We investigated characteristics of various phosphatidylcholines (PCs) used as dispersing agents and emulsifiers. Six PCs with different lengths of acyl hydrocarbon chains and different degrees of unsaturated acyl hydrocarbon chains were selected to examine influences of a lipophillic part of phosphatidylcholines in emulsion and dispersion systems. Vesicles and oil-in-water emulsions were prepared by sonication under several ambient temperature conditions. Mean diameters of vesicles and oil droplets in emulsions were measured by a submicron particle sizer. In vesicles that are generated by hydration of the PCs and have a bilayer form, particle size was influenced by length and degree of unsaturation of acyl hydrocarbon chains of a PC. PCs with shorter acyl hydrocarbon chains or unsaturated bonds are considered more potent dispersing agents. Preparation temperature of the PC is also a factor affecting potency of dispersion. In O/W emulsions in which PCs were absorbed at water-oil interfaces and which have a single layer form or liquid-crystal layer form, particle size was also influenced by length and degree of unsaturation of acyl hydrocarbon chains of a PC. PCs with shorter and saturated acyl hydrocarbon chains are considered more potent emulsifiers. Unsaturation of acyl hydrocarbon chains weaken the ability of emulsification due to vulnerable double bonds. For stable emulsions, it is considered beneficial for PCs to form small oil droplets and lamellae liquid-crystal phase. From this perspective, saturated PCs with short hydrocarbon chains, i.e., DLPC and DMPC, may have advantages in preparing a stable emulsion not only by giving a smaller droplet size but also by forming lamellae liquid-crystal phase. When considering characteristics of PCs as emulsifiers, their characteristics as dispersing agents is also useful information.  相似文献

The decomposition of hydrogen peroxide (H2O2) was examined in aqueous solution (50 mM Tris-HCl buffer, pH 7.4, containing 100 mM NaCl) at 25 degrees C in pure buffer or in the presence of either vesicles or micelles formed from various phosphatidylcholines (PCs). In the absence of PCs, more than 90% of the initially added H2O2 (1.0 mM) remained intact after incubation for 120 h. The effect of the PCs on the decomposition of H2O2 was studied by using different PCs that varied in terms of number of carbon atoms in the two acyl chains n as well as in terms of the degree of unsaturation. PCs with short hydrocarbon chains (n = 4, 6-8) were dissolved in the buffer solution in the form of nonassociated monomers or as micelles in equilibrium with monomers at a fixed PC concentration of 10 mM. The presence of these short-chain PCs slightly enhanced the H2O2 decomposition rate. Micelles formed by non-lipid detergents (sodium cholate, Triton X-100, and sodium dodecylsulfate) had a similar effect. In marked contrast, PCs with long hydrocarbon chains (n > or = 10) dispersed in buffer solution as vesicles (liposomes) significantly enhanced the rate of H2O2 decomposition, with the most effective PC being 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) at 25 degrees C. This indicates that the packing density of the PC molecules influences the reactivity, presumably through the direct interaction of the PC assemblies with H2O2 molecules. Furthermore, in the case of vesicles formed from PCs with unsaturated acyl chains (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine, POPC; 1,2-dioleoyl-sn-glycero-3-phosphocholine, DOPC), carbon-carbon double bond oxidation did not occur extensively under the conditions used. This indicates that the observed effect of PCs on the decomposition of H2O2 is indeed related to the assembly structure (vesicle vs micelles vs monomers) and is clearly not related to the presence of unsaturated hydrocarbon chains. Fluorescence polarization measurements of two fluorescent probes embedded either in the acyl chain region of the vesicles (DPH, 1,6-diphenyl-1,3,5-hexatriene) or on the surface of the vesicles (TMA-DPH, 1-(4-trimethylammoniumphenyl)-6-phenyl-1,3,5-hexatriene iodide) show that the presence of H2O2 leads to a decrease in the fluidity of the lipid-water surface and not to a change in the fluidity of the hydrophobic region of the vesicle bilayer. This indicates that the decomposition of H2O2 is triggered through interactions between H2O2 and the polar head group area of PC vesicles.  相似文献

