A new automated spectrophotometric method for the determination of total sulfite in white and red wines is reported. The assay is based on the reaction of o-phthalaldehyde (OPA) and ammonium chloride with the analyte in basic medium under SI conditions. Upon on-line alkalization with NaOH, a blue product is formed having an absorption maximum at 630 nm. The parameters affecting the reaction - temperature, pH, ionic strength, amount concentration and volume of OPA, amount concentration of ammonium chloride, flow rate and reaction coil length - and the gas-diffusion process - sample and HCl volumes, length of mixing coil, donor flow rate - were studied. The proposed method was validated in terms of linearity (1-40 mg L−1, r = 0.9997), limit of detection (cL = 0.3 mg L−1) and quantitation (cQ = 1.0 mg L−1), precision (sr = 2.2% at 20 mg L−1 sulfite, n = 12) and selectivity. The applicability of the analytical procedure was evaluated by analyzing white and red wine samples, while the accuracy as expressed by recovery experiments ranged between 96% and 106%.  相似文献

Three fluorescent probes were constructed by incorporating an α,β-unsaturated ketone to a coumarin fluorophore. The selective addition of sulfite to the alkene of TSP assisted by cetyltrimethyl ammonium bromide (CTAB) micelle can be visualized by dramatic color and ratiometric fluorescence changes. In CTAB–PBS system, the fluorescence intensity ratio at 465 nm and 592 nm (I465/I592) and the absorbance ratio at 390 nm and 470 nm (A390/A470) were linearly proportional to sulfite concentration in the range of 0.5–150 μM, and the detection limit was 0.2 μM. Good selectivity and competition of TSP1 towards sulfite over several anions and biological thiols were acquired. Probe TSP1 was used to detect sulfite in three realistic samples (mineral water, sugar and white wine) with good recovery.  相似文献

A novel spectrofluorimetric method for the determination of sulfite using rhodamine B hydrazide as fluorogenic reagent in the presence of polyoxyethylene sorbitan monooleate (Tween 80) surfactant micelles is described. The method is based on the mixture of sulfite with rhodamine B hydrazide, a colorless, non-fluorescent substance in Tween 80 surfactant micelles which gives rhodamine B-like fluorescence emission. The fluorescence intensity increase is linearly related to the concentration of sulfite in the range 5-800 ng ml−1 with a detection limit of 1.4 ng ml−1 (3σ). The optimal conditions for the detection of sulfite are evaluated and the possible detection mechanism is also discussed. The method has been successfully applied to the determination of total sulfite in wines and compares well with the standard iodimetric method.  相似文献

A new capillary electrophoretic (CE) method was developed for the rapid and simple determination of glutaraldehyde in photographic developing solutions. The proposed system is based on the selective pre-capillary derivatization of glutaraldehyde with sulfite and CE determination of anionic glutaraldehyde-bisulfite adduct formed. Since the developing solutions contain large amounts of sulfite ions, the only procedure required for derivatization is a dilution of the sample with carrier electrolyte. For separations of sulfite, acetate, glutaraldehyde-bisulfite and hydroquinone sulfonate anions, capillaries coated with poly(diallyldimethylammonium chloride) prior to analysis were used. Separation of four anions was accomplished in less than 3 min in 5 mmol l−1p-hydroxybenzoate-imidazole (p-HB) electrolyte at pH 6.0 using indirect UV detection at 254 nm. The optimal conditions for the derivatization reaction were established by varying concentration of sulfite, pH and reaction time. The recovery tests established for photographic developer samples were within the range 94.0-98.4%.  相似文献

In the work described here, a biosensor was developed for the determination of sulfite in food. Malva vulgaris tissue homogenate containing sulfite oxidase enzyme was used as the biological material. M. vulgaris tissue homogenate was crosslinked with gelatin using glutaraldehyde and fixed on a pretreated Teflon membrane. Sulfite was enzymatically converted to sulfate in the presence of the dissolved oxygen, which was monitored amperometrically. Sulfite determination was carried out by standard curves, which were obtained by the measurement of consumed oxygen level related to sulfite concentration. Several operational parameters had been investigated: the amounts of plant tissue homogenate and gelatin, percentage of glutaraldehyde, optimum pH and temperature. Also, some characterization studies were done. There was linearity in the range between 0.2 and 1.8 mM at 35 °C and pH 7.5. The results of real sample analysis obtained with the biosensor agreed well with the enzymatic reference method using spectrophotometric detection.  相似文献

