The self-aggregation behavior of two amphiphilic poly(ethylene oxide)-poly[(R)-3-hydroxybutyrate]-poly(ethylene oxide) (PEO-PHB-PEO) triblock copolymer samples with nearly identical PHB block lengths but different PEO block lengths, PEO-PHB-PEO(2000-810-2000) and PEO-PHB-PEO(5000-780-5000), was studied with dynamic and static light scattering (DLS and SLS), in combination with fluorescence spectroscopy and transmission electron microscopy (TEM). The formation of polymeric micelles by the two PEO-PHB-PEO triblock copolymers was confirmed with fluorescence technique and TEM. DLS analysis showed that the hydrodynamic radius (R(h)) of the monodistributed polymeric micelles increased with an increase in PEO block length. The relative thermostability of the triblock copolymer micelles was studied by SLS and DLS at different temperatures. The aggregation number and the ratio of the radius of gyration over hydrodynamic radius were found to be independent of temperature, probably due to the strong hydrophobicity of the PHB block. The combination of DLS and SLS studies indicated that the polymeric micelles were composed of a densely packed core of hydrophobic PHB blocks and a corona shell formed by hydrophilic PEO blocks. The aggregation numbers were found to be approximately 53 for PEO-PHB-PEO(2000-810-2000) micelles and approximately 37 for PEO-PHB-PEO(5000-780-5000) micelles. The morphology of PEO-PHB-PEO spherical micelles determined by DLS and SLS measurements was further confirmed by TEM.  相似文献

Amphiphilic triblock copolymers of poly(methyl methacrylate)-b-poly(ethylene oxide)-b-poly(methyl methacrylate) (PMMA-b-PEO-b-PMMA) with well-defined structure were synthesized via atom transfer radical polymerization (ATRP) of methyl methacrylate (MMA) initiated by the PEO macroinitiator. The macroinitiator and triblock copolymer with different PMMA and/or PEO block lengths were characterized with 1H and 13C NMR and gel permeation chromatography (GPC). The micelle formed by these triblock copolymers in aqueous solutions was detected by fluorescence excitation and emission spectra of pyrene probe. The critical micelle concentration (CMC) ranged from 0.0019 to 0.016 mg/mL and increased with increasing PMMA block length, while the PEO block length had less effect on the CMC. The partition constant Kv for pyrene in the micelle and in aqueous solution was about 105. The triblock copolymer appeared to form the micelles with hydrophobic PMMA core and hydrophilic PEO loop chain corona. The hydrodynamic radius Rh,app of the micelle measured with dynamic light scattering (DLS) ranged from 17.3 to 24.0 nm and increased with increasing PEO block length to form thicker corona. The spherical shape of the micelle of the triblock copolymers was observed with an atomic force microscope (AFM). Increasing hydrophobic PMMA block length effectively promoted the micelle formation in aqueous solutions, but the micelles were stable even only with short PMMA blocks.  相似文献

Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxy-butyrate)-poly(ethyl ethylene phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a core-shell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. MTT assay also demonstrates that the block copolymers are biocompatible, rendering these copolymers attractive for drug delivery. 采用耗散粒子动力学(Dissipative particle dynamics, DPD)模拟方法研究了三嵌段共聚物聚氧乙烯-聚氧丙烯-聚氧乙烯(PEO-PPO-PEO)的胶束化和凝胶化行为. 通过模拟得到了F127(EO99PO65EO99)水溶液的临界胶束浓度和临界凝胶浓度. 结果发现, 在298 K、 质量分数低于40%时, F127水溶液中形成的胶束形状均为球形. 此外,进一步研究了亲水嵌段长度对胶束结构及凝胶形成浓度的影响, 结果发现, 亲水嵌段越短, 越有利于长椭球状胶束的形成, 而临界凝胶浓度随着亲水嵌段PEO长度的增加而降低.  相似文献

Two new poly(ethylene oxide)-poly(styrene oxide) triblock copolymers (PEO-PSO-PEO) with optimized block lengths selected on the basis of previous studies were synthesized with the aim of achieving a maximal solubilization ability and a suitable sustained release, while keeping very low material expense and excellent aqueous copolymer solubility. The self-assembling and gelling properties of these copolymers were characterized by means of light scattering, fluorescence spectroscopy, transmission electron microscopy, and rheometry. Both copolymers formed spherical micelles (12-14 nm) at very low concentrations. At larger concentration (>25 wt%), copolymer solutions showed a rich phase behavior, with the appearance of two types of rheologically active (more viscous) fluids and of physical gels depending on solution temperature and concentration. The copolymer behaved notably different despite their relatively similar block lengths. The ability of the polymeric micellar solutions to solubilize the antifungal drug griseofulvin was evaluated and compared to that reported for other structurally-related block copolymers. Drug solubilization values up to 55 mg g−1 were achieved, which are greater than those obtained by previously analyzed poly(ethylene oxide)-poly(styrene oxide), poly(ethylene oxide)-poly(butylene oxide), and poly(ethylene oxide)-poly(propylene oxide) block copolymers. The results indicate that the selected SO/EO ratio and copolymer block lengths were optimal for simultaneously achieving low critical micelle concentrations (cmc) values and large drug encapsulation ability. The amount of drug released from the polymeric micelles was larger at pH 7.4 than at acidic conditions, although still sustained over 1 day.  相似文献