Abstract— Phospholipid vesicles of saturated and unsaturated phosphatidylcholines have been prepared with phenanthrenequinone incorporated into the hydrocarbon region of the lipid bilayer. Blue or ultraviolet light exposure of these vesicles causes a loss of quinone absorbance and the corresponding formation of new absorbance bands due to addition products. In vesicles composed of unsaturated phospholipids, cycloaddition is preferred over R-H addition. The quantum yield for quinone disappearance is -0.25 in vesicles which contain unsaturated phospholipids and decreases to 0.08 when the photolysis is conducted below the lipid characteristic temperature.  相似文献

Giant vesicles generated from synthetic and natural lipids such as phosphatidylcholines are useful models for understanding mechanical properties of cell membranes. Line tension is the one-dimensional force enabling the closing of transient pores on cell membranes. Transient pores were repeatedly and reproducibly formed on the membrane edge of giant vesicles generated from synthetic and natural phosphatidylcholines employing a nitrogen-pumped coumarin dye laser (440 nm). Line tension was determined at room temperature from closing of these pores that occurred over several seconds when the radius of the vesicle could be considered to be constant. The value of line tension depends on the nature of the lipid for single lipid systems, which, at room temperature, yielded a vesicle bilayer region in the gel, fluid, or mixed gel and fluid phases. The line tension for vesicles generated from phosphatidylcholines with saturated acyl chains of lengths of 12-18 carbon atoms ranges from 1 to 12 pN, exhibiting an increase with chain length. Vesicles generated from the natural Egg-PC, which is a mixture of lipids, are devoid of phase transition and exhibited the largest value of line tension (32 pN). This value is much larger than that estimated from the line tensions of vesicles obtained from lipids with homologous acyl chains. This study, to our knowledge, is the first to employ laser ablation to generate transient pores and determine line tension from the rate of pore closure and demonstrate a relationship between line tension and acyl chain length.  相似文献

Photon correlation spectroscopy and freeze-fracture electron microscopy have been used to determine the ability of a range of micelle-forming, polyoxyethylene (20) sorbitan monoesters (Tweens) to solubilise vesicles prepared from phosphatidylcholines of different acyl chain lengths and degrees of saturation with a view to rationalising (in terms of their membrane toxicity) which of the micelle-forming surfactants to use as drug delivery vehicles. The phosphatidylcholines used were dimyristoyl-, dipalmitoyl-, distearoyl- and dioleoylphosphatidylcholine (DMPC, DPPC, DSPC and DOPC, respectively) while the nonionic polyoxyethylene sorbitan monoesters studied were polyoxyethylene (20) sorbitan monolaurate (Tween 20), a 9:1 weight ratio mixture of polyoxyethylene (20) sorbitan monopalmitate and monostearate (Tween 40), a 1:1 weight ratio mixture of polyoxyethylene (20) sorbitan monopalmitate and monostearate (Tween 60), and polyoxyethylene (20) sorbitan monooleate (Tween 80). The ability of the Tween micelles to solubilise phospholipid vesicles was found to depend both upon the length of the surfactant acyl chain and the length of the acyl chains of the phospholipid comprising the vesicle. Vesicles composed of long saturated diacyl chain phospholipids, namely DSPC and DPPC, were the most resistant to solubilisation, while those prepared from the shorter acyl chained DMPC were more readily solubilised. In terms of their solubilisation behaviour, vesicles made from phospholipids containing long, unsaturated acyl chains, namely DOPC behaved more akin to those vesicles prepared from DMPC. None of the Tween surfactants were effective at solubilising vesicles prepared from DPPC or DSPC. In contrast, there were clear differences in the ability of the various surfactants to solubilise vesicles prepared from DMPC and DOPC, in that micelles formed from Tween 20 were the most effective solubilising agent while those formed by Tween 60 were the least effective. As a consequence of these observations it was considered that Tween 60 was the surfactant least likely to cause membrane damage in vivo and, therefore, is the most suitable surfactant for use as a micellar drug delivery vehicle.  相似文献

