Abstract

The phytochemical examination of the polar constituents of Sambucus ebulus L. leaves led to the identification of patrinoside (1) and of a new diglycoside iridoid, patrinoside-aglycone-11-O-[β-D-glucopyranosyl-(1→6)-2′-deoxy-β-D-glucopyranoside] (trivially named as sambuloside) (2). Summary Some components of the lipidic fraction of the drupes of Sambucus ebulus were separated and studes using TLC and GC. Among the fatty acids, unsaturated ones were present in very hihg percentage (93.4%). The sterols were identified as campesterol (9.5%), stigmasterol (3.1%), -sitosterol (84.6%), 7-stigmastenol (2.7%). The C18, C20, C22, C24, C28 aliphatic alcohols were present. Among the triterpenic alcohols cycloartanol, 24-methylene-cycloartanol and - and -amyrin were detected (the last two were very abundant).  相似文献

Chemical investigation of Chinese folk medicine Sambucus williamsii Hance has resulted in the isolation and characterisation of seventeen compounds from the n-BuOH extract of its fruits, including two new phenylethanoid glycosides and fifteen known compounds. Structures of new compounds were elucidated primarily on the basis of their extensive spectroscopic data including 2D NMR. In addition, the n-BuOH extract from the fruits of S. williamsii was found to show a protective effect on D-galactosamine (D-GalN)-induced cytotoxicity in primary cultured mouse hepatocytes. So the hepatoprotective effects of principal constituents from it were tested by MTT assays. The results showed that Compounds 13, 16 and 17 displayed hepatoprotective effects.  相似文献

An LC-MS/MS method was developed for simultaneous determination of voriconazole and its main metabolite voriconazole N-oxide in human plasma. Plasma pretreatment was simple one-step protein precipitation with acetonitrile, and separation was achieved on a Hypersil GOLD aQ column. A triple-quadrupole tandem mass spectrometer with electrospray ionization source was operated by multiple reaction monitoring (MRM) in positive ion mode, and the precursor–product ion pairs used for MRM were m/z 350.1→281.0, 365.8→224.1, and 531.3→489.2 for voriconazole, voriconazole N-oxide, and internal standard, respectively. The total analytical run time was 6?min. The proposed method was linear over the range 0.001–1?µg/mL for voriconazole and voriconazole N-oxide. The lower limit of quantification was 1?ng/mL for both of them. The specificity, accuracy, precision, recovery, matrix effect, stability as well as dilution integrity of this method were within acceptable limits during validation period. This method was successfully applied to therapeutic drug monitoring of voriconazole and to evaluate its metabolite profile.  相似文献

A hydrophilic interaction high performance liquid chromatography-tandem mass spectrometric method has been developed and validated for simultaneous quantification of dacarbazine (DTIC) and its terminal metabolite, 5-amino-4-imidazole-carboxamide (AIC) in human plasma. The plasma samples are first extracted by a C8 + SCX mixed-mode 96-well plate to extend the extraction stability of DTIC and AIC. The extracted residues are further cleaned by a primary and secondary amine (PSA) adsorbent for minimization of matrix effect. Analyses are done on an Amide-80 HPLC column coupled to a tandem mass spectrometer fitted with an atmospheric pressure turbo ion spray ionization interface in the positive-ion mode. Both DTIC and AIC have reproducible retention times on the Amide-80 HPLC column. This type of column not only has an excellent column life (over 4000 injections), but also has zero carryover effect. The injection volume should be limited at 10 μL or less to avoid the peak splitting. The validated concentration ranges are from 0.5 to 500 ng/mL for DTIC and from 2.0 to 500 ng/mL for AIC. The validated method has been successfully applied to determine the pharmacokinetic profiles for human patients receiving DTIC infusions.  相似文献

Digoxin, a commonly prescribed cardiac glycoside with a narrow therapeutic window, is routinely used in pharmacokinetic studies to assess the in vivo activity of the drug efflux pump P-glycoprotein. To minimize adverse events, a sub-therapeutic dose of digoxin is usually administered, producing low plasma concentrations requiring a sensitive detection technique. Commonly available immunoassay techniques do not provide the required sensitivity to measure these low plasma concentrations and are potentially non-specific in certain subject populations. Previously published mass spectrometric techniques require either large plasma volumes or a tandem mass spectrometer. To overcome these challenges we have developed a sensitive and specific LC-MS method for the quantification of digoxin in small volumes of human plasma and urine. Plasma (1 mL) was extracted with methyl t-butyl ether under basic conditions followed by LC-MS detection of the sodium adducts of digoxin (803.4 m/z) and digitoxin (787.4 m/z, internal standard). Linearity and accuracy were demonstrated across a wide range of digoxin plasma concentration (0.05-1.5 ng/mL). This specific, sensitive, validated digoxin LC-MS assay can be used to quantify sub-therapeutic digoxin plasma concentrations in men and women (pregnant and non-pregnant).  相似文献