A simple, rapid and sensitive chemiluminescence method for the determination of sulfite has been developed by combining flow-injection analysis and its sensitizing effect on the known chemiluminescence emission produced by the oxidation of luminol in alkaline medium; in this work permanganate has been proposed as oxidizing reactive. The optimum conditions for the chemiluminescence emission were established. The chemiluminescence was proportional to the sulfite concentration over the range 1.6 × 10−5 and 4.0 × 10−4 mol L−1. The detection limit was 4.7 × 10−6 mol L−1 of sulfite. The method has been satisfactorily used for the determination of free and bound sulfite in wines.  相似文献

Artificial neural networks (ANNs) are proposed for the determination of sulfite and sulfide simultaneously. The method is based on the reaction between Brilliant Green (BG) as a colored reagent and sulfite and/or sulfide in buffered solution (pH 7.0) and monitoring the changes of absorbance at maximum wavelength of 628 nm. Experimental conditions such as pH, reagents concentrations, and temperature were optimized and training the network was performed using principal components (PCs) of the original data. The network architecture (number of input, hidden and output nodes), and some parameters such as learning rate (η) and momentum (α) were also optimized for getting satisfactory results with minimum errors. The measuring range was 0.05-3.6 μg ml−1 for both analytes. The proposed method has been successfully applied to the quantification of the sulfite and sulfide in different water samples.  相似文献

An efficient electrocatalytic biosensor for sulfite detection was developed by co-immobilizing sulfite oxidase and cytochrome c with polyaniline sulfonic acid in a layer-by-layer assembly. QCM, UV–Vis spectroscopy and cyclic voltammetry revealed increasing loading of electrochemically active protein with the formation of multilayers. The sensor operates reagentless at low working potential. A catalytic oxidation current was detected in the presence of sulfite at the modified gold electrode, polarized at +0.1 V (vs. Ag/AgCl 1 M KCl). The stability of the biosensor performance was characterized and optimized. A 17-bilayer electrode has a linear range between 1 and 60 μM sulfite with a sensitivity of 2.19 mA M−1 sulfite and a response time of 2 min. The electrode retained a stable response for 3 days with a serial reproducibility of 3.8% and lost 20% of sensitivity after 5 days of operation. It is possible to store the sensor in a dry state for more than 2 months. The multilayer electrode was used for determination of sulfite in unspiked and spiked samples of red and white wine. The recovery and the specificity of the signals were evaluated for each sample. A novel amperometric sensor for the determination of sulfite was fabricated based on multiwalled carbon nanotubes (MWCNTs)/ferrocene-branched chitosan (CHIT-Fc) composites-covered glassy carbon electrode (GCE). The electrochemical behavior of the sensor was investigated in detail by cyclic voltammetry. The apparent surface electron transfer rate constant (Ks) and charge transfer coefficient (α) of the CHIT-Fc/MWCNTs/GCE were also determined by cyclic voltammetry, which were about 1.93 cm s−1 and 0.42, respectively. The sensor displayed good electrocatalytic activity towards the oxidation of sulfite. The peak potential for the oxidation of sulfite was lowered by at least 330 mV compared with that obtained at CHIT/MWCNTs/GCE. In optimal conditions, linear range spans the concentration of sulfite from 5 μM to 1.5 mM and the detection limit was 2.8 μM at a signal-to-noise ratio of 3. The proposed method was used for the determination of sulfite in boiler water. In addition, the sensor has good stability and reproducibility.  相似文献