Interaction of amphiphilic poly(ethylene oxide)-b-poly(epsilon-caprolactone) copolymers with anionic sodium dodecyl sulfate (SDS) has been investigated in aqueous solution. Formation of mixed micelles has been confirmed by surface tension measurements, whereas the influence of the surfactant on the copolymer self-assembling has been studied by measurement of the 1H NMR self-diffusion coefficients and by small-angle neutron scattering. As a rule, the surfactant decreases the heterogeneity of the micellar structures formed by the copolymer in water. Moreover, increasing the content of SDS results in the increasingly more important extension of the poly(ethylene oxide) (PEO) corona chains and the copolymer micelle deaggregation. The stability of the micelles against SDS increases with the length of the hydrophobic block. Preliminary two-dimensional NMR measurements with nuclear Overhauser enhancement have confirmed the spatial vicinity between SDS and the constitutive blocks of the copolymer.  相似文献

Biodegradable, amphiphilic, four‐armed poly(?‐caprolactone)‐block‐poly(ethylene oxide) (PCL‐b‐PEO) copolymers were synthesized by ring‐opening polymerization of ethylene oxide in the presence of four‐armed poly(?‐caprolactone) (PCL) with terminal OH groups with diethylzinc (ZnEt2) as a catalyst. The chemical structure of PCL‐b‐PEO copolymer was confirmed by 1H NMR and 13C NMR. The hydroxyl end groups of the four‐armed PCL were successfully substituted by PEO blocks in the copolymer. The monomodal profile of molecular weight distribution by gel permeation chromatography provided further evidence for the four‐armed architecture of the copolymer. Physicochemical properties of the four‐armed block copolymers differed from their starting four‐armed PCL precursor. The melting points were between those of PCL precursor and linear poly(ethylene glycol). The length of the outer PEO blocks exhibited an obvious effect on the crystallizability of the block copolymer. The degree of swelling of the four‐armed block copolymer increased with PEO length and PEO content. The micelle formation of the four‐armed block copolymer was examined by a fluorescent probe technique, and the existence of the critical micelle concentration (cmc) confirmed the amphiphilic nature of the resulting copolymer. The cmc value increased with increasing PEO length. The absolute cmc values were higher than those for linear amphiphilic block copolymers. © 2004 Wiley Periodicals, Inc. Hierarchical mesostructures of poly(ε-caprolactone)-b-poly(ethylene oxide)-b-poly(ε-caprolactone) (PCL-PEO-PCL) triblock copolymers have been grown from evaporation-induced self-assembly directed by alkali metal ions. The self-assembly process began with a dilute homogeneous solution of the triblock copolymers in a mixture of tetrahydrofuran (THF) and water. THF preferentially evaporated under reduced pressure and induced the formation of amphiphilic polymer micelles. The spherical polymer micelles formed both in deionized water and NaOH aqueous solution. However, different mesostructures were discovered during the film depositing process for scanning electron microscopy observation. The polymer micelles were observed for the deposition sample in deionized water while sisal-like hierarchical mesostructures resulted from the film deposition of polymer micelles in NaOH aqueous solution. The sisal-like mesostructures and their formation process were observed through scanning electron microscopy, transmission electron microscopy, fluorescent microscopy, X-ray diffraction, and Fourier transform infrared spectroscopy. Detailed study revealed that during evaporation-induced self-assembly of PCL-PEO-PCL amphiphilic triblock copolymer directed by alkali metal ions, the sodium ions and polymer micelles increasingly concentrated in NaOH aqueous solution and the solvent quality for the diblock progressively decreased, which resulted in the stronger coordination between alkali metal ions and PEO ligands in the block copolymer and PEO segment crystallization.  相似文献

