The feasibility of a technique to confirm the presence of residues from seven beta-lactam antibiotics in bovine milk has been demonstrated. The technique makes use of electrospray ionization and tandem ion trap mass spectrometry. Residues are first extracted from milk by reversed-phase solid phase extraction. Target analytes are separated by on-line reversed-phase liquid chromatography and ionized in the electrospray interface. The product ion mass spectra are acquired following collision-induced dissociation of protonated molecules. Confirmation is based on comparison of full scan spectra between unknowns and bona fide standards. The feasibility of this technique has been demonstrated for the six beta-lactams currently approved for use in lactating dairy cattle (penicillin G, ampicillin, amoxicillin, cloxacillin, cephapirin and ceftiofur) and a drug not approved for animal use, cefazolin. The technique has been applied to control milk fortified at 5 ng/mL of penicillin G and 10 ng/mL of the other six drugs.  相似文献

This method was developed and validated to detect and quantitate tilmicosin residues in cow milk over a range of 0.010-10 microg/mL, and in sheep milk over a range of 0.025-0.5 microg/mL. The procedure involves extracting the milk sample with acetonitrile and using a C18 cartridge to perform a solid-phase extraction cleanup of the extract. A reversed-phase gradient liquid chromatography method with detection at 280 nm is used to separate the tilmicosin from matrix components in a 30 min run time. The limit of quantitation of the method is 0.010 microg/mL for cow milk, and 0.025 microg/mL for sheep milk. Average percentage recoveries for milk samples ranged from 82 to 94%. Percentage relative standard deviation values ranged from 3.1 to 17.2%.  相似文献

A simple and rapid high-performance liquid chromatographic (HPLC) method with ultraviolet detection has been developed and validated for the simultaneous determination of rifampicin and sulbactam in mouse plasma. Plasma samples were deproteinized with acetonitrile and separated by HPLC on a RP-18 (125 x 4 mm, 5 microm) column and gradient elution with potassium dihydrogen phosphate solution (pH 4.5; 50 mm) and acetonitrile at a flow-rate of 1.0 mL/min. Rifampicin and sulbactam were monitored at 230 nm and confirmed by means of their UV spectra using a diode-array detector. The method was linear at plasma levels from 1 to 100 microg/mL for rifampicin and from 5 to 200 microg/mL for sulbactam. The limits of quantification were 0.6 microg/mL for rifampicin and 4.2 microg/mL for sulbactam. The intra- and inter-day precisions of the method (RSD) were lower than 5% for both compounds. Average recoveries of rifampicin and sulbactam from mice plasma were 98.2 and 89.3%, respectively. The developed method was successfully applied to the determination of the pharmacokinetic profile of both compounds in mice.  相似文献

Two recently developed surface plasmon resonance biosensor assays for detection of beta-lactams in milk were used to screen raw producer milk samples. Both assays use a beta-lactam receptor protein with carboxypeptidase activity for detection. The results of the biosensor assays were compared with those of various commercial screening tests, i.e., the Delvotest SP, Penzym S, Beta-STAR, SNAP, and Parallux. The results of the 2 biosensor assays showed good agreement with those of the other screening tests. Of 195 analyzed milk samples, the results of only 5 samples differed between the assays. Additionally, 30 milk samples with both negative and positive results in the screening assays were analyzed by liquid chromatography for identification and quantification of any beta-lactam residues. All screening tests showed 0% false-negative results with 15 incurred samples containing between 4.0 and 268 microg/kg penicillin G. The biosensor assays showed 27% positive results (false violatives) with 15 producer milk samples containing penicillin G concentrations between 0 and 3.6 microg/kg, i.e., below maximum residue limit. This figure varied between 27 and 53% for the other screening tests.  相似文献

