A method was developed for the synthesis of 6,7-benzo-3-borabicyclo[3.3.1]nonane and 6,7-benzo-3-azabicyclo[3.3.1]nonane derivatives based on intramolecular cyclization of 2-allylphenyl(diallyl)borane. Intramolecular arylboration of the double bond in 1,5-diallyl-2,3-benzo-1-boracyclohexane was carried out for the first time. Conventional oxidation (H2O2-OH) of 6,7-benzo-3-methoxy-3-borabicyclo[3.3.1]nonane afforded cis-1,3-di(hydroxymethyl) tetralin. New pyridine, pyrazoloyridine, and furopyridine derivatives substituted with naphthyl and thienyl moieties were designed and synthesized starting from 6-(naphthalen-2-yl)-2-oxo-4-(thiophen-2-yl)-1,2-dihydropyridine-3-carbonitrile (1). The chloro, methoxy, cholroacetoxy, imidazolyl, azide, and arylamino derivatives were prepared to obtain the pyridine-C2 functionalized derivatives. The derived pyrazolpyridine-N-glycosides were synthesized via heterocyclization of the C2-thioxopyridine derivative followed by glycosylation using glucose and galactose. The furopyridine derivative 14 and the tricyclic pyrido[3′,2′:4,5]furo[3,2-d]pyrimidine 15 were prepared via heterocyclization of the ester derivative followed by a reaction with formamide. The newly synthesized compounds were evaluated for their ability to in vitro inhibit the CDK2 enzyme. In addition, the cytotoxicity of the compounds was tested against four different human cancer cell lines (HCT-116, MCF-7, HepG2, and A549). The CDK2/cyclin A2 enzyme inhibitory results revealed that pyridone 1, 2-chloro-6-(naphthalen-2-yl)-4-(thiophen-2-yl)nicotinonitrile (4), 6-(naphthalen-2-yl)-4-(thiophen-2-yl)-1H-pyrazolo[3,4-b]pyridin-3-amine (8), S-(3-cyano-6-(naphthaen-2-yl)-4-(thiophen-2-yl)pyridin-2-yl) 2-chloroethanethioate (11), and ethyl 3-amino-6-(naphthalen-2-yl)-4-(thiophen-2-yl)furo[2,3-b]pyridine-2-carboxylate (14) are among the most active inhibitors with IC50 values of 0.57, 0.24, 0.65, 0.50, and 0.93 µM, respectively, compared to roscovitine (IC50 0.394 μM). Most compounds showed significant inhibition on different human cancer cell lines (HCT-116, MCF-7, HepG2, and A549) with IC50 ranges of 31.3–49.0, 19.3–55.5, 22.7–44.8, and 36.8–70.7 μM, respectively compared to doxorubicin (IC50 40.0, 64.8, 24.7 and 58.1 µM, respectively). Furthermore, a molecular docking study suggests that most of the target compounds have a similar binding mode as a reference compound in the active site of the CDK2 enzyme. The structural requirements controlling the CDK2 inhibitory activity were determined through the generation of a statistically significant 2D-QSAR model.  相似文献

(S)-4-Chloro-2-fluorophenylalanine and (S)-(α-methy)-4-chloro-2-fluorophenylalanine were synthesized and labeled with no carrier added (n.c.a.) fluorine-18 through a radiochemical synthesis relying on the highly enantioselective reaction between 4-chloro-2-[18F]fluorobenzyl iodide and the lithium enolate of (2S)-1-(tert-butyloxycarbonyl)-2-(tert-butyl)-3-methyl-1,3-imidazolidine-4-one for (S)-4-chloro-2-[18F]fluorophenylalanine and (2S,5S)-1-(tert-butyloxycarbonyl)-2-(tert-butyl)-3,5-dimethyl-1,3-imidazolidine-4-one for (S)-(α-methyl) -4-chloro-2-[18F] fluorophenylalanine. Quantities of about 20–25 mCi were obtained at the end of sy nthesi s, ready for injection after hydrolysis and high performance liquid chromatography (HPLC) purification, with a radiochemical yield of 17%–20% corrected to the end of bombardment after a total synthesis time of 90–105 min from [18F] fluoride. The enantiomeric excesses were shown to be 97% or more for both molecules without chiral separation and the radiochemical and chemical purities were 98% or better.  相似文献