The micelle formation of a series of amphiphilic block copolymers in aqueous and NaCl solutions was studied by a fluorescent probe technique using pyrene as a "model drug". These copolymers were synthesized from poly (ethylene glycol) (PEG) and l-lactide by a new calcium ammoniate catalyst. They had fixed PEG block lengths (44, 104 or 113 ethylene oxide units) and various poly(l-lactide) (PLLA) block lengths (15–280 lactide units). The critical micelle concentration (cmc) was found to decrease with increasing PLLA content. The distinct dissimilarity of the cmc values of diblock and triblock copolymers based on the same block length of PEG provided evidence for the different configurations of their micelles. It was also observed that the introduction of NaCl salt significantly contributed to a decrease in the cmcs of the copolymers with short PEG and PLLA blocks, while it had less influence on the cmcs of copolymers with long PEG or PLLA blocks. The dependence of partition coefficients ranging from 0.2×105 to 1.9×105 on the PLLA content in the copolymer and on the micelle configuration was also discussed. The contribution of NaCl salt to increasing the partition of pyrene into a micellar phase was observed.  相似文献

The adsorption of amphiphilic poly(ethylene oxide)-b-poly(epsilon-caprolactone) and poly(ethylene oxide)-b-poly(gamma-methyl-epsilon-caprolactone) copolymers in aqueous solution on silica and glass surfaces has been investigated by flow microcalorimetry, small-angle neutron scattering (SANS), surface forces, and complementary techniques. The studied copolymers consist of a poly(ethylene oxide) (PEO) block of M(n) = 5000 and a hydrophobic polyester block of poly(epsilon-caprolactone) (PCL) or poly(gamma-methyl-epsilon-caprolactone) (PMCL) of M(n) in the 950-2200 range. Compared to homoPEO, the adsorption of the copolymers is significantly increased by the connection of PEO to an aliphatic polyester block. According to calorimetric experiments, the copolymers interact with the surface mainly through the hydrophilic block. At low surface coverage, the PEO block interacts with the surface such that both PEO and PCL chains are exposed to the aqueous solution. At high surface coverage, a dense copolymer layer is observed with the PEO blocks oriented toward the solution. The structure of the copolymer layer has been analyzed by neutron scattering using the contrast matching technique and by tapping mode atomic force microscopy. The experimental observations agree with the coadsorption of micelles and free copolymer chains at the interface.  相似文献

A block copolymer of hydrophilic poly(ethylene oxide) and a hydrophobic poly(alkylene oxide) can associate in dilute aqueous solution to form micelles. The results of recent investigations of the micellisation behaviour and micelle properties of such copolymers are described. Copolymers of ethylene oxide with propylene oxide, 1,2‐butylene oxide or styrene oxide are considered, including aspects of their preparation. Experimental methods for determination of critical conditions for micellisation, micelle association number and spherical‐micelle radius are summarised. Effects of temperature, composition, block length and block architecture (diblock, triblock and cyclic‐diblock) are described and, where possible, related to the predictions of theory. Brief consideration is given to the dynamics of micelle formation/dissociation, to cylindrical micelles, and to effects of added salts.  相似文献

Amphiphilic triblock copolymers of poly(3-hydroxybutyrate)-poly(ethylene glycol)-poly(3-hydroxybutyrate) (PHB-PEG-PHB) were directly synthesized by the ring-opening copolymerization of β-butyrolactone monomer using PEG as macroinitiator. Their structure, thermal properties and crystallization were investigated by 1H NMR, differential scanning calorimetry (DSC) and X-ray diffraction. It was found that both PHB and PEG blocks were miscible. With the increase in the PHB block length, the triblock copolymers became amorphous because amorphous PHB block remarkably depressed the crystallization of the PEG block. Biodegradable nanoparticles with core-shell structure were prepared in aqueous solution from the amphiphilic triblock copolymers, and characterized by 1H NMR, SEM and fluorescence. The hydrophobic PHB segments formed the central solid-like core, and stabilized by the hydrophilic PEG block. The nanoparticle size was close related to the initial concentrations of the nanoparticle dispersions and the compositions of the triblock copolymers. Moreover, the PHB-PEG-PHB nanoparticles also showed good drug loading properties, which suggested that they were very suitable as delivery vehicles for hydrophobic drugs.  相似文献

The acid effect on the aggregation of poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) block copolymers EO(20)PO(70)EO(20) has been investigated by transmission electron microscopy (TEM), particle size analyzer (PSA), Fourier transformed infrared, and fluorescence spectroscopy. The critical micellization temperature for Pluronic P123 in different HCl aqueous solutions increases with the increase of acid concentration. Additionally, the hydrolysis degradation of PEO blocks is observed in strong acid concentrations at higher temperatures. When the acid concentration is low, TEM and PSA show the increase of the micelle mean diameter and the decrease of the micelle polydispersity at room temperature, which demonstrate the extension of EO corona and tendency of uniform micelle size because of the charge repulsion. When under strong acid conditions, the aggregation of micelles through the protonated water bridges was observed.  相似文献