A simplified method for routine monitoring of 7 residual sulfonamides (SAs) (sulfadiazine (SDZ), sulfamerazine (SMR), sulfadimidine (SDD), sulfamonomethoxine (SMM), sulfamethoxazole (SMX), sulfadimethoxine (SDM), and sulfaquinoxaline (SQ)) in milk using high-performance liquid chromatography (HPLC) with a photodiode array detector is described. The spiked and blank samples were cleaned up by using an Ultrafree-MC/PL centrifugal ultrafiltration unit. For determination/identification, a Mightysil RP-4 GP column and a mobile phase of 25% (v/v) ethanol in water with a photodiode array detector were used. Average recoveries from milk samples spiked with 0.05, 0.1, 0.2, and 0.5 microg mL(-1) of each drug were >82%. The inter- and intra-assay variabilities were 2.0-3.1%. The practical detection limits for 7 SAs were 0.005-0.02 microg mL(-1). The total time and amount of solvent required for the analysis of one sample were <40 min and <6 mL of ethanol, respectively. No toxic solvents were used.  相似文献

Beta-lactam antibiotics (penicillins, cephalosporins) are still the most commonly used antibiotics for dairy cows in Germany. In routine milk testing, according to the German milk quality regulation, a positive result obtained for bulk tank milk by microbiological inhibitor tests needs no further confirmation, but results in reduced milk payment of 0.05 euros kg(-1) for one month. In some cases, however, further identification of the causative agent can be of interest, either if antimicrobial drugs have not knowingly been used recently, or if improper use of such drugs is denied. As a service for milk producers, our laboratory offers further analyses of violative milk samples, aiming at the identification and quantification of the inhibitor(s). In this program, a panel of microbiological inhibitor tests, receptor tests, and enzyme immunoassays (EIA) is used in a step-by-step analysis, which primarily focusses on beta-lactams, but also includes other compounds such as sulfonamides or tetracyclines, respectively. Here we report results for violative milk samples (n=63) analysed between 2003 and 2005. In most cases (95%), beta-lactam antibiotics could be identified, although not always at levels exceeding the respective MRL values. Penicillin G (mostly together with benzylpenicilloyl metabolites) could be identified in 74.6% of all samples. Other compounds identified were, in decreasing order, ceftiofur (11%), ampicillin/amoxicillin (6.3%), isoxazolyl penicillins (3.2%), and sulfonamides (1.6%). The results indicate that penicillin G is still the predominant antibiotic responsible for violative bulk tank milk samples as detected during regulatory control.  相似文献

A method for the simultaneous determination of eight beta-lactam antibiotics (nafcillin, cloxacillin, oxacillin, dicloxacillin, ampicillin, amoxicillin, and penicillin G) in fortified milk samples of different origins has been proposed by using CZE with diode-array detection (CZE-DAD). Optimum separation was obtained on a 64.5 cm x 75 microm bubble cell capillary using 175 mM Tris buffer with 20% ethanol at pH 8.0. Methylparaben has been used as an internal standard (IS). Taking into account the lack of sensitivity of the UV-Vis detection, a solvent extraction/SPE method was applied for off-line preconcentration and sample cleanup, and also an on-line preconcentration methodology, such as large-volume sample stacking (LVSS) with polarity switching, was developed, providing LODs ranging from 2 to 10 microg/L. The method permits the quantification of these residues below the levels established in milk by the EU Regulation. Satisfactory recoveries ranging from 86 to 93% were also obtained in milk samples of different origins.  相似文献

