Cyclopropanone derivatives have long been considered unsustainable synthetic intermediates because of their extreme strain and kinetic instability. Reported here is the enantioselective synthesis of 1-sulfonylcyclopropanols, as stable yet powerful equivalents of the corresponding cyclopropanone derivatives, by α-hydroxylation of sulfonylcyclopropanes using a bis(silyl) peroxide as the electrophilic oxygen source. This work constitutes the first general approach to enantioenriched cyclopropanone derivatives. Both the electronic and steric nature of the sulfonyl moiety, which serves as a base-labile protecting group and confers crystallinity to these cyclopropanone precursors, were found to have a crucial impact on the rate of equilibration to the corresponding cyclopropanone. The utility of these cyclopropanone surrogates is demonstrated in a mild and stereospecific formal [3+1] cycloaddition with simple hydroxylamines, leading to the efficient formation of chiral β-lactam derivatives.  相似文献

Abstract

The cationic polymerization of β-phenylvinyl alkyl ethers (alkyl: methyl, ethyl, n-propyl, and n-butyl) was examined in toluene and in methylene chloride with boron fluoride etherate and with stannic chloride at ?78°C. Despite the fact that these monomers have bulky substituents on the α –and β-carbons, they could be homopoly-merized easily even in nonpolar sovent at ?78°C. The general features of β-phenylvinyl alkyl ethers in homopolymerization and co-polymerization with n-butyl vinyl ether suggested that β-phenylvinyl alkyl ethers behaved as derivatives of vinyl ethers, not as derivatives of styrenes. The polymerization products were white powders having a high softening point.  相似文献

The preparation of a stabilized monofluorobenzyl carbanion by means of a remote homochiral sulfinyl group and its completely stereoselective reactions with N-p-tolylsulfinylimines are described. The use of these reactions followed by the simultaneous removal of both chiral auxiliaries with tBuLi, which occurs without epimerization at the benzylic position, provides the quickest entry to enantiomerically pure β-fluorinated β-phenylethylamines.  相似文献

Synthesis of mono- and bis(1,1′-bi-2-naphthocrown)ethers containing bis(2,6-methylene)anisyl subunit in the crown ring were developed. These chiral macrocycles are suitable precursors to introduce a chromogenic function, as exemplified by two novel crowned azophenol chromoionophores. Their coloration process induced by various achiral and chiral amines was studied by UV–vis spectrophotometry.  相似文献

The reactions of chiral four-coordinated nickel(Ⅱ) complex [Ni(RR-L)](ClO4)2/[Ni(SS- L)](ClO4)2 with β-dl-phenylalanine in acetonitrile/water gave a six-coordinated enantiomer of [Ni(RR-L)(β-d-HPhe)](ClO4)2(1) and [Ni(SS-L)(β-l-HPhe)](ClO4)2(2), respectively(L = 5,5,7, 12,12,14-hexamethyl-1,4,8,11-tetraazacyclotetradecane, HPhe = phenylalanine). Single-crystal X-ray diffraction analyses revealed that the nickel(Ⅱ) atom displays a distorted octahedral coordination geometry by coordinating with four nitrogen atoms of L in a folded configuration, and two carboxylate oxygen atoms of β-d/l-HPhe in cis-position in both complexes. The monomers of [Ni(RR-L)(β-d-HPhe)]2+ and [Ni(SS-L)(β-l-HPhe)]2+ are connected through intermolecular hydrogen bonds to generate one-dimensional zigzag chains, respectively. The homochiral natures of 1 and 2 are confirmed by the results of CD spectroscopy.  相似文献

Direct liquid chromatographic methods were developed to investigate the enantioseparation of 19 β-lactams on three cyclodextrin-bonded chiral stationary phases: permethyl-β-cyclodextrin, β-cyclodextrin and R,S-hydroxypropyl-β-cyclodextrin, prepared by a novel synthetic route. 17 of the 19 β-lactam stereoisomers were partially or baseline-separated on at least one of the tested chiral stationary phases. The influence of the structures of the β-lactams (the positions and types of the substituents, the size of the attached rings) on the enantiomer separation is discussed. The permethylated β-cyclodextrin selector proved to be the most effective one for the tested analytes.  相似文献

Axially chiral diaryl ethers, a distinguished class of atropisomers possessing unique dual C−O axis, hold immense potential for diverse research domains. In contrast to the catalytic enantioselective synthesis of conventional single axis bearing atropisomers, the atroposelective synthesis of axially chiral ethers containing flexible C−O axis remains a significant challenge. Herein, we demonstrate the first N-heterocyclic carbene (NHC)-catalyzed synthesis of axially chiral diaryl ethers via atroposelective esterification of dialdehyde-containing diaryl ethers. Mechanistically, the reaction proceeds via NHC-catalyzed desymmetrization strategy to afford the corresponding axially chiral diaryl ether atropisomers in good yields and high enantioselectivities under mild conditions. The derivatization of the synthesized product expands the utility of present strategy via access to a library of C−O axially chiral compounds. The temperature dependency and preliminary investigations on the racemization barrier of C−O bonds are also presented.  相似文献

