Abstract

Imidazoles, thiazoles, and oxazoles have been identified as valuable molecules in pharmaceutical and agriculture chemistry. Molecules containing these scaffolds are excited in many natural products and biologically active molecules. During the last decade, magnetic nanomaterials have appeared as highly efficient catalysts in chemical science in general organic chemistry, because of their simple preparation, modification, and large surface area ratio. This article assists to review the application of magnetically reusable nanocatalysts in synthesis of biologically active imidazoles, thiazoles, and oxazoles.  相似文献

Selective formation of 2H‐2‐imidazolines and 2‐substituted oxazoles by using a multicomponent reaction of amines, either aldehydes or ketones, and α‐acidic isocyano amides or esters is described. By selecting the appropriate solvent, AgI or CuI catalyst, or by employing a weak Brønsted acid, the product formation can be fully controlled and directed quantitatively to the desired heterocyclic scaffold. The described experimental procedures not only significantly increase the scope of compatible inputs for this complexity‐generating three‐component reaction, but also allow for considerable chemical diversity: At least four diversity points in two distinct scaffolds can be exploited in this way.  相似文献

A novel synthetic route to substituted esters of imidazoles, oxazoles, thiazoles, and diethyl pyrazine-2,5-dicarboxylates via C-formylation of glycine ethyl ester hydrochloride is reported. This methodology is simple, robust, and gives good yields of different heterocyclic esters in one or two steps from a common acyclic precursor and is amenable to large scale synthesis.  相似文献

Acyclic and cyclic amino acid derivatives of 2-phosphinophenols have been synthesised by reaction of primary phosphinophenols (4-R-2-H2PC6H3OH; R=H, Me, OMe) 1a–c with formaldehyde and amino acids (o- and p-aminobenzoic acid, -lysine) via in situ formed hydroxymethyl species 2a–c. Condensation reactions with glycine did not afford defined products except when the methoxymethyl and tetrahydropyranyl ethers of 1d,e were used instead of the hydroxy compounds. o-Aminobenzoic acid gives rise to linear bis(o-carboxyphenylaminomethyl)phosphines 3a–e. p-Aminobenzoic acid, dependent on the molar ratio, affords bis(p-carboxyphenylaminomethyl)phosphines 4a,d as well as eight-membered heterocyclic 1,5,3,7-diazadiphosphacyclooctanes 5a–e. The aliphatic amino acids glycine and -lysine form six-membered heterocyclic 1,3,5-diazaphosphorinanes 6d and 7a–e, respectively, in presence of excess formaldehyde. -lysine differs from glycine by reaction at the terminal amino group. The structures of the compounds have been elucidated by multinuclear NMR spectroscopy. The salts of the phosphino amino acids are soluble in water. Water solubility increases with the number of hydrophilic groups, i.e. free phenols are more soluble than their ethers. Ligand concentrations in water from 0.1 to 1 M were observed.  相似文献

[reaction: see text] The base-promoted cyclization of optically pure N-(p-methoxybenzyl)-N-(2-chloro)propionyl amino acid derivatives resulted in a diastereo- and enantioselective approach to valuable 1,3,4,4-tetrasubstituted beta-lactams. A convenient and efficient strategy for the synthesis of 2-acyl benzothiazoles/thiazoles has been developed. The treatment of benzothiazole/thiazole with allylic Grignard reagents readily generates the corresponding 2-Grignard reagents, which is followed by a reaction with N,N'-carbonyldiimidazole activated carboxylic acids to afford various 2-acyl benzothiazoles/thiazoles products. The synthetic method is applicable to a wide array of carboxylic acids and allows easy access to 2-acyl benzothiazoles/thiazoles with considerable yields under mild reaction conditions.  相似文献

NMR study of the reactivity of multifunctional ligand cis,cis-C6H9(NHCH2C6H4-o-PPh2)3 (1) with GaMe3 and Zr(NMe2)4 was carried out, yielding [cis,cis-(κN-NHCH2C6H4-o-PPh2)(κN-NCH2C6H4-o-PPh2)2C6H9]GaMe (2) and [cis,cis-(NCH2C6H4-o-PPh2)3C6H9]Ga2Me3 (3), and [cis,cis-(NCH2C6H4-o-PPh2)3C6H9]Zr(NMe2) (4), respectively. The spectral properties of 2 and 3 are very similar to that observed for the equivalent aluminum species already reported, but form at a much slower rate which allows for the observation of a GaMe31 adduct. Species 4 undergoes coordination/displacement of one of the phosphine arms, which was observed using both NMR spectroscopy and DFT analyses.  相似文献

