Melamine is a toxic triazine, illegally used as an additive in milk to apparently increase the amount of protein. A chromatographic procedure using a C18 column and a micellar mobile phase of sodium dodecyl sulphate (0.05 M) and propanol (7.5%), buffered at pH 3, and a detection set by absorbance at 210 nm, was reported for the resolution and quantification of melamine in liquid and powdered milk samples. In this work, samples were diluted with a SDS solution and were directly injected, thus avoiding long extraction and experimental procedures. Melamine was eluted in nearly 9.3 min without overlapping the protein band or other endogeneous compounds. The optimal mobile phase composition was taken using a chemometrical approach that considers the retention factor, efficiency and peak shape. Validation was performed following the European Commission's indications (European Decision 2002/657/EC), and the main analytical parameters studied were: linearity (0.02-100 ppm; r2 = 0.999), limit of detection (5 ppb), intra- and inter-day precision (R.S.D. <7.6% and <9.7%, respectively) and robustness (R.S.D. <7.4% for retention time and <5.0% for area). Sensitivity was adequate to detect melamine under the safety limits proposed by the US FDA. Finally, recoveries for several milk samples were found in the 85-109% range.  相似文献

Multi-walled carbon nanotube (CNT) was mixed with epoxy to fabricate microdisc electrode used as a detector for a specially designed miniaturized capillary electrophoresis (CE)-amperometric detection system for the separation and detection of several bioactive thiols. The end-channel CNT amperometric detector offers favourable signal-to-noise characteristics at a relatively low potential (0.8 V) for detecting thiol compounds. Factors influencing the separation and detection processes were examined and optimized. Four thiols (homocysteine, cysteine, glutathione, and N-acetylcysteine) have been separated within 130 s at a separation voltage of 2000 V using a 20 mM phosphate running buffer (pH 7.8). Highly linear response is obtained for homocysteine, cysteine, glutathione, and N-acetylcysteine over the range of 5-50 μM with detection limits of 0.75, 0.8, 2.9, and 3.3 μM, respectively. Good stability and reproducibility (R.S.D. < 5%) are obtained reflecting the minimal adsorption of thiols at the CNT electrode surface. The new microchip protocol should find a wide range of bioanalytical applications involving assays of thiol compounds.  相似文献

Alkaline-phosphatase (ALP) catalyses the hydrolysis of 1-naphthyl phosphate to fluorescent 1-naphthol (λex=346 nm, λem=463 nm). This enzymatic reaction was investigated in presence of inhibitors: organochlorine (tetradifon), carbamate (metham-sodium) and organophosphorus pesticides (fenitrothion), heavy metal (Ag+) and CN. The fluorescent signal, which is inversely dependent on the inhibitor concentration, is related to the amount of the inhibitor. Detection limits between 4.1 μM for tetradifon and 91.2 μM for metham-sodium were found. The relative standard deviation (R.S.D.) was between 2.6 and 6.2%.Sol-gel matrices derived from tetramethyl orthosilicate were doped with ALP using microencapsulation. The response of the biosensor based ALP sol-gel encapsulated to 1-naphthyl phosphate was reproducible (R.S.D.=6.6%). Inhibition plots obtained for test pesticides (metham-sodium and tetradifon) display linear calibration in the ranges 194-774 μM and 3.5-28 μM, detection limits of 4.9 and 292.3 μM and R.S.D. of 3.9 and 7.3% for metham-sodium and tetradifon, respectively. The results show that the system is able to detect class compounds such as pesticides and inorganic compounds.  相似文献

The development of a rapid method for the determination of perchlorate in rain and drinking waters is presented. In the optimised method, an on-line preconcentration technique was employed utilising a 10 mm × 4.6 mm Phenomenex Onyx monolithic guard cartridge coated with (N-dodecyl-N,N-dimethylammonio)undecanoate for selective preconcentration, with subsequent elution into a fixed volume injection loop (‘heart-cut’ of the concentrator column eluate) and separation using an IonPac AS16 (250 mm × 2 mm) anion exchange column and a potassium hydroxide concentration gradient. Off-line optimisation studies showed that the coated monolith displayed near quantitative recovery up to 50 μg/L perchlorate level from standards prepared in reagent water. On-line preconcentration of perchlorate obtained detection limits down to 56 ng/L in reagent water, between 70 and 80 ng/L in rainwater samples and 2.5 μg/L in non-pretreated drinking water. After an additional sample sulphate/carbonate removal step, low ng/L perchlorate concentrations could also be observed in drinking water. The complete on-line method exhibited reproducibility for n = 10 replicate runs of R.S.D. ≤ 3% for peak height/area and R.S.D. = 0.08% for retention time. The optimised method, of 20 min total duration, was applied to the determination of perchlorate by standard addition in 10 rainwater samples and one drinking water sample. Concentrations of perchlorate present ranged from below the detection limit for four rainwater samples, with another three samples showing perchlorate present at between 70 and 100 ng/L, and one sample showing perchlorate present at 2.8 μg/L. Levels of 1.1 μg/L in the drinking water sample were also recorded.  相似文献