The compositions of regioisomeric mixtures of tricarbonyliron complexes of 2-methyltropone (1a,b), 3-methyltropone (2a,b) and 2,6-dimethyltropone (3a,b) are studied and compared with the results of ab initio computations. The structures, frontier orbitals, and population analysis are evaluated by means of density functional theory. The thermodynamic and kinetic parameters of regioisomerizations are determined using dynamic 1H NMR technique. The influence of methyl-substituent(s) on the equilibrium ratio of regioisomers resulting from the haptotropic rearrangement is discussed. Significant differences in the reactivity of C-protonized methyl- and dimethyl-substituted tricarbonyl(tropone)iron complexes 46 in nucleophilic additions and corresponding O-trimethylsilylated complexes 79 in [3+2] cycloadditions are explained in terms of electronic and steric effects of the methyl group(s). Various hydroazulenone cycloadducts of tricarbonyl(η4-2,6-dimethyltropone)iron 3a,b have been prepared by stereoselective [3+2] cycloaddition with Fp-reagents 1214 and characterized. Formerly proposed mechanism of [3+2] cycloaddition was approved.  相似文献

3,4-Dimethylene-1,6-hexane diols2 obtained by allylboration of ketones with 2,3-dimethylene-1,4-bis(dipropylboryl)butane1 undergo cyclization on treatment with I2/NaHCO3 to give 1,5-cis-di(iodomethyl)-2,6-dioxabicyclo[3.3.0]octanes (4) and/or 3,8-dioxabicyclo[4.4.0]dec-1(6)-enes (5). 4,4,9,9-Tetramethyl-3,8-dioxacyclodeca-1,6-dione (9) was synthesized by ozonolysis of bicyclic compound5b. From the reaction mixtures in the uncatalyzed polybromination of [2.2]paracyclophane by the action of excess Br2 in CCl4, there have been found along with the known products — 4,15- and 4,16-dibromo[2.2]paracyclophanes — two new aromatic tribromides of this series, which have been isolated in pure form: 4,12,15- and 4,15,16-tribromo[2.2]paracyclophanes. Special experiments demonstrated that the mixtures of these tribromides are formed as a result of competitive monobromination of 4,15-dibromo[2.2]paracyclophane; the 4,15,16-tribromo[2.2]paracyclophane, together with still another newly isolated isomer of this series — 4,8,12-tribromo[2.2]paracyclophane — is formed as a result of competitive monobromination of 4,16-dibromo[2.2]paracyclophane. As an explanation of the features of the orienting effect of substituents in these competing reactions, a rule was proposed: On the conventional orientation (from the electronic point of view) of entry of the bromine atom into the substituted ring (para > ortho > meta), a steric limitation is imposed on its attack in the pseudo-gem-position, owing to the bulky bromine atom that is transannularly positioned above it in the neighboring aromatic ring. The structures of all of the tribromides were established on the basis of elemental analyses, mass spectrometry, and1H NMR spectrometry (including PMR using the homonuclear Overhauser effect). The data obtained in this work indicate that the 4,12,15-tribromo[2.2]paracyclophane and 4,15,16-tribromo[2.2]paracyclophane are predecessors of the two tetrabromides previously obtained by Cram — 4,7,12,15- and 4,5,15,16-tetrabromo[2.2]paracyclophanes; and the 4,8,12-tribromo[2.2]paracyclophane is a possible predecessor of 4,8,12,16-tetrabromo[2.2]paracyclophane, which is unknown up to the present time.A. Summary Arylbiguanides2 a–e react with benzoin (1) at thepH of the base to two different products.1 undergoes in presence of the base2 a–e oxidation to benzil and benzoic acid, which reacts fast with the arylbiguanides2 a–e to yield N-[4-(arylamino)-6-phenyl-1,3,5-triazine-2-yl]benzamides3 a–d. After lowering thepH of the reaction mixture, the bases2 b–e react with benzil to yield 2-[1-aryl-5-oxo-4,4-diphenyl-2-imidazoline-2-yl]guanidine4 b–e. The mechanism of the formation is discussed. The structure of4b was established from a single crystal x-ray structure analysis. The analysis was carried out at 100K: C23H21N5O,M r=383.5, monoclinic, C 2/c,a=15.842(6),b=8.419(3),c=30.223(10) Å, =98.44(3)°,V=3 987.3(9) Å3,Z=8,d x=1.277 g/cm3, =0.81 cm–1,R=5.89%R w=4.97% (1 537 observations, 233 parameters).

A new thiazolylazo chromogenic reagent, 2-[2-(6-methylbenzo-thiazoly)azo]-5-diethylaminobenzoic acid (6-Me-BTAEB), has been synthesized. Its chromogenic reaction with microamounts of nickel in the presence of sodium dodecylsulfate (SDS) has been thoroughly studied. 6-Me-BTAEB reacts with nickel(II) in weak acid medium containing appropriate amounts of SDS to form a blue-violet complex with high sensitivity, good selectivity and high stability. The composition is found to be 1:2 (nickel to 6-Me-BTAEB) and its absorption maximum is at 650 nm with an apparent molar absorptivity of 1.67 × 105l mole–1 cm–1. Beer's law is obeyed over the range 0-0.4 g of nickel per ml. The proposed method has been applied to the direct determination of nickel in aluminium alloys, pure magnesium and low alloy steels at the 0.2–0.3% (w:w) level with satisfactory results.  相似文献