A fast, simple and very selective liquid chromatography-mass spectrometry (LC-MS) method for the detection of isopropylthioxanthone (ITX) in dairy products has been developed and validated. After addition of an ITX-d(3) as internal standard and a simple extraction from the sample with acetonitrile, the extract was centrifuged and directly injected into the LC-MS system. Chromatographic separation was achieved by means of a Gemini C18 column (100 mm x 2.0 mm i.d. 5 microm) using a gradient of aqueous 20 mM ammonium formiate at pH 4.5 and methanol as the mobile phase, at a flow rate of 0.25 mL min(-1). The method was validated according to the guidelines laid down by the Commission Decision 2002/657/EC using the parent ion [M+H](+) (m/z 255) as quantification ion, and the fragment ion (m/z 213) obtained by in-source collision-induced dissociation (IS-CID) as confirmation ion. Absolute and relative recoveries rates were verified at 5, 10, 15 microg kg(-1) in yoghurt samples and at 5 microg kg(-1) in milk and pudding: mean absolute recoveries were 77% in yoghurt, 50% in pudding and 67% in milk; relative recoveries (after internal standard correction) were always >97% in each matrix. The detection limit (CCalpha) and the detection capability (CCbeta) of method were 6.2 and 7.2 microg kg(-1), respectively.  相似文献

A multiresidue analytical method based on matrix solid-phase dispersion was developed to analyze liquid milk for 22 organochlorine pesticides (OCPs) and 6 polychlorinated biphenyls (PCBs). Initial extraction is performed by loading 3 mL milk onto a 2.0 g octadecyl (C18)-bonded silica cartridge with n-hexane as the eluant. Neutral alumina column chromatography with sodium sulfate as the drying agent is used for further cleanup. The eluate is concentrated to 0.5 mL, and target analytes are determined by capillary gas chromatography with electron-capture detection. The optimized method was validated by determining accuracy (recovery percentages), precision (repeatability and reproducibility), and sensitivity (detection and quantitation limits) from analyses of milk samples fortified at 10 and 1 microg/L levels. Average recoveries were between 74 and 106% for all residues except beta-HCH, beta-endosulfan, and endosulfan sulfate. Both repeatability and reproducibility relative standard deviation values were < 22% for all residues. Detection limits ranged from 0.02 to 0.12 microg/L and quantitation limits were between 0.02 and 0.62 microg/L. The proposed analytical method may be used as a fast and simple procedure in routine determinations of OCPs and PCBs in milk.  相似文献

A high-performance liquid chromatographic method was developed for the determination of five penicillins: penicillin G (PENG), penicillin V (PENV), oxacillin (OX), cloxacillin (CLO), and dicloxacillin (DICLO), in bovine muscle. Samples were macerated with a mixture of H(2)O/CH(3)CN (1:1) and purified using RP-8 Adsorbex SPE cartridges after centrifugation, with mean recovery from spiked samples higher than 89%. The separation of the examined penicillins was achieved on an analytical column, an Inertsil C8 5 microm, 250x4 mm(2), at ambient temperature. The mobile phase consisted of 0.1% TFA/ACN 50:50 v/v delivered isocratically at a flow rate of 1.1 mL/min. Analytes were monitored at 240 nm. The procedure was validated according to the European Union Decision 2002/657/EC by means of selectivity, stability, decision limit, detection capability, accuracy, and precision. Method's LOQ values achieved were 54 microg/kg for PENG and DICLO, 46 microg/kg for PENV, 16 microg/kg for OX, and 43 microg/kg for DICLO. The detection capabilities (CC(beta)) were 73.6 microg/kg for PENG, 29.1 microg/kg for PENV, 350.6 microg/kg for OX, 379.9 microg/kg for CLO, and 355.8 microg/kg for DICLO. The method was applied to various samples from the local market. Two penicillins were identified by photodiode array (PDA) detection and quantified.  相似文献

A high-performance liquid chromatographic method was optimized and validated for the determination of atenolol and chlorthalidone (CT) in human breast milk. The milk samples were extracted and purified using ACN and phosphoric acid for precipitation of proteins followed by removal of ACN and milk fats by extraction with methylene chloride. The samples were applied, after an extraction procedure, to a cyanide column using a mobile phase consisting of ACN/water (35:65 v/v) and buffered at pH 4.0 with flow rate of 1.0 mL/min. Quantitation was achieved with UV detection at 225 nm using guaifenesin as the internal standard. The effectiveness of protein precipitation and clean up procedure were investigated. The method was validated over the range of 0.3-20 microg/mL for atenolol and 0.25-5 microg/mL for CT.  相似文献