A simple coating procedure employing a sol-gel process to modify the inner surface of a bare fused-silica capillary with a positively charged quaternary ammonium group is established. Scanning electron microscopic studies reveal that a smooth coating with 1 to approximately 2 microm thickness can be obtained at optimized coating conditions. With 40 mM citrate as a running electrolyte, the plot of electroosmotic flow (EOF) versus pH shows a unique three-stage EOF pattern from negative to zero and then to positive over a pH range of 2.5 to 7.0. At pH above 5.5, the direction of the EOF is from the anode to the cathode, as is the case in a bare fused-silica capillary, and the electroosmotic mobility increases as the pH increases. However, the direction of the EOF is reversed at pH below 4.0. Over the pH range of 4.0 to 5.5, zero electroosmotic mobility is obtained. Such a three-stage EOF pattern has been used to separate six aromatic acids under suppressed EOF and to separate nitrate and nitrite with the anions migrating in the same direction as the EOF. The positively charged quaternary ammonium group on the coating was also utilized to minimize the adsorption problem during the separation of five basic drugs under suppressed EOF and during the separation of four basic proteins with the cations migrate in the opposite direction as the EOF. Also, the stability and reproducibility of this column are good.  相似文献

Anion-exchange (AEX) columns were prepared by on-column polymerization of acrylates and methacrylates containing tertiary amino or quaternary ammonium groups on monolithic silica in a fused silica capillary modified with anchor groups. The columns provided a plate height (H) of less than 10 μm at optimum linear velocity (u) with keeping their high permeability (K = 9–12 × 10−14 m2). Among seven kinds of AEX columns, a monolithic silica column modified with poly(2-hydroxy-3-(4-methylpiperazin-1-yl)propyl methacrylates) (HMPMA) showed larger retentions and better selectivities for nucleotides and inorganic anions than the others. The HMPMA column of 410 mm length produced 42 000–55 000 theoretical plates (N) at a linear velocity of 0.97 mm/s with a backpressure of 3.8 MPa. The same column could be employed for a fast separation of inorganic anions in 1.8 min at a linear velocity of 5.3 mm/s with a backpressure of 20 MPa. In terms of van Deemter plot and separation impedance, the HMPMA column showed higher performance than a conventional particle-packed AEX column. The HMPMA column showed good recovery of a protein, trypsin inhibitor, and it was applied to the separation of proteins and tryptic digest of bovine serum albumin (BSA) in a gradient elution, to provide better separation compared to a conventional particle-packed AEX column.  相似文献

A urease optical biosensor for the determination of heavy metals based on sol-gel immobilization technique was developed. A fluorescent dye, FITC-dextran, was encapsulated and parameters including optical properties of the probe, relative enzyme activity, initial pH value and the buffer concentration for substrate preparation were investigated. In sol-gel immobilization, 1 mM Tris-HCl at pH 7.1 provided a sufficient buffer capacity for metal ion analysis as well as the enzyme activity maintenance. Also, two analytical procedures, incubated and un-incubated systems, were compared to understand the sensitivity and applicability to heavy metal analysis. The developed optical biosensor showed high reproducibility and the relative standard deviation (R.S.D.) of 5.1% (n=10) was obtained. Also, eight measurements can be completed automatically within 36 min. The biosensor has high sensitivity to Cu(II) and Cd(II) and an analytical range of 10-230 μM with a detection limit of 10 μM was achieved. Moreover, biological and environmental samples were examined to evaluate the applicability of the developed biosensor. A 19-82% of inhibition was observed when 20-45 μM metal ions were amended into tested samples, revealing that the developed system has the potential for the determination of heavy metals in real samples.  相似文献