We report here on the effects that the solution properties of poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) block copolymers have on the reduction of hydrogen tetrachloroaurate(III) hydrate (HAuCl4.3H2O) and the size of gold nanoparticles produced. The amphiphilic block copolymer solution properties were modulated by varying the temperature and solvent quality (water, formamide, and their mixtures). We identified two main factors, (i) block copolymer conformation or structure (e.g., loops vs entanglements, nonassociated polymers vs micelles) and (ii) interactions between AuCl4- ions and block copolymers (attractive ion-dipole interactions vs repulsive interactions due to hydrophobicity), to be important for controlling the competition between the reactivities of AuCl4- reduction in the bulk solution to form gold seeds and on the surface of gold seeds (particles) and the particle size determination. The particle size increase observed with increased temperature in aqueous solutions is attributed to enhanced hydrophobicity of the block copolymer, which favors AuCl4- reduction on the surface of seeds. The lower reactivity and higher particle sizes observed in formamide solutions are attributed to the shielding of ion-dipole interaction between AuCl4- ions and block copolymers by formamide, which overcomes the beneficial effects of formamide on the block copolymer conformation (lower micelle concentration).  相似文献

The thermoreversible gelation of Pluronic [poly(ethylene oxide) (PEO)–polypropylene oxide (PPO)–PEO] aqueous solutions originates from micelle formation and micelle volume changes due to PEO–water and PPO–water lower critical solution temperature behavior. The micelle volume fraction is known to dominate the sol–gel transition behavior of Pluronic aqueous solutions. Triblock copolymers of PEO and aliphatic polyesters, instead of PPO, were prepared by hexamethylene diisocyanate coupling and dicyclohexyl carbodiimide coupling. Through changes in the molecular weight and hydrophobicity of the polyester middle block, the hydrophobic–hydrophilic balance of each block was systematically controlled. The following aliphatic polyesters were used: poly(hexamethylene adipate) (PHA), poly(ethylene adipate) (PEA), and poly(ethylene succinate) (PESc). With the hydrophobicity and molecular weight of the middle block increasing, the critical micelle concentration at the same critical micelle temperature decreased, and the absolute value of the micellization free energy increased. The micelle size was rather insensitive to temperature but slightly decreased with increasing temperature. PEO–PHA–PEO and PEO–PEA–PEO triblock copolymers needed high polymer concentrations to form gels. This was ascribed to the tight aggregation of PHA and PEA chains in the micelle core due to strong hydrophobic interactions, which induced the contraction of the micelle core. However, because of the relatively hydrophilic core, a PEO–PESc–PEO aqueous solution showed gelation at a low polymer concentration. © 2003 Wiley Periodicals, Inc. Interaction of urea with pluronic block copolymers by 1H NMR spectroscopy   总被引:1,自引:0,他引:1  
Solution 1H NMR techniques were used to characterize the interaction of urea with poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) triblock copolymers. The urea was established to interact selectively with the PEO blocks of the block copolymer, and the interaction sites were found not to change with increasing temperature. Such interactions influence the self-assembly properties of the block copolymer in solution by increasing the hydration of the block copolymers and stabilizing the gauche conformation of the PPO chain. Therefore, urea increases the critical micellization temperature (CMT) values of PEO-PPO-PEO copolymers, and the effect of urea on the CMT is more pronounced for copolymers with higher PEO contents and lower for those with increased contents of PPO segments.  相似文献

We report the detailed characterization of micelles formed by two nonionic, amphiphilic ABC triblock copolymers. Poly(ethylene oxide)-b-poly(styrene)-b-1,2-poly(butadiene) (PEO-b-PS-b-PB) triblock copolymer "OSB" forms core-corona spherical micelles in aqueous solution, and the two hydrophobic blocks S and B are mixed homogeneously within the micelle core. PEO-b-PS-b-PB:C6F13I triblock copolymer "OSF" was prepared by selective fluorination of the B block in OSB with n-perfluorohexyl iodide. Fluorination of the B block induces internal segregation into an inner F core and an intermediate S shell. Furthermore, the strong incompatibility that results from fluorination drives a shape change into an oblate ellipsoid. These micellar morphologies are confirmed by combined light, neutron, and X-ray scattering measurements, as well as TEM imaging.  相似文献