High-performance liquid chromatography with diode-array detection (HPLC-DAD) was optimised and validated for the determination of tetracyclines in bovine milk and tissues. Milk and tissue samples were extracted and purified using a solid-phase extraction HLB Oasis cartridge and analysed using HPLC-DAD set at 365 nm. The analyses were carried out using the mobile phase of 0.01 M oxalic acid-acetonitrile-methanol (60:25:15, v/v/v) on a C8 column (250 x 4.6 mm I.D., 5 microm). Recoveries of tetracyclines from spiked samples at the three concentrations (0.5, 1 and 1.5) of the maximum residues limits (corresponding to 100 microg/kg for milk and the muscle) were higher than 81.1% in milk and 83.2% in muscle. The method was successfully validated for bovine milk and muscle in compliance with requirements set by draft SANCO/ 1805/ 2000 European Decision. The decision limit (CCalpha) was in the range 113.2-127.2 microg/kg and 107.7-129.9 micro/kg for all compounds in milk and muscle, respectively. The detection capability (CCbeta) was in the range 117.2-131.3 microg/kg and 114.9-133.1 microg/kg for all compounds in milk and muscle, respectively.  相似文献

Fifty-four milk samples from commercial sources that tested positive for beta-lactam antibiotics were analyzed by a multiresidue liquid chromatographic (LC) procedure based on LC fractionation. Penicillin G and cephapirin were the beta-lactam antibiotics found most frequently. Some samples did not contain detectable beta-lactam antibiotics. In a few, the presence of a beta-lactam antibiotic was suspected because certain LC fractions tested positive for antimicrobial activity, which was no longer present in a replicate treated with beta-lactamase. However, the unknowns could not be identified by LC analysis.  相似文献

A liquid chromatographic (LC) method with ultraviolet-visible photodiode array (UV-VIS PDA) detection was developed to measure penicillin G in bovine milk. A liquid chromatographic-mass spectrometric (LC-MS) procedure was divised to confirm the LC method. The method involved diluting milk with a drug-releasing solvent consisting of acetonitrile-methanol-water and ultrafiltration through a 10,000 dalton cutoff filter. Penicillin G was separated from other components in the ultrafiltrate by ion-paired LC using a reversed-phase microbore column eluted with a 25% acetonitrile solution. The LC method was confirmed by thermospray LC-MS. The detection limit for penicillin G determination in milk was estimated to be 10 ppb for LC with UV-VIS PDA and 100 ppb for LC-MS.  相似文献

The Charm Safe-Level beta-Lactam Test was evaluated by a U.S. Food and Drug Administration (FDA) test protocol administered by the AOAC-Research Institute. The sensitivity and selectivity of the test were evaluated with >800 negative raw commingled and drug-fortified milk samples by the manufacturer and an independent laboratory. Probit analysis by the independent laboratory determined the following 90% positive levels with 95% confidence: amoxicillin, 5.6 ppb; ampicillin, 8.5 ppb; cephapirin, 13.7 ppb; ceftiofur, 46.2 ppb; and penicillin G, 3.6 ppb. These values were within a range of +/- 20% of the manufacturer's data. Selection of negative samples met confidence specifications. Ruggedness parameters were studied and defined, and the stability of frozen milk was verified. There were no interferences from somatic cells (1,000,000 somatic cell count/mL) or bacteria (300,000 colony-forming units/mL), or from 27 other non-beta-lactam animal drugs. Test performance with raw milk samples containing incurred penicillin, ampicillin, and amoxicillin was consistent with the dose responses determined with fortified milk samples. Incurred cephalosporin in raw milk samples was detected at lower levels than was cephalosporin in fortified milk samples, presumably because of the presence of metabolite, as verified by other test methods. Quality control data support consistency in manufacture between batches and the stability of refrigerated test reagents for up to 1 year. Successful fulfillment of these criteria led to FDA certification of the test when used with a reader in U.S. milk testing programs.  相似文献