The first use of the kinetic plot method to characterise the performance of ion-exchange columns for separations of small inorganic anions is reported. The influence of analyte type (mono- and divalent), particle size (5 and 9 μm), temperature (30 and 60 °C) and maximum pressure drop upon theoretical extrapolations was investigated using data collected from anion-exchange polymeric particulate columns. The quality of extrapolations was found to depend upon the choice of analyte, but could be verified by coupling a series of columns to demonstrate some practical solutions for ion chromatography separations requiring relatively high efficiency. Separations of small anions yielding 25–40,000 theoretical plates using five serially connected columns (9 μm particles) were obtained and yielded deviations of <15% from the kinetic plot predictions. While this approach for achieving high efficiencies results in a very long analysis time (t0 = 21 min), separations yielding approximately 10,000 theoretical plates using two serially connected columns (t0 < 5 min) were shown to be more practically useful for isocratic separations when compared to use of a single column operated at optimum linear velocity (t0 > 10 min).  相似文献

A polar polymethacrylate‐based monolithic column was introduced and evaluated as a hydrophilic interaction CEC stationary phase. The monolithic stationary phase was prepared by in situ copolymerization of a neutral monomer 2‐hydroxyethyl methacrylate and a polar cross‐linker N,N′‐methylene bisacrylamide in a binary porogenic solvent consisting of dodecyl alcohol and toluene. The hydroxyl and amino groups at the surface of the monolithic stationary phase provided polar sites which were responsible for hydrophilic interactions. The composition and proportion of the polymerization mixture was investigated in detail. The mechanical stability and reproducibility of the obtained monolithic column preformed was satisfied. The effects of pH and organic solvent content on the EOF and the separation of amines, nucleosides, and narcotics on the optimized monolithic column were investigated. A typical hydrophilic interaction CEC was observed on the neutral polar stationary phase. The optimized monolithic column can obtain high‐column efficiencies with 62 000–126 000 theoretical plates/m and the RSDs of column‐to‐column (n = 9), run‐to‐run (n = 5), and day‐to‐day (n = 3) reproducibility were less than 6.3%. The calibration curves of these five narcotics exhibited good linearity with R in the range of 0.9959–0.9970 and linear ranges of 1.0–200.0 μg/mL. The detection limits at S/N = 3 were between 0.2 and 1.2 μg/mL. The recoveries of the separation of narcotics on the column were in the range of 84.0–108.6%. The good mechanical stability, reproducibility, and quantitation capacity was suitable for pressure‐assisted CEC applications.  相似文献

This work examines in deep the analytical performance of an example of “first-generation” microdevices: capillary electrophoresis microchip (CE) with end-channel electrochemical detection (ED). A hydroquinone and arbutin separation strategically chosen as route involving pharmaceutical-clinical testing, public safety and food control scenes was carried out. The reproducibility of the unpinched electrokinetic protocol was carefully studied and the technical possibility of working indiscriminately and/or sequentially with both simple cross-injectors was also demonstrated using a real sample (R.S.D.'s less than 7%). The robustness of the injection protocol allowed checking the state of the microchip/detector coupling and following the extraction efficiency of the analyte from real sample. Separation variables such as pH, ionic strength and, separation voltage were also carefully assayed and optimized. Analyte screening was performed using borate buffer (pH 9, 60 mM) in less than 180 s in the samples studied improving dramatically the analysis times used for the same analytes on a conventional scale (15 min), with good precision (R.S.D.'s ranging 5-10%), accuracy (recoveries ranging 90-110%) and acceptable resolution (Rs ≥ 1.0).In addition, the excellent analytical performance of the overall analytical method indicated the quality of the whole analytical microsystem and allowed to introduce the definition of robustness for methodologies developed into the “lab-on-a-chip” scene.  相似文献

A new approach to synthesis and the possibility of application of solid gel matrices containing the target analyte, a cancer drug—doxorubicin for the calibration of UV-vis spectrometers is presented. The doxorubicin-loaded gels were prepared by the acid-catalyzed sol-gel pre-doping method. The efficiency of doxorubicin encapsulation in gels by the sol-gel process was 100 ± 1.2% (w/w). The amount of doxorubicin encapsulated per unit mass of the gel (1 g) was found to be independent of the mass of the gel taken for analysis, which demonstrates the homogeneity of encapsulation of the drug in the gel matrix. The gelation process of doxorubicin in the sol was found to be both repeatable (R.S.D. = 11.2% for n = 80, confidence level P = 95%) and reproducible (largest value of R.S.D. = 4.8% for n = 27, P = 95%).The prepared doxorubicin-loaded gel matrices are characterized primarily by their lack of toxicity as compared to the toxic free form of doxorubicin as well as by high stability over a long-time span.  相似文献