The entrapment of sulfite oxidase (SOx) into ultrathin polypyrrole (PPy) films of 27–135 nm thickness has been successfully used for amperometric biosensing of sulfite with considerably improved performance. Optimum galvanostatic entrapment was accomplished in an electrolyte‐free solution which contained 0.1 M pyrrole and 5 U/mL of SOx with a polymerization period of 120 seconds and an applied current density of 0.2 mA cm?2. Evidence of the incorporation and retention of SOx in the ultrathin PPy film was obtained by scanning electron microscopy, cyclic voltammetry and amperometric measurements. Entrapment of the enzyme in a 54 nm thick PPy‐SOx film gave optimum amperometric response for sulfite and enabled the detection of as little as 0.9 μM of sulfite with a linear concentration range of 0.9 to 400 μM. The successful application of the biosensor to the determination of sulfite in beer and wine samples is reported. Comparison with a spectrophotometric method indicates that the biosensor was more superior for the determination of sulfite in red wine.  相似文献

Butyltin compounds are widespread contaminants which have also been found in some wines, determined by liquid-liquid extraction followed by alkylation with a Grignard reagent and gas chromatography-mass spectrometric (GC-MS) analysis. A promising alternative to this extraction/derivatization method is the one-step tetraethylborate in situ ethylation/solid-phase micro-extraction (SPME) method. In this work, a SPME-GC-MS method for the determination of butyltin compounds in wine was optimised. The optimised parameters concerned the pre-treatment with tetramethylammonium hydroxide, matrix modification with sodium chloride, tetraethylborate concentration, extraction time and temperature, and the GC separation program. The analytical figures of merit of the optimised method (range, limit of detection (LOD) and reproducibility) were evaluated. The sensitivity (range 20-1421 kcounts μg−1 l−1 as Sn) and LOD (range, 0.01-0.2 μg l−1 as Sn) depended greatly on the butyltin species to be measured and on the type of wine. For the tested species (monobutyltin, dibutyltin and tributyltin) the highest sensitivities were achieved for Port wine samples, followed by red wine>white wine>white Verde wine. The method allowed acceptable repeatability (relative standard deviation (R.S.D.), 6-8%; n=4) and reproducibility (R.S.D., 8-9%; n=3).  相似文献

The electrocatalytic oxidation of sulfite was investigated at carbon ionic liquid electrode (CILE). This electrode is a very good alternative to previously described electrodes because the electrocatalytic effect is achieved without any electrode modification. Comparative experiments were carried out using carbon paste electrode (CPE) and glassy carbon electrode (GCE). At CILE, highly reproducible and well-defined cyclic voltammograms were obtained for sulfite with a peak potential of 0.55 V vs. Ag/AgCl. Sulfite oxidation at CILE does not result in deactivation of the electrode surface. The kinetic parameters for this irreversible heterogeneous electron transfer process were determined. Under optimal experimental conditions, the peak current response increased linearly with sulfite concentration over the range of 6-1000 μM. The detection limit of the method was 4 μM. The method was applied to the determination of sulfite in mineral water, grape juice and non-alcoholic beer samples.  相似文献

The use of cyclic voltammetry to characterize wines and wine polyphenols in a pH 3.3 model wine solution has been extended to take into account the effects of sulfur dioxide and polyphenol adsorption processes. A good correlation was obtained between a cyclic voltammetric measure, based upon the response produced before and after acetaldehyde additions, and the concentration of free sulfur dioxide in eight white wines (r2 = 0.974). By the addition of acetaldehyde to the white wines, an important new step in the methodology, the area under the anodic scan in the potential range from −100 to 1200 mV (Ag/AgCl) closely matched the spectroscopic measure of total polyphenols (absorbance at 280 nm) for the white wines, when both were measured in terms of caffeic acid equivalents (r2 = 0.949). The anodic peak area accounted for about 70% of the 280 nm total phenols measure, in catechin equivalents, for the red wines, and a good linear correlation was also obtained (r2 = 0.942). The level of catechol and galloyl-containing polyphenols in the wines was calculated by measuring the size of the first anodic peak at around 450 mV after treatment of the wines with acetaldehyde; the peak current correlated well with the total caffeic acid derivatives in the white wines determined by HPLC (r2 = 0.982). The concentration of flavonols was estimated by selective adsorption of these compounds onto the carbon electrode and determining the anodic peak current at 1120 mV, with good correlations obtained when compared to total flavonols as measured by HPLC (r2 = 0.984 for the red wines, and r2 = 0.987 for the white wines).  相似文献