Sambucus nigra flowers (elderflower) have been widely used in traditional medicine for the relief of early symptoms of common cold. Its chemical composition mainly consists of polyphenolic compounds such as flavonoids, hydroxycinnamic acids, and triterpenes. Although the antioxidant properties of polyphenols are well known, the aim of this study is to assess the antioxidant and protective potentials of Sambucus nigra flowers in the human neuroblastoma (SH-SY5Y) cell line using different in vitro approaches. The antioxidant capacity is first evaluated by the oxygen radical absorbance capacity (ORAC) and the free radical scavenging activity (DPPH) methods. Cell viability is assessed by the crystal violet method; furthermore, the intracellular ROS formation (DCFH-DA method) is determined, together with the effect on the cell antioxidant defenses: reduced glutathione (GSH) and antioxidant enzyme activities (GPx, GR). On the other hand, mTORC1 hyperactivation and autophagy blockage have been associated with an increase in the formation of protein aggregates, this promoting the transference and expansion of neurodegenerative diseases. Then, the ability of Sambucus nigra flowers in the regulation of mTORC1 signaling activity and the reduction in oxidative stress through the activation of autophagy/mitophagy flux is also examined. In this regard, search for different molecules with a potential inhibitory effect on mTORC1 activation could have multiple positive effects either in the molecular pathogenic events and/or in the progression of several diseases including neurodegenerative ones.  相似文献

The use of quick, easy, cheap, effective, rugged and safe method followed by liquid chromatography tandem mass spectrometry (LC-MS/MS) was found to be the best combination for multiresidue determination of eight acidic herbicides in fruits and vegetables in terms of high recovery, short time of analysis, low cost and safety. Recent few articles were published for determination of different classes of acidic herbicides in single multiresidue method. In the present study, mass spectrophotometric conditions were individually optimised for eight acidic herbicides, namely 2,4-dichlorophenoxyacetic acid, bentazone, bromoxynil, fluazifop, fluroxypyr, imazethapyr, ioxynil and triclopyr to achieve maximum sensitivity and selectivity in multiple reaction monitoring (MRM) mode allowing simultaneous identification and quantification in a single run. Identity confirmation and quantitation were attained by using negative electrospray ionisation LC-MS/MS (ESI?) in MRM mode. Due to LC-MS/MS signal suppression, determination of pesticide residues was based on matrix-matched standard calculations. Most of the evaluated compounds showed a recovery ranging from 81% to 113% with relative standard deviations less than 16 % indicating acceptable precision. The precision and accuracy of the method were determined from recovery experiments on six replicates of spiked blank strawberry and green beans samples at 0.01, 0.05 and 0.1 mg/kg. The developed assay was linear over concentration range of 0.01–0.5 µg/mL, with correlation coefficient greater than 0.99 at the limit of quantitation 0.01 µg/mL. The proposed assay was successfully applied for the analysis of the studied acidic herbicides residues in two proficiency test samples. This wide scope assay protocol is applicable for monitoring acidic herbicides residues in fruits and vegetables by national regulatory authorities and accredited labs in order to help ensuring the safety of such widely used food products.  相似文献

