Due to the great importance of new therapeutic routes for morphine in pain treatment, several investigations are under development. In this way, the design of a liquid system for the oral administration of morphine would be of great help, especially in patients with difficulties in swallowing (children and elderly people). The systems studied in this work are kollidon® SR microparticles, a biodegradable polymer classically used as excipient in the design of solid dosage forms, as vehicles for morphine. A detailed investigation of the capabilities of the polymer particles to load this drug at their surface is described. Electrophoretic mobility and optical absorbance determinations were used with this aim. The main factors determining the drug incorporation, after incubation of the microparticles in the morphine solutions, were the adsorption time, the type of electrolyte and its concentration, and the drug concentration. The optimum loading conditions were used to perform morphine release evaluations, finding that the release profiles were biphasic since the drug adsorbed was slowly released during 24 h after an initial burst release phase.  相似文献

Janus particles with differentially degradable compartments were prepared by electrohydrodynamic (EHD) co‐jetting and subsequent controlled crosslinking. These bicompartmental particles are composed of an interpenetrating polymer network of poly(ethylene oxide) and poly(acrylamide‐co‐acrylic acid) in one hemisphere and a crosslinked copolymer of dextran and poly(acrylamide‐co‐acrylic acid) segments in the second compartment. The compositional anisotropy caused differential hydrolytic susceptibility: Although both compartments were stable at pH 3.0, selective degradation of the PEO‐containing compartment pH 7.4 was observed wtihin 5 days. Janus particles with differentially degradable polymer compartments may be of interest for a range of oral drug delivery applications because of their propensity for decoupled release profiles.  相似文献

The correlation between erosion and drug (lidocaine and 6‐mercaptopurine, 6‐MP) release from amorphous poly(thioether anhydrides), which are synthesized using radical‐mediated thiol‐ene polymerization, is reported. Cytotoxicity studies of the polymer toward human fibroblast human dermal fibroblasts adult, melanoma A‐375, and breast cancer MCF‐7 cells are conducted, and drug efficacy of a cancer and autoimmune disease drug (6‐MP) when released from the poly(thioether anhydrides) is examined against two cancerous cell types (A‐375 and MCF‐7). Erosion and drug release studies reveal that lidocaine release is governed by network erosion whereas 6‐MP is released by a combination of erosion and diffusion. The cytotoxicity studies show that all three cell types demonstrate high viability, thus cytocompatibility, to poly(thioether anhydrides). Toxicity to the material is dose dependent and comparable to other polyanhydride systems. The 6‐MP cancer drug is shown to remain bioactive after encapsulation in the poly(thioether anhydride) matrix and the polymer does not appear to modify the efficacy of the drug.  相似文献

The purpose of the study is to obtain multicomponent polyelectrolyte hydrogels with optimal synergistic properties by combining a modified starch with a synthetic one. Thus, new low‐cost and biocompatible semi‐interpenetrating polymer network (semi‐IPN) hydrogels of carboxymethyl starch and poly(2‐dimethylaminoethyl methacrylate) are prepared and investigated. The synthesized hydrogels are studied with respect to the specific characteristics of the gels: swelling kinetics, thermal analysis, viscoelastic characteristics, and their ability to be used as a matrix in drug delivery systems. Therefore, the semi‐IPN gels are loaded with ibuprofen, followed by additional tests to assess the in vitro drug release. The cytocompatibility of the hydrogels with respect to their composition is evaluated in vitro on fibroblast cell culture. The investigations confirm the obtainment of new semi‐IPN hydrogels with pH and temperature responsiveness, good mechanical strength, and potential for use as drug delivery systems or transdermal patches.  相似文献

Because of the fundamental importance of new therapeutic routes for cancer treatment, a number of systems based on colloidal particles as vehicles for the delivery of the anticancer drug 5-fluorouracil have been devised. The target is always to provide the proper dose of the antitumor agent only at the desired locus of action, thus reducing the unwanted side effects. In this review, the main strategies and the more significant results in the development of 5-fluorouracil carriers for cancer treatment are discussed.  相似文献

Microgel particles are cross-linked polymer particles. When dispersed in a good solvent for the polymer concerned, they are able to respond to a range of external stimuli by changing volume. Hence, microgel particles are suited to numerous applications (for example, controlled uptake and release) in the pharmaceutical, coatings, and water treatment industries. In this work, pH-sensitive, 0.5 wt % cross-linked poly(2-vinylpyridine) (PVP) microgel particles have been prepared and characterized. When the dispersion pH is decreased below 4.5, the pyridine groups become protonated and the microgel network becomes positively charged, causing the particles to expand. To investigate the possibility of using light as a trigger for effecting volume changes, the interaction of these microgel particles with a photodegradable anionic surfactant, 4-hexylphenylazosulfonate (C(6)PAS), has been investigated using dynamic light scattering and electrophoretic mobility measurements. The electrostatic attraction between the positively charged microgel network (at solution pH 3) and the negatively charged headgroups on the surfactant molecules caused a dramatic decrease in particle volume, and charge-reversal of the particles occurred with increasing surfactant concentration. The UV irradiation of phenylazosulfonate surfactants destroys the anionic headgroup of the molecules, and the microgel particles re-swell. The irradiation of PVP dispersions in the presence of C(6)PAS, along with mixed surfactant systems of sodium dodecyl sulfate plus C(6)PAS, has been investigated.  相似文献

