicted accurately with the modified kNN regression method developed in this article. A comparison with other machine learning methods (sequential minimal optimization regression, decision table, reduced error pruning tree, M5 pruned model tree) proves the superiority and efficiency of the proposed algorithm, not only for its results, but for its simplicity and flexibility in manipulating incomplete experimental data.   相似文献

A rapid and sensitive spectrophotometrc method has been developed for the microdetermination of some phenothiazine derivatives as the pure substances and in different dosage forms. The method depends on the formation of stable donor-acceptor complexes between phenothiazines and chloranilic acid in an acetonitrile-2-propanol solvent mixture. The resulting intensely purple chloranilic acid radical anion possesses a characteristic absorption maximum at 515 nm. Beer's law is obeyed over the concentration ranges 1-6, 1-10 and 5-30 mug ml for prochlorperazine dimaleate, trifluoperazine dihydrochloride and thiethylperazine dihydrochloride, with apparent molar absorptivities of 7.76 x 10(4), 1.95 x 10(4) and 6.64 x 10(3) 1. mole(-1).cm(-1), respectively. Statistical comparison of the results with those of an official method shows excellent agreement and indicates no significant difference in precision.  相似文献

An automated column-switching system for determination of vanillylmandelic acid in urine is described. The liquid chromatographic system was composed of two separation columns with different selectivity properties, an octadecyl column coated with tributyl phosphate as stationary liquid phase and a silica-based anion exchanger. Urine samples were injected directly onto the first column, where vanillylmandelic acid was separated from the main part of the sample matrix. The internal standard isovanillylmandelic acid was co-eluting with vanillylmandelic acid, and a fraction of the eluate containing both substances was switched to the second column, where separation was performed. To assess peak purity, detection was performed with dual working electrodes in parallel mode. A relative standard deviation of 3.5% was obtained for determination of human urine samples containing 3 microM vanillylmandelic acid, and less than 0.1 microM could be detected.  相似文献

Summary 5-S-Cysteinyldopamine, a new metabolite of dopamine, was determined in urine by high-performance liquid chromatography with electrochemical detection. The catechol was detected in 14 of 21 melanoma patients and 7 of 21 normal subjects; the highest values were 657 μg/day for melanoma patients and 44 μg/day for normal subjects. These results suggest that the cysteine conjugate may arise from autoxidation of dopamine but tyrosinase may also participate in the oxidation.  相似文献

A reflection anisotropy microscope (RAM) creates the contrast in its images from a change in the polarization orientation of reflected light from a surface. This may stem from local variations of the reconstruction of a surface, being initiated by changes in an adsorption layer. The advantages and disadvantages of a recently improved RAM versus other optical imaging techniques are discussed. We demonstrate the unique features of RAM and present the first experimental findings of so called 2pi phase kinks in the globally coupled CO-oxidation on Pt(110).  相似文献

An analytical method based on microwave decomposition and flow injection analysis (FIA) coupled to hydride generation atomic absorption spectrometry (HGAAS) is described. This is used to differentiate arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA) from organoarsenic compounds usually present in seafood. Without microwave digestion, direct analysis of urine by HGAAS gives the total concentration of As(III), As(V), MMA and DMA because organoarsenic compounds such as arsenobetaine, usually found in most seafood, are not reducible upon treatment with borohydride and therefore cannot be determined by using the hydride generation technique. The microwave oven digestion procedure with potassium persulfate and sodium hydroxide as decomposition reagents completely decomposes all arsenicals to arsenate and this can be measured by HGASS. Microwave decomposition parameters were studied to achieve efficient decomposition and quantitative recovery of arsenobetaine spiked into urine samples. The method is applied to the determination of urinary arsenic and is useful for the assessment of occupational exposure to arsenic without intereference from excess organoarsenicals due to the consumption of seafood. Analysis of urine samples collected from an individual who ingested some seafood revealed that organoarsenicals were rapidly excreted in urine. After the ingestion of a 500-g crab, a 10-fold increase of total urinary arsenic was observed, due to the excretion of organoarsenicals. The maximum arsenic concentration was found in the urine samples collected approximately between 4 to 17 hr after eating seafood. However, the ingestion of organoarsenic-containing seafoods such as crab, shrimp and salmon showed no effect on the urinary excretion of inorganic arsenic, MMA and DMA.  相似文献

The average degree of polymerisation (DP) and distribution of oligosaccharides in partially acid hydrolysed mannuronans were quantitatively evaluated by 1H NMR, electrospray ionisation mass spectrometry (ESI-MS), micellar electrokinetic capillary chromatography with UV detection (MEKC-UV), and high-pressure anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD). Our investigation shows that 1H NMR, MEKC-UV and, in particular, HPAEC-PAD can be used as quantitative tools to aid the investigation of polysaccharide structure, function and synthesis. For the latter two techniques, especially, this represents a significant new development as it enables calculation of the quantity of individual oligomers of nominal DP by direct analysis of a defined oligomer mixture. Appropriate statistical averages of number and weight distributions were also calculated and found to fit very well to predicted Kuhn distributions that assume random depolymerisation.  相似文献

Capillary electrophoresis–electrospray tandem mass spectrometry (CE‐ESI/MS/MS) is a simple and highly sensitive method for quantifying seven urinary androgen glucuronides. The urine samples were diluted and filtered through a membrane filter, and the filtrate was injected into a CE‐MS/MS system without further sample preparation steps such as extraction and derivatization. The calibration ranges were 0.01–5 µg/mL for glucuronides of androsterone and 11β‐OHAn‐3G, and 5–500 ng/mL for glucuronides of 11‐ketoAn, DHEA, testosterone, epitestosterone and DHT. The linearity of the method was 0.992–0.998, and the limits‐of‐detection at a signal‐to‐noise ratio of 3 were 5–10 ng/mL. The coefficients of variation were in the range of 4.0–9.0% for intra‐day assay and 4.1–9.8% for inter‐day assay. The proposed method may be applicable to metabolic profiling in both quantitative and qualitative analysis. Copyright © 2008 John Wiley & Sons, Ltd.  相似文献

Uric acid (C5H4N4O3) is one of the final products of purine metabolism. Its concentration balance is maintained in the kidneys, but compromised kidney function can result in its crystallization either in the renal tract or in the interstitial fluid of joints. In physiological deposits, crystalline uric acid is most frequently found either in a protonated state (anhydrous or dihydrate phases) or as a deprotonated urate ion (sodium or ammonium salts). Often these precipitates are found in association with a number of mineral phases (e.g., calcium oxalates, calcium phosphates, and magnesium phosphates). Their frequent and common coexistence suggests that synergistic relationships between these crystalline phases may exist. A comprehensive list of different heterogeneous uric acid/uric acid and uric acid/mineral interfaces that are epitaxially matched was generated with the lattice-matching program EpiCalc. Two hundred twenty-five coincident epitaxial matches and four commensurate epitaxial matches were identified using this screening procedure.  相似文献