A new, rapid, sensitive and specific LC‐MS/MS method has been developed and validated for the simultaneous quantification of tenofovir and lamivudine in human plasma using abacavir as an internal standard. An API‐4000 LC‐MS/MS with electrospray ionization was operated in multiple‐reaction monitoring mode for the analysis. The analytes were extracted from plasma by solid‐phase extraction technique using an Oasis HLB cartridge. The reconstituted samples were chromatographed on a Chromolith ROD speed C18 column using a mixture of 0.1% formic acid in water and acetonitrile (90:10 v/v) at a flow‐rate of 1 mL/min. The method was validated as per the FDA guidelines. The calibration curves were found to be linear in the range of 5–600 ng/mL for tenofovir and 25– 4000 ng/mL for lamivudine. The intra‐ and inter‐day precision and accuracy results were well within the acceptable limits. A run time of 2.8 min consumed for each sample made it possible to analyze more samples per day. A novel generic reverse phase high performance liquid chromatography (RP‐HPLC) method is developed and validated for simultaneous determination of seven pharmaceutically active ingredients, namely, acetaminophen, dextromethorphan, doxylamine, phenylephrine, guaifenesin, caffeine and aspirin. All seven ingredients were quantified in soft gel, syrup and tablet formulations of the over‐the‐counter US‐marketed products, as per the guidelines of the International Conference on Harmonization. The separation was achieved in a 16 min run time on an Agilent Zorbax Phenyl column using a gradient method with two mobile phases. Mobile phase A was 0.15% trifluoro acetic acid in purified water and while mobile phase B was a mixture of acetonitrile and methanol (750:250 v/v) with 0.02% trifluoro acetic acid. The flow rate was 1.0 mL min?1 and injection volume was 10 μL. Detection was performed at 280 nm using a photodiode array detector. As part of the method validation, specificity, linearity, precision and recovery parameters were verified. The concentration and area relationships were linear (R2 > 0.999), over the concentration ranges 20–120 μg mL?1 for acetaminophen, 75–450 μg mL?1 for dextromethorphan, 31.25–187.5 μg mL?1 for doxylamine, 25–150 μg mL?1 for phenylephrine, 25–150 μg mL?1 for aspirin, 6.5–39 μg mL?1 for caffeine and 12–72 μg mL?1 for guaifenesin. The relative standard deviations for precision and intermediate precision were <1.5%. The proposed RP‐HPLC generic method is applicable for routine analysis of cold and cough over‐the‐counter products.  相似文献

Knowledge of the presence of micropollutants such as pharmaceuticals, in coastal areas, is very limited; therefore, the main objective of this study was to optimize and validate a new analytical method for the quantitative analysis of 13 multiclass pharmaceuticals in seawater. Target compounds included antibiotics, non-steroidal anti-inflammatory drugs, β-blockers, lipid regulators and one psychiatric drug. A combination of solid-phase extraction and liquid chromatography coupled with multiple mass spectrometry enabled their detection at the low nanogram per litre level. The limits of quantification varied between 1 and 50 ng L-1, for most components the linearities were more than 0.99 and the recoveries obtained in seawater (95–108%) were satisfactory. This method was applied to seawater and estuarine water samples collected in the Belgian coastal zone, to assess the prevalence of common pharmaceuticals in this marine environment. Seven pharmaceuticals, including compounds of which the presence in marine environments had not been reported earlier, were detected, with salicylic acid and carbamazepine being the most abundant, in concentrations up to 855 ng L-1.  相似文献

Herein, a novel, rapid, reliable, simple method validation and simultaneous quantification of 11 bioactive compounds (mostly xanthones) have been described. International Conference on Harmonization guidelines were used for the analytical method validation. Good linearity, repeatability, intra- and inter-day precision, accuracy and reliability were well-illuminated in the method validation procedure. The calibration curves showed a good linear relationship (r > 0.999) within test range. Precision was evaluated by intra- and inter-day tests with relative standard deviation <2.79% and accuracy validation recovery of 74.16%–91.84%. On quantification study, the validated method described the high content of bioactive xanthone derivatives, including 1-hydroxy-3, 5-dimethoxyxanthone ( 7 ), 2-(allyloxy)-8-hydroxy-1, 6-dimethoxyxanthone ( 6 ) 1, 7, 8-trihydroxy-3-methoxyxanthone ( 9 ) and Coxanthone E ( 5 ) in Codonopsis ovata, which is advantageous given the numerous pharmacological and biological effects associated with these compounds, which mostly exhibit anti-cancerous, antioxidant, anti-inflammatory, anti-mutagenic and anti-obesity effects. The bulk abundance of these compounds can also be used for further modification to produce better lead molecules for drug discovery with low toxicity and high potency. The proposed method makes it possible to simultaneously determine all bioactive compounds in one run and can be extended to marker-based standardization of herbal formulations in medicinal and pharmaceutical industries.  相似文献