Capillary electrophoresis (CE) coupled with fiber-optic light-emitting diode-induced fluorescence detection has been developed for the separation of tyrosine (Tyr) enantiomers. R(−)-4-(3-isothiocyanatopyrrolidin-1-yl)-7-(N,N-dimethylaminosulfonyl)-2,1,3-benzoxadiazole was used as a chiral fluorescence tagged reagent for derivatization of Tyr. The effect of pH, running buffer concentration and applied voltage on enantioselectivity has been investigated. The optimum CE conditions are 15 mmol/L borate running buffer (pH 10.5) and 14-kV applied voltage. Good reproducibility was obtained with coefficient of variation (n = 7) of migration time and peak area less than 0.2 and 2.0%, respectively. The limits of detection of d- and l-Tyr derivatives were 2.9 and 2.2 μmol/L (S/N = 3), respectively. The proposed method has been successfully applied to the determination of Tyr in a commercial amino acid oral solution.  相似文献

A zwitterionic surfactant, dodecyldimethyl (2-hydroxy-3-sulfopropyl) ammonium (C12H25N+(CH3)2CH2CHOHCH2SO3-), named dodecyl sulfobetaine (DSB), was used as a novel modifier to coat dynamically capillary walls for capillary electrophoresis separation of basic proteins. The DSB coating suppressed the electroosmotic flow (EOF) in the pH range of 3-12. At high DSB concentration, the EOF was suppressed by more than 8.8 times. The DSB coating also prevented successfully the adsorption of cationic proteins on the capillary wall. Anions, such as Cl-, Br-, I-, SO4(2-), CO3(2-), and ClO4-, could be used as running buffer modifiers to adjust the EOF for better separation of analytes. Using this dynamically coated capillary, a mixture of eight inorganic anions achieved complete separation within 4.2 min with the efficiencies from 24,000 to 1,310,000 plates/m. In the presence of ClO4- as EOF adjustor, the separation of a mixture containing four basic proteins (lysozyme, cytochrome c, alpha-chymotrypsinogen A, and myoglobin) yielded efficiencies of 204,000-896,000 plates/m and recoveries of 88%-98%. Migration time reproducibility of these proteins was less than 0.5% relative standard deviation (RSD) from run to run and less than 3.1% RSD from day to day, showing promising application of this novel modifier in protein separation.  相似文献

Brazilian samples of fat products like margarines, butters and chocolates were prepared to reversed-phase high-performance liquid chromatography analysis, using a domestic microwave oven for fat hydrolysis and derivatization with phenacyl chloride. The total reaction time was 6 min, recoveries were above 94%. A fast high-performance liquid chromatography (HPLC) method was developed and validated for the simultaneous determination of mianserin (MIAN) and its metabolites desmethylmianserin (DMM), 8-hydroxymianserin (HM) and mianserin-N-oxide (MNO) in human plasma. Each compound, together with internal standard (propranolol) was extracted from the plasma matrix using solid phase extraction. Chromatographic resolution of the analytes was performed on a Chromolith Speed Rod monolithic silica column ( mm i.d.) under isocratic conditions using a mobile phase of 74:26 (v/v) 25 mM phosphate buffer (pH 5.3 adjusted with phosphoric acid): acetonitrile. The elution of the analytes were monitored at 292 mm and conducted at ambient temperature. Because of high column efficiency the mobile phase was pumped at a flow rate of 3.5 ml/min. The total run time of the assay was 5 min. The method was validated over the range of 10-200 ng/ml for MIAN, 10-150 ng/ml for DMM, 20-300 ng/ml for HM and 25-500 ng/ml for MNO. The method proved to be precise (within-run precision ranging from 1.6 to 6.9% R.S.D. and between-run precision ranging from 1.3 to 7.2% R.S.D.) and accurate (within-run accuracies ranged from 1.4 to 6.4% and between-run accuracies ranging from 1.5 to 4.5%). The mean absolute recoveries were 95.7, 94.8, 99.6, and 102.6% for MIAN, DMM, HM and MNO, respectively. The limit of quantitation (LOQ) for MIAN and DMM was 10 ng/ml and for HM and MNO were 20 and 25 ng/ml in human plasma, respectively. The limit of detection (LOD) for MIAN, DMM, HM and MNO was 5, 2.5, 10 and 15 ng/ml, respectively. The described method demonstrates the feasibility for employing monolithic columns to effect rapid bioanalytical HPLC analysis for the quantitative determination of MIAN and its major metabolites in human plasma.  相似文献

