Potassium carbonate as inorganic base catalyst was used to develop an efficient synthetic procedure for the preparation of thieno[2,3-d]pyrimidine derivatives. The molecular structure of the newly synthesized compounds were confirmed by NMR spectral data.  相似文献

In a one-pot synthesis, 1′-methyl-2,3″-dioxo-5″-aryl-1,2,5a″,7″,8″,9a″-hexahydro-5″H,6″H-dispiro[indole-3,2′-pyrrolidine-3′,2″-pyrano[2,3-d][1,3]thiazolo[3,2-a]pyrimidine]-4′-carboxylic acid methyl ester was prepared via the sequential reaction of 4-aryl-octahydro-pyrano[2,3-d]pyrimidine-2-thione, dimethyl acetylenedicarboxylate (DMAD), and a mixture of isatin and sarcosine. All the novel spiro compounds, in moderate yields, were characterized thoroughly by infrared, NMR, mass spectromentry, and elemental analysis together with x-ray crystallographic analysis.  相似文献

Oxo-and thioxopyrimidines 4a–i were synthesized using the Biginelli three-component cyclocondensation reaction of an appropriate β-diketone, arylaldehyde, and (thio)urea under microwave irradiation. Yields of products following recrystallization from ethanol were of the order of 65–90%. 1H and 13C NMR spectroscopy and elemental analysis were used for structural assignment.  相似文献

Intermolecular cyclization of pyrimidinethiol 1 with ethyl chloroacetate and chloroacetonitrile furnished thieno[2,3-d]pyrimidines 2a,b . Hydrazinolysis of o-aminoester 2a gave acid hydrazide 3, which was cyclized with various electrophilic reagents including formic acid, triethyl orthoformate, acetic anhydride, p-chlorobenzaldehyde then triethyl orthoformate, carbon disulfide, and acetylacetone affording thienopyrimidine derivatives 4 to 10 . Another thienopyrimidine series could be obtained via treatment of o-aminocarbonitrile 2b with a variety of reagents giving derivatives 11 to 17 . The fluorescent measurements for a group of the synthesized compounds at room temperature demonstrated high fluorescent properties.  相似文献

Abstract

Condensation of ethyl-5-amino-2,4-diphenylthieno[2,3-d]pyrimidine-6-carboxylate (3a) with 2,5-dimethoxy tetrahydrofurane in acetic acid gives the corresponding 5-pyrrolyl derivative4, which in turn could be easily reacted with hydrazine hydrate in ethanol yielding the carbohydrazide derivative5. Reaction of5 with aromatic aldehydes, acetylacetone, carbon disulfide or phenylisothiocyanate gave pyrrolyl derivatives6,7,8,9 respectively. On the other hand, condensation of 5-(1-pyrrolyl)-6-acetyl-2,4-diphenylthieno[2,3-d]pyrimidine11 with benzaldehyde afforded the corresponding chalcone12, which on treatment with hydrazine hydrate, phenyl hydrazine, or thiourea gave the pyrazolinyl derivatives13,14 and pyrimidinyl derivative15, respectively. Furthermore some new pyrimidothienopyrimidne16,17a–d,19,20a–c were obtained using 5-amino-carboxamide3c as starting material.  相似文献

The alkylation of 4-cyano-5,6-dimethylpyridazin-3(2H)-thione 3 with some halo compounds gave the S-alkylated products 4a–c , which upon treatment with ethanolic sodium ethoxide afforded the cyclized thienopyridazines 5a–c as products. Pyridazothienotriazines 6a–c were prepared by the treatment of compounds 5a–c with nitrous acid, while their reaction with triethyl orthoformate and with carbon disulfide gave the corresponding pyrimidothienopyridazines 7a–c , and 8a–c , respectively. S-alkylated products 9a–o were obtained by the reaction of 8a–c with some halo compounds.  相似文献

Various derivatives of thiophene, thiazoline and thienopyrimidine have been synthesized via the reaction of active methylene reagents (la-j) with benzoyl isothiocyanate in the presence of potassium hydroxide, followed by the subsequent treatment of the potassium salt intermediates 2a-j with phenacyl bromide or ethyl chloroacetate. The stability of the formed adducts 3 and 12 or their selective cyclization depends on both the nature of the active methylene reagent and the α-halocarbonyl compounds.  相似文献

In this study, Co MFA, Co MSIA and HQSAR techniques were used to study the important characteristic activities of thieno [2,3-d] pyrimidine derivatives for effective antitumor activity. The q~2 value of cross validation of CoMFA model was 0.621, and r~2 value of non-cross validation was 0.959. The best cross validation q~2 value of CoMSIA model was 0.522, while the r~2 value of non-cross validation was 0.961. The most effective HQSAR model was obtained by taking atoms and bonds as fragments: the q~2 value of cross validation is 0.535, the r~2 value of non-cross validation is 0.871, the standard error of prediction is 0.488, and the optimal hologram length is 199. The statistical parameters from the model show that the data fit well and have high prediction ability. In addition, molecular docking is used to study the binding requirements between ligands and receptor proteins, including several hydrogen bonds between thieno [2,3-d] pyrimidine and active site residues. The results obtained from these QSAR modeling studies can be used to design promising anticancer drugs.  相似文献