We have developed a simple, sensitive, and robust liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) method to determine free 3-nitrotyrosine concentrations in human plasma of patients receiving on-pump coronary artery bypass grafting surgery. A one-step solid-phase extraction protocol was optimized to enrich the analyte at low nanomolar concentrations. The processed samples were analyzed by LC-MS/MS with a 2.1 × 100 mm Kinetex PFP column and a triple quadrupole mass spectrometer. The method was validated for 3-nitrotyrosine concentrations close to real patient plasma levels. The relative standard deviations or relative errors of the intraday and interday determinations were all within 10%. Limit of detection and limit of quantitation were determined to be 0.034 nM and 0.112 nM, respectively, while lower limit of quantitation was below 0.625 nM. No deterioration of the column performance was noticed after running a large number of patient samples. The results showed that the 3-nitrotyrosine concentrations in coronary sinus plasma samples were elevated after cardiopulmonary bypass (CPB) procedure. The pre-CPB and post-CPB concentrations of 3-nitrotyrosine in patient plasmas were 1.494 ± 0.107 nM and 2.167 ± 0.177 nM (mean ± SEM), respectively. Application of this method to more patients in clinical studies may help validate 3-nitrotyrosine as a meaningful biomarker for nitrosative stress and link patient characteristics, clinical outcomes, and cardioprotective treatments to endogenous nitrosative stress levels.  相似文献

自制了一套液相色谱-质谱联用接口,接口包含大气压接口、离子源及连接高效液相色谱进样器和质谱采集软件的电子通讯接口。成功地将赛默飞世尔高效液相色谱戴安U3000与团队自主研制的APITOFMS10000相连。实验结果表明,自行搭建的液质联用仪有很好的稳定性(RSD=5.6%),线性相关系数(r2)为0.995 8,线性范围为2~2 000 pg,定量下限为2 pg,检出限为0.5 pg。可明显检出吡罗昔康、茶碱和泼尼松3种标准药品。实验数据表明自行研发的液相色谱-质谱联用接口满足应用要求,搭建的液相色谱-质谱联用仪在生物化学、医药分析、食品安全、环境检测等领域具有应用潜力。  相似文献

以石墨烯量子点为荧光探针,基于二氧化锰纳米片与石墨烯量子点之间发生有效的荧光共振能量转移,构建了一种茶叶中茶多酚含量的荧光检测新方法。MnO2纳米片使石墨烯量子点的荧光淬灭,而茶多酚能与MnO2发生氧化还原反应,将其还原成Mn^2+,使体系荧光恢复。茶多酚含量与荧光强度增量在20~750μg/mL范围内成线性相关,线性方程为F=1.0574c-114.9,线性相关系数为R=0.9936。将该方法用于茶叶中茶多酚含量的检测,结果与国标法接近。方法有望应用于食品中其它抗氧化物质的实时检测。  相似文献

A high-performance liquid chromatography-tandem mass spectrometric method (LC-MS/MS) has been developed and validated for the determination of daunorubicin in K3EDTA rat plasma. The 100 μL plasma samples were extracted by a methanol:acetone protein precipitation step in the presence of additional 50 μL of 70% (w/v) zinc sulfate, and subsequently analyzed by LC/MS/MS using positive turbo-ion spray ionization mode. The LC/MS/MS instrument was operated in the multiple-reaction-monitoring (MRM) mode. Doxorubicinol was better than doxorubicin as the internal standard because its recovery and absolute matrix effect data exactly matched with those for daunorubicin. In addition, HPLC gradient condition was optimized to thoroughly separate daunorubicin from the background interference. The validated concentration range was from 0.250 to 100 ng/mL. The true recoveries of daunorubicin and doxorubicinol were 93.2% and 93.6%, respectively. In addition, the ion-suppression data of daunorubicin and doxorubicinol were 78.2% and 78.4%, respectively. Absence of the relative matrix effect from six unique lots was confirmed. Results obtained from the GLP validation study demonstrated very good accuracy (95-105%) and precision (less than 10% CV).  相似文献

Artesunate (AS), a hemisuccinate derivative of artemisinin, is readily soluble in water and can easily be used in formulations for parenteral treatment of severe malaria. AS is rapidly hydrolyzed to the active metabolite dihydroartemisinin (DHA) and primarily eliminated by biliary excretion after glucuronidation. To investigate systematically the AS metabolism and pharmacokinetics, a novel liquid chromatography–tandem mass spectrometry (LC-MS/MS) method for the simultaneous quantification of AS and its metabolites DHA and DHA glucuronide (DHAG) in human plasma samples was developed. Compared to previous methods, our method includes for the first time the quantification of the glucuronide metabolite using a newly synthesized stable isotope-labeled analogue as internal standard. Sample preparation was performed with only 50 μL plasma by high-throughput solid-phase extraction in the 96-well plate format. Separation of the analytes was achieved on a Poroshell 120 EC-C18 column (50*2.1 mm, 2.7 μm, Agilent Technologies, Waldbronn, Germany). The method was validated according to FDA guidelines. Calibration curves were linear over the entire range from 1 to 2,500 nM (0.4–961.1 ng/mL), 165 to 16,500 nM (46.9–4,691.8 ng/mL), and 4 to 10,000 nM (1.8–4,604.7 ng/mL) for AS, DHA, and DHAG, respectively. Intra- and interbatch accuracy, determined as a deviation between nominal and measured values, ranged from ?5.7 to 3.5 % and from 2.7 to 5.8 %, respectively. The assay variability ranged from 1.5 to 10.9 % for intra- and interbatch approaches. All analytes showed extraction recoveries above 85 %. The method was successfully applied to plasma samples from patients under AS treatment.