This paper describes the optimization and validation of an analytical methodology for the determination of losartan potassium in capsules by HPLC using 25-1 fractional factorial and Doehlert designs. This multivariate approach allows a considerable improvement in chromatographic performance using fewer experiments, without additional cost for columns or other equipment. The HPLC method utilized potassium phosphate buffer (pH 6.2; 58 mmol L−1)-acetonitrile (65:35, v/v) as the mobile phase, pumped at a flow rate of 1.0 mL min−1. An octylsilane column (100 mm × 4.6 mm i.d., 5 μm) maintained at 35 °C was used as the stationary phase. UV detection was performed at 254 nm. The method was validated according to the ICH guidelines, showing accuracy, precision (intra-day relative standard deviation (R.S.D.) and inter-day R.S.D values <2.0%), selectivity, robustness and linearity (r = 0.9998) over a concentration range from 30 to 70 mg L−1 of losartan potassium. The limits of detection and quantification were 0.114 and 0.420 mg L−1, respectively. The validated method may be used to quantify losartan potassium in capsules and to determine the stability of this drug.  相似文献

An efficient direct method for measuring c9,t11- and t10,c12-conjugated linoleic acid (CLA) isomer content in human and rat milk was developed and validated using an RTX-2330 capillary column (40 m × 0.18 mm × 0.1 μm). In comparison with the commonly used 100 m × 0.25 mm × 0.20 μm columns, this new type of fast column allowed the separation of FAMEs with the same resolution but in much less time. An additional advantage for biological samples was that only a small volume of sample was needed. Two different procedures were tested in order to select the best methylation of CLA isomers, and the alkali plus acid-catalyzed procedure was selected. The precision results showed relative standard deviations (R.S.D.) of repeatability and reproducibility ranging between 0.10 and 8.71%. The application of this method to human and rat milk samples showed that it was a rapid, simple and reliable method for the analysis of biological samples.  相似文献

A novel anionic exchange stir bar sorptive extraction (SBSE) coating based on poly(2-(methacryloyloxy)ethyltrimethylammonium chloride-co-divinylbenzene) monolithic material for the extraction of inorganic anion was prepared. The effect of preparation conditions such as ratio of functional monomer to cross-linker, content of porogenic solvent on the extraction efficiencies were investigated in detailed. The monolithic material was characterized by elemental analysis, scanning electron microscopy and infrared spectroscopy. In order to investigate the extraction capacity of the new coating for inorganic anion, the new SBSE was combined with ionic chromatography with conductivity detection, Br, NO3, PO43− and SO42− were selected as detected solutes. Several extractive parameters, including pH value and ionic strength in sample matrix, desorption solvent, extraction and desorption time were optimized. The results showed that strongly ionic strength did not favor the extraction of anlaytes. Under the optimum experimental conditions, low detection limits (S/N = 3) and quantification limits (S/N = 10) of the proposed method for the target anions were achieved within the range of 0.92–2.62 and 3.03–9.25 μg/L, respectively. The method also showed good linearity, simplicity, practicality and low cost for the extraction inorganic anions. Finally, the proposed method was successfully used to detect the two different trademarks of commercial purified water with satisfactory recovery in the range of 70.0–92.6%. To the best of our knowledge, this is the first to use SBSE to enrich inorganic anions.  相似文献

Continuous-bed columns containing sol-gel bonded 3 microm silica particles with mixed-mode octadecyl and propylsulfonic acid functional groups (ODS/SCX) were prepared by first packing the ODS/SCX particles into a fused-silica capillary, then filling the packed capillary with a siliceous sol-gel, curing the sol-gel, and finally drying the column with supercritical carbon dioxide. The performance of the sol-gel bonded ODS/SCX columns was evaluated for capillary electrochromatography using acetonitrile aqueous mobile phase containing phosphate buffer. The columns were mechanically strong and permeable. Both EOF velocity and current increased linearly with elevation of the applied electric field strength. The EOF velocity was high at low pH and nearly constant over a range of pH 2-9. Higher buffer concentration resulted in higher current and lower EOF velocity. The acetonitrile content had no significant effect on the EOF. Without thermosetting the column, no bubble formation was noticed with currents up to 2.5 microA. The minimum plate height of a 25/34 cm x 75 microm I.D. sol-gel bonded 3 microm ODS/SCX column was 5.7 microm (1.75 x 10(5) plates per meter) at an optimum EOF velocity of 0.92 mm s(-1). Mixtures of test aromatic compounds and aromatic hydrocarbon homologues gave symmetrical peaks when using a low pH mobile phase. The retention and elution order of aromatic compounds represented a typical reversed-phase separation mechanism similar to conventional ODS columns. The run-to-run and column-to-column retention factor reproducibility was better than 2.5% and 8.0% RSD, respectively.  相似文献