[3+2] Cycloadditions of (1Z,4R,5R)-1-arylmethylidene-4-benzoylamino-3-oxo-5-phenylpyrazolidin-1-ium-2-ides 1a-e to methyl methacrylate gave the 1-CO2Me regioisomers 3/3′, exclusively, in 1-67% yields. Stereocontrol was dependent on the ortho-substituents at the 1′-aryl group in dipole 1: ortho-unsubstituted dipoles 1a-c gave the major (1R,3R,5R,6R)-isomers 3a-c, whilst ortho-disubstituted dipoles gave the major (1R,3S,5R,6R)-isomers 3′d,e. The structures of cycloadducts were determined by NMR and X-ray diffraction.  相似文献

In the presence of CoCl2·6H2O and dppm (bis(diphenylphosphino) methane), the reaction of TCNQ (7,7,8,8-tetracyanoquinodimethane) molecules by [2+2] cycloaddition forms a p-tricyanovinylphenyldicyanomethide ion (PCQ), which has been obtained as one anion unit in one new compound [Co(dppmdo)3][PCQ]2·H2O 1 (dppmdo = bis(diphylphospine oxide) methane). Its structure was determined by X-ray crystallography: 1 crystallizes in with a = 14.174(3) Å, b = 19.553(4) Å, c = 19.776(4) Å, α = 112.72(3)°, β = 95.43(3)°, γ = 110.79(3)°, and Z = 2. It was characterized by IR spectra, UV–Vis spectra, and cyclic voltammogram. Magnetic properties indicate that no magnetic coupling between PCQ and [Co(dppmdo)3]2+ unit.  相似文献

Zusammenfassung Aufgrund ihres Enamincharakters reagieren die Methylen-N-heterocyclen1–5 mit cyclischen 2-Ethoxymethylen-1,3-dicarbonylverbindungen6 zu den 2-[2-(Hetaryliden)ethyliden]-1,3-dicarbonylverbindungen7–14. Das Ergebnis der Reaktionen zwischen 1,2-Dihydro-1-methyl-2-methylen-chinolin (1a) und cyclischen 1,3-Dicarbonylverbindungen hängt von der Natur der zwischenzeitlich entstehenden DihydroverbindungenA/B ab. Die Intermediat-KetoneA gehen durch Dehydrierung während der Reaktion in die 2-(1,2-Dimethyl-4(1H)chinolyliden)-1,3-dicarbonylverbindungen17–21 über. Die Intermediat-EnoleB mit sechsgliedrigem Dicarbonylring bilden in intramolekularer Reaktion die 6,12-Methano-dibenz[d,g][1,3]oxazocinone22–25, deren Struktur am Beispiel der Verbindung23 durch1H-NMR sowie durch Röntgenkristallstrukturanalyse bewiesen wird.

A new, simple, rapid, and sensitive spectrophotometric method has been developed for the determination of nitrophenols [picric acid (PA); dinitrophenols (DNP)] in wastewater samples. The method is based on the reaction of nitrophenols with 2-[(E)-2-(4-diethylaminophenyl)-1-ethenyl]-1,3,3-trimethyl-3 H-indolium chloride reagent to form the colored ion associates, which are extracted by organic solvents. The molar absorptivity of the ion associates of PA with the investigated reagent ranges from 8.3×104 to 11.3×104 L mol–1 cm–1, depending on the extractant. Because only PA is extracted in an acidic medium with the investigated reagent, but both PA and DNP are extracted in an alkaline medium, it is possible to determine both substances in a mixture. Appropriate reaction conditions have been established. The absorbance of the colored extracts obeys Beers law in the range of 0.04–4.58 mg L–1 PA, 1.0–18.4 mg L–1 2,4-DNP and 1.2–14.7 mg L–1 2,6-DNP, respectively. The limit of detections, calculated from a blank test (n=10; P=0.95), are 0.05 mg L–1 PA, 0.9 mg L–1 (2,4-DNP), and 1.1 mg L–1 (2,6-DNP), respectively.  相似文献

Opioid analgesics are clinically used to relieve severe pain in acute postoperative and cancer pain, and also in the long term in chronic pain. The analgesic action is mediated by μ-, δ-, and κ-receptors, but currently, with few exceptions for k-agonists, μ-agonists are the only ones used in therapy. Previously synthesized compounds with diazotricyclodecane cores (DTDs) have shown their effectiveness in binding opioid receptors. Fourteen novel diazatricyclodecanes belonging to the 9-propionyl-10-substituted-9,10-diazatricyclo[4.2.1.12,5]decane (compounds 20–23, 53, 57 and 59) and 2-propionyl-7-substituted-2,7-diazatricyclo[4.4.0.03,8]decane (compounds 24–27, 54, 58 and 60) series, respectively, have been synthesized and their ability to bind to the opioid μ-, δ- and κ-receptors was evaluated. Five of these derivatives, compounds 20, 21, 24, 26 and 53, showed μ-affinity in the nanomolar range with a negligible affinity towards δ- and κ-receptors and high μ-receptor selectivity. The synthesized compounds showed μ-receptor selectivity higher than those of previously reported methylarylcinnamyl analogs.  相似文献