Simple and efficient microwave-assisted acid hydrolysis (MAAH) of proteins was used for rapid quantification of α-aminoadipic semialdehyde (AAS) and γ-glutamic semialdehyde (GGS) as major protein oxidation markers. The precursor amino acid residues corresponding to AAS and GGS in oxidized proteins were derivatized by reductive amination with sodium cyanoborohydride (NaCNBH3) and p-aminobenzoic acid (ABA) followed by MAAH to generate the marker derivatives AAS-ABA and GGS-ABA. The quantification was performed using electrospray ionization liquid chromatography-mass spectrometry (ESI LC-MS). The important parameters for hydrolysis were optimized, which include the temperature, the reaction time, the acid concentration and volume as well as the microwave power. Compared to the conventional acid hydrolysis of 18-24 h using 6-12 M HCl at 110 °C applied commonly in the literature and also in this work, MAAH of proteins can be completed as fast as in only 2-10 min and, additionally, with a 3-5 times higher yield of the final derivatization products. Furthermore, a better agreement between the ratio of the detected derivatization products and the theoretical yields from the studied protein has also been achieved, which indicates that MAAH may serve as a more reliable method of acid hydrolysis for this purpose than that with conventional thermal heating. The MAAH method is demonstrated to be a time-saving, reproducible and efficient technique for studying AAS and GGS as protein oxidation markers using LC-MS.  相似文献

This work details a rapid analytical method using direct sample injection for the simultaneous identification/quantification of 22 drugs of abuse, including some of their major metabolites, in environmental samples. This has been developed using a hybrid triple quadrupole-linear ion trap-mass spectrometer (QqLIT). With the increasing sensitivity of today's tandem mass spectrometers, direct injection analysis of water samples has become an attractive alternative to traditional analytical protocols, which often include a preliminary pre-concentration step. What's more, this kind of analysis is in accordance with many of the main objectives of so-called green analytical chemistry, or environmentally friendly practice. The analytical performance of the LC-MS/MS method was evaluated in three different water matrices (surface water, influent and effluent wastewater). Data acquisition was carried out in selected reaction monitoring (SRM) mode under time-scheduled conditions, monitoring two SRM transitions for simultaneous identification/quantification of all target compounds in the samples. Additionally, an experiment was performed using the information-dependent acquisition (IDA) scan to carry out the identification of those analytes for which the second transition was present at a low intensity. Finally, the two methodologies developed were applied to real samples for evaluation.  相似文献

A highly sensitive and specific LC‐MS/MS method was developed for simultaneous estimation of acetyl co‐enzyme A (ACoA) and malonyl co‐enzyme A (MCoA) in surrogate matrix using n‐propionyl co‐enzyme A as an internal standard (IS). LC‐MS/MS was operated under the multiple reaction‐monitoring mode using the electrospray ionization technique. Simple acidification followed by dilution using an assay buffer process was used to extract ACoA, MCoA and IS from surrogate matrix and tissue samples. The total run time was 3 min and the elution of both analytes (ACoA, MCoA) and IS occurred at 1.28 min; this was achieved with a mobile phase consisting of 5 mM ammonium formate (pH 7.5)–acetonitrile (30:70, v/v) delivered at a flow rate of 1 mL/min on a monolithic RP‐18e column. A linear response function was established for the range of concentrations 1.09–2187 and 1.09–2193 ng/mL for ACoA and MCoA, respectively. The intra‐ and inter‐day precision values for ACoA and MCoA met the acceptance as per FDA guidelines. ACoA and MCoA were stable in a battery of stability studies viz. bench‐top, auto‐sampler and long‐term. The developed assay was used to quantitate ACoA and MCoA levels in various tissues of rat.