The benzofuran skeleton is commonly found in a wide variety of natural products that display a wide range of biological activities. For this reason, benzofuran has a high potential for use as a scaffold in both chemical and pharmaceutical syntheses of natural products. This work describes the development of a novel asymmetric synthetic method of benzofuran-derived β-amino esters via K2CO3-promoted domino reactions of ortho-hydroxyl aromatic N-tert-butylsulfinyl imines and diethyl bromomalonate. A small library of target compounds was prepared under optimized reaction conditions in moderate to good yields with high distereoselective and enantioselective properties. The newly generated chiral carbon center was of an (S)-configuration as determined by x-ray diffraction.  相似文献

Chirality is ubiquitous in nature, and homochirality is manifested in many biomolecules. Although β-double helices are rare in peptides and proteins, they consist of alternating L- and D-amino acids. No peptide double helices with homochiral amino acids have been observed. Here, we report chiral β-double helices constructed from γ-peptides consisting of alternating achiral (E)-α,β-unsaturated 4,4-dimethyl γ-amino acids and chiral (E)-α,β-unsaturated γ-amino acids in both single crystals and in solution. The two independent strands of the same peptide intertwine to form a β-double helix structure, and it is stabilized by inter-strand hydrogen bonds. The peptides with chiral (E)-α,β-unsaturated γ-amino acids derived from α-L-amino acids adopt a (P)-β-double helix, whereas peptides consisting of (E)-α,β-unsaturated γ-amino acids derived from α-D-amino acids adopt an (M)-β-double helix conformation. The circular dichroism (CD) signature of the (P) and (M)-β-double helices and the stability of these peptides at higher temperatures were examined. Furthermore, ion transport studies suggested that these peptides transport ions across membranes. Even though the structural analogy suggests that these new β-double helices are structurally different from those of the α-peptide β-double helices, they retain ion transport activity. The results reported here may open new avenues in the design of functional foldamers.  相似文献

An efficient method has been found for the preparation of cyclic monodithioacetats of 1,3-dicarbonyl compounds using boron trifluoride etherate as the acid catalyst. The syntheses of three new optically active β-mercaptoalcohols,(R)-1,1-diphenyl-2-mercapto-3-methyl-1-butanol,(R)-1,1-diphenyl-2-mercapto-4-methyl-1-pentanol,and (R)-1,1-diphenyl-2-mercapto-1-benzenepropanol from the corresponding amino acids are described.The enantiomeric excesses of these β-mercaptoalcohols were determined by ^1H NMR as their (S)-mandeloyl derivatives.  相似文献

A catalytic enantioselective amination of β-keto esters using (S)-BINOL chiral calcium phosphate has been developed. The reaction produces chiral α-amino-β-keto ester derivatives in most cases with moderate to excellent enantioselectivities (up to 99 %) and good yields (up to 99 %). This mild synthetic method highlights a low catalyst loading and high catalytic efficiency. When the substrate backbone was changed to 1-tetralone-derived β-keto esters, unexpectedly high yields of selective redox products were obtained. The practicality of the reaction was realized by a scale-up without any significant loss in the enantioselectivity and yield. Chiral calcium phosphate was successfully recovered and reused for four runs, indicating its stability and high catalytic activity.  相似文献

The chiral separation of 10 β-adrenergic blockers (acebutalol, alprenolol, bufuralol, bisoprolol, celiprolol, carazolol, indenolol, metoprolol, oxprenolol and propranolol) was achieved on CelluCoat column (250 mm × 4.6 mm, 5 μm particle size). The mobile phases used were (90:10:0.2, v/v/v) and (95:5:0.2, v/v/v) combinations of n-heptane-ethanol-diethylamine, respectively. The flow rates were 0.5, 1.0 and 2.0 mL min−1 with detection at 225 nm. The capacity (k), selectivity (α) and resolution (Rs) factors were 0.44-12.91, 1.12-2.19 and 1.00-9.50, respectively. The proposed supra-molecular models indicated that the chiral resolution were governed by π-π interactions, hydrogen bondings and steric effect.  相似文献