Polyunsaturated lipids in cellular membranes are known to play key roles in such diverse biological processes as vision, neuronal signaling, and apoptosis. One hypothesis is that polyunsaturated lipids are involved in second messenger functions in biological signaling. Another current hypothesis affirms that the functional role of polyunsaturated lipids relies on their ability to modulate physical properties of the lipid bilayer. The present research has employed solid-state 2H NMR spectroscopy to acquire knowledge of the molecular organization and material properties of polyunsaturated lipid bilayers. We report measurements for a homologous series of mixed-chain phosphatidylcholines containing a perdeuterated, saturated acyl chain (n:0) at the sn-1 position, adjacent to docosahexaenoic acid (DHA, 22:6omega3) at the sn-2 position. Measurements have been performed on fluid (L(alpha))-state multilamellar dispersions as a function of temperature for saturated acyl chain lengths of n = 12, 14, 16, and 18 carbons. The saturated sn-1 chains are therefore used as an intrinsic probe with site-specific resolution of the polyunsaturated bilayer structure. The 2H NMR order parameters as a function of acyl position (order profiles) have been analyzed using a mean-torque potential model for the chain segments, and the results are discussed in comparison with the homologous series of disaturated lipid bilayers. At a given absolute temperature, as the sn-1 acyl length adjacent to the sn-2 DHA chain is greater, the order of the initial chain segments increases, whereas that of the end segments decreases, in marked contrast with the corresponding disaturated series. For the latter, the order of the end segments is practically constant with acyl length, thus revealing a universal chain packing profile. We find that the DHA-containing series, while more complex, is still characterized by a universal chain packing profile, which is shifted relative to the homologous saturated series. Moreover, we show how introduction of DHA chains translates the order profile along the saturated chains, making more disordered states accessible within the bilayer central region. As a result, the area per lipid headgroup is increased as compared to disaturated bilayers. The systematic analysis of the 2H NMR data provides a basis for studies of lipid interactions with integral membrane proteins, for instance in relation to characteristic biological functions of highly unsaturated lipid membranes.  相似文献

The interaction of bile salts with model membranes composed of soybean phosphatidylcholine (SPC) and synthetic 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) was investigated using high sensitivity isothermal titration calorimetry (ITC). The partitioning and incorporation of the bile salts sodium cholate (NaC) and sodium deoxycholate (NaDC) from an aqueous phase (pure water or 0.1 M NaCl) into fluid bilayer vesicles was studied as a function of temperature and ionic strength. The thermodynamic parameters of partitioning were determined with a model taking electrostatic interactions into account. In addition, the solubilization of SPC and POPC vesicles with NaC and NaDC as a function of temperature was also studied by ITC and the phase diagrams for the vesicle to micelle transition at two different temperatures were established. Unsaturated phospholipids require higher amounts of detergent to be transformed into micelles compared to saturated phospholipids. In addition, the width of the coexistence region of mixed micelles and mixed vesicles is larger for phosphatidylcholines with unsaturated chains. A comparison of NaDC with NaC shows the higher solubilization effectiveness of NaDC in agreement with its lower cmc. Furthermore, increasing the ionic strength decreases the amount of bile salt necessary for the formation of mixed micelles, which is also expected from the decrease of the cmc with ionic strength due to the shielding of the charges of the bile salts.  相似文献