We developed a new analysis method for the N-methyl carbamate pesticides in juice and wine. The juice and wine were diluted with ultra pure water, and determined by electrospray ionization tandem mass spectrometry (ESI LC/MS/MS) with direct sample injection into a short column. The new method, including sample preparation and determination, is simple and rapid, and allows simultaneous determination of nine N-methyl carbamate pesticides in juice and wine within analysis time that is much shorter as compared with the traditional method. The average recoveries from juice and wine fortified at the level of 0.1 ppm ranged from 59.6 to 126.7% with the coefficients of variation ranging from 0.4 to 5.1% for intra-day (n = 5 × 3 days) and from 0.5 to 22.6% for inter-day (n = 15). At the fortified level of 0.5 ppm, the recoveries ranged from 69.3 to 127.2% with the coefficients of variation ranging from 0.4 to 6.9% for intra-day (n = 5 × 3 days) and from 0.5 to 22.6% for inter-day (n = 15). The method is considered to be satisfactory for the monitoring of the carbamate pesticides residues in juice and wine, suggesting that the present method is applicable to other pesticide residues in foods.  相似文献

In this paper, we proposed a procedure for the determination of iron(II) and total iron in wine samples employing molecular absorption spectrophotometry. The ligand used is 2-(5-bromo-2-pyridylazo)-5-(diethylamino)-phenol (Br-PADAP) and the chromogenic reaction in absence or presence of ascorbic acid (reducing agent) allows the determination of iron(II) or total iron, respectively. The optimization step was performed using a multivariate technique (Box Behnken design) involving the factors pH, acid ascorbic concentration and reaction time.The method allows the determination of iron(II) and iron(III) in wine samples, with limits of detection and quantification 0.22 and 0.72 μg L−1, respectively. The precision expressed as relative standard deviation (R.S.D.) was 1.43 and 0.56% (both, n = 11) for content of iron(II) in wine samples of 1.68 and 4.65 mg L−1, and 1.66 and 0.87% (both, n = 11) for content of total iron in wine samples of 1.72 and 5.48 mg L−1.This method was applied for determination of iron(II) and total iron in six different wine samples. In these, the iron(II) content varied from 0.76 to 4.65 mg L−1 and from 1.01 to 5.48 mg L−1 for total iron. The results obtained in the determination of total iron by Br-PADAP method were compared with those that were performed after complete acid digestion in open system and determination of total iron employing FAAS. The method of regression linear was used for comparison of these results and demonstrated that there is no significant difference between the results obtained with these two procedures.  相似文献

Bismuth was evaluated as an internal standard for the direct determination of Pb in vinegar by graphite furnace atomic absorption spectrometry using Ru as a permanent modifier with co-injection of Pd/Mg(NO3)2. The correlation coefficient of the graph plotted from the normalized absorbance signals of Bi versus Pb was r = 0.989. Matrix effects were evaluated by analyzing the slope ratios between the analytical curve obtained from reference solutions prepared in 0.2% (v/v) HNO3 and analytical curves obtained from Pb additions in red and white wine vinegar samples. The calculated ratios were around 1.04 and 1.02 for analytical curves established applying an internal standard and 1.3 and 1.5 for analytical curves without. Analytical curves in the 2.5–15 μg L− 1 Pb concentration interval were established using the ratio Pb absorbance to Bi absorbance versus analyte concentration, and typical linear correlations of r = 0.999 were obtained. The proposed method was applied for direct determination of Pb in 18 commercial vinegar samples and the Pb concentration varied from 2.6 to 31 μg L− 1. Results were in agreement at a 95% confidence level (paired t-test) with those obtained for digested samples. Recoveries of Pb added to vinegars varied from 96 to 108% with and from 72 to 86% without an internal standard. Two water standard reference materials diluted in vinegar sample were also analyzed and results were in agreement with certified values at a 95% confidence level. The characteristic mass was 40 pg Pb and the useful lifetime of the tube was around 1600 firings. The limit of detection was 0.3 μg L− 1 and the relative standard deviation was ≤ 3.8% and ≤ 8.3% (n = 12) for a sample containing 10 μg L− 1 Pb with and without internal standard, respectively.  相似文献