A new strategy has been developed for the construction of oxazoles from methyl ketones and TosMIC via self-sorting oxidative domino reaction strategy. In contrast to its usual reactivity as C–N≡C synthon in the synthesis of oxazoles, TosMIC was utilized as ammonium surrogate in the present method. The protocol is attractive in terms of readily available starting materials, metal and base free conditions, operational simplicity, C–N and C–O bond formation. A variety of 2,5-disubstituted oxazoles were prepared in moderate-to-good yields.  相似文献

A small library of 3- and 4-pyridyl-substituted pyrroles was prepared from N-acylated amino acids. Nicotinoyl or isonicotinoyl chloride was used for the N-acylation of benzyl esters of amino acids. Debenzylation by palladium-catalyzed hydrogenation gave N-acylated amino acids. Dehydration of the acylated amino acids gave cyclic intermediates, münchnones or azlactones, which were treated in situ with alkynes in 1,3-dipolar cycloadditions. The starting materials were prepared in a parallel fashion, and microwave irradiation was used to facilitate the cycloaddition reactions. The regiochemistry of the cycloaddition was studied.  相似文献

Nonpeptidic foldamers capable of displaying protein-like functionality were prepared by swapping amide bonds with 1,2,3-triazole rings. The overall conformation of these triazole oligomers is largely dictated by dipole-dipole interactions between adjacent rings. Solution NMR studies suggest that a zigzag conformation, which closely mimics the beta-strand structure, predominates in two different tetramers.  相似文献

A new, fast and solvent-free method is offers to synthesize thiazoles by condensation of 4-bromoacetylpyridine with substituted N-phenylthiourea under microwave irradiation. This approach delivers good to excellent yields of thiazoles within 5 min without using any catalyst and allows various functionalities. In comparison to the traditional heating approach, this method enables for the synthesis of different thiazole analogues with several advantages. In compliance with the Lipinski rule of five, the synthesized analogues demonstrated drug-like characteristics.  相似文献

A NBS-mediated sequential one-pot synthesis of multifunctionalized thiazoles and thiophenes from 1,3-dicarbonyl compounds and mercaptonitrile salts has been developed under mild conditions. This transformation involves sequential bromination/SN2 alkylation/Thorpe–Ziegler cyclization/regio-selective elimination of a –COR group, affording the desired products in moderate to good yields. The sequence of the leaving reactivity of –COR groups was determined and a possible mechanism was proposed.  相似文献

Removal of the protecting group was then accomplished under mild conditions allowing for a straightforward preparation of C-5 monosubstituted and C-4,5 disubstituted oxazoles. A facile and rapid synthetic approach of N-benzyl-2-substituted piperazine building-blocks via an Ugi strategy is described. This strategy is high yielding (80-92% overall yield), step-efficient and fast using microwave heating and tert-butylisocyanide as a convertible isocyanide. A highly efficient one-pot, two-step microwave procedure has been developed for the synthesis of 4-substituted 1-heteroarylpiperazines. Microwave heating of heteroaryl chlorides with 1,4-diazabicyclo[2.2.2]octane (DABCO) at 160 °C for 15 min yielded 1-heteroaryl-4-(2-chloroethyl)piperazines, which could be further reacted with various nucleophiles, again under microwave irradiation conditions, to give an array of 4-substituted 1-heteroarylpiperazines in good to excellent yields.  相似文献

Bromination and thiocyanation of 2-amino- and 2-acetylamino-4-(2-furyl)thiazoles when 1 mol of reagent is used at 10°C are directed to the 5 position. Formation of 5'-bromo-substituted derivatives when the reaction temperature is raised is the result of a secondary, thermodynamically controlled process. Monohalogenation and mononitration of 4-(2-furyl)-2-methylthiazole are directed to the 5' position. Nitration of 2-acetylamino-4-(5-nitro-2-furyl)thiazole by a nitrating mixture is accompanied by oxidative cleavage of the 5-nitrofuran moiety and leads to formation of 5,5'- and 3',5'-dinitro derivatives.  相似文献

Microwave-assisted synthesis of 2-oxazolines from carboxylic acids using the open vessel technique is described. This efficient method involves direct condensation of carboxylic acids with excess 2-amino-2-methyl-1-propanol at 170 °C to give the corresponding 2-oxazolines in moderate to excellent yields.  相似文献