Polyunsaturated phospholipids are known to be important with regard to the biological functions of essential fatty acids, for example, involving neural tissues such as the brain and retina. Here we have employed two complementary structural methods for the study of polyunsaturated bilayer lipids, viz. deuterium ((2)H) NMR spectroscopy and molecular dynamics (MD) computer simulations. Our research constitutes one of the first applications of all-atom MD simulations to polyunsaturated lipids containing docosahexaenoic acid (DHA; 22:6 cis-Delta(4,7,10,13,16,19)). Structural features of the highly unsaturated, mixed-chain phospholipid, 1-palmitoyl-2-docosahexaenoyl-sn-glycero-3-phosphocholine (PDPC), have been studied in the liquid-crystalline (L(alpha)) state and compared to the less unsaturated homolog, 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC). The (2)H NMR spectra of polyunsaturated bilayers are dramatically different from those of less unsaturated phospholipid bilayers. We show how use of MD simulations can aid in interpreting the complex (2)H NMR spectra of polyunsaturated bilayers, in conjunction with electron density profiles determined from small-angle X-ray diffraction studies. This work clearly demonstrates preferred helical and angle-iron conformations of the polyunsaturated chains in liquid-crystalline bilayers, which favor chain extension while maintaining bilayer flexibility. The presence of relatively long, extended fatty acyl chains may be important for solvating the hydrophobic surfaces of integral membrane proteins, such as rhodopsin. In addition, the polyallylic DHA chains have a tendency to adopt back-bended (hairpin-like) structures, which increase the interfacial area per lipid. Finally, the material properties have been analyzed in terms of the response of the bilayer to mechanical stress. Simulated bilayers of phospholipids containing docosahexaenoic acid were less sensitive to the applied surface tension than were saturated phospholipids, possibly implying a decrease in membrane elasticity (area elastic modulus, bending rigidity). The above features distinguish DHA-containing lipids from saturated or monounsaturated lipids and may be important for their biological modes of action.  相似文献

Mixtures of cholesterol and synthetic phospholipids, differing in saturation of phosphatidylcholine (PC) acyl chains, such as distearoyl phosphatidylcholine (DSPC), stearoyl-oleoyl phosphatidylcholine (SOPC) and dioleoyl phosphatidylcholine (DOPC) have been studied as floating Langmuir monolayers at the air/water interface. In order to examine the influence of a polar group, distearoyl phosphatidylethanolamine (DSPE) was chosen. The films were spread at room temperature on aqueous subphases and characterized by the surface pressure-area (pi-A) isotherms and compression modulus (C(s)(-1)) values. The interactions were examined by analyzing the mean molecular areas and quantified by the excess free energy of mixing values. The obtained results indicate that the affinity of cholesterol to saturated/unsaturated phosphatidylcholines does not differ significantly, and revealed strong influence of the kind of a polar group on the cholesterol-phospholipid interactions. On the other hand, the apolar group structure was found to modify the stoichiometry of sterol-PC complexes.  相似文献

We described the first scanning tunneling microscopy study of spreading unilamellar vesicles of 1,2-dimyristoyl-sn-glycero-3-phosphatidylcholine (DMPC) at a Au(111) electrode surface. At the initial stage of the film formation, the molecular resolution images revealed that DMPC molecules are adsorbed flat with the acyl chains oriented parallel to the surface. The molecules assemble into double rows by aligning the acyl chains in the nearest neighbor direction of the reconstructed Au(111) surface and assuming a 90 +/- 10 degrees angle with respect to line of the molecular row. After approximately 30 min, this film is transformed into a hemimicellar state with long rows characteristic for the formation of hemicylindrical surface micelles. At hydrophilic surfaces such as glass, spreading of vesicles involves adsorption, rupture, and sliding of a single bilayer on a lubricating film of the solvent. We have provided the first evidence that a different mechanism is involved in spreading the vesicles at gold. The molecules released by rupture of vesicles self-assemble into an ordered film, and the assembly is controlled by the chain-substrate interaction.  相似文献