Simultaneous determination of sulfite and thiosulfate at sub-ppm levels in petroleum and its distillates was investigated using a convenient, accurate and sensitive procedure. This method involved preliminary extraction of the sample followed by detection via differential pulse polarography (DPP) at a dropping mercury electrode. In this procedure, an appropriate amount of sample was shaken with a recommended volume of 0.25% (w/w) sodium acetate solution. The mixture was filtered in two steps and was then ready for DPP. The method was free from interferences from hydrogen sulfide, elemental sulfur, organic sulfides, and thiophene. Various instrumental factors such as scan rate, pulse height, initial and final potential, and purge time were optimized. The 3σ detection limits were 410 and 125 ng g−1 for sulfite and thiosulfate, respectively. At 5 μg g−1 level in samples, the relative standard deviations (n=4) were 2.51 and 1.15% for sulfite and thiosulfate, respectively. The proposed method was applied to real samples containing input feeds, distillates and fuel oils from Abadan Petroleum Refinery in the south of Iran.  相似文献

This work presents an advantageous analytical procedure for the accurate determination of free trans-resveratrol in red and white wines. The proposed method involves solid-phase extraction (SPE), acetylation of the analyte in aqueous media and further determination by gas chromatography (GC) with mass spectrometry detection (MS). The use of a mixed-mode SPE sorbent provides an improvement in the selectivity of the extraction step; moreover, the presence of several intense ions in the electron impact mass spectra of its acetyl derivative guarantees the unambiguous identification of trans-resveratrol. Considering a sample intake of 10 mL, the method provides a limit of quantification (LOQ) of 0.8 ng mL−1 and linear responses for concentrations up to 2.5 μg mL−1, referred to wine samples. The average recovery, estimated with samples fortified at different concentrations in the above range, was 99.6% and the inter-day precision stayed below 8%. Trans-resveratrol levels in the analyzed wines varied from 3.4 to 1810 ng mL−1. Cis-resveratrol was also found in all samples. In most cases, equal or higher responses were measured for this latter form than for the trans-isomer. The reduced form of resveratrol, dihydro-resveratrol, was systematically identified in red wines.  相似文献

Plasticized membranes using 1-phenyl-3-(2-thiazolyl)-2-thiourea (PTT) and 1-phenyl-3-(2-thiazolyl)-2-urea (PTU) have been prepared and explored as ytterbium ion-selective sensors. Effect of various plasticizers, viz. chloronaphthalene (CN), o-nitrophenyloctyl ether (o-NPOE), dibutylphthalate (DBP), dioctylsebacate (DOS) and anion excluders, sodium tetraphenylborate (NaTPB) and oleic acid (OA) was studied and improved membrane performance was observed. Optimum performance was noted with membrane of PTT having composition of PTT (3.5):PVC (80):DOS (160):NaTPB (1.5) in mg. The sensor works satisfactorily in the concentration range 1.2 × 10−7 to 1.0 × 10−2 M (detection limit 5.5 × 10−8 M) with a Nernstian slope of 19.7 mV decade−1 of activity. Wide pH range (3.0-8.0), fast response time (10 s), non-aqueous tolerance (up to 20%) and adequate shelf life (12 weeks) indicate the vital utility of the proposed sensor. The proposed electrode comparatively shows good selectivity for Yb3+ ion with respect to alkali, alkaline earth, transition and rare earth metals ions and can be used for its determination in binary mixtures and sulfite determination in white and red wine samples.  相似文献

A new automated method for the determination of glucose in honey is proposed. The method is based on multicommutated flow analysis (MCFA) and employs an immobilized glucose oxidase reactor and spectrophotometric detection at 505 nm of the red quinoneimine formed (Trinder's method).The calibration curve obeyed a second order equation in the range 0-0.14 g L−1 (h = −2.2199 C2 + 1.3741C + 0.0077, r2 = 0.9991, where h is the peak height (absorbance) and C the concentration in g L−1). The method was validated analyzing eight commercial samples, both by the AOAC 954.11 and 977.20 official methods. According to Student's t-test of mean values, at the confidence level of 95% the results obtained with the proposed method were in agreement with those obtained by the official methods. Precision (sr(%), n = 10) was 3% and the sampling frequency of the system was 20 samples h−1.  相似文献