建立了同时检测全血中青霉素G及其2种主要代谢产物青霉噻唑酸和脱羧青霉噻唑酸的超高效液相色谱-电喷雾串联质谱分析方法。血样经简单的蛋白沉淀提取后,目标化合物经BEH C18色谱柱(50 mm×2.1 mm, 1.7 μm),以含0.1%甲酸溶液-乙腈为流动相,梯度洗脱分离后,选用正离子电喷雾多反应监测(MRM)模式检测。方法检出限(信噪比(S/N)为3计)为0.1~2.0 ng/mL,定量限(S/N=10)为0.5~5.0 ng/mL。各被测物的线性相关系数均大于0.9974,准确度为92.3%~105.5%,日内精密度小于10%。考察了不同储存温度(18、4、-18、-80 ℃)下全血中青霉素G及其代谢物的稳定性,结果表明随着储存温度升高和时间的延长,青霉素G的质量浓度下降明显。应用建立的方法检测了给予20 mg/kg青霉素G后的大鼠血液样本,血液中青霉素G原形药物在给药后0.5 h即消除完全(低于检出限),而其代谢物的体内消除时限可延长至36 h。所建方法对司法鉴定的适用性得到了进一步扩大,同时对食品残留检测也具有一定的参考价值。  相似文献

A fast screening method is presented for detecting isopropylthioxanthone (ITX) contamination in milk. The method is based on direct high-performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS) analysis of milk samples. Sample preparation is limited to the addition of a deuterated ITX solution in acetonitrile that serves both as internal standard and to precipitate proteins. The method is highly accurate and sensitive. Isomeric specific analyses of 2-ITX and 4-ITX are possible at 6 microg/L levels with about 5% precision and accuracy. This approach has been used to check contamination in samples like milk, soy milk, baby milk, in their packaging material. Out of 37 milk samples analyzed, 16 were positive with concentrations ranging from 173-439 microg/L for 2-ITX and from <6 (lower than limit of quantification) to 25 microg/L for 4-ITX.  相似文献

The first analytical method is developed and validated for the simultaneous determination of imipenem and sulbactam in mouse plasma. Sample treatment is based on plasma stabilization with 4-(2-hydroxyethyl)piperazine-ethanesulfonic acid (HEPES) (0.5 mol/L; pH 7.0)-water-ethylene glycol (2:1:1, v/v/v), precipitation of plasma proteins with acetonitrile, centrifugation, evaporation, and reconstitution with borate buffer. Analytical determination is carried out by high-performance liquid chromatography with diode-array detection. Chromatographic separation is achieved within 11 min on a C(18) column by gradient elution with borate buffer (0.1 mol/L, pH 7.2) and methanol. Imipenem and sulbactam are monitored at 295 and 230 nm, respectively. The overall interday accuracy is in the range of 95% to 100% and from 98% and 101% for imipenem and sulbactam, respectively. Interday precision is below 8% and 4% for imipenem and sulbactam, respectively. Limits of quantitation of imipenem and sulbactam are 0.05 and 1.0 microg/mL, respectively. The mean extraction recoveries are 94.5% and 94.2% for imipenem and sulbactam, respectively. The described method allows an accurate, simple, and rapid identification and quantitation of imipenem and sulbactam in mouse plasma. This method is applied to the analysis of imipenem and sulbactam in mouse plasma after drug administration.  相似文献

Penicillin-resistant bacteria can often be treated through the co-administration of an antibiotic and a beta-lactamase inhibitor. Current inhibitors target only class A beta-lactamases. We report two new series of C3-modified penicillin sulfones, having either a simple methylene group (i.e., a homologue) or exocyclic unsaturation between the thiazolidine ring and the C3 carboxylate. The homologue has 10-fold better activity against a class C beta-lactamase than does sulbactam itself. By contrast, the exocyclic C3 unsaturated compounds are less active.  相似文献