In this work, we have investigated the condensing and ordering effect induced by cholesterol on phosphatidylcholines (PCs). To perform the studies systematically, for the experiments we have selected phospholipids differing only in the number of cis monounsaturated chains (1,2-distearoyl-sn-glycero-3-phosphocholine--DSPC, 1-stearoyl-2-oleoyl-sn-glycero-3-phosphocholine--SOPC, 1,2-dioleoyl-sn-glycero-3-phosphocholine--DOPC) or in the length (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine--POPC vs SOPC) of sn-1 acyl chain. Because the cholesterol concentration in mammalian membranes can be as high as 70 mol % of total lipids, the investigations were performed in a wide range of the sterol content. The results of the Langmuir monolayer experiments evidence that the relation between the structure of hydrophobic part of PC and the magnitude of the effects induced by cholesterol found at lower sterol content is different from that observed at higher sterol concentration. At a lower concentration of sterol (up to 30%), the condensing effect of cholesterol is stronger on saturated DSPC than on PCs containing monounsaturated chain(s), which is consistent with the conclusions drawn by other authors. However, at higher sterol content (≥50%), saturated DSPC is less susceptible to the influence of sterol than the investigated unsaturated PCs. To explain these irregularities, we have considered the strength of van der Waals interactions as well as the influence of sterol on the tilt of polar heads of PCs. It was also found that in the whole range of sterol concentration the ordering effect is stronger on saturated DSPC as compared to unsaturated phospholipids. However, at lower sterol content (up to 30%) the ordering effect induced on unsaturated PCs is rather weak, and the ordering does not change drastically in comparison with pure PCs film.  相似文献

The main gel-to-liquid-crystal (LC) phase transition temperature, T(m), of the distal lipid layer in hybrid bilayer membranes (HBMs) under water was investigated using vibrational sum frequency spectroscopy (VSFS). VSFS has unique sensitivity to order/disorder transitions in the lipid acyl chains and can determine T(m) for the lipid monolayers in HBMs. We recently reported the observation that T(m) is raised and the transition width is broadened for the overlying phospholipid monolayer in HBM systems formed on densely packed crystalline self-assembled monolayers (SAMs) as compared to that of vesicles in solution. In this report, we establish that T(m) for the lipid layer of HBMs can be controlled by proper choice of the SAM underlayer. The SAM underlayer of the HBM was systematically altered by using an alkane thiol, a saturated thiolipid, a mixed SAM of a saturated lipid-pyridine disulfide, and finally a mixed SAM of an unsaturated lipid-pyridine disulfide. T(m) was measured for two different chain length saturated phosphatidylcholine lipid overlayers on these SAMs. The values obtained show that Tm of the lipid layer of HBMs is sensitive to the composition and/or packing density of molecules in the underlying SAM.  相似文献

The main gel-to-liquid-crystal (LC) phase transition temperature, T(m), of the lipid monolayer in hybrid bilayer membranes (HBMs) was investigated using vibrational sum frequency spectroscopy (VSFS). In the gel phase, the acyl chains of the lipid molecules assume an ordered, all-trans configuration, whereas in the LC phase, the acyl chains exhibit a significant number of disordered gauche conformers. VSFS has unique sensitivity to the order/disorder transitions in the acyl chains and was used to determine T(m) for a series of saturated phosphatidylcholine lipids on octadecanethiolate self-assembled monolayers (SAMs). The values obtained for T(m) for all lipids studied are significantly higher than for the corresponding lipids in vesicles in solution. Additionally, the transition widths are broader for the lipids in HBMs. The underlying SAM clearly influences the phase behavior of the overlying lipid monolayer.  相似文献

Surface pressure-area isotherm, neutron specular reflection, and small-angle neutron scattering studies have been carried out to determine the effects of added cholesterol and distearoylphosphatidylcholine (DSPC), on the molecular structures of monolayers and vesicles containing the dialkyl polyoxyethylene ether surfactant, 1,2-di-O-octadecyl-rac-glyceryl-3-(alpha-dodecaethylene glycol) (2C18E12). Previous neutron reflectivity studies on 2C18E12 monolayers at the air/water interface have shown them to possess a thickness of approximately 24 angstoms and highly disordered structure with significant intermixing of the polymer headgroups and alkyl chains. SANS studies of 2C18E12 vesicles gave a bilayer thickness of approximately 51 angstroms. Addition of cholesterol to 2C18E12 monolayers (1:1 molar ratio), produced a marked condensing effect coupled with an increased the layer thickness of approximately 7 angstroms, and in vesicles, increased bilayer thickness by approximately 16 angstroms. Monolayers consisting of 2C18E12:DSPC:cholesterol (1:1:2 molar ratio), showed a layer thickness of approximately 31 angstroms, whereas in vesicles, three-component bilayer was found to be only approximately 9 angstroms thicker than those possessed by vesicles composed solely of 2C18E12. Mixing between the molecules in three-component monolayers was shown to be ideal through analysis of the neutron reflectivity data. These findings are discussed in relation to increased ordering and decreased headgroup/hydrophobe intermixing within both monolayers and vesicle bilayers containing 2C18E12. The inferred increase in molecular order within vesicles composed of 2C18E12 with additional cholesterol and phospholipid is used as a model for explaining theoretical differences in bilayer permeability.  相似文献