A series of compounds 5‐amino‐2‐ethylmercapto‐4‐phenyl‐6‐subistitutedthieno[2,3‐d]pyrimidines ( 8a–d ), 4‐chloro‐7‐ethylmercapto‐9‐phenylpyrimido[5′,4′:4,5]thieno[3,2‐d]triazine ( 9 ), and 2‐ethylmercapto‐8‐oxo‐4‐phenyl‐7‐(4‐chlorophenyl)pyrimido [4′,5′:4,5]thieno[2,3‐d]pyrimidine ( 10 ) were synthesized and their structures were confirmed by 1H NMR , 13C NMR, and MS . All compounds were evaluated for their IC50 values against three cancer cell lines (MCF ‐7, HUH ‐7 and BHK ) and WISH cells. The IC50 of compound ( 8d ) was calculated for each cell line. Interestingly, the IC50 for the normal human amnion WISH cell line was much higher (723 µg/mL) than those found for the tumor cell lines BHK (17 µg/mL), HUH ‐7 (5.8 µg/mL), and MCF ‐7 (8.3 µg/mL). The proliferation inhibition of normal (WISH ) and tumor (BHK , HUH ‐7, and MCF ‐7) cells by compound ( 8d ) was investigated using MTT assay, and the IC50 was calculated after 48 h of treatment for each cell line.  相似文献

Several pyrazolo[3,4-b]pyridine (3,4) and pyrazolo-[3,4-d]pyrimidine (5-13) derivatives were prepared using 5-amino-l-(5-ethyl-5H-1,2,4-triazino[5,6-b]-indol-3-yl)-lH-pyrazole-4-carbonitrile (2) . A facile one‐pot synthesis of new pyrimido[1,6‐a ]thieno[2,3‐d ]pyrimidin‐4‐ones 3a , 3b is reported via base‐catalyzed reaction of 2‐amino‐3‐cyanothiophenes 1a , 1b with 2,4‐dichloro‐5‐bromo‐6‐methylpyrimidine 2 . Subsequent treatment of the products with several amines under a mild condition gave a host of the new amino derivatives 4a , 4b , 4c , 4d , 4e , 4f , 4g , 4h , 4i , 4j , 4k , 4l . The target compounds were evaluated for antiproliferative activity by an 3‐(4, 5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyl tetrazolium assay on human breast cancer (MCF‐7) and mouse fibroblast nonmalignant (L929) cell lines. The activation was structural, concentration, and time dependent. Doses inducing 50% cell‐growth inhibition (IC50) for compounds 3a , 3b and 4a , 4b , 4c , 4d , 4e , 4f , 4g , 4h , 4i , 4j , 4k , 4l against MCF‐7 cells were explored. Compounds 4f and 4l as the two most potent analogs induced a sub‐G1 peak in the flow‐cytometry histogram of treated cells, compared with control, indicating that apoptotic cell death is involved in compound 4f ‐induced and compound 4l ‐induced toxicity. Compounds 4f and 4l exerted cytotoxic and proapototic effects on MCF‐7 cell line.  相似文献

用邻氨基噻吩腈与不同结构的二羰基化合物反应,合成了新的多取代噻吩并吡啶类衍生物4-氨基-5-取代羰基噻吩并[2,3-b]吡啶-6-酮。用元素分析、红外光谱、核磁共振氢谱和质谱等测试技术对所合成化合物的结构进行了表征。初步生物活性测试表明,该类化合物有较好的杀菌活性和一定的除草活性。  相似文献

The Knoevenagel reactions of malononitrile with acetophenone or 4‐substituted acetophenons were carried to give the corresponding 2‐(1‐aryle thylidene)malononitriles, which was further cyclized with sulfur using NaHCO3 as catalysts to generate 2‐amino‐5‐arylthiophene‐3‐carbonitrile 2 . The intermediate enamines 3 were prepared by refluxing of 2 with 5‐substituted‐1,3‐cyclohexanedione using p‐toluenesulfonic acid as catalyst. The title compounds 4‐amino‐3‐aryl ‐7‐substituted‐7,8‐dihydrothieno[2,3‐b]quinolin‐5(6H)‐one were synthesized by cyclization of 3 in the presence of K2CO3 and Cu2Cl2. The structures of all compounds were characterized by elemental analysis, IR, MS, and 1H‐NMR spectra.  相似文献
