Asymmetric radical additions of trialkylboranes to 2H-azirine-3-carboxylates

Asymmetric additions of alkyl radicals, generated from R3B, to chiral 2H-azirine-3-carboxylates offer a new entry to enantio-enriched aziridines, and proceed with high diastereoselectivity when using 8-phenylmenthol as chiral auxiliary.     

New facilities of Biginelli reaction. Synthesis of methyl 6-aryl-5-benzoyl-4-methoxy-2-oxohexahydropyrimidine-4-carboxylates

Three-component reaction of methyl benzoylpyruvate, aromatic aldehyde, and urea in methanol in the presence of sodium bisulfate stops at the stage of the formation of nondehydrated product with a methylated tertiary hydroxy group and results in methyl 6-aryl-5-benzoyl-4-methoxy-2-oxohexahydropyrimidine-4-carboxylates.     

Synthesis of alkyl 4-aryl-2-methyl-5-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylates

Reactions of alkyl acetoacetates, aromatic aldehydes, ammonium acetate, and 1,3-cyclohexanedione afford alkyl 4-aryl-2-methyl-5-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylates. The structure of compounds obtained was established with the help of IR, ¹H NMR, and mass spectra and by X-ray diffraction analysis.     

Synthesis of methyl 2-aryl-5-chlorosulfonyl-1,3-oxazole-4-carboxylates and their reactions with amines and amidines

Previously unknown methyl 2-aryl-5-chlorosulfonyl-1,3-oxazole-4-carboxylates have been synthesized. Their reactions with amines and amidines have yielded the corresponding sulfonamides and 6H,7H-[1,3]oxazolo-[5,4-d]pyrimidin-7-ones.     

Synthesis of cucurbitine derivatives: facile straightforward approach to methyl 3-amino-4-aryl-1-methylpyrrolydine-3-carboxylates

A general three- or four-step synthesis of cis- and trans-substituted cucurbitine (3-aminopyrrolidine-3-carboxylic acid) derivatives from methyl 2-nitroacetate is reported. The first step utilizes a Knoevenagel condensation with five different aromatic imines or their corresponding aldehydes to form (Z/E)-mixtures of α-nitro acrylates. The second step gives rise to the pyrrolidine-core structures of the title compounds by a 1,3-dipolar cycloaddition reaction using an azomethine ylide. The last step consists of reduction of the nitro group to yield both diastereoisomers of the corresponding 4-aryl cucurbitine methyl esters.     

Reactions of methyl 3-hydroxythiophene-2-carboxylate. Part 4. Synthesis of methyl 5-azolyl-3-hydroxythiophene-2-carboxylates

The indirect introduction of an azolyl group in position 5 of compound 1 by an easy two-step procedure taking place at room temperature is described. A similar procedure yields the 4-chloro derivatives of these 5-azolyl compounds. The same method is applied for the introduction of a 5-azido group and from the 5-azido compounds, 5-v-triazolyl derivatives are obtained by a known method.     

Thermal rearrangement of 2-amino-5-aryl-4,5-dihydro-3-cyanofurans

Synthesis and Structure of Ethyl 3-acetamido-4-aryl-6-methyl-2-oxo-3,4-dihydropyran-5-carboxylates

Two ethyl 3-acetamido-4-aryl-6-methyl-2-oxo-3,4-dihydropyran-5-carboxylates(3) were synthesized with high diastereoselectivities and characterized by IR,NMR and MS.The configuration of 3a was confirmed by single-crystal X-ray diffraction,ethyl 3-acetamido-4-(4-nitrophenyl)-6-methyl-2-oxo-3,4-dihydropyran-5-carboxylate(3a),C17H18N2O7.The crystal is of orthorhombic,space group Pbca with a = 11.7781(16),b = 9.5979(10),c = 32.115(3),V = 3630.4(7)3,Z = 8,Mr = 362.33,Dc = 1.326 Mg/m3,λ = 0.71073,μ(MoKα) = 0.104 mm-1,F(000) = 1520,the final R = 0.0646 and wR = 0.1464 for 1788 observed reflections with I > 2σ(I).The N-H and oxygen atom are involved in intermolecular hydrogen bonds which link the molecules into a one-dimensional chain and stabilize the structure.     

5-Amino-4,5-dihydroisoxazoles. Part I. 5-Amino-3-aryl-4-methylene-4,5-dihydroisoxazoles and 4-aminomethyl-3-arylisoxazoles from 5-amino-4-aminomethyl-3-aryl-4,5-dihydroisoxazoles

5-Amino-4-aminomethyl-3-aryl-4,5-dihydroisoxazoles 2 were obtained by cycloaddition of nitrile oxides to 1,3-diaminopropenes 1. On reaction with methyl iodide the corresponding 4-(quaternary)-ammoniomethyl iodides 3 were formed. These compounds, on reaction with bases, afforded 5-amino-3-aryl-4-methylene-4,5-dihydroisoxazoles 4. The acid-catalyzed deamination of compounds 2 afforded 4-aminomethyl-3-arylisoxa-zoles 5 and 3-arylisoxazoles as retro-Mannich products. The deamination of 2 to yield 5 was also obtained by base catalysis.