Statistical analysis of quantitative structure—property relationships (QSPR) for the phase transition gel—liquid crystal (main phase transition) temperatures (T m) in hydrated vesicle bilayers of diacyl phosphatidylcholines (PC) and diacyl phosphatidylethanolamines (PE) with saturated acyl chains was carried out. Multiple regression equations relating the T m values to the volumes of the hydrocarbon fragments of the sn-1 and sn-2 acyl chains show that the acyl groups influence the transiton temperatures T m in different manner, the effect of the sn-2 group being predominant. The transition temperatures T m of saturated phosphatidylethanolamines and diacyl phosphatidylglycerols can be predicted using the T m values calculated for the corresponding diacyl phosphatidylcholines.  相似文献

We report on the fluorescence lifetime and anisotropy decay dynamics of the tethered chromophore NBD in unilamellar vesicles comprised of phosphoglycerol and phosphocholine lipids with C(12) and C(18) saturated acyl chains, with or without cholesterol and/or sphingomyelin. For the phosphocholine vesicles, we use the chromophore 2-(12-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino)dodecanoyl-1-hexadecanoyl-sn-glycero-3-phosphocholine (NBD-PC), and for the phosphoglycerol vesicles, we use the chromophore 2-(12-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino)dodecanoyl-1-hexadecanoyl-sn-glycero-3-[phospho-rac-(1-glycerol)] (NBD-PG). The addition of cholesterol and/or sphingomyelin to the PC vesicles restricts the chromophore environment, in agreement with the known rigidizing effect of cholesterol on PC membranes. The PG systems do not exhibit an analogous effect with the addition of cholesterol and/or sphingomyelin. The motional freedom of the NBD chromophore is, in general, more restricted in the PC bilayers than it is in the PG bilayers, and we understand this behavior in the context of the role of the lipid headgroups in mediating bilayer organization.  相似文献

The molecular packing in triacylglycerols having different acyl chains has been examined by differential scanning calorimetry, powder X-ray diffraction and vibrational spectroscopy (infrared and Raman) techniques. In the triacylglycerols examined, the acyl chain length, unsaturation or the position of substitution on the glycerol were changed systematically to observe their influence on the molecular packing in different polymorphic forms. Variation in the 3-acyl chain length of 1,2-dipalmitoyl-3-acyl-sn-glycerols (PPX) influenced the molecular packing along the long axis in the stable polymorphic forms. Three different modes of packing were observed. If X ≤ 4, the compounds packed in a bilayer structure similar to diacylglycerols, or if X ≥ 10 and ≤ 16 the compounds packed in a bilayer structure but similar to mono acid triacylglycerols. However for intermediate 3-acyl chain lengths, as in PP6 and PP8 the stable packing can occured only through chain segregation resulting in a trilayer structure. In the triacylglycerols containing unsaturated acyl chains, 1,2-dioleoyl-3-acyl-sn-glycerols (00X) and 1,3-dioleoyl-Z-acyl-sn-glycerols (0X0) the stable polymorphic forms packed in a trilayer structure where the odd acyl chains segregated and formed a middle layer. In a metastable hexagonal packing (α-phase) the long range ordering is minimal. Because of this lack of specific chain-chain interaction the 3-short acyl chain compounds of PPX packed in a unimolecular length structure (except PP2) whereas the 3-long acyl chain compounds packed in a bilayer structure. In orthorhombic perpendicular and triclinic parallel packing where the specific chain-chain interaction is increased, the end plane methyl packing and the glycerol conformation played important roles in the formation of bi-, tri- and hexalayer structures. The driving force in the formation of these different structures is to minimize the crystal defects created by the odd acyl chains and to enhance the specific chain-chain interactions. The presence of an odd acyl chain influenced the lateral chain packing as well, e.g., the stability of the orthorhombic perpendicular packing is enhanced by the presence of an odd acyl chain and even in some cases it is favored over the triclinic parallel packing. The odd acyl chain at the 1- or 3position of -sn-glycerol stabilized the orthorhombic perpendicular packing. This indicates the glycerol conformation is probably perpendicular to the layer plane and thus is different from the monoacid triacylglycerols.  相似文献