A facile approach to synthesis of ABCDE‐type H‐shaped quintopolymer comprising polystyrene (PSt, C) main chain and poly(ethylene glycol) (PEG, A), poly(ε‐caprolactone) (PCL, B), poly(L ‐lactide) (PLLA, D), and poly(acrylic acid) (PAA, E) side chains was described, and physicochemical properties and potential applications as drug carriers of copolymers obtained were investigated. Azide‐alkyne cycloaddition reaction and hydrolysis were used to synthesize well‐defined H‐shaped quintopolymer. Cytotoxicity studies revealed H‐shaped copolymer aggregates were nontoxic and biocompatible, and drug loading and release properties were affected by macromolecular architecture, chemical composition, and pH value. The release rate of doxorubicin from copolymer aggregates at pH 7.4 was decreased in the order PAA‐b‐PLLA > H‐shaped copolymer > PEG‐PCL‐PSt star, and the release kinetics at lower pH was faster. The H‐shaped copolymer aggregates have a potential as controlled delivery vehicles due to their excellent storage stability, satisfactory drug loading capacity, and pH‐sensitive release rate of doxorubicin. © 2012 Wiley Periodicals, Inc. Polymersomes have gained much interest within the biomedical field as drug delivery systems due to their ability to transport and protect cargo from the harsh environment inside the body. For an improved drug efficacy, control over cargo release is however also an important factor to take into account. An often employed method is to incorporate pH sensitive groups in the vesicle membrane, which induce disassembly and content release when the particles have reached a target site in the body with the appropriate pH, such as the acidic microenvironment of tumor tissue or the endosome. In this paper, biodegradable poly(ethylene glycol)-poly(caprolactone-gradient-trimethylene carbonate)-based polymeric vesicles have been developed with disassembly features at mild acidic conditions. Modifying the polymer backbone with imidazole moieties results in vesicle disassembly upon protonation due to the lowered pH. Furthermore, upon increasing the pH efficient re-assembly into vesicles is observed due to the switchable amphiphilic nature of the polymer. When this re-assembly process is conducted in presence of cargo, enhanced encapsulation is achieved. Furthermore, the potency of the polymeric system for future biomedical applications such as adjuvant delivery is demonstrated.  相似文献

The kinetics of the controlled release of the antiproliferative drug dipyridamole from microspheres based on the biocompatible and biodegradable polymer poly(3-hydroxy)butyrate is studied. As carriers for dipyridamole, microspheres prepared from a solution of poly(3-hydroxy)butyrate by single emulsion method are used. Under in vitro conditions, the kinetic curves describing the release of dipyridamole from microspheres with diameters of 19, 63, and 92 μm show two characteristic regions: the region of fast drug release within a short time period and a well-pronounced continuous linear region. For microspheres with a diameter of 4 μm, the linear region is missing. Analysis of the kinetic curves illustrating controlled drug release together with the measurements on polymer degradation shows that their kinetic profiles depend on the diffusion-controlled process and hydrolytic degradation of poly(3-hydroxy)butyrate. The diffusion kinetic equation describing both linear and nonlinear regions of dipyridamole released from the microspheres involves the sum of two terms: desorption from the sphere via the diffusion-controlled mechanism and drug release via the zero-order reaction. The linear region of the drug release curve is explained by the zero-order hydrolysis of poly(3-hydroxy)butyrate. The diffusion coefficients and kinetic constants are calculated. For bigger microspheres, the existence of the continuous linear region in the corresponding kinetic curves makes it possible to use microsystems based on poly(3-hydroxy)butyrate and dipyridamole as novel systems for local prolonged drug delivery.  相似文献

Heterodimers based on the polymer-bound chromophore 4-methylcoumarin and the prodrug 1-heptanoyl-5-fluorouracil, synthesized by photochemical [2 + 2]-cycloaddition are promising photoresponsive drug depots. Drug release experiments are one possibility to deliver proof of a successful reversible drug immobilization, whereas NMR spectroscopy is a potent tool for further structural characterization of these polymer-bound heterodimers. In case of the random copolymer poly(methyl methacrylate-co-7-(2'-methacryloyloxyethoxy)-4-methylcoumarin) three dimers have been identified of which the syn head-to-tail was the predominant one. In contrast, only the syn head-to-head dimer was formed in reasonable yield when the 4-methylcoumarin monofunctionalized pMMA was used as the base polymer. 1D and 2D NMR spectroscopic techniques combined with some theoretical calculations helped in successfully closing one major gap concerning polymer bound 4-methylcoumarin/1-heptanoyl-5-fluorouracil heterodimers that are of potential use in photoresponsive drug delivery devices.  相似文献