The paper provides new insights into the structure of Pt-containing diblock and triblock copolymers based on poly(ethylene oxide) (PEO) and poly(4-vinylpyridine) (P4VP), using a combination of atomic force microscopy (AFM), X-ray diffraction (XRD), transmission electron microscopy (TEM), and anomalous small-angle X-ray scattering (ASAXS). Parallel studies using methods contributing supplemental structural information allowed us to comprehensively characterize sophisticated polymer systems during metalation and to exclude possible ambiguity of the data interpretation of each of the methods. AFM and TEM make available the determination of sizes of the micelles and of the Pt-containing micelle cores, respectively, while a combination of XRD, TEM, and ASAXS reveals Pt-nanoparticle size distributions and locations along with the structural information about the polymer matrix. In addition, for the first time, ASAXS revealed the organization of Pt-nanoparticle-filled diblock and triblock copolymers in the bulk. The nanoparticle characteristics are mainly determined by the type of block copolymer system in which they are found: larger particles (2.0-3.0 nm) are formed in triblock copolymer micelles, while smaller ones (1.5-2.5 nm) are found in diblock copolymer micelles. This can be explained by facilitated intermicellar exchange in triblock copolymer systems. For both systems, Pt nanoparticles have narrow particle size distributions as a result of a strong interaction between the nanoparticle surface and the P4VP units inside the micelle cores. The pH of the medium mainly influences the particle location rather than the particle size. A structural model of Pt-nanoparticle clustering in the diblock PEO-b-P4VP and triblock P4VP-b-PEO-b-P4VP copolymers in the bulk was constructed ab initio from the ASAXS data. This model reveals that nearly spherical micellar cores of about 10 nm in diameter (filled with Pt nanoparticles) aggregate forming slightly oblate hollow bodies with an outer diameter of about 40 nm.  相似文献

Spontaneous formation and efficient stabilization of gold nanoparticles with an average diameter of 7 approximately 20 nm from hydrogen tetrachloroaureate(III) hydrate (HAuCl4.3H2O) were achieved in air-saturated aqueous poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) block copolymer solutions at ambient temperature in the absence of any other reducing agent. The particle formation mechanism is considered here on the basis of the block copolymer concentration dependence of absorption spectra, the time dependence (kinetics) of AuCl4- reduction, and the block copolymer concentration dependence of particle size. The effects of block copolymer characteristics such as molecular weight (MW), PEO block length, PPO block length, and critical micelle concentration (cmc) are explored by examining several PEO-PPO-PEO block copolymers. Our observations suggest that the formation of gold nanoparticles from AuCl4- comprises three main steps: (1) reduction of metal ions by block copolymer in solution, (2) absorption of block copolymer on gold clusters and reduction of metal ions on the surface of these gold clusters, and (3) growth of metal particles stabilized by block copolymers. While both PEO and PPO blocks contribute to the AuCl4- reduction (step 1), the PEO contribution appears to be dominant. In step 2, the adsorption of block copolymers on the surface of gold clusters takes place because of the amphiphilic character of the block copolymer (hydrophobicity of PPO). The much higher efficiency of particle formation attained in the PEO-PPO-PEO block copolymer systems as compared to PEO homopolymer systems can be attributed to the adsorption and growth processes (steps 2 and 3) facilitated by the block copolymers. The size of the gold nanoparticles produced is dictated by the above mechanism; the size increases with increasing reaction activity induced by the block copolymer overall molecular weight and is limited by adsorption due to the amphiphilic character of the block copolymers.  相似文献

Miktoarm star triblock copolymers mu-[poly(ethylethylene)][poly(ethylene oxide)][poly(perfluoropropylene oxide)] self-assemble in dilute aqueous solution to give multicompartment micelles with the cores consisting of discrete poly(ethylethylene) and poly(perfluoropropylene oxide) domains. Tetrahydrofuran is a selective solvent for both the poly(ethylethylene) and poly(ethylene oxide) blocks, and thus in tetrahydrofuran mixed corona micelles are favored with poly(perfluoropropylene oxide) cores. The introduction of tetrahydrofuran into water induces an evolution from multicompartment micelles to mixed corona [poly(ethylethylene) + poly(ethylene oxide)] micelles, as verified by dynamic light scattering and nuclear magnetic resonance spectroscopy. A mixed solvent containing 60 wt % tetrahydrofuran corresponds to the transition point, as verified by analysis of a poly(ethylethylene)-poly(ethylene oxide) diblock copolymer in the same solvent mixtures. Furthermore, cryogenic transmission electron microscopy suggests that, as the poly(ethylethylene) block transitions from the core to the corona, the micelle morphologies evolve from disks to oblate ellipsoid micelles (with some vesicles), with worms and spheres evident at intermediate compositions.  相似文献