Mn-Anderson-C6 and Mn-Anderson-C16, A type of inorganic-organic hybrid molecules containing a large anionic polyoxometalate (POM) cluster and two C6 and C16 alkyl chains, respectively, demonstrate amphiphilic surfactant behavior in the mixed solvents of acetonitrile and water. The amphiphilic hybrid molecules can slowly assemble into membrane-like vesicles by using the POM clusters as polar head groups, as studied by laser light scattering and TEM techniques. The hollow vesicles have a typical bilayer structure with the hydrophilic Mn-Anderson cluster facing outside and long hydrophobic alkyl chains staying inside to form the solvent-phobic layer. Due to the rigidity of the POM polar heads, the two alkyl tails have to bend significantly for the vesicle formation, which makes the vesicle formation more difficult compared to some conventional surfactants. Bilayer structures are formed by approaching two liquid surfaces with phospholipid monolayers, which are brought into contact by oblique drop impact on a liquid surface. Asymmetric bilayers can be produced by the coupling of drop and target monolayers. In contrast, symmetric bilayers or multilayers are formed by collapse of the compressed target monolayer. We show that under all studied conditions bilayer/multilayer synthesis takes place. The experimental conditions for the synthesis of asymmetric or symmetric bilayers are described quantitatively in terms of the surface rheological (surface elasticity and dilational viscosity) and the hydrodynamical parameters (Weber number and impact angle). The composition and mechanical properties of the phospholipid monolayers strongly influences the patterns of drop impact and the bilayer/multilayer formation. Cholesterol stiffens unsaturated phospholipid monolayers and fluidifies saturated monolayers. All monolayers form asymmetric vesicle-like structures, which are stable in the aqueous medium. Additionally, unsaturated phospholipid monolayers without cholesterol form symmetric vesicles by folding parts of the target monolayer. Sufficient presence of cholesterol in unsaturated phospholipid monolayers inhibits the folding of the target monolayer and the subsequent formation of symmetric bilayers. The rheological properties of saturated and unsaturated phospholipid monolayers and their mixtures with cholesterol are discussed. Based on drop impact results it is shown that the state of a so far undefined region in the DPPC/cholesterol phase diagram is a fluid phase.  相似文献

The main constituents of plant oils are complex mixtures of TGs differing in acyl chain lengths, number and positions of double bonds, and regioisomerism. A non-aqueous reversed-phase HPLC method with acetonitrile-2-propanol gradient and 30 + 15 cm NovaPak C18 columns makes possible an unambiguous identification of the highest number of TGs ever reported for these oils, based on positive-ion APCI mass spectra. A new approach to TG quantitation is based on the use of response factors with three typical detection techniques for that purpose (APCI-MS, evaporative light-scattering detection, and UV at 205 nm). Response factors of 23 single-acid TGs (saturated TGs from C7 to C22, 7 unsaturated TGs), 4 mixed-acid TGs, diolein and monoolein are calculated from their calibration curves and related to OOO. Due to differences between saturated and unsaturated acyl chains, the use of response factors significantly improves the quantitation of TGs. 133 TGs containing 22 fatty acids with 8-25 carbon atoms and 0-3 double bonds are identified and quantified in 9 plant oils (walnut, hazelnut, cashew nut, almond, poppy seed, yellow melon, mango, fig, date) using HPLC/APCI-MS with a response factor approach. Average parameters and relative fatty acid concentrations are calculated with both HPLC/APCI-MS and GC/ FID.  相似文献