The overall objective of the present work was to modulate the release behavior of drug-impregnated silica particles from almost instantaneous release to a more sustained delivery, prolonged during several hours. Triflusal was chosen as a model drug of the low biodisponibility type. The process is based in the coating with Eudragit® RL 100 polymer of aerogel-like silica particles. Materials were processed in compressed CO2 by using the batch and semicontinuous antisolvent coating methods. Triflusal release from Eudragit-coated aerogel particles was compared with the dissolution profiles recorded for pristine triflusal and for triflusal impregnated into polymer or non-coated aerogel particles. The release profiles were determined by high-performance liquid chromatography. Eudragit-coated materials presented an intermediate drug-release rate between this obtained for the infused polymer and that of the impregnated aerogel particles. Diffusion-governed mechanisms were found for the studied aerogel-like systems after fitting the release data to both Korsmeyer-Peppas and Baker-Lonsdale equations. The major advantage of the compressed CO2 antisolvent approach was the ability to physically coat very fine particles (<100 nm). A series of semi-interpenetrating, polymer network (semi-IPN), hydrogel beads, composed of calcium alginate (Ca-alginate) and poly(N-isopropylacrylamide) (PNIPAAM), were prepared for a pH/temperature-sensitive drug delivery study. The equilibrium swelling showed the independent pH- and thermo- responsive nature of the developed materials. At pH=2.1, the release amount of indomethacin incorporated into these beads was about 10% within 400 min, while this value approached to 95% at pH=7.4. The release rate of the drug was higher at 37 degrees C than that at 25 degrees C and increased slightly with increasing PNIPAAM content. These results suggest that the Ca-alginate/PNIPAAM beads have the potential to be used as an effective pH/temperature sustainable delivery system of bioactive agents. Thin polymer films with the ability of both drug release and protein adsorption resistance were formed on silicon substrates and silica particles. The films were made of a block copolymer of poly(N-isopropylacrylamide) (p(AAm)) that can load and release drugs and poly(2-methoxyethyl methacrylate) (p(MEMA)) that can suppress protein adsorption. Aspirin and bovine serum albumin were respectively used as model substances for testing the abilities of the films to load and release drugs and to suppress protein adsorption. The films were immersed in a phosphate buffer saline (pH 7.4) for 100 days to evaluate their water resistance. The experimental results showed that the films have both drug release and protein adsorption resistance and are highly stable against PBS.  相似文献

The interplay of guest encapsulation and release mechanisms in nanoscale metal–organic vehicles and its effect on the drug‐delivery kinetics of these materials were investigated through a new multidisciplinary approach. Two rationally‐designed molecular guests were synthesized, which consist of a red‐fluorescent benzophenoxazine dye covalently tethered to a coordinating catechol group and a protected, non‐coordinating catechol moiety. This allowed loading of the guests into compositionally and structurally equivalent coordination polymer particles through distinct encapsulation mechanisms: coordination and mechanical entrapment. The two types of particles delivered their fluorescent cargo with remarkably different kinetic profiles, which could be satisfactorily modeled considering degradation‐ and diffusion‐controlled release processes. This demonstrates that careful selection of the method of guest incorporation into coordination polymer nanoparticles allows selective tuning of the rate of drug delivery from these materials and, therefore, of the time window of action of the encapsulated therapeutic agents.  相似文献

The aim of this study was to synthesize and characterize a novel biocompatible polymeric membrane system and demonstrate its potential use in various biomedical applications. Synthetic hydrogels based on poly(hydroxyethyl methacrylate), poly(HEMA), have been widely studied and used in biomedical fields. A novel copolymer hydrogel was prepared in the membrane form using 2-hydroxyethyl methacrylate monomer (HEMA) and a macromonomer p-vinylbenzyl-poly(ethylene oxide) (V-PEO) via photoinitiated polymerization. A series of poly(HEMA/V-PEO) copolymer membranes with different compositions was prepared. The membranes were characterized using infrared, thermal and SEM analysis. The thermal stabilities of the copolymer membranes were found to be lowered by an increase in the ratio of macromonomer (V-PEO) in the membrane structure. Because of the incorporation of PEO segments, the copolymers exhibited significantly higher hydrophilic surface properties than pure poly(HEMA), as demonstrated by contact angle measurements. Equilibrium swelling studies were conducted to investigate the swelling behavior of the membranes. The equilibrium water uptake was reached in about 4 h. Moreover, the blood protein adsorption and platelet adhesion were significantly reduced on the surface of the PEO containing copolymer membranes compared to control pure poly(HEMA). Drug release experiments were performed in a continuous release system using model drug (vancomycin) loaded copoly(HEMA/V-PEO) membranes. A specific poly(HEMA/V-PEO) membrane formulation possessing the highest PEO content (with a HEMA:V-PEO (mmol:mmol) feed ratio of 112:1 and loaded with 40 mg antibiotic/g polymer) released about 81% of the total loaded drug in 24 h at pH 7.4. This membrane composition provided the best results and can be considered as a potential candidate for a transdermal antibiotic carrier and various biomedical and biotechnological applications.  相似文献