Unusual photochemical C-N bond cleavage in the novel methyl 2-chloro-3-methyl-2H-azirine-2-carboxylate

The structure, preferred conformers, vibrational spectrum, and photochemical behavior of the novel azirine, methyl 2-chloro-3-methyl-2H-azirine-2-carboxylate (MCMAC) were investigated in low-temperature matrixes and in the neat solid amorphous state by infrared spectroscopy and quantum-chemical calculations. Two conformers of the compound were observed in argon, krypton, and xenon matrixes, in agreement with the DFT(B3LYP)/6-311++G(d,p) and MP2/6-311++G(d,p) theoretical calculations. Both conformers were found to exhibit the carboxylic ester group in the cis conformation, differing in the arrangement defined by the O=C-C-Cl dihedral angle (cis and trans, for Ct and Cc forms, respectively). The Ct conformer was found to be the most stable conformer in the gaseous phase as well as in both argon and krypton matrixes, whereas the more polar Cc conformer became the most stable form in the xenon matrix and in the neat solid amorphous phase. In situ broadband UV (lambda > 235 nm) excitation of matrix-isolated MCMAC led to azirine ring C-C and C-N bond cleavages, the latter process corresponding to the most efficient reaction channel. The photochemical cleavage of the C-N bond had never been previously observed in the case of aliphatic 2H-azirines. Two electron withdrawing substituents (methoxycarbonyl group and chlorine atom) are connected to the azirine ring in the novel MCMAC azirine. The simultaneous presence of these two groups accelerates intersystem crossing toward the triplet state where cleavage of the C-N bond takes place. The primary photoproducts resulting from the C-N and C-C ring-opening reactions were also found to undergo further photochemical decarbonylation or decarboxylation reactions.     

Lipid-lowering effects of ethyl 2-phenacyl-3-aryl-1H-pyrrole- 4-carboxylates in rodents

A series of substituted 2-phenacyl-3-phenyl-1H-pyrrole-4-carboxylates were prepared from substituted acetophenones in 6 steps. The final condensations between a chloroenal and an aminoketone were carried out under neutral conditions in parallel to yield the series listed below. Selected pyrrole derivatives proved to be potent hypolipidemic agents lowering serum triglyceride concentrations in CF-1 male mice after 14 days of I.P. administration. One agent orally lowered serum cholesterol in Sprague-Dawley male rats at 2mg/kg/day after 14 days. The agents demonstrated a lowering of mouse serum LDL- cholesterol levels and selected compounds showed an elevation of serum HDL-cholesterol levels. The cholesterol concentrations in the liver were raised while the cholesterol and triglyceride contents of the aorta were significantly lowered by the selected trisubstituted pyrrole.     

A regioselective route to 3-alkyl-1-aryl-1H-pyrazole-5-carboxylates: Synthetic studies and structural assignments

Ethyl 2,4-dioxooctanoate ( 1 ) was selectively protected as the 2-(methoxyimino) derivative 2 . When 2 was reacted with phenylhydrazine hydrochloride, ethyl 3-butyl-1-phenyl-1H-pyrazole-5-carboxylate ( 4 ) was favored over the corresponding 5-butyl-1-phenyl-1H-pyrazole-3-carboxylate product 5 by a ratio of at least 6:1, a complete reversal of the regioselectivity observed for 1 . The structures of 4 and 5 were assigned definitively by NOE difference experiments. Regiochemical and configurational assignments of the mono- and bis(methoxyimino) derivatives of 1 were also achieved by ID and 2D ¹H and ¹³C nmr methods.     

Thermal isomerization of the isoxazole ring and rearrangement processes in the mass spectra of 3-aryl-5-methylisoxazole-4-carboxylic acids

The mass spectra of 15 compounds of 3-aryl-5-methylisoxazole-4-carboxylic acid and their derivatives were investigated. Thermal isomerization of the isoxazole ring was observed for several derivatives of this series. The formation of rearranged pseudomolecular ions of the arylamines during the dissociative ionization of 3-aryl-5-methylisoxazole-4-carboxylic acids was examined on the basis of a study of the mass spectra of labeled compounds. It is assumed that there is a high probability of concerted elimination of several groups in the first step of the disintegration of the molecular ion.     

Three-component synthesis of methyl 6-alkyl-3-cyano-2-halopyridine-4-carboxylates

A procedure has been developed for the synthesis of methyl 6-alkyl-3-cyano-2-halopyridine-4-carboxylate by three-component reaction of 4-oxoalkane-1,1,2,2-tetracarbonitriles with hydrogen halides in methanol.