The reaction of [2-(dimethylaminomethyl)]cymantrenyl]lithium with ytterbium diiodide affords [2-(dimethylaminomethyl)cymantrenyl]ytterbium iodide or bis[2-(dimethylaminomethyl)cymantrenyl]ytterbium depending on the ratio of the starting reactants. Summary The keto (thio) tautomers1–4 of the hydrazono pyrazolone, thiopyrazolone derivatives underwent simultaneous diformylation, chlorination (desulphurization) and ring closure under Vilsmeier reaction conditions giving the pyrazolo[3,4-c]pyrazole aminoacroleins (5,6). Treatment of5 and/or6 with proper reagents afforded the corresponding pyrazolo[3,4-c]pyrazoles with different heterocyclic systems at the 3-position.The structures of these compounds were confirmed by elemental analysis, IR and1H-NMR spectroscopy. All synthesized compounds have been screened in vitro for their antibacterial activities against a number of Gram-positive and Gram-negative bacteria.

Zusammenfassung Die Keto-(Thio)-Tautomeren1–4 der Hydrazonopyrazolon-/Thiopyrazolon-Derivate gingen unter Vilsmeier-Bedingungen zugleich Diformylierung, Chlorierung (Entschwefelung) und Ringschluß zu Pyrazolo[3,4-c]pyrazol-aminoacroleinen5 und6 ein. Aus5 und/oder6 konnten die entsprechenden Pyrazolo[3,4-c]pyrazole mit verschiedenen heterocyclischen Systemen in 3-Position erhalten werden. Die Strukturen der Verbindungen wurden mittels Elementaranalyse, IR und1H-NMR überprüft. Alle synthetisierten Verbindungen wurden in vitro bezüglich ihrer antibakteriellen Aktivität gegenüber einer Anzahl Gram-positiver und Gram-negativer Bakterien untersucht.

The structural and spectroscopic properties of a new triazolopyridine derivative (1,2,4-triazolo[4,3-a]pyridin-3-amine) are described in this paper. Its FTIR spectrum was recorded in the 100–4000 cm−1 range and its FT-Raman spectrum in the range 80–4000 cm−1. The molecular structure and vibrational spectra were analyzed using the B3LYP/6-311G(2d,2p) approach and the GAUSSIAN 16W program. The assignment of the observed bands to the respective normal modes was proposed on the basis of PED calculations. XRD studies revealed that the studied compound crystallizes in the centrosymmetric monoclinic space group P21/n with eight molecules per unit cell. However, the asymmetric unit contains two 1,2,4-triazolo[4,3-a]pyridin-3-amine molecules linked via N–H⋯N hydrogen bonds with a R22(8) graph. The stability of the studied molecule was considered using NBO analysis. Electron absorption and the luminescence spectra were measured and discussed in terms of the calculated singlet, triplet, HOMO and LUMO electron energies. The Stokes shifts derived from the optical spectra were equal to 9410 cm−1 for the triazole ring and 7625 cm−1 for the pyridine ring.  相似文献

Selective formation of (η3-siloxyallyl)tungsten complexes by reaction of hydrido(hydrosilylene)tungsten complexes with α,β-unsaturated carbonyl compounds was reported experimentally. The mechanisms have been investigated by employing the model reaction of [Cp(CO)2(H)WSi(H)–{C(SiH3)3}] (R), derived from the original experimental complex Cp′(CO)2(H)WSi(H)–[C(SiMe3)3] (1a, Cp′ = Cp*; 1b, Cp′ = η5-C5Me4Et), with methyl vinyl ketone, under the aid of the density functional calculations at the b3lyp level of theory. It is theoretically predicted that the route involving migration of the hydride to silicon to afford a 16e intermediate [Cp(CO)2W–SiH2–{C(SiH3)3}] is inaccessible (route 2), supporting the proposition by experiments. Another route, via [2 + 4] cycloaddition followed by directly Si–H reductive elimination, is theoretically predicted to be accessible (route 1). In route 1, two possible paths with different attacking directions of the oxygen of methyl vinyl ketone at Si (WSi) are put forward. The attack at the Si atom from the hydride (H1) side of the plane W–Si–H1 in R is found to be preferred kinetically. The regioselectivity for formation of (η3-siloxyallyl)tungsten complexes, where only the exo-anti isomer was obtained, is discussed based on the consideration of thermodynamics and kinetics.  相似文献