The functionality of a new class of monolithic systems for the controlled release of drugs is discussed. The systems consist of uniformly dispersed particles of osmotically active therapeutic agents (drugs) in biocompatible polymeric matrices. The drug particles are encapsulated by polymers to form a multiplicity of microcapsules throughout the matrix. These osmotic film systems display zero-order drug delivery kinetics. The principal energy source governing the release of agents is osmotic in nature. When such a film is placed in an aqueous infinite sink, the film imbibes water into the outermost layer of the dispersion at a rate dictated by permeability of the polymer. Water transport into the film continues until volumetric rupture of the drug-containing capsules occurs, after which time saturated drug solution is pumped through channels created by the rupture. This process repeats itself in a serial fashion until the system is exhausted of agent. Due to the osmotic functionality of these systems, reduction of the thermodynamic activity of water outside the system can proportionally reduce the release of agent. In this paper the effects of varying drug particle size, osmotic pressure gradients, system area, drug type, polymer type, and temperature upon the drug release kinetics are presented. Application of this new technology has allowed the fabrication of several useful drug therapeutic systems.  相似文献

Composite gels based on polyacrylamide and poly(N-isopropyl acrylamide) with incorporated sodium bentonite particles are synthesized. It is shown that the presence of hydrophobic isopropyl groups in a polymer molecule promotes the subsequent formation of highly ordered aggregates of clay and cetylpyridinium chloride in a gel composite. An increase in temperature results in the collapse of composite gels based on poly(N-isopropyl acrylamide); however, no marked changes in the structure of lamellar aggregates of clay and surfactant are observed. It is revealed that the gel can stabilize lamellar structures formed in organoclay suspension prior to the incorporation into swollen polymer network.  相似文献

The present investigation describes a novel method for preparing beads based on crosslinked chitosan with glutaraldehyde interpenetrating glycine polymer network. Four type of beads, viz., CHI1 (composed of chitosan, glycine and glutaraldehyde); CHI2 (composed of chitosan and glutaraldehyde); CHI3 (composed of chitosan and glycine) and CHI4 (only chitosan) were prepared and their release characteristics were studied using thyamine hydrochloride (Thy-HCl) as a model drug. Structural changes during swelling of CHI1 beads in solutions of different pH were studied using IR and UV spectroscopy.  相似文献

Drug forms based polymer carriers of prolong action were created for toxicologic effect of drug to be reduced in spite of long treatment of diseases. In present work a number of synthesis and natural polymers have been studied as carriers of antituberculous drugs for controlled delivery application. Following as drugs as isoniazid and ethionamide were incorporated into polymeric matrix (segmented polyurethanes, polyvinyl alcohol) and chemically bound with the polymer chain by covalent or electrostatic forces (aldehyde- and carboxymethylderivatives of polysaccharides). Biodegradation of polymeric systems and the release of drugs were studied by various physico-chemical methods. It was shown that the drug release depends of method of the immobilization, type of the drug/polymer bonding, drug loading. The bacteriostatic activity of obtained systems was determined. The possibility of tuberculosis treatment was proved in experiments of animals.  相似文献

合成了三种亲疏水性不同的温度及pH敏感的PAAc/P(NIPAAm-co-BMA)、PAAc/PNIPAAm和PAAc/P(NIPAAm-co-AAm)互穿网络(IPN)水凝胶,以水杨酸钠和水杨酸为模型药物,研究了温度、pH值及药物和凝胶的亲疏水性相互作用对模型药物释药性能的影响。研究结果表明,随着凝胶亲水性的增强,水杨酸钠的载药率提高,释药率也越大;相反疏水性增强也有利于提高水杨酸的载药率;IPN凝胶在水中的释药过程属于溶胀支配型释放,药物释放率随凝胶的亲水性增强而增强,同时,载药凝胶在45℃水中的释药率大于25℃时的释药率。在25℃时,水杨酸在pH=2.2的缓冲溶液中几乎不释放,而在pH=7.4的缓冲溶液中能以较快的速率释放。  相似文献