Protein functions are enabled by their perfectly arranged 3D structure, which is the result of a hierarchical intramolecular folding process. Sequence-defined polypeptide chains form locally ordered secondary structures (i.e., α-helix and β-sheet) through hydrogen bonding between the backbone amides, shaping the overall tertiary structure. To generate similarly complex macromolecular architectures based on synthetic materials, a plethora of strategies have been developed to induce and control the folding of synthetic polymers. However, the degree of complexity of the structure-driving ensemble of interactions demonstrated by natural polymers is unreached, as synthesizing long sequence-defined polymers with functional backbones remains a challenge. Herein, we report the synthesis of hybrid peptide-N,N-Dimethylacrylamide copolymers via radical Ring-Opening Polymerization (rROP) of peptide containing macrocycles. The resulting synthetic polymers contain sequence-defined regions of β-sheet encoding amino acid sequences. Exploiting the pH responsiveness of the embedded sequences, protonation or deprotonation in water induces self-assembly of the peptide strands at an intramacromolecular level, driving polymer chain folding via formation of β-sheet secondary structures. We demonstrate that the folding behavior is sequence dependent and reversible.  相似文献

Organocatalytic asymmetric Michael addition of 3-(OTBS)-propanal to β-nitrostyrenes catalyzed by chiral sulfamides was investigated. Good d.r. (up to 80:20) and excellent enantioselectivities (up to >99% ee) were achieved. Both the N-[(1R,2R)-2-aminocyclohexyl]-N’-(phenylmethyl)sulfamide 7b and the novel chiral N-[(1R,2R)-2-aminocyclohexyl]-N’-[3,5-bis(trifluoromethyl)phenyl]sulfamide 7a were identified as efficient primary amine organocatalysts.  相似文献

Chiral separation of dausyl amino acids by capillary electrophoresis using mixed selectors of Mn(ll)-L-alanine complex and β-cyclodextrin (β-CD) was studied. Resolution was considerably superior to that obtained by using either Mn (Ⅱ)-L-alanine complex or β-CD alone. The effects of separation parameters, such as pH value of buffer solution, capillary temperature, the concentration of Mn (Ⅱ)-L-alanine complex, the types of CD and ligand on the migration times and resolutions were investigated. Six different transition metal complexes,Cu(Ⅱ), Zn(Ⅱ), Co(Ⅱ), Ni(Ⅱ), Hg(Ⅱ) and Cd(Ⅱ)-L-alanine complexes have been employed and compared with Mn(Ⅱ)complex. Differences in retention and selectivity were found.The substitution of Cu(Ⅱ), Zn(Ⅱ), Co(Ⅱ) and Ni(Ⅱ) for Mn(Ⅱ) resulted in a better chiral resolution while Hg(Ⅱ) and Cd(Ⅱ) showed poorer resolution abilities. The chiral separation mechanism was also discussed briefly.  相似文献

Abstract

The N-halosuccinimide glycosylation is a highly selective reaction that leads to trans-configured 1-alkoxy-2-halo-glycosides (halo = bromo, iodo).23 As an exception to the generally observed exclusive formation of α-linked glycosides in such reactions,2 we obtained a 3 / 1 mixture of α- and β-disaccharide 6 and 7, when we treated the silylated glycal 4 with NIS (N-iodosuccinimide) and the glycoside 5.4 The similarly protected arabino glycal 9, on the other hand, gave exclusively the expected α-linked saccharide 11, when treated with NIS and the alcohol component 10.5 Silylated glycals 4 and 9 were obtained from L-digitoxal 1 6 and L-rhamnal 8 7 by treatment with tert-butyldiphenylchlorosilane8 and tert-butyldimethylchlorosilane,9 respectively. In both cases the 3-O-silylated derivatives formed in high yields. Only in case of the ribo-configuration minor amounts of a 4-O-silylated product 3 were identified.  相似文献

Vancomycin-capped (3-(2-O-β-cyclodextrin)-2-hydroxypropoxy)-propylsilyl-appended silica particles (VCD-HPS), a new type of substituted β-cyclodextrin-bonded chiral stationary phase (CSP) for liquid chromatography (LC), have been synthesized by treatment of bromoacetate-substituted-(3-(2-O-β-cyclodextrin)-2-hydroxypropoxy)-propylsilyl-appended silica particles (BACD-HPS) with vancomycin in anhydrous methanol. The stationary phase is characterized by elemental analysis. This new CSP has a chiral selector with two recognition sites: vancomycin and β-cyclodextrin (β-CD), which can provide multiple interactions with the solutes. The chromatographic performance of VCD-HPS was studied with several disubstituted benzenes and some chiral drug compounds as solutes in reversed-phase LC. The results show that VCD-HPS has excellent selectivity for the separation of aromatic positional isomers and enantiomers of chiral compounds.  相似文献
