Conclusions Two phospholipases A with molecular weights of 11,000±1000 and 18,000±1000, consisting of individual compounds according to rechromatography and disk electrophoresis, have been isolated by filtration through Sephadex G-75 gel and by ion-exchange chromatography on CM-cellulose from the venom of the Central Asian cobra.V. Conclusions 1. The amino-acid compositions of two neurotoxins from the venom of the Central Asian cobra have been established. The N-terminal amino-acid residue of toxin (I) is isoleucine and that of toxin (II) leucine. There is the same sequence from the C-end of both toxins — Cys-Asn-Asn-COOH.2. It has been shown that the selectivity of the direct hemolytic effect of the cytotoxin on different erythrocytes correlates with its influence on the ionic permeability of the membranes of these erythrocytes. It has been established on bilayer phospholipid membranes that the influence of the cytotoxin on their ionic permeability depends on the amount of cholesterol in them. It has been shown by the spin probe method that the action of the cytotoxin of the venom of the central asian cobra on artifical liposomes extends at least to the depth of localization of the iminoxyl head of stearic acid spin-labeled at C-16. The interaction of cytotoxins Vc1, Vc5, and Vc6 of the venom of the Central Asian cobra with liposomes having a negative surface charge prepared from a mixture of phosphatidylcholine and palmitic acid (1:1, molar) has been investigated with the aid of a pyrene fluorescent probe. It has been shown that on interacting with liposomes a cytotoxin increases the microviscosity of the hydrophobic region of membranes. This effect depends on the phase state of the lipids. Observations on the kinetics of the transfer of energy between pyrene probes and diphenylhexatriene have shown that on the addition of a cytotoxin to samples of liposomes with a negative surface charge an aggregation of the liposomes takes place without a disturbance of their integrity.Institute of Bioorganic Chemistry, Academy of Sciences of the Uzbek SSR, Tashkent.
